Immunohistochemical observation on substance P in regenerating taste buds of the rat.

The present study was performed to investigate the relationship between substance P-positive (SP-positive) nerve fibers and regeneration of taste buds in the foliate papillae of the rat by means of immunohistochemistry. It was confirmed by neurotomy that taste buds in the foliate papillae of the rat were innervated mostly (90%) by the glossopharyngeal (IXth) nerve and partly (10%) by the chorda tympani. In this experiment, the IXth nerve was sectioned distal to the petrosal ganglion. The rats were sacrificed at various intervals from 20 days to 80 days after the operation. In the course of degeneration and disappearance of taste buds, both SP-positive fibers and taste buds disappeared completely from the posterior folds of the foliate papillae 7 days after the operation. Within 22 days, regenerated SP-positive fibers began to appear in the lamina propria, and following the penetration of the fibers into the epithelium, taste bud anlagen reappeared at the bottom of the trench, and in the posterior folds at 24 days. The process of new taste bud formation extended toward the apex of the trench and to the anterior folds, which seemed to follow the regeneration in the nerve trunk. Quantitative data showed a gradual increase in the number of taste buds and taste buds containing SP-positive fibers. These findings indicate that SP might have a role in regeneration of taste buds.

The distribution and origin of substance P-like immunoreactivity in the rat molar pulp and periodontal tissues.

Rat mandibles were fixed in Zamboni fixative and demineralized in a mixture of EDTA and fixative. Substance P-like immunoreactivity was demonstrated by indirect immunofluorescence in molar pulp, periodontal ligament and gingiva. Substance P (SP) containing nerve fibres with varicosities were observed in the pulp horn and root pulp in general located around blood vessels. Some SP-containing fibres penetrated into the predentine and dentine. In the periodontal ligament, SP fibres were localized along the blood vessels in the middle and apical regions. Many SP-containing fibres were associated with the blood vessels in the lamina propria of gingiva. After inferior alveolar nerve section, SP-positive nerve fibres in the pulp and periodontal ligament disappeared completely. In gingiva the number of SP fibres decreased but not all fibres disappeared. Removal of the superior cervical ganglion did not affect the distribution of SP-containing nerve fibres.

Biochemical characteristics of cardiac myosin: the pH dependence of Ca-ATPase activity, and that of the absorption spectrum of 2,4,6-trinitrophenyl groups attached to myosin.

For both cardiac and skeletal myosin, the Ca-ATPase activity of myosin at acidic pH was shown to be different from that at alkaline pH, in the susceptibility to heat-inactivation, the effects of organic solvents, and the effect of trinitrophenylation of the myosin. It is therefore suggested that there are two different types of Ca-ATPase of both cardiac and skeletal myosin. Differences in the Ca-ATPase activity were also found between cardiac and skeletal myosins. (a) The Ca-ATPase activity of cardiac myosin was more susceptible to heat-inactivation at alkaline pH than at acidic pH. In contrast, the activity of skeletal myosin was more susceptible to heat-inactivation at acidic pH than at alkaline pH. (b) Dioxane weakly stimulated the activity of cardiac myosin at acidic pH, but strongly activated that of skeletal myosin at acidic pH. Acetone very strongly inhibited the activity of cardiac myosin at alkaline pH, but not so strongly that of skeletal myosin at alkaline pH. (c) Trinitrophenylation of the myosin resulted in loss of the activity optimum at acidic pH with skeletal myosin but not with cardiac myosin. As reported by Srivastava et al. (J. Biochem. 86, 725-731, 1979), 1 mol of lysine residue per mol of cardiac myosin quickly reacted with 2,4,6-trinitrobenzene sulfonate (TNBS) either in the absence or presence of inorganic pyrophosphate (PPi). However, trinitrophenyl (TNP) groups bound to cardiac myosin in the presence of PPi were significantly different, in the pH dependence of the absorption spectrum, from those bound (to cardiac myosin) in the absence of PPi.(ABSTRACT TRUNCATED AT 250 WORDS)

[Evaluation of 164 cases of ocular injuries caused by various sports].

