Response and remission rates in different subpopulations with major depressive disorder administered venlafaxine, selective serotonin reuptake inhibitors, or placebo.

BACKGROUND: Most examinations of the clinical efficacy of drugs used to treat depression pool subjects across gender and age groups. This investigation compared these patient subpopulations on the basis of remission and response rates associated with venlafaxine and selective serotonin reuptake inhibitor (SSRI) treatment. METHOD: A meta-analysis of original data from 8 comparable double-blind, active-controlled, randomized clinical trials (4 also placebo-controlled) was conducted. Antidepressant efficacy was assessed for patients (N = 2,045) aged 18 to 83 years (subgroups: < or = 40, 41-54, 55-64, and > or = 65 years) who met DSM-III-R criteria for major depression or DSM-IV criteria for major depressive disorder and were randomly assigned to receive venlafaxine (immediate release, N = 474; extended release, N = 377), one of several SSRIs (N = 748), or placebo (N = 446) for up to 8 weeks. Symptoms of depression were assessed using the Hamilton Rating Scale for Depression (HAM-D). Remission was defined as a HAM-D-17 score < or = 7, response was defined as > or = 50% decrease in HAM-D-21 score, and absence of depressed mood was defined as a HAM-D depressed mood item score of 0. RESULTS: We detected no significant age-by-treatment, gender-by-treatment, or age-by-gender-by-treatment interactions; men and women of different ages within a given antidepressant treatment group exhibited similar rates of remission, response, and absence of depressed mood. Regardless of age or gender, remission rates during venlafaxine therapy were significantly higher than during SSRI therapy (remission rates at week 8: venlafaxine, 40%-55% vs. SSRI, 31%-37%; p < .05). Regardless of patient age or gender, onset of remission was more rapid with venlafaxine than with SSRI treatment. By contrast, rates of absence of depressed mood with venlafaxine (34%-42%) and SSRIs (31%-37%) did not differ significantly and tended to be similar for all patient subgroups. CONCLUSION: These data suggest that men and women have comparable responses to SSRIs and venlafaxine across various age groups. Moreover, patients exhibited a more rapid onset and a greater likelihood of remission with venlafaxine therapy than with SSRI therapy regardless of age or gender.

Keratin 23 (K23), a novel acidic keratin, is highly induced by histone deacetylase inhibitors during differentiation of pancreatic cancer cells.

Sodium butyrate (NaBu) was shown to induce differentiation and apoptosis in the human pancreatic cancer cell line AsPC-1. A suppression subtractive hybridization-based technique was used to identify genes induced by NaBu. A novel cDNA was found to be highly up-regulated in AsPC-1 cells in response to NaBu. The gene expresses a 1.65-kb mRNA encoding a protein with an open reading frame of 422 amino acids. It has an intermediate filament signature sequence and extensive homology to type I keratins. Sequence comparison with known keratins indicated that the gene shares 42-46% amino acid identity and 60-65% similarity within the alpha-helical rod domain. The gene is named K23 (for human type I Keratin 23, KRT23). K23 mRNA was highly induced by NaBu in different pancreatic cancer cells. Trichostatin A (TSA), a potent and specific inhibitor of histone deacetylase, similarly induced K23 mRNA expression. Treatment with either actinomycin D or cycloheximide efficiently blocked the induction of K23 mRNA by NaBu/TSA. These results indicate that K23 mRNA induction by NaBu or TSA is a downstream event of histone hyperacetylation. We also demonstrated that expression of p21(WAF1/CIP1) antisense RNA efficiently blocked the induction of K23 mRNA induced by NaBu. Our results suggest that K23 is a novel member of the acidic keratin family that is induced in pancreatic cancer cells undergoing differentiation by a mechanism involving histone hyperacetylation. p21(WAF1/CIP1) serves as an important mediator during the induction process of K23 by NaBu.

[The effect of EB virus bhrfl gene expressing on the topoisomerase I expression in nasopharyngeal carcinoma cell line].

OBJECTIVE: To study the effect of bhrfl expressing on the topoisomerase I expression in nasopharyngeal carcinoma cell line CNE2. METHODS: bhrfl recombinant expressing vector was constructed and used to transfect the nasopharyngeal carcinoma cell line CNE2. The CNE2 cells were then tested for changes of the topoisomerase I expression and anti-apoptotic ability under the treatment of camptothecin. RESULTS: bhrfl expressing could enhance the topoisomerase I expression in CNE2 cells, that in end reduced cell sensitivity to camptothecin increased cell survival span and decreased apoptotic rate(P < 0.01). CONCLUSION: By increasing the expression of topoisomerase I, bhrfl expressing can enhance the resistance of CNE2 cells to apoptosis induced by camptothecin.

