Photopic electroretinogram B-wave implicit time in monkey eye.

Photopic electroretinograms were elicited in anesthetized monkey eyes with stimulus light of various intensities under background light of different luminances. The b-wave implicit time was prolonged with increasing stimulus light intensity and shortened with brighter background luminance. While the change of anesthetic conditions produced by the addition of halothane (0.5% or 1.0%) mildly prolonged the implicit time, it affected neither the waveforms nor the relationship between the b-wave implicit time and background luminance.

Fundus flavimaculatus with severely reduced cone electroretinogram.

An 11-year-old girl complaining of progressive visual loss in both eyes showed atrophic macular degeneration with yellowish white flecks around the fovea in both eyegrounds. Decreased vision, central scotoma with normal peripheral visual field, and angiographic dark choroid except for transmitted hyperfluorescence in the macula and at the flecks confirmed the diagnosis of fundus flavimaculatus with atrophic macula. Electroretinograms, however, revealed severely reduced cone responses associated with almost normal rod response, which led to the diagnosis of cone dystrophy. The rare combination of angiographic evidence of fundus flavimaculatus and electroretinographically diagnosed cone dystrophy was discussed.

[The effect of stimulus intensity and background luminance on the b-wave implicit time of photopic electroretinogram in normal human eyes].

The authors studied the effect of stimulus intensity and background luminance on the b-wave implicit time of photopic electroretinogram in normal human eyes. Photopic electroretinograms were recorded at various stimulus intensities and background luminances in the range of 2.5 log scale using an integrating sphere of 30 cm diameter in the first experiment. The results from two normal eyes showed that with increasing stimulus intensity or with decreasing background luminance the b-wave implicit time of photopic electroretinogram prolonged and one or three wavelets superimposed on the ascending limb of the b-wave. The second experiment was performed using a half dome sphere of 60 cm diameter to study the b-wave implicit time at three different stimulus intensities. The results from 30 normal eyes disclosed a statistically significantly longer implicit time at the greater stimulus intensities. These results suggest that b-wave implicit time in photopic electroretinogram becomes delayed with increasing stimulus intensity and becomes shorter with brighter background luminance.

[Static perimetry and temporal aspects of the photopic electroretinogram in sector retinitis pigmentosa].

Six cases of sector retinitis pigmentosa were studied by automated static perimetry and photopic electroretinogram. Four cases out of six revealed elevated thresholds in the lower half of the visual field, which corresponded to the normally appearing upper eyeground. They also showed delayed implicit time on the photopic electroretinogram. These results show that sector retinitis pigmentosa is not necessarily a regional retinal dystrophy.

Human substance P receptor (NK-1): organization of the gene, chromosome localization, and functional expression of cDNA clones.

The gene for the human substance P receptor (NK-1) was cloned using cDNA probes made by the polymerase chain reaction from primers based on the rat sequence. The gene spans 45-60 kb and is contained in five exons, with introns interrupting at sites homologous to those in the NK-2 receptor gene. Analysis of restriction digests of genomic DNA from mouse/human cell hybrids indicates the NK-1 receptor is a single-copy gene located on human chromosome 2. Polymerase chain reaction using primers based on the 5' and 3' ends of the coding sequence was used to generate full-length cDNAs from human lung and from IM9 lymphoblast cells. When transfected into COS-7 cells, the NK-1 receptor binds 125I-BHSP with a Kd of 0.35 +/- 0.07 nM and mediates substance P induced phosphatidylinositol metabolism. The receptor is selective for substance P; the relative affinity for neurokinin A and neurokinin B is 100- and 500-fold lower, respectively. Human IM9 lymphoblast cells express relatively high levels of the NK-1 receptor, and Northern blot analysis indicates modulation of mRNA levels by glucocorticoids and growth factors, suggesting that this cell line may be useful as a model for studying the control of NK-1 receptor gene expression.

Stereoselective total synthesis of glycopeptides bearing the dimeric and trimeric sialosyl-Tn epitope.

The dimeric and trimeric sialosyl-Tn epitopes, [alpha-D-Neup5Ac-(2----6)-alpha-D-GalpNAc-(1----3)-L-Ser]n-L-Val (n = 2 and 3), which represent part of a clustered carbohydrate region of glycophorin A, a human erythrocyte glycoprotein, have been synthesised stereoselectively. 2-Azido-3-O-benzyl-4,6-O-benzylidene-2-deoxy-D-galactopyranosyl fluoride (GalpNAc unit), Fmoc-L-serine phenacyl ester (Ser unit), and benzyl 5-acetamido-4,7,8,9-tetra-O-benzyl-5-deoxy-3-S-phenyl-3-thio-D-erythro-L - gluco-2-nonulopyranosylonate bromide (Neup5Ac unit) were the key intermediates for stereoselective glycosylation. 2-Ethoxy-1-ethoxycarbonyl-1,2-dihydroquinoline-promoted elongation of the peptide chain and then hydrogenolysis afforded the title compounds.

