Effect of corticosterone on beta-adrenoceptor density in rat skeletal muscle.

Corticosteroid hormones increase the density of beta-adrenoceptors in some tissues and may be able to prevent the anabolic effects of beta-agonists from becoming attenuated. The aim of this study was to find a suitable dose of corticosterone that would up-regulate beta2-adrenoceptors in skeletal muscle without arresting the animal's growth. Male rats were given five daily injections of corticosterone at 0, 6.25, 12.5, 25, or 50 mg/kg. The animals were far more sensitive to the catabolic effects of this steroid than female rats used in a previous study. There was no change in food intake, liver, heart, or soleus muscle mass, but corticosterone caused a dose-related decrease in weight gain, carcass weight, omental fat pad weight, and gastrocnemius/plantaris muscle mass (P < .01). From a regression of muscle mass against dose, we calculated that 4.4 mg x kg(-1) x d(-1) would be the largest dose of corticosterone that a male rat could tolerate without any catabolic effect in skeletal muscle. Corticosterone failed to increase beta-adrenoceptor density at any of the doses tested. We conclude that corticosterone treatment is unlikely to be effective at enhancing the growth response of male rats to beta-agonists.

CT-guided stereotactic biopsy of deep brain lesions: report of 310 cases.

OBJECTIVE: To evaluate the accuracy of CT-guided stereotactic biopsy in making correct pathological diagnosis and choosing corresponding management of brain tumors. METHODS: From 1991 to 1995, CT-guided stereotactic biopsy was performed in 310 patients with intracerebral lesions which were deep-seated or located in certain main functional areas. The patients were 198 men and 112 women. Their ages ranged from 4.5 to 70 years (average: 39.3 years). The lesions were located in the deep cerebrum (74 patients), the sellar area (62), the basal ganglion (51), the posterior part of the third ventricle (38), other intraventricleular area (21), the cerebellum (17) and the brain stem (9), and intracranial multiple lesions were found in 38 patients. RESULTS: Brain tumors were diagnosed pathologically in 266 patients (85.8%); inflammatory process in 25 (8.1%), other lesions in 8 (2.6%) and uncertain cases were 11 (3.6%). The overall positive rate of biopsy was 96.4% and the positive rate for brain tumor was 85.8%. Intracranial hematomas after biopsy were found in 5 patients (1.6%). There were no deaths induced by the biopsy or other serious complications. CONCLUSIONS: The results suggest that CT-guided stereotactic biopsy is a reliable method for histopathological diagnosis of brain tumors and it is also of great help in selecting appropriate management.

Detection of the E7 transform gene of human papilloma virus type 16 in human oral squamous cell carcinoma.

OBJECTIVE: To determine, with the use of polymerase chain reaction, the prevalence of human papillomavirus (HPV) 16 in 30 patients with primary oral squamous cell carcinoma (OSCC) and 30 healthy control patients. MATERIALS AND METHODS: DNA was extracted from freshly frozen tumor tissues of 30 patients with primary oral squamous cell carcinoma and from the oral mucosa of 30 controls. A pair of specific primers of the E7 early gene of HPV 16 were designed. PCR products were run by 1.5% agarose gel and the results of electrophoresis were photographed. RESULTS: HPV 16 was detected in 36.7% (11/30) of oral squamous cell carcinoma patients and 11.1% (4/30) of controls. CONCLUSIONS: HPV 16 has a significant association with oral squamous cell carcinoma. However, the role HPV 16 plays in the tumorigenesis of oral cancer and its clinical significance remain to be investigated.

[A clinical control study on the treatment of uterine leiomyoma with gonadotrophin releasing hormone agonist or mifepristone].

OBJECTIVE: To compare the results and side effects in treating uterine leiomyoma with gonadotrophin releasing hormone agonist (GnRH-a) or mifepristone. METHODS: 75 patients with uterine leiomyoma who had clinical symptoms and diagnosed by Bcan were divided into two groups. The GnRH-a group (30 patients) was treated by injection of GnRH-a 150 micrograms/day subcutaneously for three months, and the mifepristone group (45 patients) was treated by mifepristone 12.5 mg/day po for three months. RESULTS: The clinical symptoms improved obviously in both groups. The volume of leiomyoma reduced 20.0% or more in 90.0% (27/30) of the patients in GnRH-a group, while it was 91.1% (41/45) in mifepristone group. However, the recurrent rates were 40.0% and 17.8% in the 2 groups. CONCLUSION: It suggested that mifepristone is a more practical and hopeful drug in treating uterine leiomyoma.

[The effect of Epstein-Barr virus gene BHRF1 expression on the apoptotic resistance of nasopharyngeal carcinoma cells].

