The influence of luting cements on the fermentation of sucrose by oral microorganisms.

OBJECTIVES: Three luting cements based on zinc oxide phosphate, glass ionomer or composite resin were compared with respect to their influence on the anaerobic fermentation of sucrose by human oral bacteria. METHODS: Fifteen standardized samples of each of the three luting cements were prepared according to the manufacturer's instructions and stored for 72 h at 37 degrees C in distilled water, which was also used as elution medium. After removing the samples solutions were incubated with human fasting saliva, sucrose and a bicarbonate buffer in a Warburg apparatus. Intermediary organic acids resulting from anaerobic fermentation of sucrose were quantified indirectly by manometric measurement of CO2, released from the buffer. RESULTS: The materials did not indicate either an accelerating or an inhibiting effect on the formation of organic acids from sucrose. CONCLUSION: In the case of bacterial microleakage along the interface between cavity and luting cement, an inhibition of acid induced dentin or enamel demineralisation--using the materials investigated--cannot be expected.

Detection of binding of denatured salivary alpha-amylase to Streptococcus sanguis.

Native alpha-amylase, either solubilized, or immobilized and tested with an overlay immunotechnique, was bound in a species-specific manner to Streptococcus mitis and to one of the Streptococcus gordonii strains. However, only insignificant amounts of alpha-amylase were bound to Streptococcus sanguis and all other strains tested. When alpha-amylase was denatured before immobilization, Streptococcus sanguis bound strongly to the protein while binding of other strains was insignificant.

[Caries in the permanent teeth of 7- to 15-year-old Hamburg pupils].

The dental health status of 2193 seven- to fifteen-year old pupils was evaluated in Hamburg. A total of 20.6% of these school children hat caries-free dentitions. Among the seven-year-old children this figure was 43.4% and it dropped down to 4.8% among the fifteen-year old ones. DMFS values increased from 1.0% to 13.8% between the same groups. Obviously, caries prevalence was increasing to a statistically significant degree with the lower social classes.

[Oral streptococci affecting Ca-phosphate precipitation].

The precipitation of Ca-compounds in ageing dental plaque leads to the formation of calculus. Bacterial lipoteichoic acid (LTA) has been discussed as Ca-carrier in this process (Zoon et al., 1989). Therefore in this study, LTA was isolated from oral streptococci and tested for its influence on the precipitation of Ca-phosphate. LTA-containing bacteria, LTA-free bacteria and isolated LTA were added to the reaction mixtures for Ca-phosphate precipitation and the amount of precipitated mineral was measured. LTA-containing cells and, above all, LTA, inhibited Ca-phosphate precipitation in a distinct pH-range just above the acid solubility of the mineral by complexing Ca(2+)-ions. Experiments with Ca-fluoride in place of Ca-phosphate showed that LTA did not affect the precipitation of this mineral.

[Inter- and intrapersonal coincidence of caries epidemiologic data].

For testing the reliability of caries epidemiologic data a group of 273 children, aged 7 to 15 years, was examined by two dentists. A second group of children was investigated twice by one examiner at an interval of two weeks. Neither an inter-examiner nor an intra-examiner comparison of these data did indicate any statistically significant differences in caries incidence.

Characterization of a Streptococcus mutans serotype e plasmid pJEB110.

A search for extrachromosomal elements in 300 mutans streptococcal strains, isolated from dental plaque, yielded one 5.7 kb plasmid (pJEB110) in a serotype e strain (JEB110). The strain was distinguished from a plasmid-free (B2) and a plasmid-carrying (LM7) serotype e strain, by the presence of a prominent protein band of Mr 70 K on electrophoretograms of buffer-extracted denatured cell protein. Restriction enzyme site mapping indicated that pJEB110 was more closely related to pAM7, the plasmid endogenous to LM7, than to the other Strep. mutans plasmids harboured by serotype c or f strains.

[Preventive dentistry in the Federal Republic of Germany].

Caries rates are on the decline in the Federal Republic of Germany, too. And, in some cases considerable, increase in the number of children with caries-free teeth and a clear reduction in the average number of carious teeth has been recorded, above all in kindergartens with preventive dentistry programmes. These results have to be consolidated and expanded upon. A good basis for achieving this would be a standard federal programme in which all the institutions involved would cooperate as partners.

[The adaptive responses of human oral microorganisms to sorbitol].

It was the goal of this study to evaluate the extent to which sorbitol is metabolized by the oral microflora of persons adapted to this sugar substitute (diabetics). Resting saliva of 25 test persons with juvenile diabetes mellitus was incubated with sorbitol or sucrose in the Warburg-system both under aerobic and under anaerobic conditions. Saliva of 25 healthy persons served as control. Sorbitol was metabolized faster and to a higher total amount in the diabetic group. These results indicate that if sorbitol is offered continuously, the oral microflora will adapt and will metabolize sorbitol to a considerably higher extent.

[Electrophoretic analysis of potential receptors for oral streptococci in human saliva].

Initial adhesion of oral streptococci occurs at specific binding sites present in the dental pellicle. These receptors are salivary proteins and glycoproteins which by necessity possess affinity to dental enamel as well as to the adhering microorganism. By gel electrophoresis and immunoblotting, five such proteins were shown which bound to hydroxyapatite and oral streptococci. Information on the molecular events of initial adhesion to dental surfaces by oral microorganisms may possibly open new avenues in caries prevention.

[Leucrose metabolism in the microflora of the oral cavity].

The metabolizing activity towards leucrose in comparison to sucrose and sorbitol in human saliva and plaque-borne microorganisms was assessed using the Warburg-technique. Leucrose metabolism was slightly increased initially, but the overall rate and total amount of leucrose degraded was lower than that of sorbitol. This suggests that the cariogenic potential of leucrose, if there is any at all, is very low.

[Dried fruit as sugar substitute?].

Alternative foodstuffs restrict the usage of household sugar and instead recommend sweet honey or dried fruits; in popular informative magazines raisins and dried fruit are even described as "healthy snacks". In this study, with the help of sugar clearance and lactic acid measurements, the cariogenic potential of dried fruits is to be better estimated. The results clearly show that the alternative recommendations do not promote healthy teeth. The cariogenic potential of the named foodstuffs is comparable to sucrose containing products.
