11 out of 13 N-[3-[5-nitrofuryl-(2)-propenylidene)]-benzhydrazides described by us showed an in vitro activity against T. vaginalis almost equivalent to or approaching that of the standard substances metronidazole and tinidazole. One compound was markedly more effective; two compounds exhibited much weaker activity than the two reference substances. In the model of the T. foetus infection of mice, only two compounds came close to the chemotherapeutic effect of tinidazole when administered orally. The other compounds were less effective. Metronidazole showed an activity 10 times weaker than that of tinidazole in this animal model. The in vitro efficacy of the most active substances a and d on T. foetus infection paralleled the bacteriostatic effect against different species of bacteria. In comparison, d was more effective than a against T. vaginalis, 2 Candida strains and M. tuberculosis. In the Ames test, 5 out of the 13 described N-[3-[5-nitrofuryl-(2)-propenylidene))]-benzhydrazides proved mutagenic in test strains TA 98 and TA 100; this was the case also in strain TA 1537 for the microbiologically most promising compound a. Because of liver damage observed in the test on toxicity, the substance was not taken up in clinical studies. It is interesting to note that these substances were not found to be mutagenic in the host-mediated assay. No signs of chromosome breaks were observed for substances a and n in the micronucleus test. The relevance of these findings was discussed. Statistical procedures were described for both the Ames test and the host-mediated assay.
Although there is doubt about the importance of rinsing the operative field with a solution containing locally acting antibiotics, it is frequently done. In this paper we tried to answer the question, wether intraoperative autotransfusion (IAT) with the Haemonetics Cell Saver is contraindicated during rinsing the operative area with locally acting antibiotics or vice versa. The measured serum concentrations for neomycin and bacitracin (Nebacetin) were extremely low. Therefore IAT is recommended with a system separating and washing the autologous red blood cells even under circumstances of rinsing the operative field with a solution containing locally acting antibiotics.
Treatment with neomycin sulphate 1.5 g/d for 8 weeks significantly lowered total serum cholesterol by an average of 19% in 15 out of 19 patients with primary hypercholesterolaemia and LDL-cholesterol by 21%, without significantly changing the corresponding triglyceride and HDL-cholesterol concentrations. Combined treatment with neomycin sulphate 1.5 g/d and d-thyroxine 6 mg/d for 8 weeks lowered total serum cholesterol in the same patients by an average of 30%, LDL-cholesterol by 27% and serum triglycerides by 18%, without significantly changing the HDL-cholesterol concentration. Continued treatment for 10 patients with neomycin sulphate 1.5 g/d for up to 13 months did not further change the reduced serum cholesterol level. 10 of 19 patients complained of side effects after 4-8 weeks of treatment with neomycin: 8 of recurrent diarrhoea, abdominal pain or poor appetite, and 4 of acute attacks of vertigo with nystagmus. All side effects were reversed a few days after stopping the neomycin treatment. No additional serious side effects due to d-thyroxine were observed. These serious side effects of neomycin sulphate limit its use to selected high risk patients with hypercholesterolaemia, who have failed to respond successfully to other LDL-cholesterol-lowering drugs. They stress the necessity for frequent monitoring for the side effects described.

With the application of neomycinsulfat (Nebacetin) during neurosurgical operations the authors observed an infection rate below 1%. The form of application is intracerebral, epidural or intramuscular depending on the operation itself, wound infections and serum concentrations of neomycinsulfat are reported separately.

Clinical observations show an increasing rate of psycho-drug prescriptions for preschool children. This is in contrast to only sparse information in the literature. In two Hamburg quarters parents of 790 children (aged 4 10/12 -7 10/12 years) were asked for this topic on the occasion of the school beginners medical examination. 17.3% of the children had taken a psycho-drug once or more up to the day of the examination. Psycho-drugs had been given in all ages from 1-6 years. Pediatricians had prescribed 90% of these psychopharmaca. 18% of the prescriptions served as accompanying medication in somatic diseases. 50% of the patients got neuroleptics, 20% the antidepressant imipramine. 20% of these children were treated with a psycho-drug for more than four weeks. The wanted effect of the medication was noted by 70% of the parents. It was concluded, that an intensive instruction in psychopharmacology is necessary not only for child psychiatrists but especially for pediatricians.

The natural occurrence of sterigmatocystin (S) in in-shell pecans is reported. Aspergillus versicolor was not isolated from contaminated samples. Incidence of A. flavus and A. glaucus, species known to produce sterigmatocystin under laboratory conditions, was high (43 and 35%, respectively). Isolation data suggest sterigmatocystin may have been produced by one or both of these species.
The effects of glucose concentrations in the perfusion medium ranging from 5 to 15 mM and thiopental, on cerebral energy metabolism were studied using the isolated perfused rat brain. After a perfusion time of 30 min brain levels of the following substrates and metabolites were determined: P-creatine, ATP, ADP, AMP, glycogen, glucose, glucose-6P, fructose-6-P, pyruvate, lactate, alpha-ketoglutarate, glutamate, ammonia. In control experiments increasing the glucose concentration in the perfusion medium produced an increase of intracellular brain glucose concentration only, revealing a linear relationship between glucose content in brain and blood. Neither high-energy phosphates nor glycolytic intermediates were markedly affected by the changes in blood glucose. With an anesthetic dose of thiopental (0.15 mM) in the perfusion medium identical metabolic alterations occured in all experiments: P-creatine and glucose were significantly increased whereas ADP, AMP, lactate and pyruvate were diminished. Also with thiopental brain glucose was linearly related with the glucose concentration in the perfusion medium. The calculated regression line was apparently parallel with that from control experiments; that means thipental always caused an elevation of brain glucose by the same amount of 0.9 mumoles/g--irrespective of the initial cerebral glucose content. The results yield further evidence that glucose transport is not the rate-limiting step in glycolysis. The action of thiopetal on glycolytic pathway is discussed.
Two tremorgenic mycotoxins were isolated from Aspergillus caespitosus, and identified as verruculogen and fumitremorgin B. They were produced at the rate of 172 and 325 mg per kg, respectively, on autoclaved cracked field corn.
