Monoclonal antibody SP-21 defines a sialosyl-Tn antigen expressed on carcinomas and K562 erythroleukemia cells.

Monoclonal antibody (mab) SP-21 resembles the well established mab B72.3 by high affinity binding to peptide linked sialosyl-Tn disaccharide, NeuAc alpha 2-6 GalNAc alpha, and non-reactivity to the related trisaccharide NeuAc alpha 2-6 (Gal beta 1-3)GalNAc alpha. Although mab SP-21 may be classified, accordingly, into the family of mabs defining the sialosyl-Tn antigen, its fine specificity is distinct from the reference antibody B72.3 by binding of mab SP-21 to fetal mucins from human meconium or amniotic fluid. The distinct immunoreactivity of this mab is also documented by its specific cell staining of the erythroleukemia cell line K562, where it binds to a 105 KDa glycoprotein. Moreover a subpopulation of normal lymphocytes stains to this mab SP-21.

Antitumor effect of K18 on metastatic models.

K18 is a newly synthesized antitumor agent which is the conjugate form of human immunoglobulin with p-di (2-chloroethyl)-amino-L-phenylalamine (melphalan). The antitumor effect of K18 on two animal metastatic models was investigated: (a) Lewis lung carcinoma was transplanted into thigh muscle of mice, followed by the reaction of the primary lesion, or the tumor was transplanted through the tail vein: (b) a renal model was also created, where colon-26 was inoculated into the renal capsule of BALB/c mice. Oral administration of K18 resulted in the decrease in the number of nodules in the lung in the case of tumor transplantation through tail vein.

Effect of PSK on prohibited immunity of splenectomized mice.

The effect of PSK (Krestine, an anti-tumor drug prepared from Coriolus versicolor) on splenectomized experimental animals was investigated. Splenectomy was performed on both a tumor-free control group and a tumor-bearing group. The administration of PSK on the splenectomized control group significantly increased the immune state of the host. In the case of the tumor-bearing group, administration of PSK resulted in restoration of the immune function as observed in the control group. Recovery of the immunological function was accelerated when tumor-bearing animals were splenectomized at the terminal stage. The results suggest that the immunomodulating effects of PSK developed at the time of the splenectomy resulted in anticancer activity.

Marked induction of HSP47, a collagen-binding stress protein, during solid tumor formation of ascitic Sarcoma 180 in vivo.

HSP47, a heat shock-inducible glycoprotein of Mr = 47,000, is assumed to be a molecular chaperone specific for collagen. In addition to its stress inducibility, HSP47 has transformation-sensitivity; that is, the synthesis of HSP47 decreases after malignant transformation. We developed a rat monoclonal antibody against mouse HSP47, which we then used to examine the amounts of HSP47 in transplanted tumors. HSC70 (HSP73) was expressed constitutively in these ascites as well as solid tumors. While HSP47 was not expressed in ascitic form of Sarcoma 180, its expression was markedly induced during solid tumor formation of ascitic Sarcoma 180 cells after subcutaneous transplantation. The average survival period of ascites Sarcoma 180-bearing mice was 20.3 +/- 3.6 days (mean +/- SD), and was much shorter than that of solid Sarcoma 180-bearing mice. These findings suggest an important role of HSP47 in solid tumor formation as well as a possible marker for tumor malignancy.

Inhibition of HSP47 during the transition from solid to ascitic form of Sarcoma 180 in vivo.

HSP47, a collagen-binding stress protein, has transformation-sensitivity; that is, the synthesis of HSP47 decreases after malignant transformation. We examined the expression of HSP47 in transplanted tumours by Western blotting. While HSP47 was not detected in ascites tumours including Ehrlich, P388 and Sarcoma 180, marked expression of HSP47 was observed in solid tumours such as B16, B16-BL6 and Sarcoma 180. The expression of HSP47 in Sarcoma 180 cells disappeared completely after these solid tumour cells were dispersed and inoculated intraperitoneally. These findings suggested an important role of HSP47 as a marker for tumour malignancy.

The role of neutrophils as cytotoxic cells in lung metastasis: suppression of tumor cell metastasis by a biological response modifier (PSK).

We aimed to determine the role of neutrophils and the usefulness of a protein-bound polysaccharide (PSK) in the suppression of tumor cell metastasis in the lung in vivo. Circulating neutrophils collected frm tumor-bearing animals (Line-10 hepatocarcinoma) induced a marked decrease in the size and number of metastatic foci in the lung. Although pulmonary macrophages (PAMs), lymphocyte and eosinophil in bronchoalveolar lavage (BAL) fluid increased following tumor cell inoculation, in addition to these findings we found that PSK caused an increase in BAL neutrophil levels causing increased of target cell toxicity and a marked decrease in the size and the number of lung metastatic foci. Superoxide anion generation of blood neutrophils collected from PSK-treated animals with metastasis showed forward acceleration. The presence of neutrophil chemotactic factors was confirmed in the BAL fluid of PSK-treated animals with metastasis, but not leukotriene B4. The results suggest that modulation of the tumor cell microenvironment by activation of neutrophils may prove to be an additional modality in treatment strategy by combining PSK as a biological response modifier with conventional therapies for lung metastasis.
