Heparin-binding growth factor type one and platelet-derived growth factor are required for the optimal expression of cell surface low density lipoprotein receptor binding activity in human adult arterial smooth muscle cells.

Purified heparin-binding growth factor-1 (HBGF-1) stimulated low density lipoprotein binding, internalization, and degradation in isolated human adult arterial smooth muscle cells. Exposure of quiescent cells to HBGF-1 in serum-free, defined medium increased both low density lipoprotein (LDL) receptor activity and de novo cholesterol biosynthesis. Both events preceded the onset of DNA synthesis by 6 to 9 h. HBGF-1 acted additively with platelet-derived growth factor (PDGF) to maximally stimulate cell surface LDL receptor binding activity and DNA synthesis in the smooth muscle cells. The presence of LDL was required for maximal mitogenic activity of HBGF-1 and PDGF. In the presence of LDL, growth factor-stimulated, proliferating human smooth muscle cells accumulated cholesterol ester and triglycerides. The results suggest that HBGF-1, PDGF, and LDL act together to promote the maximal proliferation of smooth muscle cells in culture. Chronic exposure to the three growth promoters may contribute to the smooth muscle cell hyperplasia and lipid accumulation observed in atherosclerotic lesions.

Comparative endocrinology-paracrinology-autocrinology of human adult large vessel endothelial and smooth muscle cells.

Endothelial and smooth muscle cells were isolated from human adult large blood vessels to compare their proliferative response to hormones and growth factors. Neural extracts and the medium from differentiated hepatoma cells were used as concentrated sources of required hormones and growth factors that supported both cell types. Active hormones and growth factors were identified from the neural extracts and hepatoma medium by substitution or direct isolation and biochemical characterization. Epidermal growth factor, lipoproteins, and heparin-binding growth factors elicited growth-stimulatory effects on both endothelial and smooth muscle cells. Both types of human vascular cells displayed 7600 to 8600 specific heparin-binding growth factor receptors per cell with a similar and apparent dissociation constant (Kd) of 200 to 250 pM. Heparin modified the response of both endothelial and smooth muscle cells to heparin-binding growth factors dependent on the type of heparin-binding growth factor and amount of heparinlike material present. In addition, heparin exerted a growth factor-independent inhibition of smooth muscle cell proliferation. Platelet-derived growth factor, insulinlike growth factors, and glucocorticoid specifically supported proliferation of smooth muscle cells with no apparent effect on endothelial cell proliferation. Growth-factorlike proteinase inhibitors had an impact specifically on endothelial cell proliferation. Transforming growth factor beta was a specific inhibitor of endothelial cells, but had a positive effect on smooth muscle cell proliferation. The results provide a framework for differential control of the two vascular cell types at normal and atherosclerotic blood vessel sites by the balance among positive and negative effectors of endocrine, paracrine and autocrine origin.

Gallium-67 scintigraphy in patients with hemochromatosis treated by deferoxamine.

Gallium scintigraphy was performed as an aid for determining the presence or absence of malignant neoplasm in two patients with hemochromatosis treated by deferoxamine. However, gallium scan images could not be obtained. So gallium scintigraphy was performed once more to investigate the cause of low activity. Both patients had heavy urinary excretion of gallium in the first 24 hrs after the injection, and activity was very low on the day of examination. This phenomenon may be attributed to the effect of deferoxamine which is highly bound to the gallium.

Biodistribution of N-isopropyl-p-iodoamphetamine in mice.

Biodistribution of N-isopropyl-p-iodoamphetamine (IMP) was evaluated in mice. After synthesis of IMP and exchange labeling with radioactive iodine, IMP was injected intravenously in male ddY mice. Activity in the brain reached 8.0 (%dose/g), at 10 min after injection and it was almost constant till 120 min. Activities in the lung and heart were high just after injection, decreased rapidly and was almost constant from 30 to 120 min. Activity in the liver increased slowly compared with other organs, and reached peak level at 60 min. In autoradiography, almost the same activity in the spinal cord as the brain was observed. Uptake and excretion of IMP were shown in kidneys and stomach, which may be the main route of the excretion of IMP and their metabolites. We concluded IMP showed good uptake in the central nervous system.

Induction of lymph follicle formation with endotoxin lipopolysaccharide in the draining lymph node of athymic nude mice.

