Characterization of hydrocarbons, halocarbons and carbonyls in the atmosphere of Hong Kong.

Ambient air quality measurements of 156 species including 39 alkanes, 32 alkenes, 2 alkynes, 24 aromatic hydrocarbons, 43 halocarbons and 16 carbonyls, were carried out for 120 air samples collected at two sampling stations (CW and TW) in 2001 throughout Hong Kong. Spatial variations of volatile organic compounds (VOCs) in the atmosphere were investigated. Levels of most alkanes and alkenes at TW site were higher than that at the CW site, while the BTEX concentrations at the two sites were close. The BTEX ratios at CW and TW were 1.6:10.1:1.0:1.6 and 2.1:10.8:1.0:2.0, respectively. For major halogenated hydrocarbons, the mean concentrations of chloromethane, CFCs 12 and 22 did not show spatial variations at the two sites. However, site-specific differences were observed for trichloroethene and tetrachloroethene. Furthermore, there were no significant differences for carbonyls such as formaldehyde, acetaldehyde and acetone between the two sites. The levels of selected hydrocarbons in winter were 1-5 times that in summer. There were no common seasonal trends for carbonyls in Hong Kong. The ambient level of formaldehyde, the most abundant carbonyl, was higher in summer. However, levels of acetaldehyde, acetone and benzaldehyde in winter were 1.6-3.8 times that in summer. The levels of CFCs 11 and 12, and chloromethane in summer were higher than that in winter. Strong correlation of most hydrocarbons with propene and n-butane suggested that the primary contributors of hydrocarbons were vehicular emissions in Hong Kong. In addition, gasoline evaporation, use of solvents, leakage of liquefied petroleum gas (LPG), natural gas leakage and other industrial emissions, and even biogenic emissions affected the ambient levels of hydrocarbons. The sources of halocarbons were mainly materials used in industrial processes and as solvents. Correlation analysis suggested that photochemical reactions made significant contributions to the ambient levels of carbonyls in summer whereas in winter motor vehicle emissions would be the major sources of the carbonyls. The photochemical reactivity of selected VOCs was estimated in this study. The largest contributors to ozone formation were formaldehyde, toluene, propene, m,p-xylene, acetaldehyde, 1-butene/i-butene, isoprene and n-butane, suggesting that motor vehicles, gasoline evaporation, use of solvents, leakage of LPG, photochemical processes and biogenic emission are sources in the production of ozone. On the other hand, VOCs from vehicles and gasoline evaporation were predominant with respect to reactions with OH radical.

Source apportionment of ambient non-methane hydrocarbons in Hong Kong: application of a principal component analysis/absolute principal component scores (PCA/APCS) receptor model.

Receptor-oriented source apportionment models are often used to identify sources of ambient air pollutants and to estimate source contributions to air pollutant concentrations. In this study, a PCA/APCS model was applied to the data on non-methane hydrocarbons (NMHCs) measured from January to December 2001 at two sampling sites: Tsuen Wan (TW) and Central & Western (CW) Toxic Air Pollutants Monitoring Stations in Hong Kong. This multivariate method enables the identification of major air pollution sources along with the quantitative apportionment of each source to pollutant species. The PCA analysis identified four major pollution sources at TW site and five major sources at CW site. The extracted pollution sources included vehicular internal engine combustion with unburned fuel emissions, use of solvent particularly paints, liquefied petroleum gas (LPG) or natural gas leakage, and industrial, commercial and domestic sources such as solvents, decoration, fuel combustion, chemical factories and power plants. The results of APCS receptor model indicated that 39% and 48% of the total NMHCs mass concentrations measured at CW and TW were originated from vehicle emissions, respectively. 32% and 36.4% of the total NMHCs were emitted from the use of solvent and 11% and 19.4% were apportioned to the LPG or natural gas leakage, respectively. 5.2% and 9% of the total NMHCs mass concentrations were attributed to other industrial, commercial and domestic sources, respectively. It was also found that vehicle emissions and LPG or natural gas leakage were the main sources of C(3)-C(5) alkanes and C(3)-C(5) alkenes while aromatics were predominantly released from paints. Comparison of source contributions to ambient NMHCs at the two sites indicated that the contribution of LPG or natural gas at CW site was almost twice that at TW site. High correlation coefficients (R(2) > 0.8) between the measured and predicted values suggested that the PCA/APCS model was applicable for estimation of sources of NMHCs in ambient air.

