[Diagnosis of breast-duct disease by breast duct endoscope].

OBJECTIVES: To study the pathological characteristics of breast-duct disease under breast duct endoscope in diagnosis and treatment of breast diseases. METHODS: The images of breast duct of 62 patients with nipple discharge were obtained by hard breast duct endoscope (Olympus Inc) and computer assistant analysis system. RESULTS: Of the 62 patients, malignant tumor was found in 4, premalignancy in 6, intradutal papilloma in 15, and mammary duct or plasma cell mastitis in 37. CONCLUSIONS: Intraductal lesions of patients with nipple discharge can be observed and be followed up with breast duct endoscope. The accuracy of etiological diagnosis of patients with nipple discharge, after the use of breast duct endoscope can partly modify the indications for operation in patients with nipple discharge.

[A primary study on seed production of medicinal plant Sinopodophyllum hexandrum].

The study on the seed production of five Sinopodophyllum hexandrum (Berberidaceae) populations demonstrated that the average ovule number, average seed number and seed set ratio of these populations were varied, but all positively related with the altitude of the population. The average ovule number of a flower ranged from 58.79 to 87.97, average seed number of a fruit ranged from 40.02 to 80.58, and average seed set ratio of population ranged from 61.29% to 91.60%. The average weight per seed in a fruit was negatively related with the altitude and seed number in the fruit. The result also showed that seed production of a population had no obvious difference in different years. Although seed production was mainly influenced by nutrition condition of the individual, it was still influenced by altitude and other factors. Seed number in a fruit was positively related with fruit size(including weight, length and width) and total seed weight of the fruit, but average single seed weight of a fruit had no obvious relation with fruit size and total seed weight of the fruit.

Modified cold storage of rat livers with self-made HYD solution.

OBJECTIVE: To investigate the cold preservation effect on rat livers of a modified storage method with self-made HYD solution. METHODS: The vascular bed of rat livers was expanded with an additional 20 to 40 ml self-made HYD solution/100 g liver. After resection of the liver, the extra HYD solution (expressed as % liver weight) was entrapped via portal infusion by tying off the supra- and infra-hepatic inferior vena cava. Forty rats were randomly divided into four groups including control group with conventional storage method, and 20%, 30% and 40% groups according to the amount of extra HYD solution. We compared the preservation effect of the modified storage method with that of the conventional storage method using an isolated perfused rat liver model. RESULTS: Bile production and all the indices of hepatic microcirculation including portal perfusion pressure, endothelin-1 in the effluent, trypan blue distribution time and histology were significantly superior in the modified method groups compared to those in the control group (P < 0.05). The contents of dihydroxybenzoic acid (DHBA) in the modified method groups were significantly lower than those in the control group (P < 0.05). Liver enzymes activities in the 30% group were markedly lower than those in the control group (P < 0.05). The preservation effect on rat liver in the 30% group was the best among the modified method groups. CONCLUSION: The modified cold storage method is effective and may have potential for clinical application in liver preservation.

Low power density microwave radiation induced early changes in rabbit lens epithelial cells.

OBJECTIVE: To determine whether low power density microwave radiation can induce irreversible changes in rabbit lens epithelial cells (LECs) and the mechanisms of the changes. METHODS: One eye of each rabbit was exposed to 5 mW/cm2 or 10 mW/cm2 power density microwaves for 3 hours, while the contralateral eye served as a control. Annexin V-propidium iodide (PI) two-color flow cytometry (FCM) was used to detect the early changes in rabbit lens epithelial cells after radiation. RESULTS: Lots of rabbit LECs were in the initial phase of apoptosis in the 5 mW/cm2 microwave radiation group. A large number of cells became secondary necrotic cells, and severe damage could be found in the group exposed to 10 mW/cm2 microwave radiation. CONCLUSION: Low power densities of microwave radiation (5 mW/cm2 and 10 mW/cm2) can induce irreversible damage to rabbit LECs. This may be the non-thermal effect of microwave radiation.

[Influence of specimen processing method on Mycobacterium tuberculosis detection by PCR-reversed dot hybridization].

