Regulation of expression of the galactose gene cluster in Saccharomyces cerevisiae. Isolation and characterization of the regulatory gene GAL4.

The GAL4 gene positively regulating the expression of the gene cluster GAL7-GAL10-GAL1 in the yeast Saccharomyces cerevisiae was isolated for its ability to suppress a recessive mutation in that gene. When the isolated gene was incorporated into a multi-copy plasmid, the GAL cluster genes in the host chromosome partially escaped the normal control; a yeast that harbors the plasmid bearing the GAL4 gene synthesized the galactose-metabolizing enzymes encoded by the GAL cluster genes at a low but significant level in the absence of galactose. If the GAL7 gene was amplified along with GAL4 on the multi-copy plasmid, the constitutive synthesis of Gal-1-P uridylyl transferase encoded by GAL7 was further pronounced and the enzyme activity reached the level of the fully induced wild-type yeast. Such an escape synthesis of the GAL enzymes was not detected if GAL4 or both GAL4 and GAL7 were carried by a single-copy plasmid. The results suggest that the escape synthesis of GAL enzymes observed in the GAL4-amplified yeast was a consequence of overproduction of the GAL4 protein. The GAL80 gene negatively regulating the GAL cluster genes was also isolated, and when amplified together with GAL4, no escape synthesis of the GAL enzymes was observed, suggesting that the balanced synthesis of two regulatory proteins was essential to maintain the repressed state of the GAL cluster genes.

Myxoid malignant fibrous histiocytoma versus myxoid liposarcoma. A comparative ultrastructural study.

An ultrastructural comparison of 7 examples of myxoid malignant fibrous histiocytoma (myxoid MFH) with 6 of myxoid liposarcoma is described. Despite certain histological differences between the two, electron microscopy was more valuable in differentiating MFH from liposarcoma. Electron microscopically, MFH contained a variety of cell types including histiocyte-like, fibroblast-like, intermediate-type, xanthomatous, multinucleated tumor giant cells and undifferentiated cells. Liposarcoma was composed mainly of lipoblasts at various stages of differentiation with a minority of undifferentiated cells and fibroblast-like cells. In contrast to the component cells of the MFH, these lipoblasts were characterized by abundant cytoplasmic glycogen, numerous pinocytotic vesicles and a discontinuous basal lamina in addition to large lipid droplets.

Pancreatic somatostatinoma.

A 66 year old woman was admitted for right hypochondriac pain and an abdominal tumor. A tumor of the pancreatic head with duodenal infiltration and liver metastases was found by laparotomy, and pancreaticoduodenectomy was performed. In addition, there was associated cholelithiasis and a goiter. Histopathologically, a tumor of the islets of Langerhans was suspected. The tumor tissue was positive by the Hellman-Hellerström silver impregnation method, and electron microscopy confirmed the presence of secretory granules similar to D cells. The tumor tissue contained a large quantity of somatostatin (1.3 micrograms/g). Finally, the diagnosis of somatostatin-producing tumor originating in the pancreas was made.

Intravascular papillary endothelial hyperplasia. A clinicopathologic study of 91 cases.

Ninety-one cases of intravascular papillary endothelial hyperplasia were studied clinically and histologically. This peculiar benign process, occasionally resembling hemangiosarcoma, was subgrouped in the following categories: a pure form that occurs within a dilated vascular space (30 cases), a mixed form that appears as a focal change in a hemangioma (55 cases), a third form (six cases) that belongs to neither of the first two. In the pure form, the lesions were most frequently situated in the subcutis of fingers (14 cases), of the head and neck (seven cases), and in the region between the elbows and hands (six cases). In the mixed form, half of the accompanying hemangiomas were intramuscular in no particular predilective sites. Papillary proliferation of endothelial cells was commonly found to be closely associated with thrombotic material and seemed to be an unusual feature of a thrombus undergoing organization.

Malignant neuroepithelioma (peripheral neuroblastoma). A clinicopathologic study of 15 cases.

A clinicopathologic study of 15 cases of malignant neuroepithelioma (peripheral neuroblastoma) of soft tissues is reported. The patients were chiefly young Japanese adults with a median age of 21 years. The tumors arose mainly in the soft tissues of the lower extremity (seven cases) and the trunk (four cases). Microscopically, there were sheets of closely packed, small round or oval cells, and Homer Wright-type rosettes were seen in all cases, one of which also had Flexner-type rosettes. Immunohistochemical cytoplasmic localization of neuron-specific enolase (NSE) was demonstrated in six of the eight cases, using the peroxidase-antiperoxidase (PAP) method. In no case, however, was there any staining reaction for S-100 protein. Of the 14 patients for whom follow-up information could be obtained, nine died within a period of 2 years and two were alive and well for over 5 years after the initial treatment. Differential diagnosis from other soft-tissue round-cell sarcomas, such as embryonal or alveolar rhabdomyosarcoma, extraskeletal Ewing's sarcoma, and others, are briefly discussed, on a clinicopathologic basis.