Pathogenesis and pathology of delayed xenograft rejection in pig-to-rhesus monkey cardiac transplantation.

UNLABELLED: It has been recognized that delayed xenograft rejection (DXR) is the major barrier to the acceptance of xenotransplantation after overcoming hyperacute rejection. OBJECTIVES: This study sought to investigate the pathogenesis and pathology of delayed xenograft rejection following pig-to-rhesus monkey heart xenotransplantation. METHODS: Heterotopic xenogeneic heart transplants in the abdominal cavity were performed using piglet donors to four monkey recipients. Complete complement depletion was achieved in the recipients with repetitive doses of high-activity cobra venom factor (Y-CVF). The recipients were immunosuppressed with a combination of cyclosporine, cyclophosphamide, and steroids. Sera were analyzed for C3 and C4 levels and complement activity and anti-pig endothelial xenoantibody. The grafts were examined histopathologically and immunohistochemically for C3, C4;C5b-9, IgM, IgG, tumor necrosis factor-alpha (TNF-alpha), intercellular adhesion molecule-1(ICAM-1), CD57(NK cells), CD68 (macrophages), CD4, and CD8. RESULTS: Xenografts survived 8, 10, 13, and 13 days respectively, all developing DXR. Venous thrombosis was the outstanding feature within DXR xenografts, complicated by interstitial edema, local hemorrhage, myocardial necrosis, and mild to moderate cellular infiltration. The serum C3 levels and complement activity decreased to almost 0 from the day of transplantation due to treatment with Y-CVF. The C4 level began to decrease 2 to 4 days before the cardiac xenografts lost their function. Anti-pig endothelial xenoantibody also decreased after transplantation, slightly increasing during DXR. All rejected xenografts showed C3, C4, C5b-9, IgG, and IgM deposits to various degrees. Large numbers of macrophages (50% of total leukocytes) infiltrated the entire xenograft with a few natural killer cells (8% to 10%), as well as some CD4+ T cells (15%) and CD8+ T cells (25%). Upregulation of ICAM-1 on graft endothelial cells and TNF-alpha in the interstitium were also demonstrated in the rejected heart. CONCLUSION: Both humoral and cell-mediated immunologic reactions may play important roles in the pathogenesis of DXR. Besides C3, C4, C5b-9, IgM, and IgG destroying the xenograft, NK cells, macrophages, and CD4+ and CD8+ T cells may further aggravate the development of DXR.

Risk assessment of exposure to volatile organic compounds in different indoor environments.

The lifetime cancer risks of exposure of cooks and food service workers, office workers, housewives, and schoolchildren in Hong Kong to volatile organic compounds (VOCs) in their respective indoor premises during normal indoor activities were assessed. The estimated cancer risk for housewives was the highest, and the second-highest lifetime cancer risk to VOC exposure was for the groups of food service and office workers. Within a certain group of the population, the lifetime cancer risk of the home living room was one to two orders of magnitude higher than that in other indoor environments. The estimated lifetime risks of food service workers were about two times that of office workers. Furthermore, the cancer risks of working in kitchen environments were approximately two times higher than the risks arising from studying in air-conditioned classrooms. The bus riders had higher average lifetime cancer risks than those travelling by Mass Transit Railway. For all target groups of people, the findings of this study show that the exposures to VOCs may lead to lifetime risks higher than 1 x 10(-6). Seven indoor environments were selected for the measurement of human exposure and the estimation of the corresponding lifetime cancer risks. The lifetime risks with 8-h average daily exposures to individual VOCs in individual environments were compared. People in a smoking home had the highest cancer risk, while students in an air-conditioned classroom had the lowest risk of cancer. Benzene accounted for about or more than 40% of the lifetime cancer risks for each category of indoor environment. Nonsmoking and smoking residences in Hong Kong had cancer risks associated with 8-h exposures of benzene above 1.8 x 10(-5) and 8.0 x 10(-5), respectively. The cancer risks associated with 1,1-dichloroethene, chloroform, methylene chloride, trichloroethene, and tetrachloroethene became more significant at selected homes and restaurants. Higher lifetime cancer risks due to exposure to styrene were only observed in the administrative and printing offices and air-conditioned classrooms. Higher lifetime cancer risks related to chloroform exposures were observed at the restaurant and the canteen.