OBJECTIVE: To evaluate specimen processing with sodium dodecyl (lauryl) sulfate (SDS) on the detection of Mycobacterium tuberculosis by PCR-reversed dot hybridization. METHODS: Totally 266 specimens were collected from sputum, pleural fluid, ascites, urine or blood of patients with active tuberculosis and were treated with SDS or routine method. All specimens handled were tested for detecting M. tuberculosis by PCR-reversed dot hybridization. RESULTS: The detection rate of M. tuberculosis from specimens prepared by SDS method was 65.0% (173/266) as compared with 51.5% (136/168) by routine method. Results from two methods were considered statistically significant (chi(2) = 35.03, P < 0.001). CONCLUSION: The SDS processing method may be used to detect M. tuberculosis from clinical specimens by PCR and enhances the detection rate of M. tuberculosis.

[Relationship between dopamine and iron contents in the brain of parkinsonian rats].

Using fast cyclic voltammetry (FCV), atomic absorption/flame emission spectrophotometry and high performance liquid chromatography for electrochemical detection, we studied the change in iron content in the substantia nigra (SN) of 6-hydroxydopamine (6-OHDA) lesioned Parkinsonian (PD) rats and the toxic effect of intranigral injection of iron on DA neurons. The neuroprotective effect of desferrioxamine mesylate was also observed. The results are as follows. (1) The iron content in SN on the lesioned side of 6-OHDA-lesioned PD rats was about three times as high as that in nonstandard PD rats. (2) The iron content in caudate putamen (CPu) on the lesioned side of PD rats was not different from that on the unlesioned side. (3) DA release as well as the content of DA and its metabolites were significantly decreased on the lesioned side of PD rats. (4) In the rats pretreated with intracerebroventricular desferrioxamine mesylate before 6-OHDA injection, the release and content of DA on the lesioned side were not significantly different from those on the unlesioned side. (5) Intranigral injection of 40 micrograms FeCl3 resulted in a dramatic reduction of both DA release and content in CPu. The above results strongly suggest that 6-OHDA reduces the DA release from CPu, in which iron plays an important role. Elevation of iron content in SN is one of the mechanisms responsible for the reduction of DA content. Desferrioxamine mesylate may exert a protective action on dopaminergic neurons.

[Profile of telomerase and telomerase RNA expression in nasopharyngeal carcinogenesis of rats induced by N, N'dinitrosopiperazine (DNP)].

OBJECTIVE: To investigate the profile of telomerase and telomerase RNA expression in nasopharyngeal carcinogenesis (NPC) induced by N, N'dinitrosopiperazine (DNP) and examined histolofically. METHODS: Nasopharyngeal carcinomas of rats were induced by DNP and examined histologically. PCR-ELISA and nested RT-PCR were used to assay telomerase and telomerase RNA expression at different stages in the nasopharyngeal tissues of rats. RESULTS: During the carcinogenesis process, telomerase activity increased along with the formation of a nasopharyngeal carcinoma. Telomerase expression was positively related with nasopharyngeal carcinogenesis. Telomerase RNA expression was present and did not change during the NPC process. Expression of telomease RNA was earlier than telomerase activation. Telomerase activation and telomerase RNA expressin were also detected in the pre-cancerous nasopharyngeal lesions. CONCLUSION: Telomerase activation may participate in the onset and progression of NPC, and is an early step in NPC.

[A study on the mechanism of the biological roles of danshensu on fibroblast].

OBJECTIVE: To explore the effects of danshensu on fibroblast apoptosis and the expression of procollagen gene. METHODS: Danshensu was added to in vitro culture of human cutaneous fibroblasts, and the nuclear proteins and total RNA were harvested from the cells. Electrophoretic mobility shift assay (EMSA) was adopted to determine the combining activity of nuclear transcription factors NF-kB and NF-1. Cell apoptosis was analyzed by DNA ladder fragmentation. The procollagen gene expression was analyzed by RT-PCR. RESULTS: Eight hours after the action of danshensu on the culture fibroblasts, the NF-kB combining activity was almost inhibited completely and the NF-1 combining activity decreased by about 50%. In addition, ladder-like DNA fragments were revealed clearly by sepharose electrophoresis. The mRNA levels of type I procollagen alpha1 and alpha2 decreased by 56% and 59%, respectively. CONCLUSION: Danshensu could inhibit the nuclear transcription factor NF-kB activity of fibroblast and induce the occurrence of its apoptosis. Furthermore, danshensu could also inhibit the nuclear transcription factor NF-1 activity of fibroblast and modulate the synthesis and secretion of collagen.