Drug resistance and R plasmids in Escherichia coli strains isolated from broilers.

In 1978, 1,021 Escherichia coli strains were isolated from 105 field broilers (F) and 1,058 strains from 106 broilers in a zootechnical experiment station (Z), and their drug-resistance patterns and the presence of conjugative R plasmids were compared. The resistance markers examined were tetracycline (TC), chloramphenicol (CM), streptomycin (SM), sulfonamides (SA), kanamycin (KM), and ampicillin (APC). The populations of individuals that excreted resistant strains were 100% in F and 58% in Z. Frequencies of isolation of drug-resistant strains among the total isolates were 93% in F and 36% in Z, indicating that the resistant strains are a rather high proportion of the intestinal flora in F but are slightly less prevalent in Z. The resistance pattern to (TC.SM.SA.KM) was seen at the highest frequency in both groups. Conjugative R plasmids were demonstrated more frequently in field broilers (F). The results reflect the wide use of antibiotics in the livestock industry, resulting in the appearance of drug-resistant strains mostly due to the presence of R plasmids.

Conjugal transferability of multiple resistance in Shigella strains.

Shigella strains isolated in Japan between 1971 and 1979 were surveyed for drug resistance and distribution of R plasmids. Of 2,510 strains, 89.3% were resistant to either one or various combinations of four drugs, tetracycline, chloramphenicol, streptomycin, and sulfanilamide. About 66% of the Shigella isolates were quadruply resistant. The frequency of isolation of R plasmids from quadruply resistant Shigella strains was the highest when compared with other strains resistant to various combinations of the four drugs. The conjugal transferability of 204 quadruply resistant strains isolated between 1977 and 1979 was tested by various mixed-culture methods. Among the total strains examined, 70.6% carried transferable resistance when tested by the conventional broth culture method, 90.2% transferred their resistance when, in addition the replica-plating method was used and 97.5% could transfer their resistance when the membrane filter method was also used. Although the remaining five strains could not transfer their resistance by any of the mixed culture methods, the drug resistance of four of the five strains was mobilized by the concomitant presence of F-tet or T-kan plasmid. These results indicate that almost all of the quadruple resistance in Shigella isolates was mediated by plasmid.

Drug resistance and conjugative R plasmids in fecal Escherichia coli strains isolated from healthy younger animals (chickens, piglets, calves) and children.

A total of 11,777 Escherichia coli strains were isolated from 90 chickens, 103 piglets, 96 calves, and 104 children in 1979 in Gunma Prefecture and tested for drug resistance and the presence of conjugative R plasmids. The percentages of individuals that excreted drug-resistant strains were: chickens, 100%; piglets, 99%; calves, 100%; and children, 64%. The frequency of isolation of drug-resistant strains among the total isolates was: chickens, 98%; piglets, 93%; calves, 94%; and children, 41%. Frequency of isolation of R plasmids among the strains tested was: chickens, 48%; piglets, 33%; calves, 38%; and children, 10%. Resistance patterns of the strains isolated most frequently among the four groups were tetracycline (TC), sulfonamides (SA) in single resistance, TC.SA in double resistance, TC.streptomycin (SM).SA in triple resistance and TC.SM.SA. kanamycin (KM) in quadruple resistance. R plasmids were isolated frequently from animals (over 33%) but infrequently from children (about 10%). The high frequency of isolation of drug-resistant strains and R plasmids from animals was caused by the heavy use of chemicals in the period of growth of younger animals.

Metabolism of D-phenylalanine and its effects on concentrations of brain monoamines and amino acids in rats--a basic study on possibility of clinical use of D-phenylalanine as an antidepressant.

The effect of D-phenylalanine on the concentrations of brain catecholamines, serotonin, beta-phenylethylamine and amino acids was examined using rats injected intraperitoneally with 200 mg/kg of D-phenylalanine. The contents of these monoamines in the rat brain were not affected by the administration of D-phenylalanine. No spectacular change was observed in the concentrations of brain amino acids except phenylalanine, which increased about four times during 30-60 minutes after the injection. This increase was attributed to the administered D-phenylalanine. To confirm the finding that D-phenylalanine did not affect the content of beta-phenylethylamine, the metabolism of D-phenylalanine was examined using D-[14C]-phenylalanine. It was proven that D-phenylalanine did not convert to beta-phenylethylamine. On the basis of these findings the antidepressant effect of D-phenylalanine was critically discussed.

Age-dependent differences in the positive inotropic effect of phenylephrine on rat isolated atria.

