Comet assay and cytokinesis-blocked micronucleus test for monitoring the genotoxic effects of X-ray radiation in humans.

OBJECTIVE: To assess the genotoxic effects of X-ray radiation on human populations. METHODS: The single cell gel electrophoresis (SCGE) and cytokinesis-blocked micronucleus (CBMN) test were applied as biological dosimeters to detect DNA damage and abnormalities in human peripheral lymphocytes of subpopulation exposed to X-ray radiation. The subjects were divided into four groups: 12 radiation-patients; 13 intervention-radiation-therapy doctors; 32 radiation-diagnostians; 28 controls. RESULTS: The average comet lengths of the four groups were 128.17 +/- 4.49 microns, 88.09 +/- 5.39 microns, 72.68 +/- 2.57 microns and 32.87 +/- 0.57 microns, respectively. The difference in average comet length between any two groups was highly significant (P < 0.01). The average micronucleated cell (MNC) rates (@1000) of the four groups were 12.33 +/- 0.85, 9.75 +/- 1.02, 8.48 +/- 0.66 and 3.18 +/- 0.36, respectively. The difference of MNC rates of Group 1 vs 3, 1 vs 4, 2 vs 4 and 3 vs 4 was highly significant (P < 0.01), and the difference of Group 1 vs 2 was significant (P < 0.05), but there was no difference of MNC rate in Group 2 vs 3 (P > 0.05). CONCLUSIONS: This study showed that both the comet assay and the CBMN test could be used to monitor populations exposed to X-ray radiation, but the comet assay seems to be more sensitive than the CBMN test.

[Effect of kintop on gastrointestinal movement and its significance].

OBJECTIVE: To study the effect of obesity-reducing drug Kintop on gastrointestinal movement in rats. METHODS: Eighteen Wistar rats were divided into high dose (200 mg/100 g) Kintop group, low dose (125 mg/100 g) Kintop group, and control group. After anesthesia, stomach emptying time was directly observed using methylene blue as indicator. The frequency and amplitude of intestinal movement, and movement speed of intestinal content were detected. The serum tumor necrosis factora (TNF alpha) level was measured by ELISA. RESULTS: After administration of Kintop, the stomach emptying time was prolonged markedly, movement speed of intestinal content accelerated, time of dejection increased, the intestinal movement frequency and amplitude increased. There was a negative correlation between intestinal movement frequency and serum TNF alpha level (r = -0.91, P < 0.05). CONCLUSIONS: Kintop could significantly prolong the stomach emptying time, accelerate intestinal movement. These effects are correlated with not only the increased time of dejection but also the lowered serum TNF alpha level.

[Apoptosis in atrophic skeletal muscle induced by brachial plexus injury in rats].

OBJECTIVE: To study the role of apoptosis and the expression of apoptosis-associated genes in skeletal muscle atrophy induced by brachial plexus injury in rats. METHODS: Rat models of skeletal muscle atrophy were established by cutting off brachial plexus of one upper limb. Apoptosis of muscular cells was investigated by TUNEL, flowcytometry, DNA electrophoresis and electromicroscopic observation. The apoptosis associated genes such as Fas, FADD, Caspase 8, c-myc, P53 and Bcl-2 were detected by immunohistochemical method (ABC) and Northern-blot. RESULTS: It was found with TUNEL and flowcytometry that the percentage of apoptotic muscle cell rose obviously in atrophic skeletal muscle (P < 0.05). DNA laddering could be seen in DNA gel electrophoresis of atrophic muscle after brachial injury. Morphologic changes of early stage in apoptotic cell could be seen under eletcromicroscope, such as the aggregation of chromosome, the expansion of nucleic cistern and the contraction of nucleus. Fas, FADD and Caspase genes were expressed highly and Bcl-2 gene was expressed lowly with immunohistochemical method in atrophic muscle. The results were all significantly different with that of the controls (P < 0.01). But the expression changes in P53 and c-myc genes were not obvious. The result of Northern-blot indicated that the mRNA of Fas gene rose and that of Bcl-2 gene decreased obviously (P < 0.01) in atrophic muscle induced by brachial plexus injury. CONCLUSION: There are much more apoptotic cells in atrophic muscle induced by brachial plexus injury, and apoptosis plays an important role in its pathogenesis. The apoptotic signal maybe transmitted through Fas-->FADD-->Caspase 8, and the decrease in Bcl-2 gene expression aggravates the process.

