Spontaneous architectural organization of mammalian epidermis from random cell packing.

The cells of the stratum corneum in the epidermis of some mammalian species are precisely stacked in columns in a honeycomb fashion. The epidermis constantly loses surface cells, which are replaced by basal cells that have differentiated during migration to the surface. The path of this migration is seen as precisely defined columns of cells that are in compressed Kelvin's tetrakaidecahedral form. We present a computer simulation of this architectural organization based on the assumption that the cells that migrate upward occupy less crowded regions. The simulation not only explained the mechanism by which the architecture is maintained during the process of cell replacement, but also showed that the architecture was spontaneously organized from initial cells supplied at random. Living organisms consist of self-organizing systems at various levels; however, self-organizing systems have been investigated mostly at the molecular level. The present computer simulation clarified the self-organizing system at the cellular level.

Intracellular hyperthermia for cancer using magnetite cationic liposomes: in vitro study.

'Magnetite cationic liposomes (MCL)' were developed as a means to generate intracellular hyperthermia. Affinity of the MCL to glioma cells was ten times higher than that of magnetite 'neutral' liposomes due to the electrostatic interaction based on the positive charge of the MCL. Heat generation of the MCL was studied using agar phantoms and small pellets of rat glioma cells. When a high-frequency magnetic field, 118 kHz, 384 Oe was applied to glioma cells in the presence of MCL, the glioma cell pellet of 80 microl (5.4 mm in diameter) was heated to over 43 degrees C and all the cells died after 40 min irradiation owing to the hyperthermic effect. The terminal temperature of the cell pellet was proportional to the pellet volume when other parameters were constant. It thus appears that the MCL can heat a tumor of more than 80 microl in volume to above 42 degrees C.

Cumulative incidence and prevalence of childhood autism in children in Japan.

BACKGROUND: An epidemiological survey of childhood autism as defined in ICD-10 Research Criteria was conducted in the northern part of Yokohama, Japan. METHOD: The routine health checkup for 18-month-old children served as the initial mass-screening, and all facilities which provide child care services function to detect all cases with childhood autism and refer them to the Yokohama Rehabilitation Centre. RESULTS: Cumulative incidence of childhood autism up to 5 years of age among the birth cohort of 1988, and prevalence on 1 January 1994, among residents born in 1988 were estimated RESULTS: Cumulative incidence and prevalence were 16.2 per 10,000 and 21.1 per 10,000, respectively. Children with high-functioning autism who had IQs of 70 and over constituted approximately half of all the children with childhood autism. CONCLUSION. It was confirmed through better detection of high-functioning cases that childhood autism in Japan is more common than formerly estimated.

Elevated serum levels of manganese superoxide dismutase in polymyositis and dermatomyositis.

We studied serum concentrations of manganese superoxide dismutase (Mn SOD) and copper-zinc superoxide dismutase (Cu/Zn SOD) in 22 patients with polymyositis and dermatomyositis (PM/DM), 87 patients with four types of muscular dystrophy, 20 patients with amyotrophic lateral sclerosis, and 15 patients with collagen vascular diseases (CVD). Serum levels of Mn SOD were increased only in the patients with PM/DM and CVD, and the elevation was more prominent in those with PM/DM. Levels of Cu/Zn SOD were slightly elevated in some patients with PM/DM and Duchenne muscular dystrophy. In patients with PM/DM, the change in Mn SOD levels corresponded to disease activity as closely as or more closely than those of creatine kinase. The results indicate that serum Mn SOD may be a useful clinical marker for PM/DM.

[Effect of (-)-N-[(S)-hexahydro-l-methyl-2, 6-dioxo-4-pyrimidinylcarbonyl]-L-histidyl-L-prolinamide (TA-0910), a new TRH analog, on plasma levels of TSH and thyroid hormones in rats].