[Hypoglycemic effect of chitosan-microcapsulated insulin on the blood glucose level of streptozotocin-diabetic Wistar rats].

This study was conducted to observe the hypoglycemic effect of chitosan-microcapsulated insulin (insulin-microspheres) on blood glucose (BG), in diabetic rats by gavage. Using the scanning electron microscope, we observed the shape and distribution of insulin-microspheres in the tissues of liver, spleen, and small intestine of normal rats after gavage. The following groups of rats were studied: normal control rats (NC, n = 5); streptozotocin (STZ)-inducing diabetic control rats(DC, n = 6); STZ-diabetic rats treated with insulin-microsphere [120 u/kg] by gavage (DF, n = 6); STZ-diabetic rats treated with subcutaneous insulin injection [24 u NPH/kg] (DT, n = 6). Blood glucose was measured at 0-7 days after the treatments. The insulin-microspheres was found in small intestine, liver and spleen at 1, 2, 3 and 7 days. In DF group, the average blood glucose level decreased significantly from 24.7 +/- 3.2 mmol/L to 16.9 +/- 5.5 mmol/L during the first day of treatment and reached the lowest level (12.1 +/- 5.7 mmol/L) on the second day. From the third day on, the BG gradually elevated to the level before treatment; the maximum decrease of blood glucose level was 50.2%. In DT group, the average blood glucose level decreased from 25.2 +/- 3.8 mmol/L to 10.4 +/- 5.2 mmol/L during the first day of treatment and then went up gradually from the 2nd day on; the maximum decrease of glucose level was 58.7%. No significant difference was seen in maximum decrease between the two groups. These data demonstrate that oral chitosan-microcapsulated insulin has the antihyperglycemic effect on the blood glucose level of streptozotocin-diabetic rats.

[Primary respiratory syncytial virus infection in mice].

OBJECTIVE: To establish a mice-model of primary respiratory syncytial virus infection. METHODS: Twelve Balb/c mice (8-12 w) were divided into two groups with three different titers of RSV infection in the experimental group. Mice were anaesthetized with pentobabitone(30 mg/kg) and infected by nasal drip of 0.1 ml of RSV which was harvested from HEP-2 cell culture. Three mice in the control group received sham infection with HEP-2 cell media. Five days later, lungs were removed in aseptic condition. Right lung was weighted and homoginized with MEM(1:10 ratio), then centrifuged at 4 degrees C, 2,000 r/min for 20 minutes. Supernatants were collected for virus isolation and plaque forming assay. Left lung was collected for histopathological and electromicroscopic examination. RESULTS: (1) RSV grew with a characteristic syncytial formation in HEP-2 cells, leading to the fusion of cells with round, elliptic, map or tree like shape. (2) Virus was isolated and detected in the lung tissue in experiment group. (3) There was a positive correlation between RSV titer added and that replicated in the lung. (4) After the eclipse phase, viral replication in the lung reached maximum within 4 and 6 days. The most apparent pathological change was found between day 5 and 8. Mononuclear cell infiltration was presented in the perivascular, peribronchial and in the alveoli spacers. (5) Huge viral replication with its fuzzy coat was detected inside the cytoplasm of the type II alveolar lining cell at high magnification. CONCLUSION: An animal model of RSV infection Balb/c mice was successfully established by nasal inoculation with respiratory syncytial virus.

[Laparoscopic surgery in the treatment of early uterine malignancies: a report of 8 cases].

OBJECTIVE: To explore the feasibility of laparoscopic operation as treatment for early malignant uterine tumors. METHOD: Six patients with endometrial carcinoma underwent laparoscopic extensive total hysterectomy and bilateral adnexectomy, and 2 with cervical cancer underwent additional pelvic lymphadenectomy. RESULTS: The mean operating time of the 6 laparoscopic extensive total hysterectomy and bilateral adnexectomy was 220 min, and mean blood loss was 200 ml. The mean operating time of 2 cases underwent additional pelvic lymphadenectomy was 240 min. No organ trauma occurred, and the average post-operative hospital stay was 8 days. CONCLUSION: These results suggest that laparoscopic operation is safe and feasible in treating early stage malignant uterine tumors.

