Molecular cytogenetic discrimination and reaction to wheat streak mosaic virus and the wheat curl mite in Zhong series of wheat--Thinopyrum intermedium partial amphiploids.

Thinopyrum intermedium (2n = 6x = 42, JJJsJsSS) is potentially a useful source of resistance to wheat streak mosaic virus (WSMV) and its vector, the wheat curl mite (WCM). Five partial amphiploids, namely Zhong 1, Zhong 2, Zhong 3, Zhong 4, and Zhong 5, derived from Triticum aestivum x Thinopyrum intermedium crosses produced in China, were screened for WSMV and WCM resistance. Zhong 1 and Zhong 2 had high levels of resistance to WSMV and WCM. The other three partial amphiploids, Zhong 3, 4, and 5, were resistant to WSMV, but were susceptible to WCM. Genomic in situ hybridization (GISH) using a genomic DNA probe from Pseudoroegneria strigosa (SS, 2n = 14) demonstrated that two partial amphiploids, Zhong 1 and Zhong 2, have almost the identical 10 Th. intermedium chromosomes, including four Js, four J, and two S genome chromosomes. Both of them carry two pairs of J and a pair of Js genome chromosomes and two different translocations that were not observed in the other three Zhong lines. The partial amphiploids Zhong 3, 4, and 5 have another type of basic genomic composition, which is similar to a reconstituted alien genome consisting of four S and four Js genome chromosomes of Th. intermedium (Zhong 5 has two Js chromosomes plus two Js-W translocations) with six translocated chromosomes between S and Js or J genomes. All three lines carry a specific S-S-Js translocated chromosome, which might confer resistance to barley yellow dwarf virus (BYDV-PAV). The present study identified a specific Js2 chromosome present in all five of the Zhong lines, confirming that a Js chromosome carries WSMV resistance. Resistance to WCM may be linked with J or Js chromosomes. The discovery of high levels of resistance to both WSMV and WCM in Zhong 1 and Zhong 2 offers a useful source of resistance to both the virus and its vector for wheat breeding programs.

Effects of thermal properties and geometrical dimensions on skin burn injuries.

A one-dimensional multi-layer model is presented to characterise the skin burn process resulting from the application of a high temperature heat source to a skin surface. Transient temperatures were numerically calculated using a finite difference method to solve the Pennes bioheat equation. A damage function denoting the extent of burn injury was then calculated using the Arrhenius assumptions. The model was used to predict the effects of thermal physical properties and geometrical dimensions on the transient temperature and damage function distributions. The results show that the epidermis and dermis thicknesses significantly affect the temperature and burn injury distributions, while variations of the initial temperatures and the blood perfusion have little effect.

FTIR spectroscopic analyses of human placental membranes.

We investigated the differences in the Fourier transform infrared (FTIR) spectra of normal and abnormal human placentas. Normal placentas, placentas with infant intrauterine growth restriction (IUGR), and placentas from mothers with diabetes mellitus (DM) were used, none of which had been treated before measurement. The tissues were divided into three parts: the upper one-third portion (P1), the middle portion (P2), and the lower one-third portion (P3). Placental tissues were also investigated histochemically. The differences of the main second-derivative FTIR spectra among P1, P2, and P3 in normal placentas were observed in bands appearing between 1080 and 1090 cm(-1). Bands in P2 were observed at 1083 cm(-1), which was significantly higher than that in P3 (p < 0.05). The spectrum of P2 tissue in placentas with infant IUGR had a peak at 1081 cm(-1), which was significantly different from those of P1 and P3 (p < 0.05). In placentas with DM, the P2 band was shifted to a peak at 1088 cm(-1). These data were well correlated with the histochemical sugar-chain staining pattern of the P2 portion of the placenta. Our data suggested that this IR technique is applicable to the clinical diagnosis of diseases in the gynecological field.

[Important bio-thermal physical problems and latest advancement in laser cell engineering].

The recently emerging technique of laser microsurgery (optical tweezers, optical scissors, etc.) is providing a new precise, sterile method for the cell engineering practices such as introduction of external gene into an object cell, cell-fusion, and trapping or transportation of microscopic objects (cells or chromosomes etc.). The thermal effects thus induced usually proved to be critical factors for successful operation of this method. In order to meet the requirement for the rapid development in this territory, some important bio-thermal physical problems and the corresponding research subjects in this area were comprehensively summarized. Difficulties and critical issues were discussed. The latest advancement of the laser cell engineering was also described. This review is attempted to bridge up the gap between bioengineering and thermal science fields and then to enhance the rapid progress of laser microsurgery.

