Human apolipoprotein E receptor 2. A novel lipoprotein receptor of the low density lipoprotein receptor family predominantly expressed in brain.

Isolation and characterization of a human cDNA demonstrated a novel lipoprotein receptor designated apolipoprotein E receptor 2 (apoER2). The new receptor consists of five functional domains resembling the low density lipoprotein (LDL) and very low density lipoprotein (VLDL) receptors. LDL receptor deficient Chinese hamster ovary cells expressing human apoER2 bound apoE rich beta-migrating VLDL with high affinity and internalized. LDL was bound with much lower affinity to these cells. The 4.5- and 8.5-kb mRNAs for the receptor were most highly expressed in human brain and placenta. In rabbit tissues, multiple species of the mRNA with 4, 4.5, 5.5, 8.5, and 11 kb were detected most intensely in brain and testis and, to a much lesser extent, in ovary, but were undetectable in other tissues. In rat adrenal pheochromocytoma PC12 cells, the receptor mRNA was induced by treatment of the cells with nerve growth factor. The receptor transcripts were detectable most intensely in the cerebellar cortex, choroid plexus, ependyma, hippocampus, olfactory bulb and, to a much lesser extent, in the cerebral cortex as revealed by in situ hybridization histochemistry. In the cerebellar cortex, the receptor transcripts were densely deposited in Purkinje cell somata.

Nitrogen dioxide exposure increases airway contractile response to histamine by decreasing histamine N-methyltransferase activity in guinea pigs.

To determine the mechanism responsible for nitrogen dioxide (NO2)-induced airway hyperresponsiveness, we examined the effects of NO2 exposure on the contractile response to histamine and the level of histamine N-methyltransferase (HMT) activity, a histamine-degrading enzyme, in guinea pig trachea in vitro. Guinea pigs were divided into seven groups. Each group received continuous NO2 exposure (2.0 ppm) for either 2, 6, 12, 24, 48, or 96 h. The remaining group did not receive NO2 exposure (control). HMT activity in trachea was decreased from the control value of 70.3 +/- 7.7 pmol/min/mg protein to 34.6 +/- 6.7 pmol/min/mg protein by 12 h exposures of NO2. However, 24 and 48 h exposures of NO2 did not significantly alter HMT activity. In contrast, HMT activity exceeded the control value by 96 h exposures of NO2 (85.5 +/- 5.1 pmol/min/mg protein). Twelve hour exposures of NO2 shifted the concentration-response curves to histamine to lower concentrations and significantly reduced the median effective concentration (EC50) of histamine (log M) from the control value of -5.16 +/- 0.09 to -6.15 +/- 0.14 (P < 0.01). In contrast, the EC50 concentration of histamine (log M) increased from the control value of -5.20 +/- 0.10 to -4.90 +/- 0.11 by 96 h exposures of NO2 (P < 0.05). However, NO2 exposure did not alter the contractile response to acetylcholine. Morphologically, tracheal epithelial cells had vacuoles after 12 h exposures of NO2, but denudation of the epithelium did not occur during this experiment. In situ hybridization for HMT mRNA demonstrated that the level of HMT mRNA increased dominantly in tracheal epithelial cells after 96 h exposures of NO2. The present results indicated that the decrease in the level of HMT activity in the trachea was closely associated with the increase in the airway contractile response to histamine, suggesting that NO2-induced transient airway hyperresponsiveness to histamine is due to the decreased capacity of histamine catabolism in airway.

4,5-dichlorophthaloyl group for amino protection in carbohydrate chemistry.

Phthaloyl (Phth) is a valuable amino-protecting group for use in synthetic carbohydrate chemistry. Its strong 1,2-trans-directing nature in a glycosylation reaction, when it is introduced to the C-2 position of a glycosyl donor, makes the construction of beta-GlcNAc or beta-GalNAc glycosides quite straightforward. The Phth group can be removed by using an appropriate nucleophile, most typically hydrazine; however, this transformation often requires a long reaction time at elevated temperature when applied to a large oligosaccharide. We studied the 4,5-dichlorophthaloyl (DCPhth) group as an alternative to Phth. A thioglycoside carrying a DCPhth group at the C-2 position was reacted with alcohol by the action of PhSeNPhth-TMSOTf to selectively give the corresponding beta-glycoside. DCPhth could then be removed under mild conditions by using ethylenediamine or hydrazine/MeOH at room temperature.

Antimetastatic effect of PSK, a protein-bound polysaccharide, against the B16-BL6 mouse melanoma.