OBJECTIVE: To investigate the effect of EB virus gene BHRF1 expression on the apoptotic resistance of nasopharyngeal carcinoma cell. METHODS: Expression vector for BHRF1 was constructed and transfected into nsaopharyngeal carcinoma cell line, CNE2. The alteration of biologic behaviour of the transfected cells was tested after 60Co radiation. RESULTS: BHRF1 expression could inhibit the expression of proliferative cell nuclear antigen, redistribute cell cycle, decrease cell sensitivity to radiation, and enhance their survival ability. CONCLUSION: It is suggested that BHRF1 expression can increase the apoptotic resistance induced by radiation.

[Antisense to cyclin D1 inhibits the proliferation of glomerular mesangial cells].

OBJECTIVES: To investigate the role of Cyclin D1 in the proliferation of glomerular mesangial cells. METHODS: We first detected the expression of Cyclin D1 protein in mesangial cells by using immunohistochemical method, set up a retroviral vector mediated genetransfer system expressing Cyclin D1 antisense RNA with recombinating techniques, and assay the proliferating situation of genetransferred mesangial cells by MTT method and flow cytometry respectively. RESULTS: Cyclin D1 protein was expressed in the nuclei of cultured mesangial cells in vitro. The exgoenous gene was transducted into mesangial cells mediated by retrovirus and transcripted and expressed antisense RNA in high effiency. The response of antisense Cyclin D1 genetransferred mesangial cells to Endothelin-1 (OD570)(18 hr 0.105 +/- 0.009 vs 6 hr 0.096 +/- 0.013) was not significant (P > 0.05), but normal cells (18 hr 0.259 +/- 0.080 vs 6 hr 0.100 +/- 0.007) and only vector transferred cells (18 hr 0.278 +/- 0.020 vs 6 hr 0.097 +/- 0.008) were significant (P < 0.01). The progression of cell cycle was prevented from G1 to S phase by antisense Cyclin D1, and the percentage of cells of S phase in antisense Cyclin D1 genetransferred cells (12.70 +/- 3.39) was significantly reduced (P < 0.01) as compared with normal cells (29.98 +/- 3.26). CONCLUSIONS: Cyclin D1 is a key regulatory protein of G1 progressing of cell cycle of mesangial cells, and the cellular proliferation stimulated by Endothelin-1 can be suppressed prominently in antisense Cyclin D1 gene transducted mesangial cells.

[The study of radioresistance of BHRF1 gene].

In order to learn relationship between the expression of BHRF1 gene and radiative sensitivity of cells of nasopharyngeal carcinoma. High BHRF1 expressive vector was set up and transfected in the cell line of nasopharyngeal carcinoma, CNE2. Then the cells were radiated to test its ability of clone formation in nude mice. The results showed that the tumors formed by BHRF1-CNE2 cells were lighter before radiation and weighter after radiation (P < 0.01), and apoptotic index of BHRF1-CHE2 was less than control groups (P < 0.01). So, it suggests that the BHRF1 expression could prevent apoptosis by enhancing the resistibility of the tumor cells to radiation.

[A research for basic properties of distortion product otoacoustic emissions in normally hearing subjects].

With ILO-92 Otodynamics Analyzer, distortion product otoacoustic emissions (DPOAEs) at the 2 f1-f2 frequency were recorded from 86 normally hearing ears in response to four groups of different primary levels (L1 = L2 = 75 dB SPL; L1 = L2 = 65 dB SPL; L1 = L2 = 60 dB SPL; L1 = 65 dB SPL, L2 = 50 dB SPL). The average DPOAEs-gram demonstrated a bilobed contour with two peaks at approximately 1.5 kHz and 5 kHz and "notch" between 2.5 kHz and 3 kHz. Moreover, the contour of DPOAEs-gram had no obvious changes with sex, ear or primary stimulus levels differences. While the primary levels increased, the amplitude of DPOAEs gradually increased, which was about 55-65 dB SPL below the primary levels. At each frequency from 1-6 kHz, statistical analysis showed that there existed significantly different DPOAEs level under four groups of different primary stimulus levels.

[Effects of receptor-mediated endocytosis on macrophage membrane potential, cytosolic pH and lysosomal pH].

The effects of Con A, WGA, Zymosan A on membrane potential, cytosolic pH and lysosomal pH of cultured mouse macrophages were measured with fluorescence method. The results showed all the three kinds of ligands caused macrophage membrane potential hyperpolarization, cytosolic acidification and lysosomal alkalization, but reached their stabilized conditions at different times. The acidification of cytosl might inhibit further endocytosis. Alkalization of lysosomes was a basic factor for exocytosis of lysosomal contents. These receptor-mediated induced changes reflected cellular self-regulation and self-protection.

[Changes of prostaglandins in kidney tissue in gentamicin-induced acute renal tubular injury in rats].