[A primary study on seed production of medicinal plant Sinopodophyllum hexandrum].

The study on the seed production of five Sinopodophyllum hexandrum (Berberidaceae) populations demonstrated that the average ovule number, average seed number and seed set ratio of these populations were varied, but all positively related with the altitude of the population. The average ovule number of a flower ranged from 58.79 to 87.97, average seed number of a fruit ranged from 40.02 to 80.58, and average seed set ratio of population ranged from 61.29% to 91.60%. The average weight per seed in a fruit was negatively related with the altitude and seed number in the fruit. The result also showed that seed production of a population had no obvious difference in different years. Although seed production was mainly influenced by nutrition condition of the individual, it was still influenced by altitude and other factors. Seed number in a fruit was positively related with fruit size(including weight, length and width) and total seed weight of the fruit, but average single seed weight of a fruit had no obvious relation with fruit size and total seed weight of the fruit.

[Chelate-induced phytoextraction of copper contaminated upland red soil].

A copper spiked red soil was used in pot experiment to study the effects of EDTA and low molecule weight organic acids on Cu speciation in soil, and Cu uptake by Brassica juncea. The results showed that the mobility of Cu in soil was improved obviously when EDTA was added at the vigorous growth stage. Both water-extractable and exchangeable Cu concentration increased significantly following EDTA addition. Citric acid and malic acid only had an effect on water-extractable Cu, and no effect on Cu uptake by the plant. EDTA significantly increased the concentration of Cu in plant leaves and roots, and the Cu uptake by Brassica juncea. All of these indicated the chelate-induced phytoextraction.

[Expression of alpha-smooth muscle actin in scar fibroblasts in vitro].

OBJECTIVE: To explore the expression of alpha-smooth muscle actin (alpha-SMA) induced by transforming growth factor beta 1 (TGF-beta 1). METHODS: Five samples of hypertrophic scars and three samples of normal mature scars were collected as the experimental and control groups respectively. The fibroblasts were isolated from scars, and cultured in 2-dimension or 3-dimension culture system. The immunohistochemical staining method of LSAB were used to investigate the expression of alpha-SMA in fibroblasts in the different concentration of TGF-beta 1. RESULTS: The expression of alpha-SMA in 3-dimension culture system were markedly lower than those in 2-dimension culture system with respect to the fibroblasts in the experimental group. The expression of alpha-SMA in fibroblasts were different in response to various TGF-beta 1 concentration, it was more effective at the concentration of 5 ng/ml. The expression of alpha-SMA in the fibroblasts from hypertrophic scars seemed to be more sensitive to TGF-beta 1 compared to that of the normal mature scars. CONCLUSION: There are concentration-dependent in the expression of alpha-SMA induced by TGF-beta 1 in scar fibroblasts in vitro. The biological characteristics of the fibroblasts from hypertrophic scars and normal mature scars and their sensitivity to the inducement of TGF-beta 1 were different. The inducement of TGF-beta 1 may be depressed by extracellular matrix components and that may decrease the expression of alpha-SMA.

[Longitudinal study about the function of pituitary-thyroid axis in pregnancy].

OBJECTIVE: To evaluate the change about the function of pituitary-thyroid axis and the role of human chorionic gonadotropin (hCG) on regulation of pituitary-thyroid axis in pregnancy. METHODS: The study was designed to provide longitudinal information of parameters of thyroid function and hCG in 74 healthy women during pregnancy and after delivery. Serum free triiodothyronine (FT3), free thyroxin (FT4), TT3, TT4, hCG and thyroid hormone binding globulin (TBG) concentrations were measured with radioimmunoassary and serum TSH concentration with immunoradiometric assay. RESULTS: (1) The FT3 and FT4 values below the reference range were 5.88% and 9.80%, respectively, in third trimester and after delivery, both were 38.10%. The serum TSH level below the reference range was 31.25% in first trimester and supranormal were 11.90% after delivery. The serum hCG level at all stages of pregnancy was higher than postpartum. (2) The serum FT3 level was significantly higher in first (4.37 +/- 0.78) pmol/L and second (4.75 +/- 0.90) pmol/L trimesters than postpartum (2.96 +/- 0.84) pmol/L, decreased in third trimester (3.94 +/- 0.75) pmol/L and the lowest level after delivery. The serum FT4 level was significantly higher in first (14.07 +/- 1.44) poml/L and second (12.86 +/- 0.84) pmol/L trimesters than postpartum (10.45 +/- 1.45) poml/L, decreased in third trimester (11.29 +/- 1.00) pmol/L and the lowest level after delivery. (3) The serum TSH level was the lowest values in early pregnancy (0.88 +/- 0.83) mU/L, showed significant increase in second (1.86 +/- 1.04) mU/L and third (1.48 +/- 0.90) mU/L trimesters and reached maximal level after delivery (2.82 +/- 1.42) mU/L. (4) The serum hCG level was the highest in early pregnancy (309.05 +/- 320.02) micrograms/L, then significant decreased in second (69.11 +/- 19.18) micrograms/L and third (86.25 +/- 44.60) micrograms/L trimesters and was the lowest levels after delivery (29.92 +/- 20.91) micrograms/L. (5) A significant negative correlation between TSH and hCG levels and a significant positive correlation between hCG and FT4 and TT4/TBG were demonstrated. CONCLUSIONS: (1) hCG may be as role on regulation of pituitary-thyroid axis during pregnancy and during certain time of postpartum. (2) After delivery, part of women were transient hypothyroxinemia.

