Recombination and disjunction in female germ cells of Drosophila depend on the germline activity of the gene sex-lethal.

Gametogenesis in males and females differs in many ways. An important difference in Drosophila is that recombination between homologous chromosomes occurs only in female meiosis. Here, we report that this process relies on the correct functioning of Sex-lethal (Sxl) which is primarily known as the master gene in somatic sex determination. Certain alleles of this gene (Sxl(fs)) disrupt the germline, but not the somatic function of Sxl and cause an arrest of germ cell development during cystocyte proliferation. Using dominant suppressor mutations that relieve this early block in Sxl(fs) mutant females, we discovered additional requirements of Sxl for normal meiotic differentiation of the oocyte. Females mutant for Sxl(fs) and carrying a suppressor become fertile, but pairing of homologous chromosomes and formation of chiasmata is severely perturbed, resulting in an almost complete lack of recombinants and a high incidence of non-disjunction events. Similar results were obtained when germline expression of wild-type Sxl was compromised by mutations in virilizer (vir), a positive regulator of Sxl. Ectopic expression of a Sxl transgene in premeiotic stages of male germline development, on the other hand, is not sufficient to allow recombination to take place, which suggests that Sxl does not have a discriminatory role in this female-specific process. We propose that Sxl performs at least two tasks in oogenesis: an 'early' function in formation of the egg chamber, and a 'late' function in progression of the meiotic cell cycle, suggesting that both events are coordinated by a common mechanism.

Role of reactive oxygen metabolites in organophosphate-bidrin-induced renal tubular cytotoxicity.

Due to low toxicity to nontarget species and rapid degradation after its application, organophosphate (OP) remains a widely used class of pesticide. Suicidal or accidental overdose of OP can result in acute tubular necrosis. Experimental evidence shows little correlation between the renal tubular necrosis and the degree of OP-induced acetylcholinesterase inhibition, the main mechanism of OP's toxicity, suggesting the involvement of alternate mechanisms. Since reactive oxygen species (ROS) are known mediators of many toxin-induced renal injuries, this study was conducted to investigate whether ROS play a role in Bidrin (BD)-induced renal tubular epithelial cell (LLC-PK1) toxicity. BD is an OP insecticide formulation with dicrotophos as the active ingredient. LLC-PK1 cell death, determined by lactate dehydrogenase (LDH) release (% of total), rose concentration- and time-dependently after exposure of the cells to 1000, 1250, 1500, 1750, and 2000 ppm of BD for 6, 12, 24, and 48 h. Antioxidants 2-methylaminochroman (2-MAC; 0.3 to 2.5 microM) and desferrioxamine (DFO; 0.25 to 2 mM) reduced cell damage induced by 1250 ppm of BD over a 24-h incubation in a concentration-related manner. The greatest reductions in % LDH were produced by DFO 2 mM and 2-MAC 2.5 microM, both significantly lower than BD alone. H2O2 levels (micromol/mg protein per h) were significantly elevated after exposure to 1250 ppm of BD. Significantly increased malondialdehyde formation (nmol/mg protein) compared with control was also found in BD-exposed cells indicating enhanced lipid peroxidation. Malondialdehyde generation was significantly suppressed by 2-MAC and DFO. These results demonstrate that the organophosphate BD can cause direct tubular cytotoxicity, and implicate, at least in part, a role for ROS and accompanying lipid peroxidation in cytotoxicity. Based on these direct in vitro findings, it is hypothesized that, besides hypotension that often accompanies OP intoxication, OP-induced oxidative stress at the tubular level may play a role in the pathogenesis of acute tubular necrosis.

The putative mechanistic basis for the modulatory role of endothelin-1 in the altered vascular tone induced by Trypanosoma cruzi.

Chagas' disease, caused by Trypanosoma cruzi, is an important cause of heart disease in Latin America. T. cruzi-induced microvascular compromise, in turn, is thought to play a major role in chagasic heart disease. Previous in vitro studies have implicated endothelin-1 (ET-1) as a potentially important vasomodulator present in increased levels in the supernatant of T. cruzi infected cultured human umbilical vein endothelial cells (HUVEC). Thus, the goal of the present investigation was to further evaluate the potentially important contribution of ET-1 to T. cruzi-induced alterations in vascular tone in vitro. Bioassay studies once again documented that exposure of isolated rat aortic rings to infected HUVEC supernatants elicited contractile responses whose steady-state magnitude was significantly greater than contractile responses elicited by exposure of aortic rings to uninfected HUVEC supernatants. Furthermore, the increased aortic contractility was significantly attenuated by the presence of the ET(A) subtype selective antagonists BMS-182,874 or BQ-123. Additionally, incubation of HUVEC with either verapamil or phosphoramidon prior to infection was also associated with reduced aortic contractility, upon exposure to the supernatant. Phosphoramidon, but not verapamil, produced a significant decrease in the measured ET-1 levels in the HUVEC supernatant. Consistent with the bioassay results, preincubation of Fura-2-loaded cultured rat aortic vascular smooth muscle cells with verapamil resulted in a near complete ablation of ET-1-induced transmembrane Ca2+ flux. Taken together, these data are consistent with the hypothesis that ET-1-induced vasoconstriction may play an important modulatory role in the vascular compromise characteristic of T. cruzi infection.