[A comprehensive evaluation on psychological quality among automobile drivers].

OBJECTIVE: To study the comprehensive evaluation on the indexes of psychological quality on automobile drivers. METHODS: A index system, consisted of seven psychological indexes (including relevant physiological indexes), was developed through Delphi method and mathematical statistics. Basic data of the seven psychological and physiological indexes including mean, standard deviation and percentile, were taken from more than 8,000 occupational licensed automobile drivers and more than 6,000 trainees who were randomly sampled from 5 cities across China. The weight coefficients of indexes were determined by analytical hierarchy process (AHP). RESULTS: Comprehensive evaluation equations of the occupational automobile drivers and the trainees were established respectively. The critical values of evaluation were divided into grades by the percentiles of evaluated values. Verified results showed that there was significant difference of the values under evaluation between drivers who had accidents and who did not (t = 14.68, P < 0.01; H = 73.61, P < 0.001). The test of consistence showed that the consistence was quite good (rate of consistence = 92.99%; kappa = 0.85). CONCLUSION: Results showed that the comprehensive equations for evaluation were reasonable and feasible for detecting the potential accident-proneness drivers.

[Sarcomatoid carcinoma in breast].

OBJECTIVE: To observe the distinctive pathomorphological features of sarcomatoid carcinoma (SC) in mammary gland as well as its diagnosis and differential diagnosis. METHODS: 1 538 malignant tumors of the breast were reviewed. 15 cases (0.98%) were diagnosed as SC. AE1/AE3, carcinoembryonic antigen (EMA), vimentin, S-100 protein, actin, estrogen receptor (ER) and progesterone (PR) were performed with SP immunohistochemical staining methods. RESULTS: The SC were divided into four morphological types according to the distinctive features of their sarcomatoid components. (1) Polymorphic sarcoma type: sarcomatoid components are polymorphic; (2) Spindle cell type: sarcomatoid components consisted of spindle cells; (3) Osteoclastic giant cell type: a huge number of osteoclastic giant cells in the sarcomatoid components; (4) Myxocartilage type: myxocartilaginoid components are seen in the tumor tissue. With immunohistochemical staining, the expression rates of AE1/AE3, EMA, vimentin, S-100 protein and actins in the epithelial components were in 11, 15, 11, 12 and 8 cases respectively, and sarcomatoid components were in 10, 13, 13, 13 and 10 cases respectively, while ER and PR were positive only in 2 cases. CONCLUSION: Sarcomatoid carcinoma of the breast should be distinguished from carcinosarcoma, desmoplastic carcinoma, sarcoma, nodular fasciitis and fibromatosis.

[A study on the intrinsic compensative mechanism of craniofacial soft and hard tissue structures of Uygur adults with normal occlusion].