[Reconstruction of forearm function by fibular transfer with vascular anastomoses].

OBJECTIVE: To analyse and report the reconstruction of forearm function by fibular transfer with vascular anastomoses. METHODS: Fourteen cases of forearm function dismissed due to defects of bone were managed by fibular transfer with vascular anastomoses. RESULTS: All cases obtained satisfactory effects, and recovered their normal function. CONCLUSION: The vascularized fibular transfer is a good method to reconstruct the forearm function with defects of bone resulting from injury or tumor.

A strong source of methyl chloride to the atmosphere from tropical coastal land.

Methyl chloride (CH3Cl), the most abundant halocarbon in the atmosphere, has received much attention as a natural source of chlorine atoms in the stratosphere. The annual global flux of CH3Cl has been estimated to be around 3.5 Tg on the grounds that this must balance the loss through reaction with OH radicals (which gives a lifetime for atmospheric CH3Cl of 1.5 yr). The most likely main source of methyl chloride has been thought to be oceanic emission, with biomass burning the second largest source. But recent seawater measurements indicate that oceanic fluxes cannot account for more than 12% of the estimated global flux of CH3Cl, raising the question of where the remainder comes from. Here we report evidence of significant CH3Cl emission from warm coastal land, particularly from tropical islands. This conclusion is based on a global monitoring study and spot measurements, which show enhancement of atmospheric CH3Cl in the tropics, a close correlation between CH3Cl concentrations and those of biogenic compounds emitted by terrestrial plants, and OH-linked seasonality of CH3Cl concentrations in middle and high latitudes. A strong, equatorially located source of this nature would explain why the distribution of CH3Cl is uniform between the Northern and Southern hemispheres, despite their differences in ocean and land area.

Molecular cytogenetic analysis of intergeneric chromosomal translocations between wheat (Triticum aestivum L.) and Dasypyrum villosum arising from tissue culture.

Fluorescence in situ hybridization (FISH) was applied with total genomic DNA extracted from Dasypyrum villosum (L.) Candargy as a probe to characterize chromosome translocations arising from tissue culture in hybrids of Triticum aestivum x (T. durum - D. villosum, amphiploid). Chromosome translocations between wheat and D. villosum occurred in callus cells at an average frequency of 1.9%. Translocations existed not only in callus cells but also in regenerants. Three plants with translocation chromosomes were characterized among 66 regenerants of T. aestivum 'Chinese Spring' x 'TH1W' and 'NPFP' x 'TH1'. One of them proved to be a reciprocal translocation with an exchange of about one third of a wheat chromosome arm with about one half of a chromosome arm of D. villosum. The breakpoints of the other two translocations were located at, or near centromeres. The results are similar for both callus cells and regenerants and provide further evidence that translocations take place in tissue culture. Other structural chromosomal changes, for example, fragments, telocentrics, dicentromeres, and deletions, as well as numerical alterations including aneuploidy and polyploidy were recorded both in callus cells and regenerants.

[The function of T7 promoter as cis-acting elements for polymerase II in eukaryotic cell].

Using the chlorampheniol acetyltransterase gene as reporter, the function of phage T7 promoter in mammalian cells was studied by inhibition of transcription with alpha-amanitin. The experiment proved that the reporter under T7 promoter was transcribed by RNA polymerase II. Competitive electropho retic mobility shift assay (CEMSA) with TATA box, CAAT box, GC box and octamer showed that the TATA box was competitive molecular for synthetic T7 promoter. It is possible that T7 promoter is bound with TF II D transcription factor. The TATA box and octamer were inserted into Pvu II site upstream from the T7 promoter of pT7CAT. Two recombinant plasmids, pT7TATACAT and pT7OCTCAT, were constructed and transfected into CHO cells. CAT-activity test showed that T7 promoter strength was increased by octamer factor, not by TATA box. These results suggested that T7 promoter functions as cis-acting elements of RNA polymerase II transcriptional system in eucaryotic cells.

[Tissue culture induced translocation conferring powdery mildew resistance between wheat and Dasypyrum villosum and its marker-assisted selection].