Induction of cyclooxygenase-2 messenger RNA after transient and permanent middle cerebral artery occlusion in rats: comparison with c-fos messenger RNA by using in situ hybridization.

OBJECT: Recently, two different cyclooxygenase (COX) genes, COX-1 and -2, were identified. In this study, topographic and chronological profiles of COX-2 messenger (m)RNA and c-fos mRNA expression were investigated using in situ hybridization after focal cerebral ischemia. METHODS: Rats undergoing permanent ischemia were decapitated at 30 and 90 minutes and at 2, 4, 8, and 24 hours after middle cerebral artery (MCA) occlusion, and rats undergoing transient ischemia were decapitated at 4, 8, and 24 hours after MCA occlusion that lasted for either 30 or 90 minutes. After brief transient MCA occlusion, c-fos mRNA was induced in the whole MCA territory, adjacent cortex (cingulate cortex), and distant brain regions such as the hippocampus and substantia nigra. In contrast, COX-2 mRNA was not induced in the ischemic core (lateral striatum) but only in the penumbral area (MCA cortex). Long transient and permanent MCA occlusion did not induce c-fos and COX-2 mRNAs in the ischemic core but strongly induced both mRNAs in the penumbral area (medial striatum and periphery of MCA cortex) and adjacent cortex (cingulate cortex). In brain regions distant from the ischemic territory, although c-fos mRNA was induced in the thalamus, substantia nigra, and hippocampus after extended transient and permanent occlusion, COX-2 mRNA was only induced in the bilateral hippocampi. The induction of COX-2 mRNA persisted in all locations even at 24 hours after MCA occlusion. CONCLUSIONS: The distribution of COX-2 mRNA induction was apparently different from that of c-fos mRNA after MCA occlusion. These results pertaining to COX-2 mRNA agree well with the previous observations of changes in prostaglandin metabolism induced by focal cerebral ischemia. However, whether this induction of the COX-2 gene contributes to the histopathological outcome of cerebral ischemia remains to be elucidated.

BHRF1 antisense oligonucleotide inhibits anti-apoptosis of nasopharyngeal carcinoma cells.

Epstein-Barr virus gene BHRF1 has homology with proto-oncogene bcl-2, which can protect cells from apoptosis, thus, it may play important roles in oncogenesis and affect treatment of EBV-related cancers. We used BHRF1 antisense oligonucleotide to block its expression in nasopharyngeal carcinoma cell line, CNE2. The results showed that after blocking by BHRF1 antisense oligonucleotide, CNE2 cells had higher S-phase cell percentage, more susceptibility to radiation with weaker ability of proliferation, colony forming efficiency and tumor development in nude mice after radiation. Our results suggest that BHRF1 antisense oligonucleotide could inhibit BHRF1 anti-apoptotic ability, and may contribute to the treatment of EBV-related cancers.

Application of cardiac gated magnetic resonance imaging in murine Chagas' disease.

To evaluate the role of gated cardiac magnetic imaging resonance (MRI) in Chagas' disease, we infected mice with Trypanosoma cruzi (Brazil strain). Two models were chosen for study, the CD1 and the inducible nitric oxide synthase knockout (NOS2-/-) mice. Infection of CD1 mice was associated with a significant increase in the right ventricular inner diameter (RVID) that was reversed in some mice by verapamil. Expression of cardiac NOS2 has been associated with myocardial dysfunction. Therefore, we evaluated chagasic cardiomyopathy in NOS2-/- and syngeneic wild type (WT) mice. Infected WT mice exhibited an increase in RVID in the acute phase (< 60 days postinfection) that was more marked during chronic infection (>100 days postinfection). Chronically infected NOS2-/- mice had an increase in RVID. The RVID in infected WT mice was greater than in NOS2-/- mice. These data demonstrate that MRI is a useful tool in the serial evaluation of the heart in murine Chagas' disease. In addition, it supports the notion that the NOS2-/-/NO pathway may contribute to the pathogenesis of murine chagasic cardiomyopathy.

