Potentiation of natural killer activity of human lymphocytes by Staphylococcus aureus bacteria and its protein A.

Cytotoxic activity of human lymphocytes against the myeloid cell line K-562 was augmented greatly by 24-h incubation with Staphylococcus aureus Cowan I bacteria (SpA CoI) and its protein A. This effect was not observed when these stimulants were added after preincubation, suggesting that this activity was different from so-called lectin-induced cellular cytotoxicity. Potentiation required at least 12 to 18 h incubation of lymphocytes with these stimulants. Macrophage depletion did not affect the potentiation by protein A or SpA coI, although the potentiation by poly I:C or OK-432, an immunopotentiator of Streptococcus pyogenes was completely reduced. Further cell separation procedures revealed that neither T cells nor FcR- cells, which showed little natural killer (NK) activity, were enhanced by protein A or SpA coI. On the other hand, (a) null cells which were obtained from nylon column (NC)-passed fraction by depleting T cells and surface membrane Ig-positive cells, and (b) FcR+ E- cells which were obtained from NC-passed fraction by depleting FcR- cells and T cells, showed marked NK activity by themselves and were further augmented by these stimulants. FcR+ E+ cells failed to show NK activity even if they were stimulated by these stimulants. Thus, it was found that protein A and SpA CoI, as well as human interferon, could stimulate NC-non-adherent, FcR+, E- NK cells and potentiate markedly their NK activity.

[Studies on Pd-Co-Ni dental casting ferromagnetic alloys. (Part I) (author's transl)].

Many attempts have been made to use magnets as denture retention. But the magnet, such as alnico, ferrite or Pt-Co magnet, is not sufficient for our daily works. Recently, by using the rare earth metal-cobalt magnet, especially Sm-Co magnet, clinicians have gained good results. But magnet, itself, is not good for use in the mouth. Because it is scarcely possible to shape the magnet into suitable forms, it is easy to be broken by masticatory pressure, and its corrosion resistance is not sufficient and so on. To improve these difficulties, Pd-Co-Ni casting alloys which have soft ferromagnetic properties have been developed in this paper. Some metals were added to the alloy to improve the castability of the ferromagnetic alloy. Magnetic properties, range of solidification temperature and castability were examined. As a result, it was found that (43 Pd-27 Co-30 Ni)-10 Au-0.1 Si and (43 Pd-27 Co-30 Ni)-5 Cu-0.1 Si alloy had most excellent properties.

[Studies on Pd-Co-Ni dental casting ferromagnetic alloys. (Part II) (author's transl)].

In the previous paper, the effect of added metals such as Au, Cu, Cr, Si, etc., on the 43 Pd-27 Co-30 Ni alloy was reported. In this paper, the magnetic properties, mechanical properties, corrosion resistance and casting accuracy of the Pd-Co-Ni alloys containing Au, Zn and Si were studied. Results were as follows: 1) Mechanical properties and corrosion resistance of these alloys satisfied the standards for the dental casting alloys. 2) The corrosion resistance of the alloy containing 10 wt% Au was excellent in comparison with other specimens. 3) Casting accuracy was improved by addition of 10 wt% Au-0.1 wt% Si or 10 wt% Au-1 wt% Zn to the base 43 Pd-27 Co-30 Ni alloy.

The hexosamine concentration and output in human pure pancreatic juice in chronic pancreatitis.

Hexosamine concentration in human pure pancreatic juice was determined during wash-out phase and secretin stimulation phase. Specimens were collected by endoscopic retrograde catheterization of the papilla at one minute intervals for 20 minutes after intravenous injection of secretin (Eisai, 1 U/kg) and for 10 minutes after pancreozymin injection (Boots, 1 U/kg). In suspected and established chronic pancreatitis (calcifying or non-calcifying), hexosamine concentration was significantly raised during both wash-out phase and secretin phase. Hexosamine output was significantly raised in suspected chronic pancreatitis and non-calcifying chronic pancreatitis during both wash-out phase and secretin phase; in calcifying chronic pancreatitis, no significant increase in hexosamine output was noted during both phases because of decreased secretory volume. Significance of these findings was discussed in relation to the pathogenesis of chronic pancreatitis.

The calcium concentration in human pure pancreatic juice in chronic pancreatitis.

Calcium concentration in human pure pancreatic juice was determined during wash-out period, secretin stimulation and pancreozymin stimulation. Specimens were collected by endoscopic retrograde catheterization of the papilla at one minute intervals for 20 minutes after intravenous injection of secretin (Eisai, 1 U/kg) and for 10 minutes after pancreozymin injection (Boots, 1 U/kg). The determination was also made in duodenal juice obtained during the ordinary pancreozymin-secretin test. Calcium concentration in duodenal juice was significantly raised in patients with chronic pancreatitis (suspected, definite or calcifying), confirming the results of previous investigators. Calcium concentration and calcium per mg of protein in human pure pancreatic juice were significantly raised in patients with chronic pancreatitis (suspected, definite or calcifying). Calcium per mg of protein in normal human pure pancreatic juice was quite similar to that reported in zymogen granules of the guinea-pig pancreas. Calcium output in human pure pancreatic juice was also significantly raised in patients with suspected chronic pancreatitis; it was within normal limits in patients with definite or calcifying chronic pancreatitis due to decreased volume output. Significance of these findings were discussed in relation to pathogenesis and early detection of chronic pancreatitis.

Significance of levator muscle sling formation in cleft palate surgery: evaluation by electrical stimulation.

The effect of levator muscle sling formation in cleft palate surgery was evaluated by electrical stimulation of the muscle at surgery. Palatal movement before and after sling formation was recorded through a force transducer. After muscle sling formation, the soft palate was elevated more effectively toward the retropharynx and its mobility increased remarkably. The surgical procedure, which had been proposed on the basis of anatomic considerations, was proved to be effective for improving velopharyngeal function.

Low responsiveness of human neonatal lymphocytes to a B-cell mitogen, Staphylococcus aureus Cowan I (SpA CoI).

Responses of neonatal and adult lymphocytes to various mitogens were studied. Lymphocytes from umbilical cord blood (UCB) responded well to both phytohemagglutinin and concanavalin A, and also to pokeweed mitogen and Staphylococcus aureus Protein A. The responses of UCB lymphocytes to these mitogens were not significantly lower than those of adult peripheral blood lymphocytes (PBL). In contrast, UCB lymphocytes showed only a minimal response to killed Staphylococcus aureus Cowan I (SpA CoI), a potent B-cell mitogen for human PBL, although the proportion of B cells in UCB was not less than that in PBL. The low level of response of lymphocytes from UCB to SpA CoI was not ascribed to differences in dose response or kinetics. Purified B cells from UCB were not stimulated by SpA CoI either, suggesting tht the low responsiveness was not due to the suppressive effect of T cells or macrophages, but to some intrinsic defect in B cells in UCB. These results suggest that the B cells in neonates may be more immature than the T cells.

Clinical effects of HC 20-511 (ketotifen) in bronchial asthma and its inhibitory effect on antigen-induced morphological changes of basophils.

Sixty-four patients with confirmed bronchial asthma were treated with HC 20-511 (Ketotifen). HC20-511 was evaluated to be very effective in 6.3%, effective in 50.0% and slightly effective in 10.9% of these patients. The appearance of reactive basophils was inhibited by HC 20-511 in 5 out of 6 cases of reaction to house dust, in all three cases with buckwheat allergy to their allergen and in 7 out of 11 cases to anti-IgE. These results confirm that HC 20-511 inhibits type I allergic reactions induced by specific allergen and IgE.