En bloc staining available for stereoscopic observation of epoxy resin Quetol 651-embedded thick sections under a high voltage transmission electron microscope.

This method has been devised for easy en block staining for stereoscopic observation of thick sections under a high voltage transmission electron microscope (HVTEM). It uses carbohydrazide as an osmium bridging agent and both osmium tetroxide and uranyl acetate as electron staining agents. Osmium tetroxide-fixed and en bloc-stained tissue blocks are embedded in a Quetol 651 resin mixture. Thick sections (2-3 microns thick) without double staining are observed at an accelerating potential of 300 kV and a tilt angle of +/- 10 degrees by an H-9000 TEM with a side-entry goniometer. Stereoscopic electron micrographs can be obtained.

[Recovery of visual field defects in ischemic optic neuropathy and idiopathic optic neuritis].

Fifty-four eyes of 41 patients with optic nerve disease demonstrating acute visual field defects without any traumatic, compressive, or other known etiology were classified into four categories. Those showing poor recovery of visual field defects were ischemic optic neuropathy which was subclassified into either anterior ischemic optic neuropathy (AION) or posterior ischemic optic neuropathy (PION) according to the ophthalmoscopic changes in the optic nerve head. Those showing good recovery of visual field defects were idiopathic optic neuritis which was subclassified into either papillitis or retrobulbar neuritis according to the ophthalmoscopic pathology of the optic disc. Patients with ischemic optic neuropathy were significantly older than those with optic neuritis. All eyes with optic neuritis showed good recovery of vision, whereas those with ischemic optic neuropathy showed varying outcomes of vision. With regard to the pattern of field defect, central or paracentral scotoma was predominant in all but eyes with AION in which altitude defect predominated. Pale swelling of the optic nerve head and angiographic evidence of circulatory disturbance in the optic disc or adjacent choroid were common findings in eyes with AION, whereas such findings were never observed in eyes with papillitis. The amplitude of pattern visual evoked potential was significantly lower in eyes with PION than in those with retrobulbar optic neuritis. Four patients classified as optic neuritis developed into multiple sclerosis in the follow-up study. It was concluded that poor recovery of visual field defect is one of the most convincing evidences for the diagnosis of ischemic optic neuropathy.

Electroretinogram in cone dystrophy.

Electroretinograms (ERGs) in 10 cases of cone dystrophy were studied with special respect to log(bp/bs) that represents the log of photopic ERG amplitude divided by scotopic ERG amplitude. Photopic ERGs were either greatly diminished or nonrecordable, and scotopic ERGs showed either normal or reduced amplitudes. All cases had a low value of log(bp/bs), less than the lower normal limit, indicating significantly greater impairment in cone function than in rod function. These results have proved log(bp/bs) to be helpful in confirming the diagnosis of cone dystrophy especially in cases with recordable photopic ERG and reduced scotopic ERG. Among other ERG parameters, the photopic ERG b-wave implicit time that was determined with averaging and digital amplification of the responses in 4 cases showed prolongation in 3 cases while normal in one case. Oscillatory potentials were nonrecordable or barely recordable in all cases.

[Retinal arterial obstruction and systemic disorders].

The records of 36 patients (37 eyes) with central retinal artery occlusion (CRAO, 19 patients, 19 eyes), branch retinal artery occlusion (BRAO, 15 patients, 16 eyes), and cilioretinal artery occlusion (CIAO, 2 patients, 2 eyes) were reviewed with respect to underlying systemic disorders. Hypertension was the most common disorder (58%). Half of the patients had cardiovascular disorders including ischemic heart disease, valvular disease, atrial fibrillation, internal carotid artery obstruction, and atrial myxoma. Seventeen percent of the patients had diabetes mellitus, and 8% had hyperlipemia. Cerebral infarction was detected in a quarter of the patients. The incidence of the disease was one CRAO patient per 1,000 outpatients. Age distribution showed a peak in the seventh decade. Men were twice more frequently affected than women. Right eyes were more commonly involved.

Recovery of an epitope recognized by a novel monoclonal antibody from airway lavage during experimental induction of chronic bronchitis.

Prolonged exposure of dogs to high concentrations of SO2 gas results in a syndrome with many of the characteristics of human chronic bronchitis, including cough and chronic mucous hypersecretion as well as airway obstruction. We developed and used a novel monoclonal antibody, GB-4B, raised against epithelial glycoprotein isolated from human hypersecretory mucus to probe airway lavage samples from dogs before and during prolonged exposure to SO2 gas. There were relatively low mean titers of the epitope recognized by GB-4B in airway lavage fluid as evidenced by enzyme-linked immunosorbent assay before exposure to SO2 gas. After 25 to 50 wk of SO2 exposure, the dogs showed a significant increase in pulmonary resistance and there was a significant increase in the titer of the epitope in the airway lavage fluid. Using the same antibody immunohistochemical analysis of airway tissues from SO2-exposed dogs revealed patchy staining of the mucous glands and airway secretory cells and dense staining along the airway surface; airway tissue from control dogs and one SO2-exposed dog whose lavage fluid did not contain the epitope showed little or no staining. These data demonstrate that similar mucin epitopes appear in airway lavage fluid under hypersecretory conditions in both animals and humans. The epitope may have utility as a marker of chronic mucous hypersecretion.

