[Diagnosis of basal body temperature, serum progesterone and endometrial biopsy for luteal phase defect].

Fourteen infertile women and two women with recurrent abortions were suspected of having luteal phase defect (LPD) by BBT charts. In these patients, the midluteal phase serum progesterone (P) levels were determined together with histological examination by the late luteal endometrial biopsies during the same cycle. The diagnostic criteria for LPD was BBT of luteal phase < or = eleven days, mean P level of 3 blood samples taken on three days during midluteal phase was less than 48 nmol/L (15 ng/ml) and the histological dating of endometrium was two or more days behind. The results showed that histological abnormality and low P level were 6.3% and 31.3%, respectively. When histological abnormality combined with lowered P level at the same cycle, the diagnosis of LPD increased to 87.5%. The author suggests that both midluteal phase P level and late luteal endometrial histological examination should be assessed at the same cycle in the diagnosis of LPD. BBT should also be assessed.

[A serological survey on antibodies to polio, measles, diphtheria and tetanus in Yichang human population].

A serological survey on antibodies to polio, measles, diphtheria and tetanus in 1444 normal people in Yichang City is reported. The positive rates and geometric mean titres (GMT) of neutralizing antibodies to polio types 1-3 were 95.00%, 93.61%, 94.17% and 103.57, 94.61, 83.80, respectively. Antibody to diphtheria had higher level (GMT = 0.0829 IU/ml). Antibodies to tetanus in child-bearing women had the lowest level (GMT = 0.0014 IU/ml). There was a significant positive correlation between the antibody to measles in mothers and that in their newborn babies (r = 0.7153, P < 0.0005).

Effects of semotiadil fumarate (SD-3211) on renal hemodynamics and function in dogs.

Studies were carried out to define the effect of semotiadil on renal hemodynamics, renal function and renin release in pentobarbital-anesthetized dogs. Intrarenal arterial infusion of semotiadil resulted in a significant increase in renal blood flow (RBF), glomerular filtration rate (GFR), urine flow, urinary excretion of electrolytes and renin release. Semotiadil did not affect the linear relationship between osmolar clearance and the free water reabsorption rate, and increased the urinary excretion of sodium and calcium to the same extent. These results suggest that the main tubular site of action of semotiadil is the proximal tubule. Intrarenal infusion of a potent non-peptide angiotensin II antagonist, DuP753 (15 micrograms/kg per min), resulted in an increase in RBF, GFR, urine flow and UNaV. In spite of the blockade of the intrarenal renin angiotensin system(RAS) with DuP753, semotiadil caused almost the same effects as it did in the absence of DuP753. These results suggest that the renal effects of semotiadil are independent of the intrarenal RAS.

Transient expression of somatostatin peptide is a widespread feature of developing sensory and sympathetic neurons in the embryonic rat.

Previous studies from this and other laboratories demonstrated that many embryonic sensory ganglion cells in the rat transiently express the catecholamine synthesizing enzyme tyrosine hydroxylase (TH), a trait not expressed by most mature sensory neurons. We, therefore, sought to determine whether transient expression was uniquely associated with catecholaminergic traits, or, alternatively, whether embryonic ganglion cells transiently expressed peptidergic properties as well. Of the four peptides examined (somatostatin [somatotropin release inhibiting factor] (SRIF), galanin (Gal), calcitonin gene-related peptide (CGRP), and substance P (SP)), only SRIF was found to be transiently expressed during early stages of sensory gangliogenesis. Surprisingly, SRIF immunoreactivity was observed in virtually all cranial and spinal sensory ganglion cells on embryonic day (E) 12.5. In addition to perikaryal labeling, intense SRIF immunoreactivity was also observed in the central and peripheral processes of E12.5 sensory neurons, suggesting the peptide may be released from nerve endings. The time course of SRIF appearance in cranial ganglion cells paralleled that previously described for TH, and double-labeling studies revealed extensive co-localization of these two phenotypes. By E16.5, however, the number of neurons expressing SRIF had diminished markedly, indicating that SRIF is only transiently expressed by most sensory neurons during early stages of ganglion development. An unexpected finding was that transient expression of SRIF is also a prominent feature of sympathetic ganglion cells; however, the temporal pattern of staining in the sympathetic and sensory ganglia differed substantially. Whereas virtually no SRIF staining was observed in E12.5 sympathetics, the vast majority of cells in the E16.5 superior cervical ganglion (SCG) were labeled. This contrasted sharply with the adult SCG, in which only low levels of SRIF expression were found. These findings demonstrate that SRIF peptide is transiently expressed at high levels in peripheral sensory and sympathetic neurons during embryogenesis. The time course and widespread distribution of SRIF expression indicates that the peptide may play a role in early stages of ganglion cell growth and development. Moreover, these data, in conjunction with previous studies demonstrating SRIF immunoreactivity in developing central neurons, suggest that transient expression of this peptide is a common property of diverse neuronal cell types.