We examined the effect of PSK, a protein-bound polysaccharide, upon in vivo metastasis and in vitro invasion of the B16-BL6 mouse melanoma cells. (1) PSK suppressed in vivo artificial and spontaneous lung metastases of B16-BL6 in C57BL/6 mice. (2) PSK in a dose-dependent fashion suppressed in vitro invasion and chemotaxis of the tumor cells using filters coated with a reconstituted basement membrane. (3) PSK had little effect on DNA synthesis in tumor cells in vitro, but suppressed tumor cell adhesion to, degradation of, and haptotaxis to components of the basement membrane. (4) PSK suppressed the binding of tumor cells to components of the basement membrane. These findings suggest that PSK may suppress metastasis through inhibition of tumor cell invasion and that this effect is the result of interactions between PSK and components of the basement membrane.

The involvement of transforming growth factor beta in the impaired antitumor T-cell response at the gut-associated lymphoid tissue (GALT).

We studied the antitumor immune response in gut-associated lymphoid tissue (GALT), which is the tolerance-inducing site for numerous dietary antigens. The mice inoculated with colon 26 carcinoma (C-26) into the subserosal space of the cecum (i.c.) showed a more rapid tumor growth than did the mice inoculated s.c. with C-26 into the flank. In addition, the serum of the i.c. C-26-inoculated mice showed a more potent suppressive activity, and their plasma contained a higher level of transforming growth factor than the s.c. C-26-inoculated mice. We also evaluated the tumor-specific T-cell response in the GALT by utilizing B7-transfected P815 mastocytoma (B7/P815). The rejection of i.c. inoculated B7/P815 was delayed compared to that of the s.c. inoculated B7/P815. The draining axillary lymph node (LN) cells of the s.c. B7/P815-inoculated mice exhibited a CD4+ T-cell-dependent proliferative response to in vitro restimulation, whereas the draining mesenteric LN cells of the i.c. B7/P815-inoculated mice exhibited no apparent response even with the addition of interleukin 2. However, such draining mesenteric LN cells did produce higher levels of interleukin 2 and transforming growth factor beta than the draining axillary LN without any stimulation, and their production of such cytokines depend on the CD4+ and CD8+ cells, respectively. Collectively, our results suggest the possibility that the impaired antitumor T-cell response in the GALT may be attributed to "bystander suppression" by TGF-beta-producing CD8+ T cells.

Potential role of interleukin-1 in allergen-induced late asthmatic reactions in guinea pigs: suppressive effect of interleukin-1 receptor antagonist on late asthmatic reaction.

Interleukin (IL)-1 is a pluripotential proinflammatory cytokine and is thought to be involved in the pathogenesis of bronchial asthma and late asthmatic reactions (LARs). To determine whether IL-1 plays a role in LAR, guinea pigs sensitized with Ascaris antigen were used. We evaluated IL-1 production by immunostaining with anti-IL-1 beta antibody and elucidated the action of IL-1 in LAR with recombinant IL-1 receptor antagonist. Immunostaining revealed that IL-1 beta-like immunoreactivity-positive cells increased in the airway walls and in bronchoalveolar lavage fluid after the antigen challenge. IL-1 receptor antagonist protein pretreatment reduced the generation of LAR in terms of pulmonary resistance. IL-1 receptor antagonist protein pretreatment did not change cellular components but reduced the percentage of hypodense eosinophils in bronchoalveolar lavage fluid. We also studied the direct effect of recombinant human IL-1 beta on pulmonary resistance and eosinophil activity measured as released eosinophil peroxidase activity. Recombinant human IL-1 beta did not change pulmonary resistance but primed eosinophils to release eosinophil peroxidase activity in response to platelet activating factor. Therefore these results suggest that IL-1 was produced in sensitized pulmonary tissue of guinea pigs by allergen exposure and played a role in the generation of LAR, at least partially by modulating the activation of eosinophils.

Relation between markers for viral hepatitis and clinical features of Japanese patients with hepatocellular carcinoma: possible role of alcohol in promoting carcinogenesis.