Paclitaxel chemotherapy after autologous stem-cell transplantation and engraftment of hematopoietic cells transduced with a retrovirus containing the multidrug resistance complementary DNA (MDR1) in metastatic breast cancer patients.

The MDR1 multidrug resistance gene confers resistance to natural-product anticancer drugs including paclitaxel. We conducted a clinical gene therapy study to determine whether retroviral-mediated transfer of MDR1 in human hematopoietic cells would result in stable engraftment, and possibly expansion, of cells containing this gene after treatment with myelosuppressive doses of paclitaxel. Patients with metastatic breast cancer who achieved a complete or partial remission after standard chemotherapy were eligible for the study. Hematopoietic stem cells (HSCs) were collected by both peripheral blood apheresis and bone marrow harvest after mobilization with a single dose of cyclophosphamide (4 g/m2) and daily filgrastim therapy (10 microg/kg/day). After enrichment for CD34+ cells, one-third of each collection was incubated ex vivo for 72 h with a replication-incompetent retrovirus containing the MDR1 gene (G1MD) in the presence of stem-cell factor, interleukin 3, and interleukin 6. The remaining CD34+ cells were stored without further manipulation. All of the CD34+ cells were reinfused for hematopoietic rescue after conditioning chemotherapy with ifosfamide, carboplatin, and etoposide regimen. After hematopoietic recovery, patients received six cycles of paclitaxel (175 mg/m2 every 3 weeks). Bone marrow and serial peripheral blood samples were obtained and tested for the presence of the MDR1 transgene using a PCR assay. Six patients were enrolled in the study and four patients received infusion of genetically altered cells. The ex vivo transduction efficiency, estimated by the PCR assay, ranged from 0.1 to 0.5%. Three of the four patients demonstrated engraftment of cells containing the MDR1 transgene. The estimated percentage of granulocytes containing the MDR1 transgene ranged from a maximum of 9% of circulating nucleated cells down to the limit of detection of 0.01%. One patient remained positive for the MDR1 transgene throughout all six cycles of paclitaxel therapy, whereas the other 2 patients showed a decrease in the number of cells containing the transgene to undetectable levels. Despite the low level of engraftment of MDR1-marked cells, a correlation was observed between the relative number of granulocytes containing the MDR1 transgene and the granulocyte nadir after paclitaxel therapy. No adverse reactions to the genetic manipulation procedures were detected. Therefore, engraftment of human HSCs transduced with the MDR1 gene can be achieved. However, the overall transduction efficiency and stable engraftment of gene-modified HSCs must be improved before MDR1 gene therapy and in vivo selection with anticancer drugs can be reliably used to protect cancer patients from drug-related myelosuppression.

Exploration of the association between apoptosis and cleft palate.

OBJECTIVE: To explore the significance and effect of apoptosis in the development of cleft palate induced by dexamethasone (DEX). METHODS: Various doses of DEX (10-50 mg/kg) were injected intraperitoneally into Kunming mice on gestation day (GD) 12. The cranio-maxillary sections of samples were labeled by HRP-avidin-biotin-dUTP in situ end-labeling (ISEL) and were observed morphologically and quantitatively under a light microscope and electron microscope. RESULTS: ISEL positive cells were found in treatment groups. The means of sums and cleft palate incidence have a positive correlation with the doses of DEX (r = 0.9911/0.9978). CONCLUSION: A relationship exists between an increased level of apoptosis and the development of cleft palate induced by DEX.

[Comparison of endometrial histology and ultrastructure during different ovarian stimulation protocols in hamsters].

OBJECTIVE: To evaluate the influence of ovarian stimulation by pregnant mare's serum gonadotropin(PMSG) with or without gonadotropin releasing hormone agonist (GnRH-a) on endometrial histology and ultrastructure in hamsters. METHODS: Thirty hamsters were randomly allocated into 3 groups. (1) LA + PMSG group: Leuprolide acetate (LA) was given first for desensitizing the pituitary, then PMSG added for ovarian stimulation. (2) PMSG group was injected with PMSG only. (3) Control group was given with saline of same volume as groups LA + PMSG and PMSG on the same time schedule. Endometrial histology and ultrastructure were investigated by light microscope and electron microscope. RESULTS: Comparing to the controls, the changes of endometrial histology in group of LA + PMSG was not significantly different, but its ultrastructure had marked atterations: mitochondria swelling, undifferentiated crista and no basket-like structure. Delayed endometrial maturation was frequently found in group PMSG (P < 0.01), and the ultrastructure showed poorly developed mitochondria. CONCLUSION: The degree of endometrial maturation varied considerably with different ovarian stimulation protocols. Ovarian stimulation with GnRH-a was better than PMSG alone, because the protocol LA + PMSG, in hamsters, facilitated the formation of a physiologic endometrium. But the protocol with GnRH-a still had a negative effect on endometrial ultrastructure.