Glutamate oxaloacetate transaminase (GOT) electrophoretic analyses were performed in 175 regenerants arising from immature embryos of crosses between wheat (Triticum aestivum L.) and 6D/6V substitution stocks. The GOT-V2 coding specific enzyme band was absent in two regenerants, designated 98R149 and 98R159 respectively, originated from cross of Yi 4095 and 6D/6V substitution stock c.v. RW15. Pm21 gene linked SCARs (Sequence Characterized Amplified Regions) analysis indicated that 6VS chromosome arms existed in 98R149 and 98R159. Fluorescence in situ hybridization with total genomic DNA extracted from Dasypyrum villosum (L.) as a probe confirmed the occurrence of translocation between 6V chromosome and a wheat one in the two regenerants mentioned above. 98R149 and 98R159 were immune to powdery mildew (Erysiphe graminisDC. f. sp. tritici) inoculation with mix races collected from Hebei Province. The results of the present paper added another feasible example of useful translocations via tissue culture.

[High efficient intergeneric chromosomal translocations between wheat (Triticum aestivum L.) and Dasypyrum villosum arising from tissue culture and irradiation].

Intergeneric chromosomal translocations were discernable both in callus cells and in regenerants arising from crosses between Triticum aestivum and T. durum-Dasypyrum villosum amphiploid c.v. TH1 and TH1W by means of fluorescence in situ hybridization. There were not only reciprocal translocations, but small fragment translocations. The results proved again the feasibility of creating intergeneric translocations via tissue culture. Irradiation facilitated numerical and structural chromosome changes in callus cells. The frequency of translocations was as high as 7.4 percent in irradiated callus cells. Callus age had an important impact on numerical and structural chromosome abnormalities. During a given time of culture, the frequency of unchanged cells was declined, while those cells with chromosome losses were inclined. The duration of culture had not significant effects on cells with chromosome gains. As structural chromosome changes is concerned, the duration of culture predominantly increased the frequency of cells with telocentric chromosomes. The chromosomal changes occurred at the initiation period of tissue culture. A number of cells which were doubled their chromosome numbers (2n = 84) were observed at a certain frequency in the period of tissue culture. These cells, however, disappeared in the successive culture.

[Fluorescence in situ hybridization applied to the meiotic analysis and spontaneous chromosome translocation in the pollen mother cells of hybrids of Triticum-Haynaldia].

Fluorescence in situ hybridization was applied to the meiotic analysis and chromosome pairing in the pollen mother cells of hybrids of Triticum-Haynaldia. The results indicated that most (90.5%-93.2%) of the PMCs of two Triticum-Haynaldia hybrids contained two univalents involving one Haynaldia chromosome at metaphase I, and only 1 of 320 PMCs with homologous pairing between one wheat chromosome and one Haynaldia chromosome was observed in the hybrid of Guinong 22 x common wheat Chinese Spring, the frequency was 0.3%. The frequencies of the PMCs with mis-dividing chromosome ranged from 32.7% to 37.5% at anaphase I and from 20.5% to 24.4% at anaphase II in the hybrids between Triticum-Haynaldia. The frequency of Haynaldia chromatin translocating into Triticum chromosome was about 0.7% at anaphase I in the hybrid of Guinong 22 x common wheat Chinese Spring and was 0.8%-1.7% at anaphase II in two wheat-Haynaldia hybrids. Otherwise, the chromosome morphology involving Haynaldia chromatin was observed at preleptotene I, and the frequencies of chromatin nucleus at tetrad stage and at microspore stage were investigated. The time of chromosome translocation in meiosis and the transmitting frequencies of Haynaldia chromosome in wheat background were discussed.

Thoracoscopic retrieval of metal rods after penetrating chest injury: case report.

Thoracoscopy has been used for both diagnostic and therapeutic purposes. Its use in the trauma field is rapidly increasing. Here, we present a case of chest trauma that was successfully treated using the thoracoscopic approach. A 43-year-old male patient was brought to our emergency room with a severe right chest wall-penetrating metal-rod injury, which had occurred after falling from a height. The chest X-ray showed an upper right lung lobe injury. Video-assisted thoracoscopic surgery (VATS) was performed for diagnosis of any other associated injury and for management of the penetrating injury. The prognosis was good. We believe that minimal thoracoscopic surgery is an alternative way that provides a more rapid diagnosis, and a less-invasive and safe operation for acute chest trauma patients.

[Localization of the gene for 4 hereditary multiple exostoses families].

We investigated 11 families with hereditary multiple exostoses (EXT) by linkage analysis using 8 short-tandem-repeat (CA)n polymorphic markers on chromosomes 8, 11 and 19. The Lod score in four families indicated that the gene responsible for EXT is located in the pericentromeric region of chromosome 11.

Reduced sarcolemmal dystrophin distribution and upregulation of utrophin in the cardiac and skeletal muscles of CHF-146 dystrophic hamsters.

