Isolation of porcine, canine and feline IgG has hitherto been achieved usually by DEAE-ion exchange chromatography and gel filtration. These procedures, however, are rather time-consuming, as they involve purification of IgG. Isolation of IgG by affinity chromatography on a column of protein A-Sepharose was attempted. IgG of all three animal species was very easily isolated with a high yield. In the absence of any other method that allows isolation of IgG of three animal species from a single column, the procedure proposed would be very useful.
A three-year-ten-month old boy with the Fukuyama type of congenital muscular dystrophy (FCMD), early onset of muscular hypotonia, severe mental retardation, a slow progressive course and a characteristic fundus appearance similar to that of gyrate atrophy is described. Hyperornithinaemia and iminoglycinuria were not noted. To our knowledge, this is the first reported case of FCMD associated with atypical gyrate atrophy of the choroid and retina.
By cloning the beta-galactosidase gene of Bacillus stearothermophilus IAM11001 (ATCC 8005) into Bacillus subtilis, enzyme production was enhanced 50 times. beta-Galactosidase could be purified to 80% homogeneity by incubating the cell extract of B. subtilis at 70 degrees C for 15 min, followed by centrifugation to remove the denatured proteins. Because of its heat stability and ease of production, beta-galactosidase is suitable for application in industrial processes.

X-irradiation administered to nude mice and the ensuing pulmonary metastasis of injected human tumor cells were investigated. When human tumor cells (5 X 10(5)) were injected into the tail vein of male adult nude mice 5 days after 3 Gy whole-body X-irradiation, the incidence of pulmonary metastasis was more frequent in the irradiated mice than in the nonirradiated controls. Platelet aggregation and fibrin deposition around the tumor cells were present in the capillaries of the lung in the irradiated mice while prominent neutrophilic infiltration was noted around the tumor cells in the nonirradiated controls. Fibrinolytic activity of the lung was decreased in the irradiated mice, while serum fibrinogen level was increased after irradiation. Natural killer cell activity was decreased after irradiation. We conclude from this study that increase in serum fibrinogen levels and decrease in fibrinolytic activity of the lung, as a target organ, together with decreased infiltration of neutrophils, may be related to the trapping and lodgement of tumor cells when nude mice are subjected to irradiation.
Effect of whole body X-irradiation on the spontaneous pulmonary metastasis of human cancer cells transplanted into adult nude mice was investigated. Human cancer cells were inoculated into footpads of adult nude mice following 3 Gy whole body X-irradiation. The incidence of pulmonary metastasis was increased in the irradiated mice. Cytotoxicity of the splenocytes, particularly adherent cells, was lower in the irradiated mice than in the nonirradiated mice. Histological examinations revealed decreased mononuclear cell infiltration around the primary tumor and pulmonary metastatic foci in the irradiated mice. The suppressive effect of cytotoxicity of the splenocytes by whole body X-irradiation may thus relate to ensuing metastasis both in the phase of release and intravasation from the primary tumor and in the phase of lodgement and proliferation in the target organ.
Human tumor cells were injected intravenously into nude mice following whole body X-irradiation and the number of platelets and the serum fibrinogen level in the blood were examined. Pulmonary metastasis was present in the irradiated mice but not in the non-irradiated mice. The number of platelets decreased rapidly after intravenous injection of the tumor cells and remained low in the irradiated mice, yet there was a recovery in the number in the non-irradiated mice. Serum fibrinogen level was increased in the irradiated mice and was decreased after intravenous injection in the irradiated mice, to a greater extent than seen in the non-irradiated mice. These findings may be related to trapping and lodgement of tumor cells in the target organ.
The role of lymphography for patients with malignant lymphomas involving Waldeyer's ring was retrospectively reviewed. The materials were 97 patients who were treated at Kyushu University Hospital from January, 1966 to December 1982. Lymphography was performed in 72 patients. Abnormal retroperitoneal nodes were detected only in 13 of them (18%). Six of the 7 patients (86%) with gastrointestinal involvement as well as Waldeyer's ring had positive lymphography, whereas only 6 of the 51 patients (12%) without gastrointestinal involvement had such findings. Although there was a significant difference in survival by cervical and inguinal lymph node status, no such discrepancy was seen between 41 stage II patients staged after lymphography and 16 stage II patients staged clinically. All these results suggest that lymphography is not always essential for patients with malignant lymphomas involving Waldeyer's ring.
An alanine carrier protein was isolated from membranes of the thermophilic bacterium PS3 using ion exchange column chromatography followed by high performance liquid chromatography with a hydroxylapatite column. The final preparation consisted of a single polypeptide, with Mr = 42,500, as estimated by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. A polarity index of 33% was calculated from the amino acid analysis. Proteoliposomes reconstituted with the purified alanine carrier carried out an active alanine transport driven by either an electrochemical potential difference of protons or that of sodium ions.

Cutaneous thermoreceptors recorded from the trigeminal ganglion of cats anesthetized with urethane showed a significant reduction in thermal sensitivity when compared with neuronal responses obtained from sodium pentobarbital-anesthetized cats. The results of this study indicate that a detailed analysis of primary afferent thermoreceptor response characteristics should consider the anesthesia employed.

Bacillus stearothermophilus IAM11001 produced three beta-galactosidases, beta-galactosidase I, II, and III (beta-gal I, II, and III), which are detectable by polyacrylamide (nondenatured) gel electrophoresis. By connecting restriction fragments of the chromosomal DNA to plasmid vectors, followed by transformation of Escherichia coli, two beta-galactosidase genes (bgaA and bgaB) located close to each other on the chromosome were isolated. Identification of the gene products and Southern hybridization analyses with a 2.7-kilobase-pair EcoRI fragment containing the bgaA gene as probe revealed that a single bgaA gene exists on the genome and that beta-gal II and beta-gal III consist of a common subunit (the bgaA gene product; molecular weight, 120,000), but differ in their assembly (beta-gal II is a dimer, and beta-gal III is a tetramer). The bgaB gene product (molecular weight, 70,000) in Bacillus subtilis harboring pHG5 (a hybrid plasmid consisting of pUB110 and a 2.9-kilobase-pair EcoRI fragment) was estimated to be the beta-gal I protein from its heat stability. Southern hybridization and immunological testing indicated that the two genes have no homology.

Galactose transport activity from Escherichia coli was solubilized with octyl glucoside, and reconstituted into liposomes made from soybean or E. coli lipid. Galactose counterflow in the proteoliposomes was inhibited by glucose, talose, 2-deoxygalactose and 6-deoxygalactose, confirming that it was due to GalP and not one of the other E. coli galactose transport systems.
