Clinical and experimental evaluation of the thromboprophylactic effect of hydroxychloroquine sulfate after total hip replacement.

The rate of postoperative deep vein thrombosis was studied by the 125I-fibrinogen method in 35 patients after total hip replacement. 12 out of 18 patients treated prophylactically with hydroxychloroquine sulfate developed deep vein thrombosis compared with 11 out of 17 patients in the control group. Platelet aggregability was inhibited in vitro by high concentrations of hydroxychloroquine sulfate, but these concentrations were not obtained in vivo and platelet aggregation was not limited in patients treated with hydroxychloroquine sulfate (Plaquenil).

Antiaggregatory, physiological and clinical effects of ticlopidine in subjects with peripheral atherosclerosis.

Twelve patients with varying degrees of peripheral atherosclerotic disease were given an antiaggregatory drug, ticlopidine [5-(6-chlorobenzyl)-4,5,6,7-tetrahydrothieno-(3,2-C)-pyridine HCl] in a single blind trial for one or four months and the effects on platelet aggregation, blood coagulation, marcro- and micro-circulation and walking distance were studied. Two patients were excluded; one because of nausea attributable to the drug, one because of lack of co-operation. No statistically significant changes in circulation parameters or walking distance were noted. No changes were observed in APT-time, thrombine- and Reptilase-clotting time, platelet counts, concentrations of fibrinogen and fibrinopeptide A in plasma or serum antithrombin activity. The mean concentration of fibrinopeptide A was slightly increased in all patients. ADP-induced aggregation was inhibited in all patients. Aggregation induced by arachidonic acid was partially inhibited but not abolished in all patients. Prostaglandin G2-induced aggregation was not altered by ticlopidine but collagen-induced aggregation was inhibited. Ticlopidine, in contrast to acetyl-salicylic acid, inhibits the primary aggregation but also seems to interfere with the release action. Treatment of larger patient groups for longer periods are necessary to determine the clinical usefulness of ticlopidine.

Thallium-201 scintigraphy after acute myocardial infarction.

Fifty-five patients with acute myocardial infarction were examined with thallium-201 scintigraphy before discharge from hospital. Fifty-one showed significant scintigraphic defects. The number of abnormal Q waves in a 12-lead ECG but not the maximal ASAT value for each patient were correlated to the total estimated image defect. Mortality during the follow-up period of 14-28 months was not related to the estimated total image defect, but the two early deaths occurred among patients with the most dilated left ventricles.

Vitamin A (retinol) status in the rat after a single oral dose of 2,3,7,8-tetrachlorodibenzo-p-dioxin.

The effect of a single oral dose of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD), 10 microgram . kg-1, on the status of vitamin A (retinol) was studied in rats during an eight week period after administration. Retinol was determined spectrophotometrically after separation by means of high pressure liquid chromatography. In the liver of control animals the total storage and the concentration of retinol were found to increase linearly with time whilst in the TCDD-exposed animals both the concentration and total storage remained essentially unchanged. Differences in the storage levels were evident after 4 days, and after 8 weeks the treated animals had a total liver storage corresponding to about 30% of the controls. Retinol levels in serum were significantly higher in the treated animals after week 1 and 2.

Effects of meningococcal and Escherichia coli capsular polysaccharides on human and dog platelet aggregation in vitro.

Highly purified capsular polysaccharides from groups A, B, and C meningococci and from two strains of Escherichia coli did not aggregate human or dog platelets in vitro. Nor was there any detectable effect on platelet aggregation induced by adenosine diphosphate (ADP), epinephrine, or collagen. The results do not support the hypothesis that capsular polysaccharides are involved in the pathogenesis of thrombocytopenia often seen in severe infections with these bacteria.

Interference on the haemostasis in dogs with induced coagulopathies by drugs affecting the platelet function.

Acetylsalicylic acid and phenylbutazone increased the bleeding significantly from standardized wounds in dogs defibrinogenated with Defibrase. Administration of dipyridamole and Xylocain had no effect. The results were the same in dogs treated with warfarin sodium. The results demonstrate the value of using laboratory animals with an induced coagulopathy when establishing the effect on the haemostasis exerted by drugs known in vitro to interfere with platelet functions. Acetylsalicylic acid and dipyridamole did not alter the activity of the K-vitamin-dependent coagulation factors, factors V, VIII, platelet count or fibrinogen concentration.

Acetylsalicylic acid in a trial to diminish thromboembolic complications after elective hip surgery.

Fifty-one patients were investigated concerning thromboembolic complications in the operated leg after elective surgery of the hip. Suspicion of clinical deep venous thrombosis (DVT) arose in 11 patients, i.e. 21 per cent. Thirty-five patients were investigated with phlebography. The incidence of DVT among those patients was 15 cases, i.e. 43 per cent. It is concluded that acetylsalicylic acid, a drug which inhibits the platelet release reaction and thereby blocks the platelet aggregation, given orally in a dose of 2 gram per day from the first postoperative day, is not effective in diminishing the incidence of postoperative deep venous thrombosis.

Non-ortho PCB levels in various fish species from the east and west coast of Sweden.

Non-ortho (coplanar) PCBs 77, 126 and 169 were determined in a number of fish species from the Swedish coastal environment, using either supercritical fluid extraction (SFE) or the traditional liquid/liquid extraction followed by HPLC separation on porous graphitic carbon (PGC) column prior to GC/MS analysis. While PCB 77 showed the highest concentration level, the TEQ value for PCB 126 was dominant in virtually all the samples analysed. The toxic equivalents, however, indicated some differences in analytical data between 1990 and 1992/93 but these are not significant probably owing to the short period in between.

Levamisole residues in milk from a herd of cows suffering from lungworms.

A single intramuscular dose of 7 mg levamisole hydrochloride per kg body weight was administered to a herd of 42 milking cows suffering from lungworms. Levamisole was rapidly excreted in the milk, the peak concentration being found 1 h after administration. The half-life of levamisole in milk was about 5 h. Levamisole levels were below the limit of determination (0.04 microgram/ml) in all milk samples 29 h after treatment.