[Clinical discussion of the relationship between endometriosis and epithelial ovarian cancer].

OBJECTIVE: To analyse the relationship between endometriosis and epithelial ovarian cancer. METHODS: We analyzed retrospectively the 20 cases associated with endometriosis in 371 cases of epithelial ovarian cancer, and these 20 cases were also compared with the left 351 cases not associated with endometriosis(including endomtriod carcinoma 38 cases and clear cell carcinoma 39 cases). RESULTS: Among 20 cases, endomtriod carcinoma and clear cell carcinoma were most frequently associated with endometriosis, with 11 cases and 7 cases respectively, there was also 1 case of gland carcinoma and 1 case of serous papillary carcinoma. Cases of carcinomas associated with endometriosis had higher pathological grade and 5-year survival probability (Kaplan-Meier method) than those cases not associated with endometriosis. CONCLUSIONS: There is a close relationship between endometriosis and ovarian endomtriod carcinoma and clear cell carcinoma, the malignant transformation of endometriosis foci may be an important origin of them. Drug therapy is recommended for endomtriosis after the operation. In the treatment of endomtriod carcinoma and clear cell carcinoma, progestin can be used as an accessory method.

[The role and significance of PAF in the early pulmonary injury following burns combined with endotoxemia in rats].

OBJECTIVE: To investigate the role and significance of PAF in the early pulmonary injury following burns combined with endotoxemia in rats. METHODS: An animal model, in which rats were subjected to 20% TBSA III degree burns combined with intraperitoneal injection of lipopolysaccharide (LPS), was used for this experiment. The changes in the pulmonary morphology were observed by LM, EM and immunohistochemistry(IHC). The contents of PAF and TNF-alpha in the plasma were determined with quantitative analysis. PAF receptor antagonist(BN50739) was used to treat early pulmonary injury. RESULTS: PAF was one of the inflammatory mediators to precede TNF changes during burns combined with endotoxemia in the early stage of pulmonary injury. The contents of PAF in the plasma was positively correlated to the extent of pulmonary injury, which may be alleviated after injection of PAF receptor antagonist (BN50739). CONCLUSION: The results suggest that PAF plays an very important pathogenic role in the early lung injury resulted from burns combined with endotoxemia.

[High-level expression of 6B11 ovarian carcinoma anti-idiotypic antibody scFv genes in E. Coli].

OBJECTIVE: To express high-level 6B11 ovarian carcinoma anti-idiotypic antibody single chain Fv (scFv) genes in E. Coli. METHODS: Using PCR cloning technique, We cloned 6B11scFv genes into bacterial expression vector pKPL-3a. Recombinant plasmid vector pL-6B11scFv was transformed into E. Coli pop2136 with temperature control to produce proteins. Renaturated proteins were purified on DEAE-Sepharose Fast Flow ion exchange column with salt gradient elution. Immunoactivity was determined with ELISA and inhibition ELISA tests respectively. RESULTS: 6B11scFv genes were expressed as inclusion bodies in the cytoplasm. The purity of 6B11scFv turned out by SDS-PAGE was more than 95%. The expressed proteins after purification specifically reacted with anti-ovarian carcinoma monoclonal antibody COC166-9 and efficiently inhibited the reaction between primary ovarian carcinoma antigen OC166-9 and COC166-9. CONCLUSION: We successfully expressed high-level 6B11scFv genes in E. Coli. Expressed proteins showed pretty good immunoactivity.

[Mite-induced interleukin-5 and interleukin-4 expressions in peripheral blood mononuclear cells from asthmatic children with respiratory syncytial virus infection].

