Early tensile bond strengths of several enamel and dentin bonding systems.

Tensile bond strength tests are commonly used for the evaluation of adhesive dental materials. The majority of these tests are carried out after 24 h of storage in water. However, determination of the early tensile bond strength could be more important, especially in relation to gap formation between the cavity surface and the restorative material. This study investigated the tensile bond strengths of five enamel/dentin bonding systems and two experimental dentin bonding systems. Tensile bond strengths were obtained at one min, ten min, and 24 h after the resin composite was cured. Bond strengths at the early stages were always somewhat less than the 24-hour test results. For the enamel/dentin bonding systems, a significant difference was found between the enamel and dentin bond strengths at all time periods, except with Superbond D-liner and Liner Bond. The experimental group with glyceryl methacrylate as the primer produced a good 24-hour result (14.3 MPa), but the early bond strengths were no different from those in the non-primer-treated groups. It was concluded that this material may actually retard the polymerization of the bonding resin. Previous workers have suggested that a tensile bond strength in the order of 20 MPa is necessary for gap-free restorations to be obtained. Should this be the case, then all of the materials tested, from the aspect of early bond strength, lack the strength for prevention of gap formation, although Superbond D-liner and Liner Bond approached this hypothetical figure. These systems, Superbond D-liner and Liner Bond, also exhibit small differences between the enamel and dentin tensile bond strengths.

Effect of radiosurgery using Leksell gamma unit on metastatic brain tumor--autopsy case report.

A 77-year-old male presented with gradual development of right hemiparesis. Postcontrast computed tomography (CT) revealed enhanced lesions in the left parietal and right paraventricular areas. Stereotactic brain biopsy led to a histological diagnosis of metastatic carcinoma. The lesions were treated by radiosurgery using the Leksell gamma unit. CT showed reduced size of the lesions after irradiation. The primary lesion could not be found and he died from pneumonia. Autopsy 3 weeks after the irradiation disclosed histological tumor necrosis, with greater tumor cell kill effects toward the center of the tumor.

Effects of inhibitors of spermidine synthase and spermine synthase on polyamine synthesis in rat tissues.

Several inhibitors of aminopropyltransferases, developed recently in this laboratory, were tested for their specificity by measuring their effects on six enzyme activities related to polyamine biosynthesis and interconversion. Two of them, trans-4-methylcyclohexylamine (4MCHA) and N-(3-aminopropyl)cyclohexylamine (APCHA), selectively and potently inhibited the activities of spermidine synthase and spermine synthase, respectively. They were subjected to in vivo studies using rats. Oral administration of 4MCHA or APCHA dissolved in drinking water (0.02 and 0.1%) available ad lib. for a period of 10 days or 4 months caused a specific and marked decrease in spermidine or spermine in tissues (such as a 95% decrease) with a compensatory increase of spermine or spermidine, respectively, but without any observable change in the growth of the treated rats. Also, with extreme reduction of spermidine or spermine, when their sum was approximately constant, the activity of S-adenosyl-methionine decarboxylase in these tissues was enhanced significantly with no change in the activity of ornithine decarboxylase. These results suggested a separate role for spermidine or spermine in the in vivo enhancement of S-adenosylmethionine decarboxylase activity.

Williams syndrome. A middle-aged case of markedly delayed diagnosis.

A rare case of Williams syndrome diagnosed at the age of 41 is documented. The first subjective symptom was chest pain and the patient displayed many other features in addition to severe supravalvular aortic stenosis with a systolic gradient of 60 mmHg. The stenotic lesion had an area of 0.5 cm2, and was associated with dilated and tortuous coronary arteries. Extended aortoplasty was successfully performed and the postoperative course has been excellent without any cardiac symptoms. In spite of the severe cardiac lesions, this case had been largely asymptomatic and presented unusual features related to the diagnosis and management of this syndrome in an adult. The pattern of abnormalities found in this case suggested problems in relation to the calcitonin/calcitonin gene related peptide (CGRP) and the elastin gene occurring in embryonic organogenesis.

Tensile bond strength and curing gap formation of a dentin bonding resin.

