Bronchoalveolar lavage and histologic characterization of late asthmatic response in guinea pigs.

To elucidate the mechanisms of late asthmatic response (LAR) observed in asthmatic subjects, we have developed an animal model of LAR using guinea pigs. Fifty guinea pigs were immunized with a mixture of Ascaris suum extract and aluminum hydroxide and then challenged with an inhalation of Ascaris suum extract without anesthesia. Twenty of the 50 guinea pigs showed a dual asthmatic response in which the LAR occurred 3 to 6 h after immediate asthmatic response (IAR). Histologic studies by rapid freezing with liquid nitrogen or bronchoalveolar lavage (BAL) were performed in 14 of these 20 guinea pigs with LAR and compared with those in 10 of 18 guinea pigs with only IAR, 10 control guinea pigs, and 10 nonimmunized but challenged guinea pigs. Both the percentage and the absolute number of neutrophils in the BAL fluid of the guinea pigs with LAR were significantly greater than those of the control guinea pigs (p less than 0.02) and than those of the nonimmunized but challenged guinea pigs (p less than 0.02). However, that of guinea pigs with LAR was not significantly different from that of guinea pigs with only IAR. On the other hand, histologic examination showed that eosinophil infiltration within the airway walls of the guinea pigs with LAR was more prominent than that of the guinea pigs with only IAR, and showed that there was no significant difference in neutrophil infiltration within the airway walls between the guinea pigs with LAR and the animals with only IAR. Contraction of airway (bronchus, bronchiole) smooth muscle, submucosal edema, and mucus in airway lumen were also observed in LAR.(ABSTRACT TRUNCATED AT 250 WORDS)

[Mode of the anti-allergic action of cepharanthine on an experimental model of allergic rhinitis].

Cepharanthine, a biscouclaurine alkaloid of Stephania cepharantha, has been used for various clinical purposes. Cepharanthine was known to inhibit histamine release from mast cells obtained from sensitized animals. In vitro studies suggested that the mechanism of action of cepharanthine may be ascribed to the membrane stabilizing action. The membrane stabilization may be attained by reducing the elasticity of the membrane. However, in vivo mechanisms of the anti-allergic action of cepharanthine have not been examined. In the present in vivo study, the anti-allergic action of cepharanthine was examined using experimental allergic rhinitis in rats. The locally administered cepharanthine solution (0.1 mg/ml), by perfusing the nasal cavity, inhibited the dye leakage and an increase in lysosomal enzyme activity due to antigen stimulation. This is the reason for the membrane stabilization by cepharanthine. In the metyrapone (20 mg/kg, s.c., 5 days) pretreated rats of the rhinitis models, the anti-allergic action of cepharanthine was weaker. On the other hand, the effect of ketotifen was not altered by such an effect. The experimental results suggest that the anti-allergic mechanism of cepharanthine might be exerted by its membrane stabilizing action and by stimulation of the pituitary-adrenotropic function.

[Reduction of radiation dose to the worker in preparing the radiopharmaceutical solution by a simple shielding equipment].

In order to reduce radiation dose to the hands of examiners who prepare and aspirate radiopharmaceuticals, we made a prototype of simplified manually-operated dispense system, which the syringe and the vial shield with lead were set in the small box made of lead and lead glass. The result showed that our dispense system allowed substantial reduction of radiation dose to the hands and rapid preparation of radiopharmaceuticals compared with the conventional lead shield syringe system, and allowed closer operation, smaller dead volume and lower cost compared with the conventional automatic system.

[Antitumor effect of PSK (1). Interferon-inducing activity and intratumoral administration].