The stimulatory effect of TA-0910 on the secretions of thyroid-stimulating hormone (TSH) and thyroid hormones was investigated in male and female rats. Single intravenous administration of TA-0910 at 8.3 nmol/body acutely elevated the plasma TSH level, with delayed and moderate increases of T3 and T4 in plasma. Similar increments of plasma TSH and thyroid hormones were observed when TRH was injected at the dose of 0.83 nmol/body. Oral administration of TA-0910 at 2.75 mumol/body was equally potent or slightly more potent to secrete TSH than TRH at 0.275 mumol/body. The elevated TSH by TA-0910 decreased to the control level within 2 hr after intravenous injection or within 6 hr after oral administration; on the other hand, the higher levels of the thyroid hormones were retained for up to 4 and 6 hr after intravenous and oral administration, respectively. These findings indicate that TA-0910 and TRH stimulate the secretion of TSH and thyroid hormones by a similar manner and that the TSH-secreting activity of TA-0910 is lower by an order of magnitude compared with that of TRH.

[Fas antigen is expressed in human diseased muscles, but does not link to apoptosis].

In an attempt to determine whether the Fas-Fas-ligand mediated apoptosis occurs in the pathologic process of human myopathies, we examined the expression of Fas antigen, Fas-ligand mRNA, and chromosomal DNA fragmentation in the muscle cells of several human muscle disorders. The Fas antigen of a whole molecule was expressed on the muscle fibers of patients with muscle wasting diseases. However, there was no evidence of an apoptotic process, nor Fas-ligand synthesis in the diseased muscle tissue. Therefore, the expression of Fas antigen on muscle fibers in diseased muscle might be related to unknown biological functions other than "apoptosis" in the process of muscle fiber injury.

Laparoscopic splenectomy aided by balloon occlusion of the splenic artery: report of a case.

Laparoscopic splenectomy often involves difficulties in patients with splenomegaly because of a poor visual field and distorted and enlarged vessels secondary to the enlarged spleen. For safe and efficient laparoscopic splenectomy, we performed balloon occlusion of the splenic artery with the interventional angiographic technique in a patient with hereditary spherocytosis and splenomegaly. This new approach may be useful in terms of safety and time savings, especially in patients with splenomegaly.

Prevention of posttransfusion hepatitis by screening with second-generation anti-HCV antibody and clinical features of HCV infection.

After adaptation of the second-generation anti-hepatitis C virus (HCV) test, the incidence of post-transfusion hepatitis (PTH) resulted in 5.2% (11/208), which was significantly lower than that 10.5% (22/209) demonstrated by screening donor blood for C100-3 antibody. The 11 cases of PTH, three were classified as definite PTH and the other were as suspected one. Of two cases with definite PTH and two cases with suspected PTH, their blood samples after transfusion became positive for HCV-RNA, and three cases of those showed a second peak of alanine aminotransferase (ALT) more than 4 weeks after operation. On the other hand, of seven cases containing one definite PTH, their blood samples after transfusion became negative for HCV-RNA, and five cases of those showed ALT peaks within 4 weeks after operation, and returned to normal levels of ALT thereafter. Moreover, in cases of definite PTH, the periods of surgery and anesthesia were longer and the volume of bleeding was much more during operation than in cases of suspected PTH, although the differences were not statistically significant. These findings suggested that cases of PTH include those of transient liver disease attributable to surgery as well as those of HCV infection. In 11 cases of PTH, we consider that 2 cases is true PTH, because these are HCV-RNA positive and have second peak more than 4 weeks after operation and 5 cases is questionable by clinical date. Thus new diagnostic criteria should have established.

Epidermal remodelling in psoriasis.

Psoriatic hyperproliferative epidermis is characterized by a regular elongation of rete ridges, accompanied by altered keratinization. Another notable finding is close positioning of the vasculature to the suprapapillary epidermis. These architectural/morphological changes are naturally described by a concept of epidermal remodelling based on decreased epidermal turnover time. The recently described positioning of stem cells to the tips of dermal papillae fits nicely with this concept.

Development of a target-directed magnetic resonance contrast agent using monoclonal antibody-conjugated magnetic particles.