Characteristics, diagnosis and treatment of hepatic metastasis of pure immature ovarian teratoma.

OBJECTIVE: To analyze the characteristics of hepatic metastasis of pure immature ovarian teratoma and explore its proper diagnosis and treatment. METHODS: Eighteen cases of hepatic metastasis of pure immature ovarian teratoma were included in this study. The clinical stage, operation, chemotherapy and histopathology of primary and secondary tumors as well as the data from long term follow-ups were analyzed retrospectively. RESULTS: All of the hepatic metastatic tumors were located on the surface of the liver. 61.1% (11/18) of them were clinical stage III and 44.4% (8/18) were grade 1 at first operation. The hepatic metastatic rate was 16.7% (3/18) in the standard adjuvant chemotherapy group but increased markedly to 31.2% (15/48) in the irregular chemotherapy group. Auxiliary diagnostic methods could not indicate the correct results. The surgical resection rate of hepatic metastasis of pure immature ovarian teratoma was 94.4% (17/18). There were less complications in the group with tumor diameter less than 15 cm. The follow-up time ranged from 3 to 205 months with a mean of 20.9 months. The 3-year-survival rate was 77.8% (14/18), and mortality rate was 22.2%. The 5- and 10-year-survival rate was 55.6% (10/18) and 38.9% (7/18), respectively. The rate of loss in follow-up was 22.2% (4/18) and 38.9% (7/18), respectively, and one patient has survived for more than 17 years. CONCLUSIONS: The hepatic metastatic rate of pure immature ovarian teratoma could be decreased using standard adjuvant chemotherapy. Suitable surgical treatment could reduce complications and improve the prognosis for patients.

[Treatment of childhood genital malignancies].

OBJECTIVE: To present the clinical profile and the result of treatment in childhood genital malignancies. METHODS: Twenty seven childhood patients (< 12 years) with genital malignancies diagnosed and treated in Peking Union Medical College hospital were analyzed retrospectively focusing on the clinical characteristics and outcome of the treatment. RESULTS: Patients ranged in age from 2 to 12 years with mean 6.5 years at diagnosis. Of the 27 patients, 23 had ovarian malignancies including 21 (77.8%) germ cell tumor. Four of the patients had extragonadal tumors with 2 endodermal sinus tumor of the vagina, and 2 embryonic rhabdomysarcoma. In ovarian malignancies, abdominal mass and abdominal pain were main clinical features accounting for 77.8% and 74.1% respectively. In vaginal malignancies, vaginal bleeding and polypoid tumor of the vagina were important clinical manifestations. 91.3% (21/23) of the patients underwent only unilateral salpingo-oophorectomy with preservation of contralateral ovary and the uterus. Childhood genital malignancies were very sensitive to the chemotherapy. Postoperative vincristine, actinomycine, cyclophosphamide (VAC) or cisplatine, vincristine, blyomycine (PVB) chemotherapy was individualized according to the tumor site, International Federation of Gynecology and obstetrics stage and histopathology. Survival rate in our series was 71.4% during the follow-up time (mean 47.4 months). CONCLUSIONS: Malignant germ cell tumor of the ovary is the most common tumor in the childhood genital malignancies. Principle of primary surgery is preservation of fertility and chemotherapy is also considered as a very important treatment modality.

[Evaluation of cervical conization in diagnosis and management of cervical intraepithelial neoplasia].