Multipathway risk assessment on disinfection by-products of drinking water in Hong Kong.

The lifetime cancer risk and the hazard index of trihalomethanes (THMs) through oral ingestion, dermal absorption, and inhalation exposure from tap water in 19 districts in Hong Kong are estimated. The most dominant THMs are chloroform and bromodichloromethane (BDCM) in Hong Kong tap water. Among the three different pathways, residents have a higher risk of cancer through oral ingestion than through the other two pathways. The lifetime cancer risks through oral ingestion and dermal absorption for BDCM make the highest percentage contribution (59%) to total risks, followed by chloroform (24%). The chloroform and BDCM are at or above the negligible risk level of 10(-6) by a factor of 10 or more in most districts. Among the 19 districts, people living in Sai Kung have the highest risk of cancer due to the THM exposure through the multipathways, mainly because of the exposure to BDCM and dibromochloromethane (DBCM). The total cancer risk analysis indicates that each year approximately 10 out of the seven million Hong Kong residents could get cancer from the daily intake of water.

Indoor air quality in ice skating rinks in Hong Kong.

Indoor air quality in ice skating rinks has become a public concern due to the use of propane- or gasoline-powered ice resurfacers and edgers. In this study, the indoor air quality in three ice rinks with different volumes and resurfacer power sources (propane and gasoline) was monitored during usual operating hours. The measurements included continuous recording of carbon monoxide (CO), carbon dioxide (CO(2)), total volatile organic compounds (TVOC), particulate matter with a diameter less than 2.5 microm (PM(2.5)), particulate matter with diameter less than 10 microm (PM(10)), nitric oxide (NO), nitrogen dioxide (NO(2)), nitrogen oxide (NO(x)), and sulfur dioxide (SO(2)). The average CO, CO(2), and TVOC concentrations ranged from 3190 to 6749 microg/m(3), 851 to 1329 ppm, and 550 to 765 microg/m(3), respectively. The average NO and NO(2) concentrations ranged from 69 to 1006 microg/m(3) and 58 to 242 microg/m(3), respectively. The highest CO and TVOC levels were observed in the ice rink which a gasoline-fueled resurfacer was used. The highest NO and NO(2) levels were recorded in the ice rink with propane-fueled ice resurfacers. The air quality parameters of PM(2.5), PM(10), and SO(2) were fully acceptable in these ice rinks according to HKIAQO standards. Overall, ice resurfacers with combustion engines cause indoor air pollution in ice rinks in Hong Kong. This conclusion is similar to those of previous studies in Europe and North America.

Identification of Escherichia coli O172 O-antigen gene cluster and development of a serogroup-specific PCR assay.

AIM: To characterize the locus for O-antigen biosynthesis from Escherichia coli O172 type strain and to develop a rapid, specific and sensitive PCR-based method for identification and detection of E. coli O172. METHODS AND RESULTS: DNA of O-antigen gene cluster of E. coli O172 was amplified by long-range PCR method using primers based on housekeeping genes galF and gnd Shot gun bank was constructed and high quality sequencing was performed. The putative genes for synthesis of UDP-FucNAc, O-unit flippase, O-antigen polymerase and glycosyltransferases were assigned by the homology search. The evolutionary relationship between O-antigen gene clusters of E. coli O172 and E. coli O26 is shown by sequence comparison. Genes specific to E. coli O172 strains were identified by PCR assays using primers based on genes for O-unit flippase, O-antigen polymerase and glycosyltransferases. The specificity of PCR assays was tested using all E. coli and Shigella O-antigen type strains, as well as 24 clinical E. coli isolates. The sensitivity of PCR assays was determined, and the detection limits were 1 pg microl(-1) chromosomal DNA, 0.2 CFU g(-1) pork and 0.2 CFU ml(-1) water. The total time required from beginning to end of the procedure was within 16 h. CONCLUSION: The O-antigen gene cluster of E. coli O172 was identified and PCR assays based on O-antigen specific genes showed high specificity and sensitivity. SIGNIFICANCE AND IMPACT OF THE STUDY: An O-antigen gene cluster was identified by sequencing. The specific genes were determined for E. coli O172. The sensitivity of O-antigen specific PCR assay was tested. Although Shiga toxin-producing O172 strains were not yet isolated from clinical specimens, they may emerge as pathogens.

Shear-induced undulation of smectic-A: Molecular Dynamics simulations vs. analytical theory.