Abnormalities in the dystrophic gene product, dystrophin, have been implicated in initiating the primary membrane defect and excessive intracellular calcium accumulation (EICA), which play fundamental pathogenic roles in hereditary muscular dystrophy (HMD). Two other cytoskeletal proteins, spectrin and utrophin, bear remarkable structural and functional homologies to dystrophin. CHF-146 strain dystrophic hamsters (DH), like patients with Duchenne muscular dystrophy (DMD), die prematurely from cardiopulmonary insufficiency, focal myonecrosis, and progressive degeneration of the cardiac and skeletal muscles with EICA. Although DH present a suitable model for HMD, there are controversies concerning their dystrophin and utrophin status. Using immunocytochemistry and Western blotting, we studied dystrophin, spectrin and utrophin anomalies in the cardiac and skeletal muscles of 6-mo-old male DH. Age- and sex-matched CHF-148 strain albino normal hamsters (NH) served as controls. Sarcolemmal dystrophin staining was much weaker and interruptive in the DH. The densitometric analysis of the immunoblots revealed that dystrophin is reduced in DH by 83% in cardiac muscle (p < 0.0001), and by 50% in skeletal muscle (p < 0.0001). We conclude that sarcolemmal dystrophin distribution is markedly reduced and discontinuous in the cardiac and skeletal muscles of DH, with simultaneous upregulation of utrophin and a varied degree of spectrin labelling. This observation suggests that reduced sarcolemmal dystrophin is associated with membrane hyperpermeability, which leads to progressive muscle degeneration via EICA and segmental necrosis in DH. As in DMD, utrophin appears to play an important compensatory role in hamster dystrophinopathy.

[Some YAC contig construction and long range physical mapping at human X chromosome Xp11.3-21.3].

Human X chromosome short arm Xp11.3-p21.3 is an area, where several genetic disease gene loci are located. In this work, the YAC conting construction, long range physical mapping were done for this region. Some DNA probes and STS markers were used for YAC screening. Totally 77 YACs were obtained from the YAC libraries of CEPH, ICRF and ours. The size determination, 26 pairs of microsatelite STS analysis, single copy probe hybridization, Alu-PCR finger printing and long range physical mapping were conducted with these YACs. These results allowed us to map these YACs, and finally 6 YAC contigs were obtained in Xp11.3-21.3, covering about 15.3 Mb. This work will greatly facilitate the positional cloning of disease genes or the genome sequencing in this important region.

Percutaneous balloon valvuloplasty for pulmonic stenosis in adolescents and adults.

BACKGROUND: Percutaneous balloon valvuloplasty has been the accepted first-line treatment for congenital pulmonic stenosis in children. Its efficacy in adolescents and adults is less well defined. METHODS: Between December 1985 and July 1995 we performed percutaneous pulmonic valvuloplasty with a single Inoue balloon catheter in 53 adolescent or adult patients 13 to 55 years of age (mean [+/- SD], 26 +/- 11). Follow-up studies were performed 0.2 to 9.8 years after the procedure (mean, 6.9 +/- 3.1) by Doppler echocardiography (in all the patients) and by cardiac catheterization and angiography (in nine patients). RESULTS: After balloon valvuloplasty, the systolic pressure gradient across the pulmonic valve decreased from 91 +/- 46 mm Hg to 38 +/- 32 mm Hg (P < 0.001), and the diameter of the pulmonic-valve orifice increased from 8.9 +/- 3.6 mm to 17.4 +/- 4.6 mm (P < 0.001). In the nine patients catheterized at follow-up, the systolic gradient decreased from 107 +/- 48 mm Hg before valvuloplasty to 50 +/- 29 mm Hg after valvuloplasty and to 30 +/- 16 mm Hg at follow-up (P < 0.001 for the comparison of the gradient before and after valvuloplasty; P < 0.001 for the comparison before valvuloplasty and at follow-up; and P < 0.05 for the comparison after valvuloplasty and at follow-up). In the same nine patients, the diameter of the pulmonic valve, as measured by right ventricular angiography, increased from 8.3 +/- 1.4 mm before valvuloplasty to 17.2 +/- 2.0 mm after valvuloplasty (P < 0.001) and to 18.4 +/- 1.4 mm at follow-up (P = 0.08). Incompetence of the pulmonic valve was noted in 7 of the 53 patients (13 percent) after balloon valvuloplasty, but it had disappeared at follow-up in all of them. CONCLUSIONS: Patients with congenital pulmonic stenosis who present in late adolescence or adult life can be treated with percutaneous balloon valvuloplasty with excellent short-term and long-term results that are similar to those in young children.