To assess the relationship between hepatitis virus markers and the clinical features of hepatocellular carcinoma (HCC), we measured markers for hepatitis B virus (HBV) and hepatitis C virus (HCV) in 88 Japanese patients with HCC. Twelve (14%) patients were HBsAg-positive and 67 (76%) were anti-HCV-positive (both c100-3 and c11/c7). HCV-RNA was detected in 8 (38%) of the 21 anti-HCV-negative patients by PCR, so that 75 patients (85%) were infected with HCV. Of the HBsAg-negative patients infected with HCV with no history of blood transfusion, the mean age of the alcoholics (consumption > 80 g ethanol daily for at least 10 years) was lower than that of the non-alcoholics (60 years vs. 65 years, P < 0.05). Among the HBsAg-negative and anti-HCV (or HCV-RNA)-positive patients with a history of blood transfusion, the mean interval between the time of blood transfusion and the diagnosis of HCC in the alcoholics was shorter (21 years) than that in the nonalcoholics (27 years), but the difference was not statistically significant. We conclude that infection by both HCV and HBV may play a role in the development of HCC, and that alcohol consumption may promote carcinogenesis.

Effects of panretinal photocoagulation on photopic ERG in normal rabbit eyes.

The effects of panretinal photocoagulation (PRP) on photopic ERG were studied in normal pigmented rabbit eyes. Photopic ERGs were recorded before PRP and one day, one week and 4 weeks after PRP. Markedly reduced amplitude and delayed implicit time were observed one day after PRP. Four weeks after PRP, amplitude showed subtotal recovery while implicit time, showing tendency of reduction, was still significantly delayed. The delay in implicit time and the reduction in amplitude after PRP was probably due to serous retinal detachment and/or retinal edema caused by breakdown of the blood-retinal barrier. However, the findings at 4 weeks after PRP might be explained by the reduced number of photoreceptor cells; the mild decrease in the amplitude of photopic ERG might be caused by the reduced number of cones, while significantly delayed implicit time of the photopic ERG might be caused by reduced number of rods.

Recovery of photopic ERG from pressure-induced retinal ischemia in rabbit eyes.

Using precise experimental methods, we evaluated the recovery from pressure-induced ischemia in rabbit retina using photopic electroretinography (ERG). The recovery was evaluated and documented in terms of the increasing amplitude and the decreasing delay of implicit time in the photopic ERG after inducing 30-, 60- and 90-minute ischemia. A ganzfeld dome was utilized to stimulate both the experimental and the control fellow eye. The changes in ERG parameters in the ischemic eye were assessed by comparison with the values simultaneously obtained in the fellow eye, which could minimize the effect of anesthesia. One hour after reperfusion, the implicit time returned to almost the same value as that in the fellow eye in eyes with 30-minute ischemia, while it showed significant delay in eyes with 60-minute ischemia. The amplitude was smaller than that in the fellow eye in eyes with both 30- and 60-minute ischemia. Photopic ERG was not recorded one hour after reperfusion in more than half the eyes with 90-minute ischemia.

[Increased production of nitric oxide in the immediate and late asthmatic responses in models of guinea pig experimental asthma].

Nitric oxide is produced in mammalian airways by constitutive and inducible nitric oxide synthase, and endogenous nitric oxide can be detected as exhaled gas. Patients with asthma have large numbers of airway epithelial cells and inflammatory cells that contain nitric oxide synthase, and nitric oxide levels in exhaled air are high during immediate asthmatic responses. To examine the dynamics of nitric oxide synthase in the tracheas of guinea pigs, cumulative dose-response studies of 5-HT were done on tracheal strips, in the presence or absence of arginine and its analogues. This assay indicated that relative the activity of nitric oxide synthase was greater 6 hr after the challenge than immediately after, which suggests that nitric oxide is involved in the pathogenesis of bronchial asthma attacks.

Angiographic findings in eyes with active ocular toxoplasmosis.

Clinical findings in 5 cases of active ocular toxoplasmosis were documented by fluorescein angiography. The diagnosis was based upon the ophthalmoscopic findings of white, fluffy, retinal exudative lesions with or without overlying vitreous inflammatory cells, and positive serologic tests for toxoplasma and responsiveness to clindamycin therapy. Three cases were considered as recurrent ocular toxoplasmosis and the other 2 cases, acquired ocular toxoplasmosis. Angiographic characteristics of these 5 cases of acute ocular toxoplasmosis consisted of the following: 1) hypofluorescence in the center of the hyperfluorescent lesion, 2) black silhouette of the artery corresponding to the arterial occlusion traversing the necrotic lesion, 3) venous dilation, venous wall staining and dye leakage from the vein, 4) optic disc staining and dye leakage. These angiographic findings reflect either tissue necrosis or hypersensitivity reaction and are helpful in the correct and prompt diagnosis of acute ocular toxoplasmosis.