OBJECTIVE: To determine whether cervical conization can be instead of colposcopic multiple biopsies and to evaluate the clinical value of cervical conization in diagnosis and management of cervical intraepithelial neoplasia. METHODS: Fifty four patients with cervical intraepithelial neoplasia (CIN) diagnosed and treated at Peking Union Medical College hospital were retrospectively analyzed focusing on the comparative study in pathology between cerical conization and colposcopic multiple biopsies. RESULTS: Of the 54 patients, there was a correlation in pathology between cervical conization and colposcopic multiple biopsies in 22 cases (40.7%), but there was not much correspondence between cervical conization and colposcopic multiple biopsies in 32 cases (59.3%). In 31 cases conization performed for ruling out invasive cancer, there was a correlation in pathology between cervical conization and colposcopic multiple biopsies in only 13 cases (41.9%). After conization radical hysterectomy was performed in 2 cases (3.7%) because of early invasive carcinoma and simple hysterectomy was performed in 13 cases (24.1%) because of CINIII involved the gland and microinvasive carcinoma of the cervix. 39 cases (73.2%) were closely followed-up postoperatively with preservation of fertility. During the mean follow-up time of 18.3 months, no cytology abnormality was detected and 3 patients had pregnancy and delivery. Postoperative hemorrhage was main side effect for conization. CONCLUSIONS: Cervical conization plays a very important role in diagnosis and treatment of CIN and it can not be instead of colposcopic multiple biopsies. Cervical conization is a good choice for patient with CINIII and desired childbearing.

[Analysis of factors influencing the clinical in a frozen thawed embryo transfer program].

OBJECTIVE: To analyze the factors influencing the outcome in a frozen thawed embryo transfer (FTET) program. METHODS: Sixty FTET cycles performed in 53 cases from September 1997 to May 2000 were analyzed retrospectively. The related parameters were compared between the conceived and non-conceived cycles. RESULTS: One hundred and seventy eight out of 252 thawed embryos were survival. Embryo transfers were undergone in 57 cycles (53 patients), resulting in 17 pregnancies (30% per transfer or 28% per thawed cycle). There were no significant difference between conceived and non-conceived cycles in terms of the controlled ovarian hyperstimulation protocols, numbers of developed follicle and oocyte retrieved, fertilization rate, numbers of frozen embryo and the luteinizing hormone, estradiol, progesterone levels on human chorionic gonadotropin triggering day. The percentage of good quality embryo before freezing and after thawing and survival thawed embryos were higher in conceived cycles than those in non-conceived cycles (83.8% Vs 60.7%, 76.8% Vs 50.0%, 82.4% Vs 66.3%, respectively P < 0.05). CONCLUSIONS: Good quality of frozen-thawed embryos and the trilaminar sonographic pattern of endometrium may be reliable predictors of success in pregnancy.

Establishment of an animal model of chronic atrophic gastritis and a study on the factors inducing atrophy.

OBJECTIVE: To establish a rat model of chronic atrophic gastritis and explore the factors inducing atrophy. METHODS: In accordance with repeated orthogonal design of L8(2(7)), 60% alcohol and 20 mmol/L sodium deoxycholate (served as factor A), 0.05%-0.1% ammonia water (factor B), 0.05% indomethacin (factor C) were given, alone or in combination, to rats in three experiments for 3 months, 6 months or 9 months respectively. Then the rats were dissected, and their pathologic changes of the gastric mucosa were assessed. RESULTS: Typical signs of chronic atrophic gastritis (CAG) were found in all rats which were treated with factor A, B, C alone or in combination for 6 or 9 months. No significant difference of pathologic changes of gastric mucosa was found between the rats treated for 6 months and those for 9 months. No obvious CAG signs were found in the rats treated with factor A, B, C for 3 months. CONCLUSION: Sixty percent of alcohol, 20 mmol/L sodium deoxycholate, 0.05%-0.1% ammonia water and 0.05% indomethacin given to Sprague-Dawley rats for 6 months can successfully establish the animal model of CAG. Prolongation of the model-establishment time is not able to further facilitate the atrophy of gastric mucosa.

[A clinical analysis of 102 cases of chronic n-hexane intoxication].

