Two clones isolated from a metastatic variant of mouse colon adenocarcinoma 26; the high metastatic NL-17 and the low metastatic NL-44, induced similar platelet aggregation in vitro. Heterotypic aggregates of tumor cells and platelets were injected i.v. into mice. The lung colonization potential of the NL-17 was dependent on the extent of tumor cell platelet aggregation. This clearly indicates that the interaction of tumor cells with platelets can lead to enhanced tumor metastasis possibly through more efficient intravascular arrest of the heterotypic aggregates. The interaction of tumor cells with platelets could thus be an important determinant for successful metastasis. NL-44, however, did not form pulmonary metastasis even after the tumor cells formed heterotypic aggregates with platelets, suggesting that tumor metastasis, is dependent on the intrinsic nature of tumor cells. Lung extract enhanced the growth of NL-17 more effectively than that of NL-44. These results suggest that, in addition to the interaction of metastatic cells and platelets, host factors including growth-promoting factors might also have an important role in tumor metastasis.
Metastatic tumor burden in the lung of C57BL/6 or BDF1 mice was quantitated by measuring DNA polymerase alpha activity in the lung of tumor-bearing animals. DNA polymerase activity in the lung increased time-dependently following the inoculation of Lewis lung carcinoma (s.c.) or B16 melanoma variant B16-B2 (i.v.). In the Lewis lung carcinoma system, the number of metastatic modules and the weight of lung also increased time-dependently. Results from the B16 melanoma showed that the increase in lung nodules occurred 10 to 20 days after i.v. inoculation of tumor cells. DNA polymerase activity increased significantly during this period. Because the lung nodules were very small there was no obvious concomitant increase in lung weight. Since no significant infiltration of host cells was observed in the lung in response to metastatic foci, the rise in DNA polymerase activity should be due to tumor cells and not to infiltrating host cells. When the metastasis of Lewis lung carcinoma was inhibited by adriamycin and cyclophosphamide, decrease in DNA polymerase activity in the lung occurred. These results indicate that the degree of tumor metastasis can be quantitated by measuring DNA polymerase activity.
The effects of lung tissue extract on the cell growth of eight colon 26 tumor clones, four highly and four poorly lung-colonizing clones, were examined in vitro. Addition of lung extract to serum-free medium stimulated the growth of all four of the highly lung-colonizing clones and one of the poorly lung-colonizing clones, but it had minimal effect on the other three poorly lung-colonizing clones. These results indicate that the lung extract contains a growth stimulating activity; it selectively stimulated some of the colon 26 clones including highly lung-colonizing ones. The growth stimulating activity was not dialyzable, was partially destroyed by heating at 56 degrees C or 80 degrees C for 30 min and completely destroyed by trypsin. These results suggest that the activity resides in a protein. Gel filtration chromatography of lung extract on Sephacryl S-200 revealed that the active component was eluted in a molecular weight range of 90,000-120,000.
BACKGROUND: Preserved myocardial oxygen consumption estimated by carbon 11-acetate and positron emission tomography (PET) in myocardial regions with chronic but reversibly depressed contractile function in patients with ischemic heart disease have been suggested to be caused by repeated short episodes of acute myocardial ischemia. To evaluate this hypothesis myocardial 11C-acetate PET imaging was performed before and after acute repetitive myocardial ischemia. METHODS AND RESULTS: In open chest dogs (n = 8), the left anterior descending coronary artery was occluded 4 times for 5 minutes alternating with 5 minutes of reperfusion. Before and after repetitive coronary occlusions, oxygen 15 water/oxygen 15 carbon monoxide (blood flow), and 11C-acetate (oxygen consumption) PET imaging were performed. Left ventricular regional systolic wall thickening was measured with sonomicrometry. Forty-five minutes after the ischemic episodes, systolic ventricular wall thickening was decreased by 90%, whereas myocardial blood flow was reduced by 21% compared with baseline values (P < .05). Ninety minutes after the ischemic episodes, estimated oxygen consumption was unaltered compared with the baseline level despite a sustained 70% decrease in the regional contractile function (P < .05). CONCLUSIONS: Oxygen consumption estimated by 11C-acetate PET imaging is preserved after repeated episodes of acute myocardial ischemia despite a severe impairment of contractile function.
Thallium-201 has been used extensively as a myocardial perfusion agent and to assess myocardial viability. Unlike other 99mTc-labeled agents such as 99mTc-sestamibi and 99mTc-tetrofosmine, the regional concentration of 201Tl varies with time, and its kinetics make it a potential candidate for estimating absolute physiologic parameters with kinetic model analysis. This article outlines a strategy for quantitative assessment of regional myocardial blood flow in man using 201Tl and dynamic single photon emission computed tomography (SPECT). Quantitatively accurate SPECT images that are proportional to the true radioactivity distribution are prerequisites for model-based kinetic analysis. Our technique for quantitative SPECT includes ordered-subset maximum likelihood-expectation maximization (ML-EM) reconstruction with transmission data-based attenuation correction and transmission-dependent convolution subtraction scatter correction. A three-compartment model was found to reproduce the observed regional time-activity curves well, and dog experiments demonstrated that influx rate constant (K1) values estimated from the dynamic SPECT data correlated well with absolute myocardial blood flow determined by in vitro microspheres for a physiologically wide range of flows. Several possible strategies for simplifying the study procedures, without compromising accuracy, are also presented, which should make absolute quantitation of regional myocardial blood flow feasible using 201Tl and a conventional SPECT camera in a clinical setting.