Antibacterial activity of tilmicosin against Pasteurella multocida and Actinobacillus pleuropneumoniae isolated from pneumonic lesions in swine.

Sixty one strains of Pasteurella multocida and 35 strains of Actinobacillus pleuropneumoniae isolated from pneumonic lesions of porcine lungs during the period from 1985 to 1989 in Japan were tested for antibiotic susceptibility to chlortetracycline (CTC), thiamphenicol (TP), tylosin (TS), acetylisovaleryl-tylosin (AIV-TS), tilmicosin (TMS), mirosamycin (MRM). Most strains of both species were sensitive to CTC, TP and TMS. Growth of fifty-one strains (83.6%) and forty-six strains (75.4%) of P. multocida were inhibited with 3.13 micrograms/ml of CTC and 0.78 micrograms/ml of TP, respectively. TS showed low activity against almost all strains (MIC > or = 6.25 micrograms/ml). Fifty-eight (95.1%), twenty-three (37.7%) and fifty (82%) of P. multocida showed MICs of > or = 6.25 micrograms/ml against AIV-TS, TMS and MRM, respectively. The MICs of A. pleuropneumoniae against CTC were less than 1.56 micrograms/ml. Thirty-two strains (91.4%) and 33 strains (94.3%) of A. pleuropneumoniae were inhibited with 3.13 micrograms/ml of TP and TMS respectively. However, TS, AIV-TS and MRM showed low activity against all of A. pleuropneumoniae (MIC > or = 6.25 micrograms/ml). Three different resistance patterns were observed in P. multocida and two in A. pleuropneumoniae isolates, respectively.

[Bronchial asthma and desaturation--assessment by pulse oximetry].

In the first study, we used pulse oximetry to continuously measure SpO2 and pulse rate in inpatients with paroxysmal attacks of acute asthma. Desaturation and increases in pulse rate occurred during coughing, urination, defecation, eating, and sleeping. Desaturation was most severe and frequent when peak expiratory flow was in the red zone. When it was in the yellow zone or blue zone, desaturation was less severe and less frequent. In the second study, outpatients experiencing exacerbations of chronic asthma and inpatients experiencing acute asthmatic attacks inhaled of a beta 2-stimulant via an ultrasonic nebulizer and were monitored with pulse oximetry. In the former, SpO2 either did not change or increased, but in the latter SpO2 decreased markedly when patients were in the red zone. Therefore, when patients inhale a bronchodilator during an acute asthmatic attack they should be carefully monitored with pulse oximetry. In the third study, pulse oximetry was used to measure saturation in outpatients who were suspected of having hyperresponsive airways and had undergone an airway hyperresponsiveness test with an Astograph. Almost all of those who had desaturation of more than 4% from were markedly hyperresponsive. The degree of desaturation and the percent change in respiratory resistance were significantly correlated. In conclusion, pulse oximetry can be useful in the short-term and long-term management of asthma tic patients.

[Binswanger's type encephalopathy without alopecia and lumbago in young hypotensive patients].

Binswanger's type encephalopathy is characterized by progressive dementia and diffuse subcortical ischemic lesions associated with arteriosclerosis. Hypertension is believed to be a major pathogenic factor in causing this encephalopathy but there are some cases of the encephalopathy not suffering from hypertension. In 1985, Yamamura et al. and Fukutake et al. reported familial cases of normotensive juvenile Binswanger's type encephalopathy with alopecia and lumbago, and considered it to be possibly a new clinical syndrome. We reported three cases of relatively young-onset (under the age of 40) Binswanger's type encephalopathy with persistent hypotension. All three patients suffered from neither alopecia nor lumbago. Patient (male aged 40) had repeated episodes of ischemic stroke and had progressive dementia. Patients 2 (male aged 41) and 3 (male aged 34) were not in a state of dementia, but had a history of transient ischemic attacks, and at present are completely symptom-free. Though there were no risk factors for cerebrovascular disease in these cases, the repeated episodes of ischemic stroke and the existence of small multiple lacunes in the basal ganglia on CT and MRI suggest that the white matter damage was principally due to a vascular disorder. In these cases, persistent hypotension was characteristic and might be a factor for the induction and exacerbation of this encephalopathy. These three cases are different from the classic form of Binswanger's type encephalopathy based on hypertension. Normotensive cases have been described before, but our cases do not seem to fall into this category because the blood pressure constantly remained hypotensive.(ABSTRACT TRUNCATED AT 250 WORDS)