OBJECTIVE: To investigate the effects of respiratory syncytial virus(RSV) infection on mite-induced interleukin-5 and interleukin-4 expressions in cultures of PBMC from asthmatic children and their correlation with asthmatic symptoms. METHODS: IL-5 and IL-4 expressions in mite (D. Pteronyssinus)-stimulated PBMC were assessed ex vivo by ELISA and reverse transcription-polymerase chain reaction (RT-PCR) in 16 allergic asthmatic children with RSV infection(A) and 14 patients without RSV infection(B), as well as 10 normal controls(C) (5.1 +/- 2.4 yrs). Meanwhile their correlation with serum ECP or current wheezing was analysed in patients after RSV infection. RESULTS: (1) Stimulation of PBMC with mite antigens induced significantly higher IL-4 (34 +/- 24) ng/L vs (8 +/- 5) ng/L, (P < 0.01) and IL-5 (26 +/- 15) ng/L vs (6 +/- 5) ng/L release in allergic patients(B) as compared with healthy controls(C). The release was in a mite dose-dependent manner. Meanwhile, IL-5 level in group A (A) was the highest (73 +/- 47) ng/L, P < 0.05 compared with (B)). Similar results were obtained with IL-5 mRNA expressions. (2) Serum ECP level was significantly higher in group A (73 +/- 47) micrograms/L than in group C (18 +/- 12) micrograms/L, P < 0.01. There was a significant correlation between IL-5 release and ECP levels (r = 0.49, P < 0.05). (3) Mite-induced IL-5 release in PBMC was significantly increased in the group with wheezing symptom (71 +/- 40) ng/L compared with the asymptomatic group (34 +/- 29) ng/L, (P < 0.05). CONCLUSIONS: Our findings indicated that during RSV infection, mite-induced IL-5 expression in PBMC of asthmatic children was up-regulated, which may enhance activation of eosinophils and precipitate or exacerbate asthma symptoms.

[A preliminary study on dental-manpower forecasting model of Miyun County in Beijing].

OBJECTIVE: To explore the dental-manpower forecasting model of Chinese rural region and provide references for Chinese dental-manpower researches. METHODS: Chose rural Miyun County in Beijing as a sample, according to the need-based and demand-weighted forecasting method, a protocol WHO-CH model and corresponding JWG-6-M package developed by authors were used to calculate the present and future need and demand of dental-manpower in Miyun County. Further predications were also calculated on the effects of four modeling parameters to the demand of dental manpower. RESULTS: The present need and demand of oral care personnel for Miyun were 114.5 and 29.1 respectively. At present, Miyun has 43 oral care providers who can satisfy the demand but not the need. The change of oral health demand had a major effect on the forecast of the manpower. CONCLUSION: Dental-manpower planning should consider the need as a prime factor but must be modified by the demand. It was suggested that corresponding factors of oral care personnel need to be discussed further.

[Comparative study of the regulation of kidney tonic and promoting blood circulation recipe on apoptosis in T cells of old rats].

OBJECTIVE: To investigate the mechanism responsible for the observed reduction in T cell function of the Kidney Deficiency Syndrome. METHODS: After 18th activation with anti-CD3 mAb, the qualitative and quantitative analysis of apoptoses in T cells were performed by using DNA gel electrophoresis, TdT-mediated dUTP nick end labelling (TUNEL) and flow cytometry. RESULTS: The percentage of apoptotic cells was 47.0% in the old and 22.2% in the young group (P < 0.01). In the Kidney tonic group, the mean percentage of apoptotic cells was 31.2%, significantly lower than that in the aged group (P < 0.05). The mean percentage of apoptotic cells in the promoting blood circulation group was 43.3% and no significant difference was seen in the percentages of apoptotoic T cells from the promoting blood circulation group compared with the old group. CONCLUSION: Activation-induced excessive apoptosis of T cells is one of the mechanisms elucidating decreased T cell function of the Kidney Deficiency Syndrome and aging, the retardation of immune senescence by Kidney tonic recipe could be accounted for by down-regulation of activation-induced apoptosis in T cells.

[A clinical research for eosinophilic meningoencephalitis caused by angiostrongyliasis].

OBJECTIVE: To improve the awareness of angiostrongyliasis. METHODS: The clinical and laboratory data as well as the epidemiological informations of 18 patients with eosinophilic meningoencephalitis caused by Angiostrongylus cantonensis were analyzed. RESULTS: All the patients had a history of eating raw fresh water snail (Ampularium canaliculatus) before the onset of the disease. Incubation period ranged from 1 to 25 days. The major symptoms of the patients were severe headache and pain in the trunk and limbs. Peripheral blood eosinophilia was noted together with increased eosinophilic count in cerebrospinal fluid. With ELISA sera from patients were specifically IgG-antibody positive against Angiostrongylus cantonensis antigen, but were negative against other parasitic antigens such as Paragonimus westermani, Cysticercus, Cellulosae hominis, Echinococcus granulosus and Trichinella spiralis. Abnormal spot signal was found in 2 cases with brain magnetic resonance imaging. Electroencephalogram showed slow alpha rhythm. All the patients were effectively treated with good combined administration of albendazole and dexamethasone. CONCLUSION: Angiostrongyliasis is one of the common causes leading to eosinophilic meningoencephalitis. To our knowledge, Wenzhou is the first small outbreak site discovered of angiostrongyliasis in mainland China.