OBJECTIVE: To study the intrinsic compensative mechanism of craniofacial soft and hard tissue structures of Uygur adults with normal occlusion and establish standard cephalometric values to evaluate craniofacial soft and hard tissue structures of normal Uygur adults. METHODS: Lateral cephalometric radiographs of 93 Uygur adults with normal occlusion (43 men and 50 women) were evaluated with the same standard. Their age ranged from 18 to 25 years old, and they had normal and well-balanced faces without receiving any orthodontic treatment. Based on the study of craniofacial soft and hard tissue structures with normal occlusion, the horizontal tendency, the vertical tendency pattern and the medial pattern of craniofacial soft and hard tissue structures were compared, and the intrinsic compensative mechanism in the inner structures of the horizontal and the vertical tendency patterns was investigated. Finally 21 indexes were analysed by mean squar deviation and Newman-Keuls test. RESULTS: The obvious compensative changes in the horizontal and vertical tendency patterns were found in teeth, skeleton and soft-tissues. 1. When the horizontal tendency was compared with the medial pattern, the value of ODI and angles of FH-NA and FH-NB increased (P < 0.01), while the angle of SN-PP decreased (P < 0.01). It showed that the anterior maxilla rotated in the counter-clockwise direction, and the anterior mandible rotated in the clockwise direction. The angle of U1-L1 increased (P < 0.01), which meant that the inclination of anterior teeth decreased. 2. When the vertical tendency was compared with the medial pattern, the value of ODI decreased (P < 0.01), while the height of ANS-Me, U1-PP, L1-MP, and the thickness of LL-SnPg increased (P < 0.01); The angle of L1-MP decreased (P < 0.05), while the length of Sn-Stms increased (P < 0.05). These results showed that there was an obvious positive relationship between hard and soft tissues. The soft tissue thickness could compensate the changes of hard tissues to keep the facial patterns harmony. CONCLUSION: Normal occlusion depends on the homeostasis of teeth, skeleton and soft tissues, but doesn't only depend on the normal craniofacial soft and hard tissue structures respectively. The intrinsic compensative mechanism plays an important role in forming normal occlusion, therefore, doctors should choose the best treatment to obtain optimal individual intrinsic compensative mechanism and achieve an optimal balance between the craniofacial structures and the occlusion.

[An cephalomectric study on craniofacial structures of Uygur adults with normal occlusion in Xinjiang autonomous region].

OBJECTIVE: To study craniofacial structures of Uygur adults with normal occlusion and establish the standard cephalometric values to evaluate the craniofacial soft and hard tissue structures of normal Uygur adults. METHODS: Lateral cephalometric radiographs of 93 Uygur adults with normal occlusion (43 men and 50 women) were evaluated with the same standard. Their age ranged from 18 to 25 years old, and they had normal and well-balanced faces without receiving any orthodontical treatment. The lateral cephalogram of each normal adult was traced digitized and measured by the same computer system, then the indexes of craniofacial structures were analyzed by multifactor cluster analysis. 18 typical items were chosen to analyze the characters of the craniofacial structures of Uygur adults with normal occlusion. At the same time, Bayers discriminant analysis was used to identify and evaluate the cluster results. RESULTS: Uygur adults with normal occlusion were divided into three patterns according to the multifactor cluster analytic results: horizontal tendency pattern (40%), medial pattern (43%) and vertical tendency pattern (17%). When the horizontal tendency pattern was compared with the medial pattern, it showed that the height of S-Go increased (P < 0.05), while the height of ANS-Xi-Pm decreased (P < 0.05); The proportion of S-Go/N-Me increased (P < 0.01). The angle of Ar-Go-Me and Y axis decreased (P < 0.05), which meant that the mandible rotated in the counter-clockwise direction. SN-MP, PP-MP, OP-MP, MP-FH all decreased (P < 0.01), which meant that the SN plane, the PP plane, the occlusion plane and the MP plane all had the trend of hypodivergence. When the vertical tendency pattern was compared with the medial pattern, the height of Ar-Fo decreased (P < 0.01), while the height of ANS-Me increased (P < 0.01); The angle of Ar-Go-Me, Y axis, SN-MP, PP-MP, OP-MP and MP-FH all increased (P < 0.01). These results showed that the mandible rotated in the clockwise direction and the SN plane, the PP plane, the occlusion plane, and the MP plane all had the trend of hyperdivergence. At last, according to the different skeletal patterns, the normal cephalometric values of Uygur adults with normal occlusion were established. CONCLUSION: Significantly craniofacial structure difference exist in the adults with normal occlusion. Therefore, it is necessary to establish the normal cephalometric values of Uygur adults with normal occlusion according to different skeletal patterns.

[Differentiation of human glioma cells BT-325 induced by sodium butyrate].

