Shedding of the common acute lymphoblastic leukemia antigen (CALLA) by lymphoblastoid cell lines.

The release of common acute lymphoblastic leukemia antigen (CALLA) into culture medium by lymphoblastoid cell lines was studied. CALLA release was detected using a solid phase indirect radioimmunometric assay which combines the specificity of lectins and monoclonal antibodies and using immunoadsorption of labeled CALLA in spent medium from cells incubated with 35S-methionine. No CALLA was present in the medium of cells pulse-labeled at 37 degrees C, when they were placed at 4 degrees C, thus this is an active process. Sequential analysis of soluble CALLA revealed shedding of CALLA to be associated with cell growth.

Complete median cleft of the upper lip and alveolus with holoprosencephaly.

A case of complete median cleft of the upper lip and alveolus with holoprosencephaly is presented. An 8-month-old girl was successfully treated by closure of the median upper lip. In addition, 9 patients who have undergone cheiloplasty of complete median cleft of the upper lip with holoprosencephaly reported in the Japanese literature, are reviewed.

Development of autologous specific reactivity to human neuroblastoma.

Antigenic analysis of neuroblastoma cells was performed by autologous typing and indirect immunofluorescent test combined with absorption tests. Serum of a patient with neuroblastoma before treatment showed weak reactivity with cultured autologous neuroblastoma cells, while serum obtained from the same patient 3 months after chemotherapy exhibited strong reactivity with the same indicator cells. This reactivity was absorbed with neither cultured autologous fibroblasts nor allogeneic tumor cells from seven neuroblastoma and two melanoma patients. In addition, cultured hematopoietic cells from two subjects, normal fetal and adult tissues also failed to remove the reactivity. This reactivity, however, was completely removed by the absorption with the cultured autologous neuroblastoma cells and the homogenate, suggesting that these neuroblastoma cells may express tumor specific antigen(s), located on neither normal cells in any stage of the development nor other malignant cells, although the cells and tissues we tested may not be adequate to confirm this.

Effect of injection of CCK-8 into the nucleus caudatus on the behavior of rats.

This report deals with the effect of cholecystokinin octapeptide (CCK-8) on the regulation of the behavior stimulated by dopaminergic drugs. Bilateral injection of CCK-8 (1 microgram per side) into the nucleus caudatus significantly reduced the locomotor hyperactivity induced by methamphetamine. Stereotyped sniffing and yawning occurred after intrastriatal administration of apomorphine (20 micrograms per side). Injections of CCK-8 into the nucleus caudatus completely inhibited the sniffing, but did not affect the yawning induced by apomorphine. It also had no effect on the basal dopamine (DA) level or the methamphetamine-induced DA level in the striatum. These results suggest that the injection of CCK-8 into the nucleus caudatus selectively inhibited the function of the dopaminergic system in the striatum, and blocked post-synaptic DA receptors.

Effects of cholecystokinin tetra and octa peptides on locomotor activity in mice.

Intraperitoneal injection of cholecystokinin octapeptide (10 and 100 micrograms/kg, i.p.), but not the tetra-peptide (1 mg/kg, i.p.), into mice significantly reduced spontaneous locomotor activity in a dose-dependent manner. The increase in locomotor activity induced by methamphetamine (1 mg/kg, i.p.) and thyrotropin-releasing hormone (5 mg/kg, i.p.) was significantly reduced by cholecystokinin peptides. However, the inhibitory effects of these peptides differed. Neither the tetra nor octa peptide influenced the increased locomotor activity induced by nomifensine (5 mg/kg, i.p.), apomorphine (3 mg/kg, i.p.) or scopolamine (2 mg/kg, i.p.). Thus, these cholecystokinin peptides seem to selectively antagonize increased locomotor activity via the presynaptic dopaminergic system.

[In vitro effects of 5-FU on gynecologic malignant tumor cell lines].

