Activation of the JNK pathway is important for cardiomyocyte death in response to simulated ischemia.

Multiple signaling pathways, including the c-Jun N-terminal kinase (JNK) pathway, are activated in myocardial ischemia and reperfusion (MI/R) and correlate with cell death. However, the role of the JNK pathway in MI/R-induced cell death is poorly understood. In a rabbit model, we found that ischemia followed by reperfusion resulted in JNK activation which could be detected in cytosol as well as in mitochondria. To address the functional role of the JNK activation, we examined the consequences of blockade of JNK activation in isolated cardiomyocytes under conditions of simulated ischemia. The JNK activity was stimulated approximately sixfold by simulated ischemia and reperfusion (simulated MI). When a dominant negative mutant of JNK kinase-2 (dnJNKK2), an upstream regulator of JNK, and JNK-interacting protein-1 (JIP-1) were expressed in myocytes by recombinant adenovirus, the activation of JNK by simulated MI was reduced 53%. Furthermore, the TNFalpha-activated JNK activity in H9c2 cells was completely abolished by dnJNKK2 and JIP-1. In correlation, when dnJNKK2 and JIP-1 were expressed in cardiomyocytes, both constructs significantly reduced cell death after simulated MI compared to vector controls. We conclude that activation of the JNK cascade is important for cardiomyocyte death in response to simulated ischemia.

Sequence analysis of the knockdown resistance-homologous region of the para-type sodium channel gene from pyrethroid-resistant Boophilus microplus (Acari: Ixodidae).

Using reverse transcription polymerase chain reaction and degenerate oligonucleotide primers, a partial para-homologous sodium channel cDNA was obtained from the southern cattle tick, Boophilus microplus (Canestrini). The cDNA sequence encoded the region in which knockdown resistance (kdr)-type mutations have been identified in numerous insect species. Comparison of deduced amino acids from the cDNA sequence showed high similarity with sodium channels from other species, particularly in highly conserved repeat domains of the sodium channel. Analysis of the kdr-homologous region of the genomic DNA sequences from several susceptible and pyrethroid-resistant tick strains did not detect mutations. The result suggests novel mutations in the sodium channel gene or metabolic detoxification may be involved in the resistance to pyrethroids in this tick.

Cloning and sequencing of a putative acetylcholinesterase cDNA from Boophilus microplus (Acari: Ixodidae).

Using a strategy based on degenerate primers derived from acetylcholinesterase (AChE) from other species, we cloned and sequenced a putative AChE cDNA from the southern cattle tick, Boophilus microplus (Canestrini). The sequence has a high degree of homology to sequences of AChE from other species reported in the GenBank. The open reading frame of 1,689 bp, corresponding to a deduced sequence of 563 amino acids, has conserved regions and features shared by the AChE family, necessary for its catalytic activity. No differences were found in the putative cDNA sequences from organophosphorus acaricide (OP) resistant and susceptible strains. The results suggest that this putative AChE gene is not involved in resistance to OP compounds as a mutated gene in the resistant strain studied. However, differences were detected, with a probe derived from this cDNA, in DNA fragments after digestion of genomic DNA from different strains with restriction nucleases. This indicates polymorphism in this gene in B. microplus.

Theory of modulational instability in Bragg gratings with quadratic nonlinearity.

Modulational instability in optical Bragg gratings with a quadratic nonlinearity is studied. The electric field in such structures consists of forward and backward propagating components at the fundamental frequency and its second harmonic. Analytic continuous wave (CW) solutions are obtained, and the intricate complexity of their stability, due to the large number of equations and number of free parameters, is revealed. The stability boundaries are rich in structures and often cannot be described by a simple relationship. In most cases, the CW solutions are unstable. However, stable regions are found in the nonlinear Schrödinger equation limit, and also when the grating strength for the second harmonic is stronger than that of the first harmonic. Stable CW solutions usually require a low intensity. The analysis is confirmed by directly simulating the governing equations. The stable regions found have possible applications in second-harmonic generation and dark solitons, while the unstable regions may be useful in the generation of ultrafast pulse trains at relatively low intensities.

Cytoskeletal tumor suppressors that block oncogenic RAS signaling.

Several distinct peptides or drugs that block the Rho family GTPases-mediated pathways were found to suppress RAS-induced malignant phenotype. They include (1) C3 enzyme that selectively inactivates Rho, (2) ACK42, a peptide that blocks the interaction of CDC42 with its effectors such as ACKs, (3) PAK18, a peptide that blocks the activation of PAK and membrane ruffling, and (4) actin-binding drugs, chaetoglobosin K (CK) and MKT-077, that block membrane ruffling by capping and bundling actin filaments, respectively.

Phospholamban-to-SERCA2 ratio controls the force-frequency relationship.