After oral administration of 20 mg/kg of PSK, a significant level of interferon (IFN) activity was detected in the sera of BCG (1 mg/mouse)-sensitized mice. A low titer of IFN was also detected in the sera of all of 4 cancer patients after 3 g/day oral administration of PSK for 7 consecutive days. Intratumoral administration of PSK (5 mg/mouse) strongly inhibited the growth of Meth-A solid tumors in male BALB/c mice and led to complete regression of tumors and resistance to reinoculated tumors in the host animals. Subsequently, the antimetastatic effect of PSK was examined in a "double grafted tumor system" in which mice first received simultaneous intradermal inoculations of Meth-A in the right (10(6) cells) and left (2 X 10(5) cells) flanks and were then injected with PSK in the right tumor on day 3. PSK significantly inhibited the growth of the left, non-treated tumor. When mice received an inoculation of Meth-A (2 X 10(5) cells) in the left flank and PSK was injected subcutaneously in the right flank on day 3 (single tumor system), the growth of the tumor was not inhibited. These findings suggest that intratumoral PSK immunotherapy in one region has an effect on tumor growth in another region.

Sequence-dependent hydrolysis of RNA using modified oligonucleotide splints and RNase H.

To cleave RNA molecules using RNase H in a site-specific manner, a short deoxyoligonucleotide (3-5mer) joining with 2'-O-methyl oligonucleotide(s) was designed as a DNA splint to be used. Model experiments were carried out using ribooligonucleotide substrates (9 and 18 mer). It was found that the use of this type of splints (9 mer) causes a unique cleavage by RNase H. For example, when 3'm (GA)d(AGAA)m(GGU)5' was used as a hybridization strand, 32pUCUUUCUUCUUCCAGGAU was cleaved specifically between U11 and C12 to yield 32pUCUUUCUUCUU. This method will have a variety of applications for the study of RNA.

[Endocrinological study of Sertoli-Leydig cell tumor].

A 32-year-old woman, 3, para 2 with a testosterone producing tumor of the left ovary was studied endocrinologically. She complained of amenorrhea, hirsutism and abdominal mass. The peripheral testosterone level was 8.4 ng/ml, remarkably elevated. After the combined dexamethasone suppression/hCG stimulation test, the plasma testosterone level rose from 9 ng/ml to 15.1 ng/ml. Prior to removal of the tumor, venous samples were drawn directly from the right and left ovarian veins during surgery and simple total hysterectomy with bilateral salpingo-oophorectomy was performed. Testosterone levels were 40.5 ng/ml on the tumor side and 7.1 ng/ml on the normal side. After tumor removal, the plasma testosterone level fell from 8.4 ng/ml to less than 1.0 ng/ml within 24 hours. Histological examination of the left ovarian tumor revealed a Sertoli-Leydig cell tumor. In incubation of small specimens of tumor tissues in oxygenated Krebs bicarbonate buffer, the release of testosterone into the medium containing hCG was twice as high as that into the medium without hCG. These results of in vitro and in vivo studies suggest that this tumor was an hCG-dependent testosterone producing Sertoli-Leydig cell tumor.

Relation of ABH blood group antigen expression to prognosis in lung carcinoma.

The ABH blood group antigens were studied in resected specimens from patients with lung cancer employing monoclonal antibodies against A, B and H antigens by the immunoperoxidase technique. There were no differences between paraffin-processed and fresh-frozen materials in detectability of ABH antigens. Comparing specimens at operation with those at autopsy from the same patient, the loss of A and B antigens in the latter was demonstrated more frequently than H antigen. Concerning the relation between the prognoses and the expression of ABH antigens, the group with good prognosis preserved more ABH antigens than that with poor prognosis. In contrast, the group with poor prognosis showed less ABH antigens. There were also statistical differences in antigen detectability and prognosis, particularly in stage 1 patients, adenocarcinomas and well-differentiated lung cancers.

The active center of transport for phosphoenolpyruvate and inorganic phosphate in the human erythrocyte membrane.

Phosphoenolpyruvate is transported into erythrocytes by a carrier-mediated transport system. The transport is competitively inhibited by inorganic phosphate and pyridoxal phosphate, which are transported across the cell membrane by Band 3. The pH profiles for the transport of inorganic phosphate and phosphoenolpyruvate suggest that a histidine residue exposed at the inner surface of the membrane participates in both of the transport. A hydrophobic peptide which may be a part of the active center for the transport is purified from the CNBr fragments of Band 3.