OBJECTIVE: Chronic n-hexane exposure can result in n-hexane intoxication which is mainly characterized by a series of manifestations of peripheral nerve lesions. The disease was sometimes misdiagnosed as "unknown multiple peripheral neuropathy". 102 cases of n-hexane intoxication were investigated and reported in order to call attention to the disease. METHODS: All the 102 cases, male 30, female 72; age 17 - 29, mean (21 +/- 3) were n-hexane workers. During the hospitalization the following data were collected: medical history, neurological check up, electromyogram, electrocardiogram, serum biochemical analysis, ophthalmologic study and the metabolite of n-hexane- the urinary 2, 5-hexanedione level. The airborne n-hexane levels of the working environment were also measured. RESULTS: The average incubation period of the 102 cases was (8.3 +/- 2.6) months. The average course of the disease was (12.1 +/- 4.2) months. Airborne n-hexane concentration of the working places ranged from 188.0 to 7,848.6 mg/m(3). The most common prodromes were headache, anorexia, dizziness and weight loss. The main clinical manifestations of the disease were ascending abnormal sensation. Sensory loss and dyskinesia began from the distant parts of the limbs. The patient had reduction or disappearance of tendon reflexes, weakness and muscle atrophy in the limbs. Electromyography showed a neuropathic pattern, which was parallel to the clinical symptoms and signs. Delayed worsening of symptoms and signs after cessation of n-hexane exposure was observed in some cases. All the 102 cases recovered totally after treatment with Vitamin B, Chinese traditional medicine, physical therapy and training. CONCLUSION: The main lesion of the disease is multiple peripheral neuropathy. Diagnosis should be made according to the history of n-hexane exposure, the typical clinical manifestations of peripheral neuropathy as well as the neuropathic changes on electromyography. Therapeutic measures for peripheral neuropathy of other etiologies may be used and the prognosis is optimistic if correct diagnosis is made and further exposure stopped.

[Characterization of rpoB mutation in rifampin - resistant clinical Mycobacterium tuberculosis isolates from China].

OBJECTIVE: To elucidate the characterization of rpoB mutation in rifampin - resistant clinical isolates of Mycobacterium tuberculosis isolates from China. METHODS: 588 bp DNA fragment of rpoB gene including 81 bp code region (rifampin resistance determination region, RRDR) was sequenced. 242 strains of Mycobacterium tuberculosis, including 193 rifampin - resistant strains and 46 rifampin - susceptible strains and 3 artificially induced rifampin - resistant strains were sequenced. RESULTS: 89.1% (172/193) rifampin - resistant strains had rpoB gene mutations in RRDR, no mutation was found in rifampin - susceptible strains. 46.1% rifampin - resistant strains had mutations located at 531-Ser; 17.1% strains had mutations located at 526-His; Combinative mutation rate was 12.4%; 4 strains had synonymous mutations; No deletion or insertion mutation was found among all strains. High level rifampin resistant strains (250 microgram/ml rifampin resistant) had higher mutation frequency at 531 - Ser than low level rifampin resistant strains (50 microgram/ml rifampin resistant) (P < 0.05). CONCLUSIONS: About 90% rifampin resistant clinical strains of Mycobacterium tuberculosis from China had rpoB mutations; 531-Ser and 526 - His were the most common positions to be substituted, the added mutation rate was about 46%; High level rifampin resistant strains had a higher frequency of 531-Ser mutation than low level rifampin resistant strains; No insertion or deletion mutation was found in these specimens; DNA sequencing is significant in drug choosing for tuberculosis treatment.

[Combination of mycophenolate mofetil with cyclosporine A and methotrexate as acute GVHD prophylaxis after unrelated donor allogeneic bone marrow transplantation].

OBJECTIVE: To evaluate the efficacy and safety of mycophenolate mofetil (MMF) in combination with cyclosporine A (CsA) and methotrexate (MTX) for prevention of acute graft versus host disease (GVHD) after unrelated donor allogeneic bone marrow transplantation (allo-BMT). METHOD: Twelve cases of unrelated donor allo-BMT were evaluated in a single center trial. The acute GVHD was prevented with 1 g MMF daily in addition to CsA 3 mg x kg(-1) x (-1) and MTX 10 - 15 mg at post BMT day1, day3, day6 and day11. RESULTS: Acute GVHD was found in one case (Grade IV) at the seventh day and two cases (Grade II) at the tenth day and seventeenth day after BMT. These patients were treated with a combination of MMF, methyprednisolone and CsA. The common adverse hematologic events of MMF was leukopenia. CONCLUSION: The preliminary study showed that MMF could be used effectively and safely for prevention of acute GVHD in unrelated donor allo-BMT.