OBJECTIVE: To examine the relationship between epidermal growth factor (EGF) and prostaglandins (PGs) in recovery of acute tubular necrosis (ATN) induced by gentamicin in rats and the changes of renal tissue PGs with EGF treatment. METHODS: Female wistar rats were divided into three groups; normal(NL, n = 7); GM-treated only (Group G, n = 20); GM 200 mg/kg, i.p. x 3 d; GM and EGF-treated (Group G + E, n = 19): EGF(20 micrograms) was given after last GM injection. [3H]thymidine incorporation rate (3HTdR) of renal tissue, serum creatinine concentration (Scr), renal levels of PGE2, 6-keto-PGF1 alpha, TXB2 were measured at day 1,4,8,12 after GM administration. RESULTS: [3H]thymidine incorporation rate of renal tissue in group G + E was significantly higher than that in group G after toxic injury. The histological lesions of group G + E was less severe than that in group G. 6-keto-PGF1 alpha in group G + E was increased significantly than that in group G, and renal TXB2 in group G + E was lower than that in group G. PGE2 and 6-keto-PGF1 alpha in group G + E was positively correlated with 3HTdR, respectively. CONCLUSIONS: (1) changes of renal prostaglandins may be related to the injury/proliferation of renal tubular epithelial cells in ATN. (2) Administration of exogenous EGF may enhance the release of PGE2 and 6-keto-PGF1 alpha of renal tissue and inhibit the synthesis of renal TXB2. The results indicate that effect of ameliorating recovery of renal tubular epithelial cells of EGF could be partly related to the changes of renal PGs.

[The changes of EG1 and EG2 positive eosinophils and their clinical significance in asthmatics].

OBJECTIVE: To evaluate the significance of EG1 or EG2 positive eosinophils in relating to the severity of asthma. METHOD: EDTA-anticoagulated venous blood were treated by FOG method, and FTTC-conjugated monoclonal antibodies were added. Flow cytometry technique was used. RESULT: EG1 and EG2 positive eosinophils increased significantly in patients with acute asthma (n = 29) as compared with control (n = 13) and COPD (n = 12), P < 0.05. There were significant correlation of the mean fluorescence intensity(MFI) between EG1, EG2 and the degree of the symptom. CONCLUSION: There is a significant increase of activated eosinophils in peripheral blood in asthmatics, the expression of EG1 and EG2 positive eosinophils correlated well with the severity of symptom in asthmatics. EG1 and EG2, as the marker for eosinophils activity, may be sensitive and specific in evaluating the severity of asthmatics symptom.

[Clinical observation of conservative surgery for adenomyosis].

OBJECTIVE: To study the clinical value of conservative surgery for adenomyosis in reducing symptoms and raising pregnancy rate. METHODS: 26 women with histologically proven adenomyosis who wanted to preserve their reproductive capacity had conservative surgery (adenomyomectomy or remove part lesion). The patient's average age was 35.8 years. The main complaints were dysmenorrhea and hypermenorrhea. Of the 22 married patients, 7 had primary infertility (31.8%), 10 had secondary infertility (45.5%) and 3 had habitual abortion (13.6%). RESULTS: 10 patients had adenomyoma, the lesion were completely removed. Sixteen patients had adenomyosis, the part of lesion were removed. The rate of the severe dysmenorrhea and hypermenorrhea after operation were significantly lower than that before (P < 0.001). After operation, 7 patients with adenomyoma wanted to have a baby, 5 patients conceived, of the total 9 pregnancies, 5 ended in term pregnancy; 14 patients with diffuse adenomyosis wanted to have a baby, 3 patients had 3 pregnancies. One of which ended in term pregnancy. The rate of pregnancy and term delivery in adenomyoma patients were significantly higher than that of adenomyosis patients (P = 0.03, P = 0.002). Among 26 patients, 3 relapsed. The average relapse time was 49.6 months. CONCLUSIONS: The conservative surgery for adenomyoma can reduce symptom and raise pregnancy rate significantly, it can be accepted by young women who want to preserve their reproductive capacity. Though the pregnancy rate of conservative surgery for diffused adenomyosis was low, it still has therapeutic value.

[Significance of resistance index and pulsatility index in differential diagnosis of breast neoplasm].

OBJECTIVE: To investigate the significance of the resistance index (RI) and pulsatility index (PI) in differential diagnosis between the benign and malignant breast neoplasms. METHODS: Forty-seven cases of malignant breast tumors, sixty-nine cases of fibroadenomas, seven cases of inflammatory masses and twenty-two cases of mammiplasia were demonstrated on color Doppler flow imaging (CDFI). The measurements of RI and PI of the four groups were recorded respectively and the comparison between benign and malignant neoplasms was made by applying Doppler quantitative analysis. RESULTS: The color flow was detected in 59.4% of benign breast neoplasms and in 89% of malignant ones respectively (chi 2-test, P < 0.001). There were significant difference between benign and malignant neoplasms on the RI and PI. The malignant neoplasms were characteristic of higher RI (> or = 0.70) and PI (> or = 1.30), which were of satisfactory value for differential diagnosis, amounting to 95.5% of accuracy. CONCLUSIONS: Combination of both color Doppler analysis and 2D-US might remarkably increase the sensitivity and specificity of differential diagnosis of breast neoplasms. Our results confirmed the significance of color Doppler flow imaging and Doppler quantitative analysis on the differential diagnosis of breast neoplasms.
