S-100 protein, isolated from mammalian brain, has widely been used for immunohistochemical marker of the glia cells and the cells derived from the neural crest. In the present study, we made anti S-100 protein antibody and studied the immunoreactive distribution of S-100 protein in the cutaneous nervous system. Albino rabbits were immunized with S-100 protein and complete Freund adjuvant, and antiserum was purified by ion-exchange chromatography. Formalin-fixed normal human skin and sciatic nerve of rat were examined by the PAP method. S-100 protein was detected in Schwann cells of sciatic nerve of rat and cutaneous nerve bundles of human skin specimens. Meissner corpuscles and inner core cells of Pacinian corpuscles of human skin were S-100 protein positive. These findings suggest that the staining of S-100 protein with PAP method is a simple and reliable method to demonstrate the cutaneous nervous system. Also, lamellar cells of Meissner corpuscles and inner core cells of Pacinian corpuscles are indicated to be Schwann cell origin.
A method for the separation of guinea pig epidermal keratinocytes, in which the Feulgen-stainable material suffers minimal damage, has been investigated. The principal stage involves trypsin treatment of the epidermal sheet, stripped from the dermis with ethylenediamine tetraacetic acid. The epidermal cells thus isolated are separated into three groups by centrifugation on a continuous colloidal silica (Percoll) density gradient. The resulting arrangement of the keratinocytes in the centrifuge tube corresponds to their arrangement in situ, with basal cells at the bottom and the more differentiated cells above. By morphological examination, it can be shown that relatively pure fractions of basal cells, spinous cells, and granular cells are obtained by this method. With respect to DNA distribution pattern, there was good agreement between that of keratinocytes separated by the microdissection-ultrasonic irradiation method, or by the chymotrypsin method as reported previously by us, and that obtained by the present method.
Ten hemangioblastomas of the central nervous system were examined by an immunoperoxidase technique for glial fibrillary acidic protein (GFAP), S-100 protein, and Factor VIII-related antigen to determine the origin of the stromal cells. In seven cases scattered stromal cells were positive for GFAP, and eight cases showed variable numbers of S-100 protein-positive cells. These positive cells were present throughout the tumors. S-100 protein-positive stromal cells corresponded to GFAP-positive cells, but the former were more numerous than the latter. Staining for Factor VIII-related antigen was negative in stromal cells, except in endothelial cells lining capillaries. It was suggested that stromal cells of capillary hemangioblastomas are composed by heterogeneous cell populations.

Striated muscle cells coexisting with neuroglial tissue were found in the leptomeninges of a 3-day-old neonate. An immunohistochemical study demonstrated the presence of myoglobin in the striated muscle cells, glial fibrillary acidic protein, and S-100 protein in the heterotopic glia, S-100 protein in the Schwann cells of the heterotopic nerve bundles, and neuron-specific enolase in some striated muscle cells as well as the neurons. A possible explanation for this may be that the striated muscle cells originated from neuroectoderm.

A lesion of sporotrichosis shows a characteristic arrangement of the infiltrate in three zones: the central suppurative zone composed of polymorphonuclear leukocytes, the tuberculoid zone and the round cell zone. For investigating the pathomechanism of the suppurative zone formation, polymorphonuclear leukocyte chemotaxis to sporotrichin was assayed with the agarose plate method. Polymorphonuclear leukocytes in the patients with sporotrichosis showed an enhanced chemotactic index in comparison with that in the control subjects. However, there was no difference in non-specific chemotaxis to bacteria-derived substance between the patients and the control subjects.
The development of experimental sporotrichosis was examined in immunologically-disturbed mice. In the case of intracutaneous inoculation, the lesion in cyclophosphamide-treated mice enlarged strikingly up to 6 days, but at 12 days it was similar in size to that in non-treated mice. The lesion in irradiated mice developed slowly up to 12 days, but thereafter it developed promptly. In nude mice, the lesion was smallest during the development of the lesion. Histological examination showed that numerous organisms were present in the lesion of nude and irradiated mice. Intraperitoneal inoculation showed that dissemination of spores were easily induced in nude mice as compared with littermate mice. These results indicate an important role of cellular immunity in the defence mechanism of sporotrichosis.
We studied two unusual cases of malignant histiocytosis (MH). In one case, a solitary intracranial tumor developed in a 20-year-old man eight months before the characteristic symptoms of MH appeared. In the other, a 73-year-old man had multiple nodular skin lesions four months before malignant histiocytes appeared in his blood. In both, the tumor cells were identified as malignant histiocytes by light and electron microscopic, cytochemical, and cytoimmunological criteria.
Lymphocyte subpopulations of dermal infiltrated cells on experimental sporotrichosis of guinea pigs was performed. According to Tan's method, infected sites were incubated with collagenase and then mechanically disaggregated. The liberated cells were used for determination of lymphocytes subpopulation. The predominance of B cells was present on the inoculated site, compare with blood from control and inoculated animals. Thus, the present result suggests the important role of humoral immunity in the development of sporotrichosis lesion.
A case of localized cutaneous sporotrichosis is reported. The lesion was granulomatous, without lymphatic metastasis. Histological examination revealed numerous spores and asteroid bodies. Sporotrichin reaction was negative, but after treatment with potassium iodide became positive. The peculiar features of this case seem to be due to some disturbances of cellular immunity.