The age-dependent differences in the involvement of alpha-adrenoceptors in the positive inotropic effect of phenylephrine (Phe) were examined in isolated atria of male Wistar rats 6 weeks (6W), 10 weeks and 7 months (7M) of age. The maximal increase in tension development induced by Phe increased with age, whereas the EC50 values for the positive inotropic effect of Phe did not change with age. The inhibitory effect of phentolamine on the response to Phe increased with age. Propranolol caused only slight inhibition of the effect of Phe in both 6W and 7M rats, and the EC50 values for Phe in the presence of propranolol did not change significantly with age. The EC50 values for isoprenaline and 5-hydroxytryptamine in 7M rats were higher than those in 6W rats. In 7M rats, the duration of the tension development was only slightly affected by Phe in the presence or absence of propranolol, but it was markedly decreased by Phe in the presence of phentolamine. The dose-response curve for Phe was markedly shifted to the left by papaverine in 6W rats, but slightly in 7M rats. The dose-response curve for isoprenaline was markedly shifted to the left by papaverine in both groups. These results are consistent with effects of Phe being mediated by both alpha- and beta-adrenoceptors in both 6W and 7M rats, but there is a shift in the balance from rather more beta-receptors in the young animals to more alpha-receptors in the adults.

Effects of calcium antagonists on the electrical alternans of the ST segment and on associated mechanical alternans during acute coronary occlusion in dogs.

Effects of calcium antagonists on the ST alternans and associated mechanical alternans during acute coronary occlusion were examined in anesthetized dogs. The heart rate was fixed by atrial pacing. The intravenous administration of 0.2 mg/kg verapamil attenuated the ST alternans as did 0.5 mg/kg diltiazem. Although these drugs significantly attenuated TQ depression during occlusion, the attenuation was observed after a longer period of occlusion and when the degree of TQ depression was comparable to that during the control occlusion. Nifedipine, 0.03 mg/kg, slightly attenuated the ST alternans, but 0.5 mg/kg dipyridamole had no effect. These results support the idea that slow inward currents are involved in the ST alternans. On the other hand, the mechanical alternans was attenuated in six of 11 dogs. It is probable that factors other than the electrical alternans may also contribute to the mechanical alternans.

Effect of clonidine on blood pressure in chronic one-kidney, one-clip and two-kidney, one-clip hypertensive rats.

Oral administration of clonidine lowered the high blood pressure in two-kidney, one-clip hypertensive rats in chronic stage (three to four months), but not in one-kidney, one-clip hypertensive rats. This reduction of blood pressure was well correlated with decrease in plasma renin concentration, but not with decrease in urinary noradrenaline excretion. These results suggest that the reduction of blood pressure in two-kidney, one-clip hypertensive rats may be due to reduced renin release through the action of clonidine, while in one-kidney, one-clip hypertensive rats the sympathetic nervous activity may play little or no role in the maintenance of high blood pressure.

The relationship of renal histopathological lesions to immunoglobulin classes and complement fixation of anti-native DNA antibodies in systemic lupus erythematosus.

The relationship of renal immunohistopathological lesions to the immunoglobulin classes (Ig) and complement fixation (CF) of anti-native DNA antibodies (anti-nDNA) in sea was studied in 20 patients with systemic lupus erythematosus (SLE) on whom renal biopsy was performed. The Ig classes and CF of anti-nDNA were assessed by the indirect immunofluorescent and complement fixation methods employing kinetoplasts of Crithidia luciliae as substrate. The histopathologically active lesions of lupus nephritis showed a significant correlation with the IgG and complement fixation of anti-native DNA antibodies in sera. IgM anti-nDNA superiority was not necessarily observed in patients with inactive histological lesions, but rather in those with thickening of the glomerular basement membrane. The granular deposits of immunoglobulin or complement in the renal glomeruli showed a significant correlation with the presence of IgG and CF of anti-nDNA in sera, while the deposition of C3 in the glomeruli also showed a significant correlation with the CF of anti-nDNA.

Different mechanisms maintaining high blood pressure in chronic one-kidney, one-clip, and two-kidney, one-clip hypertensive rats.

To evaluate the difference of the blood pressure regulating mechanisms of chronic (12-14 weeks) one-kidney, one-clip (1K-1C) and chronic two-kidney, one-clip (2K-1C) hypertensive rats, we administered captopril, captopril plus indomethacin, and indomethacin to the rats. Pretreatment values of plasma renin concentration, plasma aldosterone concentration and urinary kallikrein excretion were significantly higher in 2K-1C than in 1K-1C hypertensive rats. Captopril-induced blood pressure reduction was greater in 2K-1C than in 1K-1C hypertensive rats. When captopril was administered to the rats treated with indomethacin, captopril-induced blood pressure reduction was attenuated only in 2K-1C hypertensive rats. Indomethacin produced renal impairment and further raised the blood pressure in 1K-1C hypertensive rats, but did not in 2K-1C hypertensive rats. These results suggest that the renin-angiotensin system functions to maintain high blood pressure more predominantly in chronic 2K-1C than in 1K-1C hypertensive rats. The renal kallikrein-kinin system is suppressed in chronic 1K-1C hypertensive rats but not in 2K-1C hypertensive rats. The renal prostaglandin system is more important for regulating the renal circulation in chronic 1K-1C than in 2K-1C hypertensive rats.
