Effect of clenbuterol on peripheral airway obstruction in bronchial asthma.

A study was carried out in 6 patients with bronchial asthma to investigate the effects of clenbuterol, a beta 2-sympathomimetic bronchodilator, on peripheral airway obstruction. The basal lung functions of the patients were almost within normal range in both vital capacity (VC) and forced expiratory volume in 1 second (FEV1), but their maximal flow rates were lower in effort-independent phase of both maximal expiratory flow volume (MEFV) curve and partial expiratory flow volume (PEFV) curve. Furthermore, they demonstrated marked basal frequency dependence of dynamic compliance [CL,dyn]. Oral administration of clenbuterol (40 micrograms) produced a significant increase in the maximal flow in effort-independent phase of both MEFV and PEFV curves, and markedly decreased frequency dependence of CL,dyn in comparison with the baseline values, while it improved both VC and FEV1 to a lesser extent. These results suggest that clenbuterol preferentially reduced the peripheral airway obstruction in bronchial asthma.

[Changes of lysosomal enzyme activities in experimental type I and type IV allergic models].

Experimental allergy was induced in animals: asthma, a Type I allergy, was induced in guinea pigs by sensitizing them with alpha-amylase (inhalation), and experimental allergic encephalomyelitis (EAE), a Type IV allergy, was induced in rats. Pulmonary, brain and serum lysosomal enzyme activities were measured in normal and allergic conditions. beta-Glucuronidase (beta-G) and arylsulfatase (AS) activities were determined by the fluorescent technique. During the asthmatic attack, pulmonary lysosomal enzyme activities were not different from that in the normal state in guinea pigs. However, brain lysosomal enzyme activities were elevated markedly on the 1 st day of EAE induction. Brain beta-G activity was elevated on the 2nd day, and AS activity had a tendency to be increased. On the other hand, serum lysosomal enzyme activity was not altered significantly. In the experimental allergy, lysosomal enzyme activity was altered in Type IV, but not in Type I.

Late pulmonary response in guinea pigs after ascaris challenge.

Respiratory resistance of guinea pigs immunized with Ascaris suum was measured over 6 hr after aerosol challenge of the allergen by use of the oscillation method. These animals showed a biphasic pulmonary response to the aerosol challenge with Ascaris suum. Histological analysis of these animals revealed a narrowing of bronchial lumen, peribronchial edema and neutrophil-rich mucous in bronchial lumen at the late pulmonary response.

Antitumor activity of the DNA fraction from Mycobacterium bovis BCG. II. Effects on various syngeneic mouse tumors.

MY-1, a fraction extracted from BCG and composed of 70.0% DNA and 28.0% RNA, was examined for its antitumor activity against 9 different syngeneic mouse tumors. Tumor regression was induced in almost all of the mice bearing any of five kinds of solid tumors by repeated intralesional injections of 100 micrograms MY-1. When cells of some tumors were inoculated intradermally together with MY-1, tumor growth was suppressed, lung metastases were inhibited, and the survival times of mice bearing 1 of 3 leukemic tumors were prolonged. Repeated sc injections with MY-1 in sites remote from tumor cell inoculation or repeated iv injections were more or less effective against three kinds of solid tumors. Mice inoculated with Lewis lung carcinoma cells in a hind footpad and whose legs were amputated 9 days later were given iv or sc injections of MY-1 every other day (8 times in total), resulting in substantial prolongation of survival. No direct cytotoxicity of MY-1 for these tumors could be shown in three kinds of experiments, which indicates that the antitumor mechanism of MY-1 is host mediated. MY-1 was equally effective in mice with or without presensitization with BCG, whereas BCG was much more effective in BCG-sensitized mice. This finding suggests that a delayed-type hypersensitivity reaction elicited by BCG protein is not required for the antitumor activity of MY-1.

Locking of the metacarpophalangeal joint of the thumb.

We clinically examined the hands of twenty-three patients and experimented with amputation specimens to clarify the characteristic features and pathomechanism of locking of the metacarpophalangeal joint of the thumb. In all of the patients the locking had resulted from forced hyperextension of the thumb. Every injured thumb showed a mild hyperextension deformity of the metacarpophalangeal joint, with the joint having become fixed. Manual reduction was successful in seven patients, but the other sixteen required open reduction, during which we found that the proximal palmar ligament was ruptured transversely with its distal part riding over the volar prominence of the radial condyle of the metacarpal together with the accessory collateral ligament. These dislocated parts of the ligament formed a constricting bundle on the distal joint surface of the condyle that prevented closed reduction. The locking was released by cutting the constricted ligament bundle. Our description of this mechanism is supported by the experimental evidence that we obtained from examining the amputation specimens.

[An electrophysiological study on the mechanoreceptor of canine knee joints].

Passive extending and flexing and change in intraarticular pressure were applied to canine knee joints and nerve impulses from the posterior articular nerve, an articular branch of the tibial nerve, were studied. On extending and flexing the knee, in the intermediate position the nerve impulses were of low frequency but on excessive extension high frequency nerve impulses were observed. The relationship between the angle of the joint and the frequency of nerve impulses exhibited a hysteresis phenomenon. On changing the intraarticular pressure, the frequency of nerve impulses increased with an increases in pressure and it depended directly on the intraarticular pressure. The mechanoreceptors of joints probably play a warning function for the protection of the joint by effectively informing the extreme conditions and non-physiological conditions to the central nervous system.