Experiments on a variety of systems have shown that layered liquids are unstable under shear even if the liquid layers are planes of constant velocity. We investigate the stability of smectic- A like liquids under shear using Molecular Dynamics simulations and a macroscopic hydrodynamic theory (including the layer normal and the director as independent variables). Both methods show an instability of the layers, which sets in above a critical shear rate. We find a remarkable qualitative and reasonable quantitative agreement between both methods for the spatial homogeneous state and the onset of the instability.

The pathogenetic role of immunoglobulin G from patients with systemic lupus erythematosus in the development of lupus pleuritis.

OBJECTIVE: To investigate the pathophysiological effect of immunoglobulin G (IgG) from systemic lupus erythematosus (SLE) patients on pleural mesothelial cells and related mechanisms. METHODS: Serum IgG from 28 lupus patients and 13 healthy controls was purified by protein-G affinity chromatography. The concentrations of anti-dsDNA-, anti-histone- and/or anti-nucleohistone-containing IgGs were determined by enzyme-linked immunosorbent assay (ELISA). Lupus patients were divided into an active (n = 12) and an inactive group (n = 16) on the basis of the SLE Disease Activity Index (SLEDAI). The binding of IgG to a human pleural mesothelial cell line (MeT-5A) under different conditions, including pretreatment with DNase and preincubation with exogenous histone, DNA or nucleohistone, was examined using flow cytometry and cellular ELISA. The effect of IgG on MeT-5A cell proliferation was studied using an MTT assay. Gene expression and protein synthesis for interleukin 1beta (IL-1beta), monocyte chemoattractant protein 1 (MCP-1) and transforming growth factor beta1 (TGF-beta1) in MeT-5A cells were determined using reverse transcription-polymerase chain reaction and ELISA. RESULTS: The binding of IgG to MeT-5A cells was higher in the active lupus group than the inactive lupus group (P = 0.047) and controls (P = 0.003). The binding decreased in both lupus groups following pretreatment of MeT-5A cells with DNase. The binding of IgG to MeT-5A cells was greater by 112% in the active lupus group after preincubation with histone (P < 0.001), but not with DNA or nucleohistone. Exposure of MeT-5A cells to IgG from either lupus group induced cell proliferation when compared with IgG from healthy controls (P = 0.04). Gene expression and protein synthesis of MCP-1, TGF-beta1 and IL-1beta in MeT-5A cells were significantly increased after incubation with IgG from patients with active lupus when compared with IgG from the inactive lupus and control groups (P < 0.01). The concentration of anti-dsDNA antibodies correlated with the binding of IgG to MeT-5A cells and the synthesis of cytokines by MeT-5A cells. The serum level of anti-histone antibodies in the active lupus group was higher than that in the inactive group (P = 0.015) and the serum concentration correlated with cell binding and MCP-1 production. CONCLUSIONS: IgG from lupus patients can bind to MeT-5A cells and the binding is modulated by DNA or histone. Binding of anti-dsDNA-containing IgG to MeT-5A cells induces the synthesis of proinflammatory cytokines. Our findings suggest that the binding of anti-dsDNA antibodies, particularly the IgG isotype, to pleural mesothelium plays a direct pathogenetic role in inducing inflammatory injury in the serositis of SLE.

Nitric oxide inhibits expression of cytochrome B in endotoxin-stimulated murine macrophages.

In LPS-mediated states of sepsis, inducible nitric oxide synthase (iNOS) expression and nitric oxide (NO) production inhibit cellular respiration and mitochondrial electron transport. NO has been demonstrated to inhibit mitochondrial respiration by nitrosylation of the iron-sulfur centers of aconitase, complex I (NADH-ubiquinone oxidoreductase), complex II (succinate-ubiquinone oxidoreductase), and complex IV (cytochrome c oxidase). However, little is known of the effect of NO on expression of critical proteins in the electron transport chain. In ANA-1 murine macrophages, LPS-mediated NO synthesis decreases Cyt b protein expression and steady-state mRNA levels. Mitochondrial run-on analysis demonstrates unaltered Cyt b mitochondrial gene transcription. In this study utilizing LPS-stimulated ANA-1 murine macrophages, we demonstrate that expression of the mitochondrial protein, Cyt b, is significantly decreased as the result of a unique and previously unknown, NO-dependent posttranscriptional regulatory mechanism. Alteration in the voltage dependence of NMDA receptor channels in rat dorsal horn neurones following peripheral inflammation.