We developed a novel magnetic resonance (MR) imaging contrast agent, MAb-magnetite, that was prepared by covalently linking polyethylene glycol-coated magnetite to a monoclonal antibody specific for a human glioma cell-surface antigen. When MAb-magnetite was injected intravenously into tumor-bearing nude mice at a dose of 100 mumol Fe/kg body weight, a 50% decrease in the T2 signal intensity of the tumor was observed, immediately following administration and continued for 48 h. Microscopic observation of the tumor tissue demonstrated localization of MAb-magnetite in cancer cells. These results suggest that MAb-magnetite is a promising agent for MR imaging of neoplasms.

[A case of MPO-ANCA related vasculitis with transient leukoencephalopathy and multiple cerebral hemorrhage].

Recently, it has become clear that anti-neutrophil cytoplasmic autoantibody (ANCA) can be detected in patients with idiopathic necrotizing and crescentic glomerulonephritis or necrotizing angitis, and can be used as a serological marker for vasculitic syndrome. Described is a 56-year-old woman who had peripheral neuropathy, intestinal hemorrhage, generalized tonic-clonic convulsions, during the course of a rapidly progressive glomerulonephritis syndrome due to myeloperoxidase (MPO)-ANCA related vasculitis. The cranial MRI showed transient leukoencephalopathy in the parieto-occipital lobes bilaterally. The following MRI revealed multiple cortical and subcortical hemorrhage in the left occipital and right temporal lobe. The neurologic examination showed a slight left hemiparesis and a right homonymous hemianopsia, but, she experienced no subjective symptoms. In the case of MPO-ANCA related vasculitis, there is the possibility of intracranial disorders seen as this case. Thus, MPO-ANCA in related vasculitis should be always considered as a differential diagnosis in cases of transient localized leukoencephalopathy or cortical-subcortical cerebral hemorrhage.

Decorating actin filaments with troponin T-I complexes and acceleration of their sliding movement on myosin molecules.

Actin filaments, when partially decorated with troponin T-I complexes, can slide faster on myosin heads than those with no decoration. Purified troponin T-I complexes bind to actins, and inhibit the actin activated myosin adenosine 5'-triphosphatase activity completely when the molar ratio of troponin T-I complex to actin is increased to 1 to 1. Those actin filaments decorated with troponin T-I complexes up to 20 to 50% of their molar ratio exhibit enhancement of the velocity of sliding on myosins up to 20% compared to those without such decoration. As the molar ratio of decoration further increases, the sliding velocity decreases. These results are consistent with the observation that even if some of actin monomers do not participate in the ATPase activity directly, they can interact with myosin heads and take part in the sliding movement.

Spontaneously immortalized adult mouse Schwann cells secrete autocrine and paracrine growth-promoting activities.

We established spontaneously immortalized Schwann cell lines from long-term cultures of adult mouse dorsal root ganglia and peripheral nerves. One of the cell lines, designated IMS32, responded to mitogenic stimuli by platelet-derived growth factor (PDGF)-BB, acidic and basic fibroblast growth factors (aFGF, bFGF), and transforming growth factors (TGF)-beta 1 and -beta 2, as determined by bromodeoxyuridine (BrdU) incorporation and double immunofluorescence for S100 and BrdU. Furthermore, conditioned media (CM) obtained from IMS32 cells showed mitogenic activity for both IMS32 cells and long-term cultured Schwann cells. Western blot analysis revealed TGF-beta-like molecule in the CM, and the activity was absorbed with anti-TGF-beta neutralizing antibody. Reverse transcription followed by polymerase chain reaction (RT-PCR) of IMS32 RNA revealed that these cells expressed TGF-beta 1, -beta 2, and -beta 3 transcripts. When rat pheochromocytoma PC12 cells were incubated with the CM, they developed neurite growth. Coculture of PC12 and IMS32 cells also showed neurite growth of PC12 cells. RNA transcripts of nerve growth factor (NGF), brain-derived neurotrophic factor (BDNF), neurotrophin-3 (NT3), ciliary neurotrophic factor (CNTF), and glial cell line-derived neurotrophic factor (GDNF) were detected from IMS32 cells by RT-PCR. In these, we sequenced the mouse GDNF cDNA coding region and observed 97% and 90% homologies to corresponding rat and human cDNA sequences, respectively. These results indicate that the immortalized Schwann cell line mitotically responds to various growth factors and secretes autocrine and paracrine growth-promoting activities in vitro.