[Clinical observation of the protective effect of oral feeding of glutamine granules on intestinal mucous membrane].

OBJECTIVE: To observe the protective effect of oral feeding of glutamine (Gln) granules on intestinal mucous membrane during early postburn stage. METHODS: Twenty burn patients were randomly divided into test (T) and control (G) groups with 10 patients in each group. Gln granules and placebo were administered in a dose of 10 grams three times a day for 10 days to patients in the two groups respectively. Blood levels of Gln, diamine oxidase (DAO) activity, endotoxin (LPS) and intestinal permeability were monitored before and after the administration of the agents. at the same time, the changes in blood and urine routine and hepatic and renal functional indices were also observed. RESULTS: There was no difference of the general data and the hepatic and renal functional indices between the two groups. The plasma level of Gln before the administration was much lower than that thereafter (P < 0.01) in T group, but all of the values were higher than that in C group (P < 0.05). The DAO activity, intestinal permeability and LPS levels before the administration were much higher than those after it (P < 0.01) in T group. CONCLUSION: Gln might be good to burn patients due to its protective effect on intestinal mucous membrane when given during early postburn stage.

[Clinical study of P-gp, and bcl-2 protein expression in non-Hodgkin's lymphomas patients].

OBJECTIVE: To investigate the relationship between the expression of P-gp, P26-bcl-2 and the prognosis in intermediate and high grade non-Hodgkin's lymphomas (NHL). METHODS: Sixty cases of intermediate and high grade NHL were retrospectively reviewed using immunohistochemical method. All patients were received CHOP chemotherapy over 4 courses. RESULTS: P-gp was identified in 15 and P26-bcl-2 in 25 cases. The 3-year survival rates for patients with P26-bcl-2 (+) and P26-bcl-2 (-) were 37.64% and 76.80%, respectively (P < 0.005), and for patients with both positive P-gp and P26-bcl-2 and both negative were 15.38% and 48.48%, respectively (P = 0.038). CONCLUSIONS: There is direct relationship between the P-gp, P26-bcl-2 protein expression and the prognosis in intermediate and high grade non-Hodgkin's lymphoma.

[Production of HPV 16 major capsid protein L1 with baculovirus expression system].

BACKGROUND: To obtain human papillomavirus type 16(HPV 16) major capsid protein L1 with baculovirus expression system. METHODS: Using pFb1 as a vector,a recombinant baculovirus expressing plasmid,which contains HPV16L1 gene sequence,was generated. The constructed virus was infected into an insect cell line Sf9. RESULTS: After incubating at 27 degrees for 72 hours, the infected cells were collected and total cellular poteins were extracted. SDS-PAGE assay revealed a roughly 56 000 expressed protein and Western blot confirmed that the expressed protein arose from HPV16L1. CONCLUSIONS: HPV16 later protein L1 could be efficiently expressed with baculovirus expression system,and the expressed L1 protein remains to have good immunoreactivity. This study may supply a basic work for preparing virus-like particle and prophylactic HPV subunit vaccines.

[Annexin V for flow cytometric detection of phosphatidylserine expression on lymphoma cells undergoing apoptosis].