Tensile bond strength and contraction gap tests were carried out using a new dentin bonding resin in association with various dentin conditioning and priming materials in vitro. For the tensile bond test, significant differences were found between the nonconditioned and conditioned dentin, and the primed and non-primed dentin surfaces. The strongest bond was obtained using a combination of phosphoric acid conditioning and priming with HEMA in glutaraldehyde at 151.3 kgf/cm2. Gap test results showed the presence of gaps in all of the test groups, but with variation in the extent of gap formation. A not significant, inverse correlation was determined between the tensile bond strength and gap tests (R = -0.65). The new bonding resin obtained good bond strengths in all cases, but high bond strengths can only be obtained with a combination of etching and priming. Similarly, the smallest gaps were associated with the etched and primed groups.

[Study on dental cement. Part 4. Protective effect of a newly designed light-cured varnish on the vulnerability to water of glass polyalkenoate cement restorations].

The protective effect of a newly designed light-cured varnish on the vulnerability to water of two kinds of glass polyalkenoate cement restorations was examined by determining the penetration depth of Ag ion into the set cement on the SEM pictures in comparison with those of the conventional varnish and a light-cured resin bonding agent. The protective effect of the experimental light-cured varnish was perfect, while those of the conventional varnish and the light-cured resin bonding agent were inferior to that.

Effect of aging on dentin bonding.

The purpose of this paper was to evaluate the effect of aging of dentin on the tensile bond strengths of four dentin bonding systems using proximal dentin of premolars extracted from young (aged from 9-21 years) and old (aged from 42-64 years) patients. Clearfil Linerbond System and Super Bond D-Liner showed higher bond strengths to both young and old teeth than those of Clearfil Photo Bond and Resto Bond 3. However, similar bond strengths to both young and old teeth for all four products were obtained despite the effects of aging. Furthermore, a SEM study of the site of bond failure revealed the persistence of the resin impregnated layer on the dentin surface, in spite of the difference of bonding system and dentin, indicating that adhesive failure tended to occur at the interface between the bonding resin and the resin impregnated layer.

The long term durability of bond strengths to dentin.

The aim of this study was to investigate the durability, throughout one year, of tensile bond strengths (TBS) to bovine dentin using various commercial and experimental bonding systems. Specimens were stored in a controlled solution of ion-exchanged water containing plaster chips and sodium azide. From the results it was concluded that the changes in TBS were not uniform over time, but a significant decrease was usually observed. For Super Bond D-liner and KB-100, the TBS were the highest and exhibited remarkable stability over the test period. The mode of fracture was noted to vary depending on the treatment system used, and was independent of TBS. Generally, the fracture mode tended to show increases in adhesive/cohesive failures within the resin over time. Super Bond D-liner always exhibited adhesive type failure at the tooth interface, and later involved failure in the hybrid layer. KB-100 showed very little change in failure over one year, being usually adhesive between bonding resin and resin composite. The results from this study indicate the need to carry out durability studies for the basic evaluation of all bonding systems. It was shown that the use of a controlled storage solution is important.

Remineralization of etched dentin.

To simulate a clinical cavity wall with the inner layer of carious dentin preserved, an artificially demineralized layer was made on the axial wall class V cavities of monkeys by a quick decalcifying solution. The cavities were then restored with a chemically adhesive composite resin after etching with a 40% phosphoric acid jelly etchant. Comparison of the Knoop hardness, the calcium concentration, and the scanning electron microscope observation of the dentin at the axial wall revealed that the dentin, demineralized by etching, was completely remineralized after 4 months.

Dentin thickness, hardness, and Ca-concentration vs bond strength of dentin adhesives.

The relationship between the tensile bond strength (TBS) and three dentin characteristics: remaining dentin thickness (RDT), Ca-concentration, and hardness, were investigated. Sixty-two extracted bovine incisors, divided into four groups, were prepared using 600-grit SiC paper to create flat dentinal surfaces. The materials tested in this study consisted of three commercially available and one experimental dentin bonding systems. The TBS and modes of failure were determined after storage for 24 h in 37 degrees C water. Then RDT, Ca-concentration, and hardness were measured. Data were statistically compared with TBS of each group. The mean bond strength of the commercial materials ranged from 2.5 +/- 0.9 MPa to 7.7 +/- 2.8 MPa. Correlations with Ca-concentration and RDT varied, depending on the product. The bond strength of the experimental material was 9.2 +/- 4.4 MPa and significantly correlated with RDT and hardness. With two of the dentin bonding agents, specimens showed a high number of cohesive fractures in dentin, 40% to 82%. It was concluded that the variability of TBS was influenced by dentinal characteristics in three of the four materials tested.

Enzyme labeling in steroid enzyme immunoassays. The p-nitrophenyl ester method for alkaline phosphatase and glucose oxidase labelings.