In order to conduct a fundamental investigation of the effect of UFT on malignant tumors, biological experiments were conducted. UFT, 300 mg/day, starting one week prior to surgery, was administered in 12 cases of ovarian carcinoma. During the operation, 5-FU concentrations in serum, tumor tissues and adjacent normal tissues were measured. The results were as follows: 0.009 +/- 0.005 micrograms/ml in serum; 0.157 +/- 0.214 micrograms/g in tumor tissues, and 0.042 +/- 0.028 micrograms/g in adjacent normal tissues. Based upon these values, the plating efficiency of cultured tumor cell lines was measured in the presence of 5-FU in vitro. With 5-FU concentrations of 0.01 micrograms/ml, no sign of restriction in plating efficiency was observed. Restriction was initially observed at 0.1 microgram/ml, and became more pronounced at 1.0 microgram/ml. Furthermore, experiments using a uterine cervical carcinoma cell line (SKG-3) indicated that restriction of the plating efficiency was enhanced, not only by the 5-FU concentration but also by lapse of time.

[Differential diagnosis of the pelvic masses by computed tomography and 67Ga-scintigraphy].

To differentiate between malignant and benign tumors, computed tomography (CT) and 67Ga-scintigraphy were carried out on 40 patients having pelvic masses. The uptake of 67Ga-citrate was divided into three groups: negative, suspicious and positive. Contrast enhancement (CE) was employed with CT. The increases in CT numbers after contrast enhancement were divided into three groups: no enhancement (0 to 20 HU), low enhancement (21 to 40 HU) and high enhancement (over 41 HU). The results were as follows: 67Ga negative group: out of 18 patients, there was only one patient with ovarian carcinoma. The lesion was too small to detect by CT. All the remaining 17 patients had benign tumors. The increased CT numbers in benign ovarian tumors were all within the range of no enhancement. On the other hand, the increases in CT number in myoma uteri were in the range of high enhancement. 67Ga suspicious group: in 3 of 10 patients, ovarian carcinomas were detected. Because the carcinoma tissues had low enhanced CT numbers after CE, they were easily differentiated from the benign tumors. 67Ga positive group: out of 12 patients, there were 3 patients with inflammatory disease. The increases in CT numbers in the inflammatory tissue were in the range of high enhancement, so they could be clearly differentiated from malignant tissues which ranged in low enhanced CT numbers. There were no benign tumors in this group.

[Studies on adequate intervals of cisplatin administration to ameliorate cisplatin-induced nephrotoxicity].

Experimental studies were designed in order to ameliorate cisplatin-induced nephrotoxicity. Cisplatin was injected intravenously into DS mice at a dose of 5.0 mg/kg. Plasma platinum levels declined in a biphasic fashion and were undetectable after 5 days post-infusion. Renal platinum levels decreased in the same manner as the plasma levels and revealed detectable plateau levels 5 days after single injection. Cisplatin was administer once a week for 3 consecutive weeks; serial plasma and renal platinum levels and BUN were measured 7 days after each injection. The results showed that there was no significant change in the levels of plasma platinum and BUN but that the renal platinum levels increased progressively (1.6 +/- 0.3, 3.1 +/- 0.4, 4.8 +/- 2.7 micrograms/g wet wt.). After another 6 successive weeks of cisplatin administration, 55% of mice died. The renal platinum levels and BUN of the survivors were highly increased. The renal tissue revealed histologically acute renal failure. However the renal concentration of platinum was decreased to a low level of 1.1 +/- 0.4 micrograms/g wet wt. 4 weeks after the third injection. These results suggested that cisplatin-induced nephrotoxicity could be ameliorated by adequate intervals of cisplatin administration.

Substance K, a novel tachykinin injected bilaterally into the ventral tegmental area of rats increases behavioral response.

Novel tachykinins such as substance K and neuromedin K have been identified in the mammalian CNS. Bilateral injections of substance K and related peptides as well as of substance P into the dopamine cell body area of the ventral tegmental area caused dose-dependent increases in locomotor activity and rearing, in rats. As this behavior was blocked by the pretreatment with haloperidol (0.1 mg/kg, i.p.), the activation of dopaminergic systems may be involved in the hypermotility induced by the administration of substance K.