OBJECTIVE: To quantitatively analyze apoptotic and secondary necrotic cells under apoptosis conditions. METHODS: The cells of Burkitt lymphoma cell line Raji were incubated with 1.0 mumol/L dexamethasone (DEX) for 2, 4 and 8 h, then stained with Annexin V-FITC (fluorescein isothiocyanate conjugated) which was used to detect the exposure of phosphatidylserine (PS) on the out membrane resulting from a loss of phospholipid asymmetry in the early stage of apoptosis, and also stained with propidium iodide (PI) which allows the analysis of secondary necrotic cells related with cell membrane and DNA damage, then apoptotic cells was quantified by flow cytometry (FMC). Furthermore, Annexin+/PI- and Annexin+/PI+ cells were sorted by fluoresence-activated cell sorter (FACS), and identified by electron microscopy (EM) and DNA gel electrophoresis. RESULTS: The results revealed that the percentage of apoptotic cells was increased and correlated well with incubation time (r = 0.97). The sensitivity of this method was shown by its detection limit 0.02%; the method was reproducible, and the coefficient variance (CV) was 4.2%. Meanwhile, the Annexin+/PI- and Annexin+/PI+ cells were identified as apoptotic and necrotic cells under EM, and the DNA extracted from the Annexin+/PI- cells was characterized by "ladder pattern". CONCLUSION: Annexin V assay for analyzing apoptotic cells is specific, sensitive, accurate, reproducible and quantitative for apoptosis investigation.

[Counting platelets by flow cytometry with platelet-specific antibody CD42a].

OBJECTIVE: To investigate a reference method for platelet (PLT) count. METHODS: 76 samples of daily whole blood of high, normal and low values in patients were collected and analyzed by flow cytometry (FCM) with CD42a monoclonal antibody that could combine specifically with the platelet membrane glycoprotein Ib-Ix (GPIb-Ix). PLT counts were analyzed by multiplying PLT/RBC ratio from FCM with the RBC that was counted by a calibrated blood analyzer. Meanwhile, the results of PLT from FCM were compared with that from blood analyzers SE-9500, NE-1500, CD-1600 or manual count. RESULTS: The within-replicate coefficient of variance(CV) of FCM was less than 4% for a high count(501 x 10(9)/L) and middle count(184 x 10(9)/L), the CV of low count(23 x 10(9)/L) was 11.432%, the between-replicate CV was 10.792%, and the overall reproducibility was less than 5%. Linearity was demonstrated well(r = 0.9975 P < 0.001) when the PLT counts were 20 x 10(9)/L-700 x 10(9)/L. PLT count was stable at room temperature within 6 hr. The carry-over was 0.87%. The results between FCM and SE-9500, NE-1500, CD-1600 or manual count was comparable (r > 0.975, P < 0.001). CONCLUSION: These findings confirm that FCM for PLT count is specific, accurate and precise and could be considered as a candidate of reference method for PLT count.

[The influence of labyrinthectomy on the expression of NMDA R1 mRNA].

OBJECTIVE: To study the effect of labyrinthectomy on the expression of NMDAR1 (N-methyl-D-aspartate receptor subunit R1). METHOD: The in situ hybridization was used to observe the expression of NMDA R1mRNA post-unilateral labyrinthectomy and investigate its effect on vestibular compensation. RESULT: 20 h post-unilateral labyrinthectomy, the NMDA R1mRNA positive neurons in the medial vestibular nucleus increased significantly compared with the unlesioned side(P < 0.01). 40 h-6 d, the positive cells decreased gradually. 6 d post lesion, the positive neuron counts of the lesioned side had no significant difference compared with those of the un-lesioned side (P > 0.05). CONCLUSION: The changing trend of the expression of NMDA R1mRNA in the medial vestibular nucleus is related to the recovery of the vestibular static symptoms after unilateral labyrinthectomy. It is suggested that the modulation of the efficacy of the NMDA R1 play an important role in the compensation of the static symptoms.

[Reversed-phase high performance liquid chromatography used for determination of amino acids of parasites].

The compositions and contents of amino acids of parasites in different areas of China have been determined by precolumn derivatization, reversed-phase high performance liquid chromatography (HPLC) separation and ultra-violet absorption quantitation in order to prevent diseases caused from parasites. The results show that there were some differences in contents of amino acids in B. malayi from seven different areas in China. The contents of amino acids gradually increased from the southern to the northern areas in China. The method is simple and accurate, and the results have important values to investigate the life activities of parasites.

[Effects of zinc deficiency on the distribution of elements in the tissue of pregnant rats and their fetuses].