OBJECTIVE: To study the differentiation of human glioma cells BT-325 induced by sodium butyrate in vitro. METHODS: BT-325 cells were treated with 1 mmol/L sodium butyrate. Flow cytometry was used to analyze the cell cycle. Cell differentiation was identified by flow cytometry and Western blot analysis. RESULTS: After 6-8 days of sodium butyrate treatment, the differentiation characters could be observed distinctively, such as the reduced cell density, the increased cell size and the marked increase in cell process formation and cell-to-cell connection. At the same time, Western blot showed that the amount of glial fibrillary acidic protein was elevated after the sodium butyrate treatment. CONCLUSION: Human glioma cells BT-325 could be induced to differentiation by sodium butyrate.

[Clinical and experimental studies on condyloma acuminata].

Condyloma acuminata(CA) is one of sexually transmitted diseases(STD) caused by human papillomavirus(HPV). We have done systemic study on the CA in recent years. Analyzing the morbidity of CA in our STD clinics showed that CA was in the first or second place of STD. Tissue specimens from CA were detected HPV DNA by dot blot hybridization and PCR. The result showed PCR was more sensitive than dot blot hybridization. HPV DNA were detected by PCR in tissue specimens of cervical squamous carcinoma, as well as cervical and vulvar CA. The result confirmed that HPV 16 was main type in cervical squamous carcinoma, and HPV 6 and 11 were main types in cervical and vulvar CA. To differentiate pseudocodyloma of vulva from CA, electron microscopy and HPV DNA detection were done. Certain ultrastructural changes were helpful, and detection of HPV DNA was better method in differentiating CA from pseudocondyloma. High purification of podophyllintoxin was studied and the effectiveness of podophyllintoxin was observed, compared to imported podophyllintoxin to treat CA, both cure rates were more than 80% after 3 week's therapy. Intralesional injection of different interferons for CA was effective.

Isolation and identification of cDNA fragments and full-length cDNA differentially expressed in human glioblastoma cell line BT-325 versus all-trans retinoic acid induction .

OBJECTIVE: To investigate the differentiation process of the human glioblastoma cells. METHODS: Differential display reverse transcribed-PCR (DDRT-PCR) was used to isolate the genes differentially expressed in control and all-trans retinoic acid treated human glioblastoma cell line BT-325. Routine method of cDNA library screening was performed to clone full-length cDNA. RESULTS: Thirty-six RT-PCR reactions were performed and 64 differentially expressed fragments were recovered, amplified and cloned. Of them, 46 ESTs were sequenced and delivered into the GenBank. The homology comparison using BLAST algorithm revealed that 22ESTs are highly homologous with the known genes and many of them play important roles in the cell differentiation progress. A dot-blot hybridization was conducted to certify the differentiation expression. The result showed that 27 EST clones are expressed at different level in control and all-trans retinoic acid treated BT-325 cells. A full-length cDNA was cloned using the EST-HGBB098. CONCLUSION: DDRT-PCR was a simple and effective method to serially analyze the differentially expressed genes.

Effects of VEGF on hemodynamics and cardiac function: characterization and mechanisms.

Vascular endothelial growth factor (VEGF), a mitogen specific for endothelial cells, produces beneficial angiogenesis in animal models of ischemic diseases; however, its side effects on hemodynamics and cardiac function may limit clinical use for some indications. This review focuses on the VEGF-induced hemodynamic profile, including hypotension, tachycardia, decreases in cardiac output and stroke volume, and changes in preload and afterload. The hemodynamic effects of VEGF are due to vasodilation and vascular hyperpermeability, which are mainly mediated by nitric oxide. Finally, the regimen or strategy for attenuation or prevention of these side effects is discussed.

Palmitoylation occurs at cysteine 347 and cysteine 351 of the dopamine D(1) receptor.

To determine the palmitoylation sites in the human dopamine D(1) receptor, we expressed wild type and mutant receptors in which candidate cysteines in the carboxyl tail were substituted by alanines both individually (A347, A351) and together (AA). Our results showed that palmitoylation levels of A347 and A351 were reduced substantially and that AA had no detectable signal of palmitoylation. These data indicate that cysteines 347 and 351 were both palmitoylated and that they were the only sites of palmitoylation. We introduced a cAMP-dependent protein kinase site encompassing the position 351. We predicted that a functional cAMP-dependent protein kinase site would impair receptor-G protein coupling if it is not occluded by palmitoylation. Our results demonstrated that indeed, the introduction of the cAMP-dependent protein kinase site caused reduced potency of dopamine stimulation of adenylyl cyclase, and thus confirmed that when unoccluded, the cAMP-dependent protein kinase site introduced to position 351 of dopamine D(1) receptor could confer constitutive desensitization.