The p-nitrophenyl ester method was assessed as an enzyme labeling technique. The active ester of a carboxylated testosterone derivative was treated with alkaline phosphatase and glucose oxidase to give labeled antigens, using various molar ratios of steroid to enzyme. Satisfactory immunoreactivities with an anti-testosterone antibody in an enzyme immunoassay system were obtained with the labeled antigens prepared at pH 8.5 by the use of molar ratios higher than 30 and 10, respectively, in the alkaline phosphatase and glucose oxidase labelings.

Use of the thermostable glucose-6-phosphate dehydrogenase as a label enzyme in steroid enzyme immunoassays.

A thermostable glucose-6-phosphate dehydrogenase was assessed as a label enzyme in a testosterone enzyme immunoassay system. Enzyme labeling of a steroid with the enzyme was carried out by the N-succinimidyl ester method. A dose-response curve with a satisfactory sensitivity could be obtained by the use of the enzyme-labeled antigen. This enzyme should be useful in hapten enzyme immunoassays because of its relatively high stability.

Effect of aging and caries on dentin permeability.

Extracted human teeth were collected from young (20-28 yrs) versus old (45-69 yrs) patients. The teeth were divided into carious and noncarious groups. Slabs were created from the mid-coronal occlusal dentin. Carious lesions were excavated and the smear layers on both normal and excavated carious dentin were removed with 37% phosphoric acid (1 min). The permeability (hydraulic conductance) of old normal dentin was only 20% of that obtained in young normal dentin but all of the specimens were permeable. Young carious dentin was only 14% as permeable as young normal dentin and only 1 out of 7 specimens was not permeable. All 7 specimens of old carious dentin were impermeable. Scanning electron microscopy of old and carious dentin exhibited far more intratubular crystals than normal dentin, providing a structural basis for the functional observation.

Effect of acid etching on the dental pulp in adhesive composite restorations.

Pulpal response to marginal enamel etching or both enamel and dentine etching with 37% phosphoric acid for adhesive composite restorations was studied in the teeth of the Japanese monkey (Macaca fuscata). Dentine etching irritated the pulp and caused moderate to severe initial changes along the odontoblastic layer at 3 days. When no bacteria were present along the cavity walls, the subsequent pulpal reactions at 30 and 90 days decreased with time and large amounts of irritation dentine were formed. These inflammatory reactions were less than those caused by enamel etching only, or zinc oxide/eugenol cement. Significant correlation (P less than 0.01) was found between the intensity of the inflammatory reactions and the degree of bacterial infection. The pulpal irritation caused by acid etching of dentine for composite restorations is transitory only when the fillings have a good marginal seal and wall adaptation.

Studies on newly designed tooth surface treatments for adhesive composite restoration.

Recently, problems have occurred frequently such as cervical and exposed dentin surface lesions or root caries and subsequent hypersensitivity because of further increase of the elderly population. This study was projected to device a proper and simple treatment and materials for the adhesive composite restoration for such lesions. The 10-20Ca (10% citric acid containing 20% calcium chloride) solution was found to be a comparatively mild tooth surface conditioning agent and to be effective with a single application to both dentin and enamel. In addition, the use of the SA primer (3% N-methacryloyl 5-aminosalicylic acid in 80% ethanol) and the LVR (visible light-cured, 33% microfilled low viscous Bis-GMA resin) dramatically improved the adhesion and adaptability of the composite restoration in the saucer cavity at the cervical area.

[Prophylactic effect of ulinastatin on cisplatin-induced renal disorders in lung cancer patients].

In order to determine whether ulinastatin (US) can prevent renal dysfunction due to cisplatin (CDDP) or not, we comparatively studied serum BUN, serum creatinine (Cr), creatinine clearance (Ccr), urine gamma-glutamyl transpeptidase (gamma-GTP) excretion, urine N-acetyl-beta-D-glucosaminidase (NAG) excretion, fractional excretion of Na (FENa) and urine beta 2-microglobulin excretion between one group of 10 patients receiving single treatment with CDDP and another group of 10 patients receiving combination treatment with CDDP+US, in 20 subjects with primary lung cancer. In the US combination treatment group, urine gamma-GTP excretion, urine NAG excretion, and FENa elevation were significantly inhibited. Urine beta 2-microglobulin excretion was inhibited. In summary, US was considered to have a preventive effect against renal toxicity due to CDDP.