Expression of p210bcr/abl by metallothionein promoter induced T-cell leukemia in transgenic mice.

The p210bcr/abl chimeric protein is considered to be implicated in the pathogenesis of Philadelphia chromosome-positive human leukemias. To investigate its biologic function in vivo, we generated transgenic mice expressing p210bcr/abl driven by the metallothionein enhancer/promoter. Two of six founder mice and the transgenic progeny developed leukemias several months after birth. In the leukemic tissues, the expression of the p210bcr/abl transgene product was detected and the increased tyrosine-phosphorylation of cellular proteins was observed. The expressed p210bcr/abl transgene product was shown to possess an enhanced kinase activity. The leukemic cells showed rearrangements in the T-cell receptor loci, indicating that the leukemic cells were monoclonal and committed to the T-cell lineage. Polymerase chain reaction analysis for tissue distribution of p210bcr/abl expression showed that, in the transgenic line that reproducibly developed leukemias, p210bcr/abl was expressed in the hematopoietic tissues such as thymus and spleen; on the other hand, in the transgenic lines that have not developed leukemias, p210bcr/abl expression was detected only in the nonhematopoietic tissues such as the brain and kidney. These results suggest that the tumorigenicity of the p210bcr/abl chimeric protein is restricted to the hematopoietic tissues in vivo and that an event enhancing p210bcr/abl expression contributed a proliferative advantage to hematopoietic precursor cells and eventually developed T-cell leukemia in transgenic mice.

Increased tyrosine-phosphorylation of 55KDa proteins in beta-actin/Tec transgenic mice.

Protein-tyrosine kinases are considered to play important roles in cell proliferation and differentiation. Tec is a cytoplasmic protein-tyrosine kinase expressed in liver and hematopoietic tissues. To better understand Tec function in vivo, we generated transgenic mice expressing tec driven by the cytomegarovirus enhancer and beta-actin promoter. Among six transgenic lines generated, a particular line, named 2-11, expressed tec transgene product more widely and abundantly than the other lines. In the tissues of 2-11, the kinase activity of Tec was enhanced in accordance with the high expression of tec transgene product. Interestingly, tyrosine-phosphorylation of approximately 55KDa proteins in the tissues was induced. These results suggest that cellular proteins of 55KDa might be potential substrates of Tec in vivo.

Small hepatocellular carcinoma of single nodular type: a specific reference to its surrounding cancerous area undetected radiologically and macroscopically.

A total of 128 surgically resected small hepatocellular carcinomas, measuring less than or equal to 3 cm in diameter, were studied by both macroscopic and histologic examinations. In 95 single nodular-type tumors of the 128 lesions, eight tumors were associated with the cancerous areas of well differentiated hepatocellular carcinoma around the nodule. These surrounding cancerous areas went undetected by both the preoperative radiological examinations and the gross findings of resected specimens. Based on the immunohistochemical findings, the labeling index, both of the proliferating cell nuclear antigen (PCNA) and of the Ki-67 in the surrounding cancerous areas, were lower than that of the main nodules but higher than in the nontumorous liver parenchyma in seven of eight cases. These results suggest that the main nodule was generated from the surrounding cancerous area, supporting the hypothesis of a stepwise progression of HCC. Even if the tumor seems to be a small and single nodular type, it is recommended that its surrounding areas should be closely examined and the surgical cutting margin should be made more than 1.0 cm away from the main nodule at hepatic resection.