The human family of Deafness/Dystonia peptide (DDP) related mitochondrial import proteins.

The gene responsible for the human genetic neurodegenerative disorder DFN-1/MTS encodes a small protein known as deafness/dystonia peptide (DDP). It bears a strong resemblance to a recently characterized set of zinc-binding yeast proteins (Tim8p, Tim9p, Tim10p, Tim12p, and Tim13p) that are implicated in the import of a class of transmembrane carrier proteins from the cytoplasm to the mitochondrial inner membrane. We describe here the human complement of DDP/Tim-like proteins and establish the likely orthologous relationships between sequences from human, yeast, and other organisms. We also describe the expression patterns and chromosomal locations of their genes, which are candidate loci for autosomal recessive neurodegenerative disorders.

Genome-wide expression profiling in Escherichia coli K-12.

We have established high resolution methods for global monitoring of gene expression in Escherichia coli. Hybridization of radiolabeled cDNA to spot blots on nylon membranes was compared to hybridization of fluorescently-labeled cDNA to glass microarrays for efficiency and reproducibility. A complete set of PCR primers was created for all 4290 annotated open reading frames (ORFs) from the complete genome sequence of E.coli K-12 (MG1655). Glass- and nylon-based arrays of PCR products were prepared and used to assess global changes in gene expression. Full-length coding sequences for array printing were generated by two-step PCR amplification. In this study we measured changes in RNA levels after exposure to heat shock and following treatment with isopropyl-beta-D-thiogalactopyranoside (IPTG). Both radioactive and fluorescence-based methods showed comparable results. Treatment with IPTG resulted in high level induction of the lacZYA and melAB operons. Following heat shock treatment 119 genes were shown to have significantly altered expression levels, including 35 previously uncharacterized ORFs and most genes of the heat shock stimulon. Analysis of spot intensities from hybridization to replicate arrays identified sets of genes with signals consistently above background suggesting that at least 25% of genes were expressed at detectable levels during growth in rich media.

Human neuronal gamma-aminobutyric acid(A) receptors: coordinated subunit mRNA expression and functional correlates in individual dentate granule cells.

gamma-Aminobutyric acid(A) receptors (GABARs) are heteromeric proteins composed of multiple subunits. Numerous subunit subtypes are expressed in individual neurons, which assemble in specific preferred GABAR configurations. Little is known, however, about the coordination of subunit expression within individual neurons or the impact this may have on GABAR function. To investigate this, it is necessary to profile quantitatively the expression of multiple subunit mRNAs within individual cells. In this study, single-cell antisense RNA amplification was used to examine the expression of 14 different GABAR subunit mRNAs simultaneously in individual human dentate granule cells (DGCs) harvested during hippocampectomy for intractable epilepsy. alpha4, beta2, and delta-mRNA levels were tightly correlated within individual DGCs, indicating that these subunits are expressed coordinately. Levels of alpha3- and beta2-mRNAs, as well as epsilon- and beta1-mRNAs, also were strongly correlated. No other subunit correlations were identified. Coordinated expression could not be explained by the chromosomal clustering of GABAR genes and was observed in control and epileptic rats as well as in humans, suggesting that it was not species-specific or secondary to epileptogenesis. Benzodiazepine augmentation of GABA-evoked currents also was examined to determine whether levels of subunit mRNA expression correlated with receptor pharmacology. This analysis delineated two distinct cell populations that differed in clonazepam modulation and patterns of alpha-subunit expression. Clonazepam augmentation correlated positively with the relative expression of alpha1- and gamma2-mRNAs and negatively with alpha4- and delta-mRNAs. These data demonstrate that specific GABAR subunit mRNAs exhibit coordinated control of expression in individual DGCs, which has significant impact on inhibitory function.
