Antitumor activity of siwenmycin: a novel anthracycline antibiotic.

A novel anthracycline antibiotic, siwenmycin, isolated from the culture of Streptomyces galilaeus var. siwenesis, was examined for its antitumor activities against P388, K562, B16-F10, HeLa, HEp-2 and Lewis lung carcinoma cell lines. The results showed that siwenmycin was effective against P388, K562, HeLa and HEp-2 tumor cell lines in vitro, and significantly inhibited the growth of the Lewis lung carcinoma cell line in vivo. Siwenmycin could also suppress spontaneous and artificial pulmonary metastases of B16-F10 and Lewis lung carcinoma cell lines in C57BL/6 mice. The inhibitory effect of siwenmycin on spontaneous pulmonary metastasis of Lewis lung carcinoma in C57BL/6 mice was even stronger than that of adriamycin (ADM), which is, at present, commonly used in clinical practice. Furthermore, the double-labeling test used in this study has verified that siwenmycin can inhibit cellular RNA synthesis at about one tenth the concentration required to inhibit DNA synthesis to the same degree, indicating that the antitumor mechanism of siwenmycin also differs from that of ADM. The acute toxicity of siwenmycin was very low, and it was as effective in vivo as in vitro, suggesting that this newly found antibiotic should be studied for possible clinical antitumor applications.

[DNA repair capacity measurement and possibility of heterozygote detection using nucleoid sedimentation technique among members taken from 3 xeroderma pigmentosum (XP) families].

The nucleoid sedimentation test was used to analyse the DNA repair capacity in 35 members taken from 3 xeroderma pigmentosum (XP) families after their peripheral blood lymphocytes being exposed to UV irradiation or N-methyl-N-nitro-N-nitrosoguanidine (MNNG) at a dose of 2.5 microJ/mm2 and 2 micrograms/ml respectively. The results showed that the 6 XP patients all failed to repair their damaged DNA even after 20-hour incubation at 37 degrees C. The repair ratios were 0.61 +/- 0.13 for UV and 0.44 +/- 0.15 for MNNG while those of non-related normal members usually reached 0.96 +/- 0.07 and 0.71 +/- 0.07 for both UV and MNNG after 11-hour incubation. It is interesting to note that 9 obligatory heterozygotes (parents and grandparents of XP patients) and 4 highly possible heterozygotes (blood relatives of XP patients) also showed very poor DNA repair capacity: they could not accomplish their repair process even after 20-hour incubation. It has been showed that the nucleoid sedimentation test can probably be used to pick out the heterozygotes in XP families.

In vivo cloning of proline genes and its expression in Escherichia coli.

The wild type proA+, B+ genes of E. coli were cloned in vivo using a plasmid containing a mini-Mu replicon, pEG5005. The cloning frequency was about 1.46 x 10(-3)/Kanr transductant. Genetic and biochemical analysis of these clones indicated that the proA+, B+ genes are on the plasmid pEG5005. The secretion of proline were assayed for 500 Pro+ clones. However, no proline accumulation was detected. A Pro+ clone pPR3 was mutagenized in vivo by NTG and the mutants resistant to D-proline were obtained. One of the Dpr mutants pPR7 was found to produce 0.35 mg/ml proline in a proA B deletion strain. When pPR7 was transferred into a proline producing strain, the yield of proline increased up to 2.5 mg/ml, which is 7 and 2.5 times higher than that of the donor and recipient respectively. The physical maps of pEG5005 and pPR3 were roughly established.