An optical biosensor for lysine based on the use of lysine decarboxylase and a cadaverine-sensitive membrane.

We describe an optical biosensor for lysine based on the use of lysine decarboxylase and an optical transducer for detection of cadaverine which is formed as a result of enzymatic action. A plasticized PVC (polyvinyl chloride) membrane containing a lipophilic tartrate as the amine carrier acts as the optical cadaverine sensor. The transport of the cadaverine cation into the membrane is coupled to a transport of a proton (of the indicator dye) out of the membrane. This causes a spectral change of the indicator dye which can be related to the cadaverine concentration, provided the pH is kept constant. The enzymatic reaction is performed in an enzyme reactor which is part of a flow-through system. The dynamic range is from 0.1 to 100 mM for both cadaverine and lysine. While the cadaverine sensor is moderately selective (ethylamines, for example, interfere), the whole sensor system is highly specific for lysine, nicotine being the only major interferent. Unlike other enzyme-based detection schemes where the production of CO2 (in case of decarboxylases) or consumption of oxygen (in case of oxidases) is measured, this scheme is based on the measurement of the organic ammonium ion (cadaverin cation) formed in the enzymatic reaction. The major advantage of this approach is that in many real samples there is a rather low and fairly constant background of organic amines.(ABSTRACT TRUNCATED AT 250 WORDS)

Regional hemodynamic effects of betaxolol, a new selective beta 1-blocker, and atenolol in conscious spontaneously hypertensive rats.

Betaxolol is a new beta-blocker that has been reported to have beta 1-selectivity, and it is devoid of both membrane stabilizing action and intrinsic sympathomimetic action. The effects of betaxolol on systemic and regional hemodynamics were examined in conscious spontaneously hypertensive rats (SHR) by a microsphere method and compared with the effects of atenolol. A single oral administration of betaxolol at 1 and 10 mg/kg decreased the mean arterial pressure in a dose-dependent manner. At the same doses, atenolol also showed a similar but weak hypotensive effect. Both of these drugs at the high dose decreased cardiac output and heart rate and at the low dose, did not. Total peripheral resistance decreased by only betaxolol at the low dose. Betaxolol showed a tendency to normalize the hemodynamic abnormalities which were observed in the kidney, spleen and gastrointestinal tract of SHR, while atenolol did not. It should be noted that betaxolol increased the flow rate in the kidney, which may be explained by its direct vasodilatory action on renal blood vessels. In conclusion, betaxolol showed an antihypertensive action at the doses of 1 and 10 mg/kg, exhibited the characteristics of a beta 1-blocker and produced preferable effects on regional hemodynamics in SHR.

[Sensitivity in vitro of Plasmodium falciparum to chloroquine, pyronaridine, artesunate and piperaquine in south Yunnan].

The sensitivity of P. falciparum to chloroquine, pyronaridine, artesunate and piperaquine (CQ, PD, AT, PQ) was assayed using in vitro microtechnique in south Yunnan in 1990. The resistance rates were 98.7% (75/76), 27.6% (16/58), 13.8% (9/65) and 97.7% (43/44) respectively, and ID50 were 125.0, 19.0, 4.7 and 243.3 nmol/L, respectively. The resistance rate against CQ showed no change as compared to the rates against CQ 5 and 9 years ago; but the ID50 was lower. CQ-resistant P. falciparum showed a marked cross-resistance to PQ, but not to PD and AT. AT-resistant P. falciparum exhibited cross-resistance to the above-mentioned three drugs. PD-resistant P. falciparum showed no cross resistance to AT, but showed cross resistance to CQ and PQ. In comparison with chloroquine-coated plates, the plates coated with pyronaridine, artesunate or piperaquine gave similar results as the former, which were shown by the rise in schizont inhibition rates along with the rise in drug concentration. It indicates that pyronaridine-, artesunate-, and piperaquine-coated plates can be used in the assay of sensitivity of P. falciparum to the three drugs.