Formation of lymph follicles in draining popliteal lymph nodes in athymic nude mice and their phenotypically normal littermates (PN mice) was investigated after footpad injection with either endotoxin lipopolysaccharide (LPS) or phytohemagglutinin in soluble and precipitated forms (PHA). The dose of LPS injected ranged from 10 to 100 micrograms, and that of PHA was 50 micrograms. After any dose of LPS, draining nodes of nude mice produced new lymph follicles in the peripheral cortex outside pre-existing follicles, though the number of follicles induced was somewhat less than in the case of PN mice treated with LPS. After injection of PHA in soluble or precipitated form, PN mice developed a significant number of new follicles in the draining nodes, but nude mice failed to do so. The present results are consistent with the view that nude mice have the ability to develop lymph follicles by way of a thymus-independent mechanism in response to exogenous stimuli.

Phorbol ester reduces number of heparin-binding growth-factor receptors in human adult endothelial cells.

Heparin-binding growth factors (HBGF) are essential and key mitogens for human adult large vessel endothelial cells. At 170 pg/ml, the tumor promoter 12-O-tetradecanoylphorbol-13-acetate (TPA) caused a 50% inhibition of heparin-binding growth factor type one (HBGF-1)-stimulated DNA synthesis in human adult large vessel endothelial cells. TPA at 1 ng/ml completely inhibited HBGF-1-stimulated proliferation. TPA at 5 ng/ml reduced specific HBGF-1 receptor sites from 6600 per cell to 3200 per cell without affecting receptor affinity. Since phorbol esters are potent activators of protein kinase C, desensitizes both animal capillary and human adult large vessel endothelial cells to the mitogenic effects of HBGF by down-regulation of specific HBGF receptors.--HOSHI, H; KAN, M.; MIOH, H.; CHEN J.-K.; McKEEHAN, W. Tc-99m hexamethylpropylene-amine oxine (HM-PAO) uptake in a bone metastasis.

Uptake of Tc-99m Hexamethylpropylene-amine Oxine (HM-PAO) was seen in bone metastases from carcinoma of the lung. The uptake was prominent when compared to Tc-99m MDP, I-123 IMP, and Ga-67 citrate. Brain imaging with Tc-99m HM-PAO and N-isopropyl-p-[I-123] iodoamphetamine (IMP) is now frequently performed. Uptake of these agents has been reported in brain tumors and melanomas. In this report, uptake of Tc-99m HM-PAO in a metastatic lesion in bone is discussed.

Unusual uptake and retention of I-123 IMP in brain tumors.

Ten patients with unusual uptake and retention of I-123 IMP in their brain tumors were studied. Of these ten patients, five had meningiomas and the remaining five had glioblastoma, malignant astrocytoma, malignant lymphoma, metastatic brain tumor, and cellular blue nevus. Six tumors (five meningiomas and the glioblastoma) showed short term retention of I-123 IMP. Three tumors (the malignant astrocytoma, the malignant lymphoma, and the metastatic brain tumor) showed a delayed increase of radioactivity and the cellular blue nevus showed a gradual increase and long term retention of the tracer.

[Measurement of regional cerebral blood flow by 133Xe inhalation method in patients with ruptured aneurysm].

Regional cerebral blood flow (rCBF) was measured in 20 patients with ruptured aneurysm. rCBF images are obtained by 133Xe inhalation method using a ring type gamma camera (SET-020, Shimazu Co.) with high sensitivity collimator. Examinations are performed at acute stage (1-7 days after onset) and at subacute stage (8-14 days after onset). In acute stage, rCBF in the affected cerebral hemisphere has tendency to be low in 12 patients with angiospasm after the aneurysmal rupture comparing to those in patients without angiospasm. Low rCBF areas in cerebral hemisphere at acute stage were apt to become normal in subacute stage. rCBF measurement by 133Xe inhalation method seemed to be an useful method to evaluate rCBF in patients with ruptured aneurysm.

Dynamic SPECT with iodine-123 IMP in meningiomas.

Dynamic studies of single photon emission computed tomography (SPECT) were performed with intravenous administration of N-isopropyl-p-[123I]iodoamphetamine ([123I]IMP) in 12 patients with meningiomas. Their vascularities were also assessed by angiography. Regions of interest (ROIs) were placed over the lesion and the contralateral presumed normal area, the symmetrical one if possible. Time-activity curves (TACs) were then constructed and the counting-rate ratio for the lesion to the contralateral presumed normal area (L/N ratio) was calculated in the first image of the dynamic SPECT. TACs were classified into three types: type I, an initial high uptake and rapid washout type, type II, an initial high uptake and slow washout one, and type III, an initial low uptake one. L/N ratios of angioblastic meningiomas were over 300%, while those of many fibroblastic ones were under 100%, at most 105%. We compared L/N ratios with angiographic vascularities and histologic types and conclude that dynamic SPECT with [123I]IMP is the examination of choice for evaluating the vascularity of lesions.