1. It has been proposed that the activation of NMDA receptors and upregulation of protein kinase C (PKC) underlie the exaggerated and persistent pain experienced in the inflammatory state. However, there is no direct evidence to show that inflammation alters the function of NMDA receptors. 2. We examined the voltage-dependent properties of NMDA receptor channels in rat dorsal horn neurones that receive sensory inputs from an inflamed hindpaw. 3. Peripheral inflammation was induced by injections of complete Freund's adjuvant (CFA). Membrane currents were measured using the perforated patch-clamp technique. 4. After CFA treatment, the current-voltage relationship of NMDA receptor channels was shifted in the hyperpolarized direction. This resulted in enhanced NMDA responses at negative potentials. 5. The change was mediated by PKC because the voltage shift was blocked by the selective PKC inhibitors chelerythrine and bisindolylmaleimide I. 6. Furthermore, the Mg(2+) blockade of NMDA receptors was reduced. This reduction could account for the shift in the voltage dependence of NMDA receptor channels. 7. These results indicate that NMDA receptor channel characteristics in the dorsal horn are altered by inflammation, and that the changes observed could contribute to the hyperalgesia and allodynia associated with tissue injury.

Potassium silver tin selenide, K(2)Ag(2)Sn(2)Se(6).

The title compound was synthesized by a reactive salt reaction at 773 K over a period of 5 d. It has a one-dimensional chain structure consisting of K(+) cations and one-dimensional [Ag(2)Sn(2)Se(6)](2-) anions. The chain is constructed by edge-sharing bitetrahedral [Sn(2)Se(6)] units connected in a 1:2 ratio via linear Ag(+) ions.

[The protective effect of low molecular weight heparin on early nephropathy in diabetic rats].

OBJECTIVE: To investigate the protective effect of low molecular heparin on early nephropathy in diabetic rats and to explore its possible mechanism. METHODS: The rats were randomly divided into following groups: normal control rats, streptozotocin (STZ, 65 mg/kg)-induced diabetic rats and diabetic rats treated with low molecular weight heparin (Fraxiparine, Nadroparin calcium 205 AXaIU/kg). The activity of anti-Xa factor and the activated partial thromboplastin time (APIT) in plasma were assayed, the renal function and profile of kidney hypertrophy were measured, and the renal tissue were observed by light microscopy after 1, 2, 4 weeks of the treatment respectively. RESULTS: Compared with the untreated diabetic group, except for higher kidney hypertrophy index, LMWH treated diabetic rats presented less urinary albumin excretion, lower creatinine clearance rate (Ccr) and longer APTT than diabetic group without any treatment. Meanwhile, renal pathologic changes in LMWH treated group were improved. CONCLUSION: Low molecular weight heparin possesses a renal protective effect on early diabetic nephropathy and its possible mechanism may be associated with the beneficial effect of LMWH on regulating abnormal diabetic renal hemodynamics.

CD147 is a signaling receptor for cyclophilin B.

Cyclophilins A and B (CyPA and CyPB) are cyclosporin A binding proteins that can be secreted in response to inflammatory stimuli. We recently identified CD147 as a cell-surface receptor for CyPA and demonstrated that CD147 is an essential component in the CyPA-initiated signaling cascade that culminates in ERK activation and chemotaxis. Here we demonstrate that CD147 also serves as a receptor for CyPB. CyPB induced Ca(2+) flux and chemotaxis of CD147-transfected, but not control, CHO cells, and the chemotactic response of primary human neutrophils to CyPB was blocked by antibodies to CD147. These results suggest that CD147 serves as a receptor for extracellular cyclophilins.

Excess Kondo resonance in a quantum dot device with normal and superconducting leads: the physics of Andreev-normal co-tunneling.

We report on a novel Kondo phenomenon of interacting quantum dots coupled asymmetrically to a normal and a superconducting lead. The effects of intradot Coulomb interaction and Andreev tunneling give rise to Andreev bound resonances. As a result, a new type of co-tunneling process, which we term Andreev-normal co-tunneling, is predicted. At low temperatures, coherent superposition of these co-tunneling processes induces a Kondo effect in which Cooper pairs directly participate formation of a spin singlet, leading to four Kondo resonance peaks in the local density of states, and enhancing the tunneling current.