The force-frequency relationship (FFR) describes the frequency-dependent potentiation of cardiac contractility. The interaction of the sarcoplasmic reticulum Ca2+-adenosinetriphosphatase (SERCA2) with its inhibitory protein phospholamban (PLB) might be involved in the control of the FFR. The FFR was analyzed in two systems in which the PLB-to-SERCA2 ratio was modulated. Adult rabbit cardiac myocytes were transduced with adenovirus encoding for SERCA2, PLB, and beta-galactosidase (control). After 3 days, the relative PLB/SERCA2 values were significantly different between groups (SERCA2, 0.5; control, 1.0; PLB, 4.5). SERCA2 overexpression shortened relaxation by 23% relative to control, whereas PLB prolonged relaxation by 39% and reduced contractility by 47% (0.1 Hz). When the stimulation frequency was increased to 1.5 Hz, myocyte contractility was increased by 30% in control myocytes. PLB-overexpressing myocytes showed an augmented positive FFR (+78%), whereas SERCA2-transduced myocytes displayed a negative FFR (-15%). A more negative FFR was also found in papillary muscles from SERCA2 transgenic mice. These findings demonstrate that the ratio of phospholamban to SERCA2 is an important component in the control of the FFR.

Serodiagnosis of Hansen's disease/leprosy by enzyme-linked immunosorbent assay using cord factor (trehalose 6,6'-dimycolate) as an antigen.

IgG and/or IgM antibodies against mycobacterial cord factor (trehalose 6,6'-dimycolate, TDM) in sera of 65 patients of Hansen's disease (21 cases with smear-positive and 44 cases with smear-negative) and 60 healthy individuals were tested by enzyme-linked immunosorbent assay (ELISA) with TDM purified from Mycobacterium tuberculosis H37Rv as an antigen. Of 65 patients with Hansen's disease, 58 cases (89.2%) had positive results (21 samples from 21 patients, 100% with acid-fast bacilli positive in the lesion, and 37 samples from 44 patients, 84.0% with acid-fast bacilli negative Hansen's disease diagnosed clinically). The sensitivity and specificity of anti-cord factor ELISA were higher than those of anti-phenolic glycolipid-I (PGL-I) agglutination test. Among the total, 34 patients were classified clinically into three types of the disease, lepromatous leprosy (LL), borderline lepromatous (BL) and borderline tuberculoid (BT). The antibody titer showed LL > BL > BT, indicating that the elevation of anti-cord factor antibody titers appeared to be parallel with the degree of humoral immune response against M. leprae. By using semisynthetic cord factor consisting of a single subclass of mycolic acid from M. tuberculosis, it was revealed that sera from patients with Hansen's disease were highly reactive against alpha-mycoloyl cord factor (alpha-TDM) and less reactive against methoxy mycoloyl TDM (methoxy TDM), differed from sera of tuberculosis patients, which were highly reactive against both methoxy and alpha-mycoloyl cord factor (alpha-TDM). Most of sera from patients with Hansen's disease were more reactive against TMM than TDM, differed from sera of tuberculosis patients which were highly reactive against TDM. ELISA using TDM as an antigen is simple, reproducible and useful for the rapid serodiagnosis of Hansen's disease, especially for smear-negative cases.

Phorbol ester-induced mononuclear cell differentiation is blocked by the mitogen-activated protein kinase kinase (MEK) inhibitor PD98059.

The purpose of this study was to evaluate whether the mitogen-activated protein kinase (MAPK) signaling pathway contributes to 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced mononuclear differentiation in the human myeloblastic leukemia ML-1 cells. Upon TPA treatment, the activity of ERK1 and ERK2 rapidly increased, with maximal induction between 1 and 3 h, while ERK2 protein levels remained constant. The activity of JNK1 was also significantly induced, with JNK1 protein levels increasing moderately during exposure to TPA. Treatment of cells with PD98059, a specific inhibitor of mitogen-activated protein kinase kinase (MEK), inhibited TPA-induced ERK2 activity. Furthermore, PD98059 completely blocked the TPA-induced differentiation of ML-1 cells, as assessed by a number of features associated with mononuclear differentiation including changes in morphology, nonspecific esterase activity, phagocytic ability, NADPH oxidase activity, mitochondrial respiration, and c-jun mRNA inducibility. We conclude that activation of the MEK/ERK signaling pathway is necessary for TPA-induced mononuclear cell differentiation.

Erigeron breviscapus prevents defective endothelium-dependent relaxation in diabetic rat aorta.

We examined the endothelium-dependent relaxation response to acetylcholine (Ach) in streptozotocin-induced diabetic rat aorta at the stages of 2- and 6-wks' duration in vitro, and compared with another two groups which were treated with dietary supplement of 0.1% Aminoquanidine (AG) and 0.5% Erigeron breviscapus (EB) from 1-week of diabetes induction. At the stage of 2-wks' duration of diabetes, relaxation responses to lower concentrations of Ach in 0.3 uM phenylepherine-precontracted aortas were diminished significantly (P<0.05) compared with age-matched control, but the maximal relaxation of Ach remained unchanged. At the stage of 6-wks' duration, diabetes caused an approximately 60% (P<0.001) deficit in maximum relaxation, and this was significantly (P<0.001) prevented in AG and EB treated groups. There was an approximately 40% enhancement in the maximum contractile response to phenylepherine with diabetes (P<0.05), which was unaffected significantly by AG and EB treatments. The data suggest that the defective endothelium-dependent relaxation in diabetic rat aorta occurred as early as 2-wks' duration of diabetes, and the treatments of AG and EB could protect vascular endothelium although the deficits in vascular smooth muscle contractile responses were not protected.

