Formation of 2-nitro-3-methylimidazo[4,5-f]quinoline, a directly mutagenic product, by near-ultraviolet irradiation of a mixture of 2-amino-3-methylimidazo[4,5-f]quinoline and N-nitrosodimethylamine.

Direct-acting mutagens to Salmonella typhimurium TA98 were found to be formed from heterocyclic amines on exposure to near-ultraviolet light in the presence of N-nitrosodialkylamines. We have isolated the mutagenic photoproduct formed from 2-amino-3-methylimidazo[4,5-f]quinoline (IQ) and N-nitrosodimethylamine, and the product was identified as 3-methyl-2-nitroimidazo[4,5-f]quinoline (IQ(NO2]. The yield of IQ(NO2) from IQ was estimated to be 17%. Similar light-dependent activation of IQ was noted with 4 different nitrosodialkylamines other than nitrosodimethylamine. Furthermore, MeIQ and MeIQx were also activated with nitrosamine and light. These reactions represent an example of interaction between 2 different classes of mutagens.

Mutagenicity of smoked, dried bonito products.

Mutagens have been found in smoked, dried bonito products, popular items in Japanese foods. The mutagens were isolated by means of blue cotton, an absorbent cotton preparation with covalently bound trisulfo-copper-phthalocyanine residues, and by means of XAD-2 resin. The mutagenicity was positive in Salmonella typhimurium strain TA98 with metabolic activation. The mutagens are produced during the process of smoking-and-drying bonito (a process called baikan). The activity was much higher than that expected from the content of benzo[a]pyrene. In contrast to benzo[a]pyrene, the mutagens were not inhibited by ellagic acid. The mutagenicity was not abolished by treatment with nitrite. Thin-layer and high-performance liquid chromatographic analysis gave two mutagenic fractions, both of which were distinguishable from benzo[a]pyrene and from the pyrolysis products Trp-P-1, Trp-P-2, Glu-P-1, Glu-P-2, A alpha C and MeA alpha C. The major mutagenic component was not chromatographically distinguishable from IQ and MeIQx, and the minor one was very similar to MeIQ. The smoked, dried bonito products contained free fatty acids, which were inhibitory to the mutagenicity of the bonito products.

Potentiation of bleomycin cytotoxicity in cultured mammalian cells by polyacrylic acid. III. Induction of DNA strand breakage and suppression of cellular heat production.

The 2E cytotoxicity of bleomycin toward cultured mammalian cells was synergistically enhanced by vortex-stirring in the presence of a low dose of high molecular weight polyacrylic acid. Cellular DNA isolated immediately after the above treatment suffered from severe single strand breaks. Heat production of the treated cells also decreased sharply immediately after the treatment, indicating that some functional disorder was probably induced on the cell membrane leading to cell death, possibly resulting from enhanced DNA strand breaks.