A high performance positron emission tomograph, HEADTOME III, with 480 bismuth germanate oxide detectors (13.4 X 25.0 X 40.0 mm), arranged in three rings of 750 mm diameter, with independent shadow masks and septa, is described. Image plane resolution at the center of the field of view (FOV) is 7.9 and 6.5 mm full width at half maximum (FWHM) in low resolution (LR) and high resolution (HR) mode, respectively. Axial resolution is 13.1, 11.3, and 9.1 mm FWHM at the center of FOV for direct planes of low quantitation (LQ), cross planes of LQ, and high quantitation (HQ) mode, respectively. Sensitivities evaluated by true events for a 68Ga 20 cm diameter cyclindrical pool were 34.5 (56.7), 16.4 (27.5), 19.4 and 9.5 kcps (muCi/ml)-1 with the direct (cross) planes of LQLR, direct (cross) planes of LQHR, and direct planes of HQLR and HQHR, respectively. The fraction of scattered coincidence in a cold spot phantom after software correction is 2.5% in LRLQ mode. Count rate linearity after software correction is within 1% up to 50 X 10(3) true events per second per plane with LRLQ mode.
A new encoding scheme was developed that is applicable to bismuth germanate (BGO) detector units for a high resolution positron emission tomograph. The detector unit is composed of eight equal-size, 3 mm wide BGO crystals coupled to a dual-module square photomultiplier tube (PMT) through a pair of light guides. The light guides divide the scintillation light between the two modules of the PMT, with a ratio dependent on the crystal of origin. Mean detector pair spatial resolution as measured by the point spread function was 3.9 mm full width at half maximum (FWHM) and reconstructed image spatial resolution was 4.8 mm FWHM at the center of the field of view. Timing resolution between two detector units was 6.0 ns FWHM. Energy resolution was 24% FWHM for 511 keV gamma rays. The finest spots of the Derenzo phantom were clearly resolved.
We have developed a method that allows us to place regions of interest on X-ray CT (XCT) images that are automatically adjusted to positron emission tomography (PET) images from the same patient. A face mask with landmarks was used during PET and XCT studies for matching slice positions between PET and XCT. Anatomical locations in different images of the same slice can be accurately adjusted using the landmarks and a video system. In our clinical experience misadjustments of the slice position are on the average less than 2 mm in axial distance and 1.0 degrees in slice angle. The method is easily applicable to any PET or XCT device.
OBJECTIVE: A fully automatic multimodality image registration algorithm is presented. The method is primarily designed for 3D registration of MR and PET images of the brain. However, it has also been successfully applied to CT-PET, MR-CT, and MR-SPECT registrations. MATERIALS AND METHODS: The head contour is detected on the MR image using a gradient threshold method. The head region in the MR image is then segmented into a set of connected components using the K-means clustering algorithm. When the two image sets are registered, the segmentation of the MR image indirectly generates a segmentation of the PET image. The best registration is taken to be the one that optimizes the segmentation induced on the PET image. In this article, the K-means minimum variance criterion is used as a cost function, and the optimization is performed using the method of coordinate descent. RESULTS: The algorithm was tested on 80 H2 15O PET and MR image pairs from 10 subjects. Qualitatively correct results were obtained in all cases. With use of external markers visible in both image modalities, the average registration error was estimated to be < 3 mm. CONCLUSION: The algorithm presented in this article requires no user interaction and can be applied to a wide range of registration problems. Quantitative and qualitative evaluations of the algorithm indicate a high degree of accuracy.
The role of bradykinin potentiation in the antihypertensive effect of captopril was assessed in conscious rabbits with two-kidney, one-clip hypertension. On the 35th day after clipping the renal artery, a bolus injection of bradykinin 0.2 microgram/kg was repeated at 15-min intervals three times before and for 180 min after the administration of captopril 0.5 mg/kg. The magnitude and duration of captopril-induced hypotension were more marked in hypertensive rabbits than in controls. The bradykinin-induced hypotension was markedly enhanced by captopril. However, the bradykinin potentiation by captopril was not correlated with its vasodepressive effect. On the other hand, a close relationship (r = 0.825, p less than 0.001) was observed between plasma renin activity and the maximum reduction in blood pressure induced by captopril. The experiment performed after sodium depletion for 7 days revealed that the bradykinin potentiating effect of captopril was not affected by sodium depletion. Therefore, it is concluded that the vasodepression by captopril does not necessarily depend on its bradykinin potentiating action and the latter is not influenced by the renin-angiotensin system or fluid volume depletion.
A 49-year-old male, with a relatively uncommon multiple neurilemmoma, exhibited symptoms of cauda equina compression. Following a diagnosis of multiple tumor of the cauda equina and extradural cyst, based on myelography and CTM, the tumors at L1-L2, the left S1 nerve root, and left hip were resected. Three years after the operation, the patient is asymptomatic apart from slight numbness in the right hip and there is no evidence of tumor recurrence. The rarity of such a case prompted this report.