Results of analysis on 164 cases of ocular trauma produced by various kinds of sports have been presented. The baseball constitutes a cause of such accidents in 46,3 % of cases. The presence of specific relation has been suggested between a given type of sports and ocular manifestations that follow this.

Abnormalities of cone photopigments in genetic carriers of protanomaly.

Anomaloscopic color matching was performed in 57 protanomalous boys. The relative luminous efficiencies of their mothers were measured by flicker photometry to clarify the characteristics of protanomaly carriers. The sensitivity loss of protanomaly carriers in the long wave-length region had a highly significant correlation with the anomalous quotients ( AQs ) of their protanomalous sons. This correlation means that both the luminous efficiencies of the protanomaly carriers and the AQs of their sons are determined by the same "anomalous" cone pigments.

Bile secretion in regenerating liver--a comparison of hepatic resection and ligation of the portal vein branch in dogs.

To elucidate the mechanism of bile secretion in the regenerating liver, changes of bile secretion were investigated in dogs during 12 weeks or more after 70% hepatectomy and after ligation of the portal vein branch supplying 70% of the area of the liver. The remnant liver weight increased remarkably and reached a normal level at 2 weeks after 70% hepatectomy. Liver weight increased in the nonligated lobes and decreased in the ligated lobes after ligation of the portal vein branch, but the whole liver weight was unchanged. Bile flow increased and continued for 12 weeks or more in the remnant liver except for a period of 2 weeks in the nonligated lobes. It decreased markedly in the ligated lobes. The taurocholic acid-independent bile flow was higher in the remnant liver than in nonligated lobes in both of which it was higher than normal liver. In the ligated lobes, taurocholic acid-independent bile flow was lower and taurocholic acid-dependent bile flow was higher than normal liver. When gallbladder bile was infused into the terminal ileum, bile flow increased in both the remnant liver and the nonligated lobes, except for 2 weeks in the remnant liver. Therefore it is suggested that in the regenerating liver enterohepatic circulation of bile acid and the new balance of the bile acid pool size participates in not only in bile flow but also in liver regeneration.

Substance P-like immunoreactivity in the pulp-dentine zone of human molar teeth demonstrated by indirect immunofluorescence.

Freshly-extracted human third molars were fixed in Zamboni fixative, demineralized with the mixture of EDTA and the fixative; substance P-like immunoreactivity (SPLI) was revealed by the indirect immunofluorescence technique of Coons. Substance P (SP) was observed in the pulp-dentine zone and the dental pulp. Some of SP-containing fibres ended at the odontoblast layer and did not reach the predentine; others terminated at the predentine surface or penetrated into the predentine. In the predentine, some of SP fibres accompanied odontoblast processes and ended near the mineralized dentine; others changed course transversely at various levels.

Ultrastructure of granular materials in rat incisor enamel organ at early maturation stage.

With scanning electron microscopy, globules were seen in inter-cellular spaces between ameloblasts with ruffled borders and papillary cells at early maturation stage. Numerous globules were located on longitudinal folds between parallel ridges on the sides of ameloblasts, but only a few in the papillary layer. In previously-scanned specimens sectioned for transmission electron microscopy, the globules were composed of finely granular material. Vacuoles containing material identical to extra-cellular granules were observed in intra-cellular portions of the ameloblasts and papillary cells.

Vitreous fluorophotometry in patients with cone-rod dystrophy.

Eight patients with cone-rod dystrophy were examined by means of vitreous fluorophotometry. The data in three patients with an early stage of retinitis pigmentosa (rod-cone dystrophy) served for comparison. In contrast with the abnormal findings on vitreous fluorophotometry in patients with an early stage of retinitis pigmentosa the value of vitreous fluorophotometry in cone-rod dystrophy was within the normal range in all patients except the two with advanced stages of this disease. Our results suggest that, although both diseases are based on diffuse dystrophy of photoreceptors, and the final stages of the two diseases may show several similar features, the process of breakdown of the blood retinal barrier differs greatly in the two diseases.
