[Eugenic significance of linkage between the gene loci for dystrophia myotonica (Steiner's disease) and ABH-secretor].

36 relatives of 2 patients with myotonic dystrophy (M.d.) were investigated during a field study. All were neurologically examined at home and their ABO blood groups and ABH-secretor phenotypes in parotid saliva were determined for linkage analyses. Additional investigations such as EMG or slit-lamp examinations were carried out by specialists in doubtful cases. Nine additional patients presenting with varying clinical manifestations of M.d. were found. In most of the secretor-positive individuals the corresponding genotypes could be identified as heterozygotes (Sese) through the segregation pattern of their first-degree relatives. In both families the allele for M.d. was linked to the dominant allele Se, the probability being greater than 99%. A recombination was observed in 1 patient of either family. This is, however, fairly consistent with the expected recombination fraction of 0.08. No further linkage information was obtained from determinations of the Lutheran blood groups carried out in family no. 1 only, since all individuals were of the most frequent type Lu (a-/b+). In 5 out of 8 matings between M.d. patients and healthy spouses the secretor genoypes were potentially informative for the estimation of risk of their children being affected or for prenatal diagnoses. The secretor analyses and special tests for the detection of preclinical cases (e.g. by slit-lamp) and their practical value for genetic counseling are discussed, especially with regard to the limitations, which derive partly from low probabilities in the predictions even in some of the so-called informative matings, and partly also from the recombinations to be expected.

[Horners syndrome. A complication of the lateral recumbent position (author's transl)].

The appearance of a unilateral Horner's syndrome after an anesthetic in the lateral recumbent position is reported. It was caused by a damage to the sympathetic nerve at the level of Th 1 probably due to the extreme lateral flexion of the cervical vertebral spine. The importance of correct positioning of the head during anesthesia is stressed.

Bursa of Fabricus: uptake of radioactive particles and radiotoxic "sealing" of bursal follicles.

Making use of the fact that the avian bursa is uniquely equiped for uptake, degradation and storage of particulate matter from the cloacal lumen, we administered 239Pu(IV) perorally or via the cloaca to cause radiotoxic damage to the bursal tissue. This resulted in a involution of the follicles, reduction of the lymphocyte mass, predominance of smaller lymphoid cells in the medulla as compared to controls, disappearance of tuft epithelia, formation of a sub-epithelial fibrous layer in many follicles and impairment or an almost complete loss of the capacity of the bursa to take up particulate matter. Concomitant with these changes was a reduction in the concentration of serum IgG. These findings are discussed in relation to the immunologic contact role of the bursa.

Studies on the regulation of the neutrophil chemotactic response using a rapid and reliable method for measuring random migration and chemotaxis of neutrophil granulocytes.

An economic and sensitive test system for measuring random and directional migration of human neutrophils is described. The technique, based on a modified Boyden chamber equipped with a two-filter system, permits a substantial reduction of both incubation time and sample volume. The influence of various technical factors such as the neutrophil concentration in the cell suspension, the incubation time of the chambers, the test concentration of activated plasma or serum, the presence of heparin, and the procedure for separating neutrophils from human peripheral blood, was investigated. Standardized procedures for measuring and reporting neutrophil chemotaxis are proposed. The method has been used to study the significance of factors regulating neutrophil migration such as cytotaxin inactivators and neutrophil immobilizing factors (NIF). Activity of cytotaxin inactivators as assessed in undiluted serum or plasma at pH 7.4, 6.0 or 4.0 was very low. In contrast, potent neutrophil immobilizing activity was found in human serum or diluted plasma. These factors which inhibit migration were accordingly termed neutrophil immobilizing factors of plasma (NIF-P) and neutrophil immobilizing factor of serum (NIF-S). These factors are heat-stable, non-dialysable and of high molecular weight.

[Studies on the thickness of the hard tissue coating in the 1st upper deciduous molars with special reference to the shape of the crown].

The report is on metric examinations of the thickness of the hard substance coating of the first upper milk molars based on 122 tooth cuts (longitudinal and transversal) and on the shape of the crowns of these teeth. The measurements were made by means of a stereomicroscope SM XX number 551 190 (VEB Carl Zeiss, Jena), and the results were statistically evaluated.

The cells of equine blood and their development.

Based on morphologie studies on bone marrow, lymphnode and blood preparations the cells of equine blood and their developmental stages are described and illustrated with 32 coloured photographs.

[Adherence of pathogenic Escherichia coli to epithelial cells isolated from the intestinal mucosa of the rabbit, inhibiting effect of hyperimmune bovine colostrum and of various carbohydrates (author's transl)].

In order to study the mode of action of a bovine anti-Escherichia coli lactoserum (BLS), we have used a new test measuring the adherence of pathogenic E. coli on epithelial cells isolated from the small intestine of rabbit. A mixed suspension of E. coli and of epithelial cells is incubated for 15 min and the number of bacteria adhering to the cells counted under the microscope. The BLS at a concentration of 3.5 mg/ml IgG is able to reduce this number by a factor of 3-5. After absorption of the BLS with formaldehyde-treated bacteria, this factor is smaller than 2. At a concentration of 5 mg/ml, D-mannose and alpha-methylmannoside are as efficient inhibitors of adherence as BLS; at the same concentration, L-mannose is ineffective. The cultures of E. coli strongly agglutinating guinea pig erythrocytes, adhere to a larger extent to the epithelial cells. The last two observations confirm the important role played by fimbriae for the adhesive properties of E. coli. The presence of fimbrial antibodies would partially explain the inhibiting effect of BLS on adherence.