Effects of the calcium antagonist manidipine on renal hemodynamics and function in dogs: comparison with nifedipine.

The renal effects of manidipine, a new dihydropyridine calcium antagonist, were compared with those of nifedipine in anesthetized dogs. Intrarenal administration of manidipine 1 and 5 micrograms/kg resulted in prolonged (120 min) increases in renal blood flow (RBF), glomerular filtration rate (GFR), urine flow (UF), and urinary excretion of electrolytes. In contrast, the renal effects of nifedipine disappeared within 5 to 10 min after administration. These results demonstrate that manidipine is a long-acting calcium antagonist. Manidipine proportionally increased RBF and GFR. As a result, filtration fraction was kept constant. The changes in afferent and efferent arteriolar resistance suggested that manidipine increases GFR via dilation of afferent arterioles with subsequent elevation of filtration pressure in the glomeruli. Manidipine also proportionally increased Tc H2O and Cosm; Tc H2O/Cosm did not change throughout the experiments. In addition, manidipine increased the fractional excretion of sodium from the distal tubule which was calculated from the lithium and sodium clearance. The mode of diuretic action of manidipine most likely involves inhibition of sodium reabsorption in the proximal and distal tubules, except the medullary portion of the ascending limb of Henle. The renal vascular site of action of manidipine might be the afferent arteriole.

Skin test inhibition by astemizole.

For the purpose of evaluating the therapeutic effect of antihistamines, we have set up an assay method called the "Skin Test Inhibition Index" (STII). Twenty subjects with hay fever were given astemizole (10 mg/d) for 7 days. Skin titration tests were carried out before and after treatment. Significant inhibition of the skin test reaction by astemizole was demonstrated (STII = 91). Another group of 6 hay fever patients was given astemizole (10 mg/d) for 10 days, and STII was determined on days 5 and 10 and on days 7, 14 and 21 after treatment. STII were calculated as 12, 108, 90, 10 and 7, respectively. These results demonstrate that astemizole is a long-acting antihistamine.

Rapid serodiagnosis of human mycobacteriosis by ELISA using cord factor (trehalose-6,6'-dimycolate) purified from Mycobacterium tuberculosis as antigen.

IgG antibodies against purified cord factor (trehalose-6,6'-dimycolate, TDM) in sera of 99 patients infected with mycobacteria (42 patients with tuberculosis excreting tubercle bacilli in the sputum, 11 patients with non-tuberculous mycobacteriosis excreting acid-fast bacilli in the sputum, and 46 patients without bacilli in the sputum but diagnosed as having pulmonary tuberculosis by chest X-ray films and physical examination), five patients with lung cancer, and 100 healthy controls which included subjects positive and negative for the tuberculin test were tested by the ELISA with TDM purified from Mycobacterium tuberculosis H37Rv as the antigen. Of the 99 cases of mycobacteriosis, 83 patients (83.8%) had positive results (48 samples from 53 patients, or 90.5%, with bacilli in the sputum, and 35 samples from 46 patients (76%) with tuberculosis diagnosed clinically). The sera of the five patients with lung cancer and the 100 controls all gave negative results. Thus, the sensitivity and specificity were 83.8% and 100%, respectively. ELISA with TDM as the antigen is simple, reproducible, and useful for the rapid serodiagnosis of general mycobacterial infections including tuberculosis, because it does not involve the cultivation of bacteria.

[Quantitative analysis of midecamycin by HPLC].

A reversed phase high performance liquid chromatographic method for the assay of midecamycin was developed. The method used a Hitachi Gel 3050 column at 50 degrees C and a mobile phase of methanol--0.01 mol/L phosphate buffer solution at pH 5.8 (45:55). The column effluent was monitored Z at 231 nm. SF-837A1, leucomycin A6 and minor components could be separated in less than 15 min. The retention times of SF-837A1 and leucomycin A6 are 7 and 10 min respectively. The method is very simple and rapid.

[Effect of PGR-1 (plant growth regulator-1) on variations in mineral content and nutritive value of Areca catechu L].

The investigation was conducted to find out the effect of PGR-1 (spurting) on variations in the mineral contents and nutritive values of the leaf, flower and root of Areca catechu in different phenophases of th year. The result shows that PGR-1 helps to expand th leaf area of different leaf segments by 20.8 to 37.47 cm2 and increase the dry matter by 4.66 to 6.56g, so as to promote the nutritive value of inflorescence and thus raise the flowering and fruitication rate.