[Acute lymphoblastic leukemia complicated with tumor lysis syndrome--four cases report].

OBJECTIVE: To report four acute lymphoblastic leukemia(ALL) patients who developed tumor lysis syndrome(TLS) after initial chemotherapy. METHODS: The clinical features and blood biochemical changes of four patients with TLS were analysed. RESULTS: TLS was characterized by hyperuricemia, hyperkalemia, hyperphosphatemia and hypocalcemia. Three cases had renal dysfunction and the other died of acute renal failure. After allopurinol therapy and alkalinization of urine, the blood biochemical parameter became normalization in all the three cases. CONCLUSIONS: TLS can be effectively controlled by early recognition and administration of allopurinol therapy and alkalinization of urine.

[Study of low-fusing porcelain fused to Ti-75 alloy].

OBJECTIVE: To study the feasibility of low-fusing porcelain fused to Ti-75 alloy. METHODS: The bond strength and interfacial state of porcelain-titanium were observed by EPM analysis and shear test. RESULTS: The results showed that the shear bond strength of low-fusing porcelain fused with Ti-75 alloy and the conventional porcelain fused to base metal alloy (NC-VMK68) respectively were 47.38 +/- 7.95 MPa and 48.50 +/- 7.60 MPa. There were no significant difference in the shear bond strength between the two results (P > 0.05). The analysis of electron microprobe showed that there was no crack in the interface, the low-fusing porcelain permeated Ti-75 alloy fairly, and titanium diffused from Ti-75 alloy to porcelain, and silicon, stannum diffused from porcelain to Ti-75 alloy. CONCLUSION: Ti-75 alloy can be regarded as a suitable alloy for low-fusing porcelain.

[Effect of fodder components on the product of obese gene expression--leptin in rats].

OBJECTIVE: To study the effects of different fodder components and levels of caloric uptake on the product of obese gene expression -- leptin and in vivo secretion of insulin and growth hormone, and to study the relationship between them in rats. METHODS: Plasma leptin was determined by Western blot, and serum insulin and growth hormone were determined by radioimmunoassay (RIA). RESULTS: There was no significant effects of fodder components, including high protein, fat and carbohydrate, on the levels of leptin, insulin and growth hormone in rats, with the same caloric uptake in them. However, with the higher caloric uptake, level of obese gene expression increased, showing increased levels of plasma leptin and serum insulin and decreased level of serum growth hormone. CONCLUSION: There could existed an interaction between expression of obese gene and secretions of leptin, insulin and growth hormone, which could be modified by the same metabolic signal.

[Factors affecting Agrobacterium-mediated genetic transformation of Poncirus trifoliata (L.) Raf].

OBJECTIVE: Establishing an effective system for the Agrobacterium-mediated genetic transformation of Poncirus trifoliata, so as to lay a foundation for improving breeds and introducing foreign genes. METHOD: The explants used for the genetic transformation were the epicotyls from Poncirus trifoliata. The Agrobacterium tumefaciens strain was EHA101, containing vector plasmid PGA482GG. The GUS gene and NPT II gene were introduced into the transformation plasmid. RESULTS: 20-day epicotyls were suitable for transformation. 2-day precultivation enhanced shoot regeneration. Shoot regeneration frequency was high when the cocultivation lasted 2-3 days. With the presence of acetosyringone during cocultivation, the efficiency of transformation could be greatly enhanced, and the frequency of shoot regeneration rose as high as 27.5%. After the explants were cocultivated with Agrobacterium, 2-day delayed selection could stimulate shoot regeneration. CONCLUSION: An effective genetic transformation system has been established for Poncirus trifoliata on the basis of factors affecting Agrobacterium-mediated genetic transformation.

[Study on cultivation of Dendrobium flexicaule by habitat imitation].

Dendrobium drug, a rare medicinal herb, reproduces differcut for growth environment to lead rare resources. By studying of cultivation for many many years in Fuliu mountain area, authors had found out the cultivated method simulating the habitat of wild Dendrobium flexicaule. The results could supply a basis for its development and utilization.

[Software package for self-constructed CCD-ICP-OES. 1. Software system and design idealogy].

A software package and its design idealogy for the self-constructed CCD-ICP-OES are described. The MS Windows 95TR was used as the operating platform. The Software package written in MS Visual C + + TR V 4.2 included the control of grating and CCD, the dynamic monitoring, the treatment of spectral signal, the fixing of wavelength position, the automatic fixing of spectral line, the quantitative analysis, the stripping method, the Kalman filter and the artificial neural network.