The influence of zinc(Zn) deficiency on the distribution of elements in tissues of pregnant rats and their fetuses was investigated. Pregnant Wistar rats were divided into zinc deficiency (ZD), pair-fed(PF), zinc supplement (ZS) and control (C) groups. ZD rats were fed for 21 days a zinc deficient (0.7 mg/kg) diet and other rats were fed a similar diet supplemented with zinc (100 mg/kg). On the 21st day of gestation, blood was drawn from heart, laparotomy was performed, and liver, spleen, kidney, placentas, fetuses were removed for analysing on zinc, copper (Cu), iron (Fe), manganese (Mn) and calcium (Ca). The results showed that the element levels in blood, organs and fetuses from ZD rats were significantly lower than those from ZS and C rats. Furthermore, the levels of Cu, Fe, Mn in blood, liver, spleen, kidney of ZD rats were significantly lower than those of PF rats. It suggests that there are some coordination relationships in intestinal absorption and tissue distribution between Zn and other elements, and the decrease of elements in the body does not mainly result from the reduction of dietary intake caused by zinc deficiency. The Fe content in placentas and Fe, Ca content in fetuses of ZD rats showed a significant increase as compared with those of rats in other groups. There may be a competitive mechanism on the transportation of Zn and Fe in the placenta of rats. In brief, the distribution of minerals in maternal tissues induced by zinc deficiency might be different from those in fetuses.

[Investigation of the effects of various factors affecting apoptosis of chronic lymphocytic leukemic cells mediated by P2Z receptors].

OBJECTIVE: To explore the effects of various factors affecting apoptosis of chronic lymphocytic leukemia (CLL) cells mediated by P2Z purinoceptor. METHODS: In vitro exposure of CLL cells with P2Z receptors to 1 mmol/L ATP or 0.1 mmol/L benzoylbenzoic ATP (BzATP) for 8 h in the presence of 1.0 mmol/L Mg2+ or other bivalent cations, 150 mmol/L choline in media, 1, 2, 4 mmol/L EDTA or EGTA, or under different temperature. ATP-induced apoptosis was detected by agarose gel electrophoresis, TdT assay and flow cytometric analysis (FCA). RESULTS: The results showed that 1. Extracellular Mg2+ or Ca2+ stimulated ATP-induced DNA fragmentation in a dose-dependent manner, and the compatible evidence was provided by the inhibition of ATP-induced DNA fragmentation in the presence of EGTA or EDTA; 2. ATP-induced DNA fragmentation was completely inhibited by 1.0 mmol/L Zn2+; 3. ATP-induced DNA breaks were not affected by Ba2+, Sr2+, Co2+ when they were substituted for extracellular Mg2+ or Ca2+; 4. Choline, an inhibitor of phospholipase D (PLD) stimulated by ATP through P2Z receptor in human lymphocytes, was also a partial inhibitor of ATP-induced DNA fragmentation, and the results were confirmed by flow cytometric analysis (FCA); 5. ATP-induced DNA fragmentation was completely obliterated when the temperature was lower than 10 degrees C. CONCLUSION: These results suggest that the endonuclease and PLD may be involved in ATP-induced apoptosis in human lymphocytes via P2Z receptor.

[Improved APAAP immunocytochemical staining-trace cytotoxicity test plate method for detection of cell surface antigens].

OBJECTIVE: To improve the conventional slide method of alkaline phosphatase-anti-alkaline phosphatase (APAAP) immunocytochemical staining for detection of cell surface antigens. METHODS: Trace cytotoxicity test plates were utilized in the detection of CD3, CD4, CD8, CD11a, CD11b, CD18 and CD54 on lymphocytes by APAAP immunocytochemical staining. The percentages of positive cells were counted. Meanwhile, slides were used in the same objects and procedure. RESULTS: The results of trace test plate method were similar to those of slide method (P > 0.05, respectively). The reproducibility was very excellent. The plate method decreased the volume of blood from 10 ml to 2 ml to detect 7 norms, and each reagent from 50 microliters to 5 microliters. CONCLUSION: Trace test plate method may not only attain the result similar to that of the slide method, but also save blood and reagents; it is easy to do, so it is more suitable for clinical examination and scientific research in hospitals or institutions at the basic level.