Competitive action of a biological response modifier, PSK, on a humoral immunosuppressive factor produced in tumor-bearing hosts.

We investigated the effect of PSK, a protein-bound polysaccharide obtained from the basidiomycetes Coriolus versicolor, on an immunosuppressive factor produced in tumor-bearing animals. Oral administration of PSK suppressed the growth of the tumor in C3H/He mice bearing X5563 plasmacytoma or MH134 hepatoma, but affected mice bearing MM102 mammary tumor little. PSK prevented the reduction in splenic lymphocyte blastogenesis caused by phytohemagglutinin that occurs in mice bearing X5563 tumors or MH134 hepatoma. The lymphocyte blastogenesis affected little by tumor or PSK in mice bearing MM102 tumors. The effect of sera on the blastogenesis of lymphocytes caused by phytohemagglutinin was different with different tumors in the C3H/He mice. Serum of mice bearing X5563 tumors inhibited blastogenesis, but serum of mice bearing MH134 hepatoma or MM102 tumors promoted it. The sera of mice bearing MH134 hepatoma contained both inhibitory and promotive factors; those of mice bearing X5563 tumors contained an inhibitory factor, and those of mice bearing MM102 tumors contained a promotive factor. The oral administration of PSK reduced the inhibition caused by the sera of mice bearing X5563 tumors. The promotive activity of sera from mice bearing MH134 hepatoma was augmented by PSK; that of sera in mice bearing MM102 tumors was not affected by PSK. Living Bacillus Calmette-Guérin did not have such effects in any of these mice. Serum immunosuppressive activity was also reduced by PSK in various tumor lines of rodents. These results suggest that PSK acts by reducing the activity of immunosuppressive factors produced in tumor-bearing hosts.

[Coats' disease and familial retinal arteriolar tortuosity].

The first Japanese autosomal dominant pedigree of familial retinal arteriolar tortuosity was presented with photographical documentation. The proband, a 7-year-old girl also had typical Coats' disease in her left eye. The fluorescein fundus angiogram revealed retinal telangiectasis and microaneurysms with dye leakage in her left peripheral eyeground and marked arteriolar tortuosity without dye leakage in posterior portions of both eyegrounds. Her father showed prominent retinal arteriolar tortuosity bilaterally, and her sister and paternal grandmother showed mild tortuosity of retinal arterioles in both eyes. The recently proposed hypothesis that the familial retinal arteriolar tortuosity is caused by the retinal microcirculatory disturbance consisting of increased resistance in the retinal capillary bed might explain the coincidental occurrence of retinal arteriolar tortuosity and Coat's disease in this report.

[Adaptational changes in cone electroretinograms in man].

Changes of amplitude and implicit time of human cone electroretinogram (ERG) were studied during dark adaptation and succeeding light adaptation. Dark-adapted cone ERG was isolated by subtracting scotopic blue response from matched scotopic red response. The former represented the rod-mediated b-wave, while the latter consisted of both rod-mediated b-wave and cone-mediated b-wave or x-wave. The b-wave amplitude of dark-adapted cone ERG remained unchanged during dark adaptation, while the implicit time increased systematically, reaching a plateau. Light-adapted cone ERG was obtained by red stimulus lights under a bright background light. The amplitude of light-adapted cone ERG was markedly suppressed through dark adaptation but it recovered gradually during light adaptation, reaching the base line level. The implicit time was unchanged during light adaptation.

Antibacterial effects of Listerine on oral bacteria.

To evaluate the efficacy of Listerine, a solution for washing the oral cavity consisting of essential oils (thymol, methanol, eukalyptol) and methyl salicylate, minimum concentrations inhibiting the growth of various microorganisms in the oral cavity and the bactericidal effects on bacteria in the saliva and dental plaque were evaluated in vitro. Listerine inhibited the growth of microorganisms over a very broad range. The minimum concentration inhibiting growth of Listerine was a 4 to 32 fold dilution, 2-4 times as potent as the solution after elimination of active ingredient components, in 38 of 54 bacterial strains, indicating the efficacy of the active ingredient in the inhibition of the growth of microorganisms. Bactericidal action of Listerine against from bacteria isolated from saliva and dental plaque from 5 healthy normal subjects was tested. Listerine exhibited a potent bactericidal effect on bacteria in saliva and dental plaque. Most of the bacteria died after a 30 second exposure to Listerine. According to these results, Listerine appears to be effective as a solution used for cleansing the oral cavity and dentures.