Isolation and characterization of hemagglutinins from the sponge Dysidea herbacea.

The sponge Dysidea herbacea (Keller) was found to possess hemagglutinins. The major component, DHA-I, is a protein with a mol. wt of 26,000, which dissociates into subunits of equal size (14,000). It contains large amounts of glutamic acid and aspartic acid residues, but no half-cystine, methionine or histidine residues. DHA-I reacted with rabbit and human AB0 erythrocytes. D-galactose and lactose were effective inhibitors of DHA-I. The sponge also contained a minor component(s) which reacted preferentially with rabbit erythrocytes but not with human AB0 erythrocytes.

Purification of antineoplastic factor from eggs of a sea hare.

An antineoplastic factor inducing tumor lysis was purified to apparent homogeneity from the supernatant of a homogenate of eggs of the sea hare Aplysia kurodai. On gel filtration, the purified substance gave a single band of material of 250 kDa containing 3 different subunits and this band coincided with cytolytic activity. The purified factor was half-maximally active at 10-60 ng protein/ml and lysed all tumor cells tested, but not normal cells.

Effect of a novel tachykinin, substance K, on salivation in rats.

A study of the effects of substance K (SK) and its synthetic fragments on salivation was performed. The rank order of potency of tachykinins that elicited salivation was as follows: physalaemin greater than substance P greater than eledoisin greater than kassinin greater than Arg-SK-(1-10) greater than SK-(1-10) greater than SK-(3-10). SK-(6-10) showed no activity. Atropine (1 mg/kg i.v.) had no effect on SK-induced salivation.

Enhanced production of intracellular ara-CTP after sequential use of methotrexate and 1-beta-D-arabinofuranosylcytosine.

Synergistic antiproliferative effect has been proven in vitro when mouse leukemic cells were sequentially treated with MTX and ara-C. The mechanism of this combination effect not well elucidated but the intracellular uptake of ara-C was higher when cells were pre-exposed to MTX. In this experiment, the intracellular ara-CTP was measured by HPLC after MTX and ara-C were sequentially administered to BDF1 mice bearing L1210 leukemic cell, either being sensitive or resistant to MTX. When MTX at the dose of 12 mg/kg was preceded 6 h and 3 h to ara-C at the dose of 25 mg/kg, the intracellular levels of ara-CTP were found to be significantly higher as compared with those of ara-C alone as control group. At 1 h after ara-C, ara-CTP was measured about 165 and 130% of the control levels, respectively, and at 12 h, ara-CTP was over 4 times higher of control level with group of mice to which MTX was preceded 6 h prior to MTX. On the contrary, the enhancement of ara-CTP production was definitely diminished with MTX-resistant cells in the same administrative model. From our present experiment, the time sequential modulation of intracellular ara-CTP production by MTX was reconfirmed in vivo, and this modulation might depend upon the sensitivity of MTX of leukemic cells.

Antitumor activity of aqueous extracts of marine animals.

Aqueous extracts (macromolecule fractions in particular) of 25 species of well-known marine animals were tested for their antitumor activity against transplanted sarcoma 180 solid form in ICR mice. All of the macromolecule fractions from the extracts of animals in the shellfish category except that of Mytilus edulis inhibited the growth of sarcoma 180. The use of aqueous extracts from Strongylocentrotus nudus, Halocynthia hilgendorfi f. ritteri, Styela plicata, Ecteinascidia turbinata, and Megabalanus rosa also revealed relatively high inhibition ratios of over 60%.

Varicella vaccine: case studies.

A live varicella vaccine (Oka strain) has been developed. Clinical trials in normal and high-risk children have been carried out successfully in Japan, the USA and Europe, leading to recent licensing of this vaccine in several countries. Characterization of the vaccine and the results of clinical trials are reviewed in this article.
