Structural and functional characterization of potent antithrombotic oligonucleotides possessing both quadruplex and duplex motifs.

We report the results of a selection for single-stranded DNA oligonucleotide ligands to the serine protease thrombin using recently developed methods. This selection yielded a family of DNA sequences that conform to a consensus structure comprised of a unimolecular quadruplex motif and complementary flanking sequences capable of forming an additional Watson-Crick duplex motif. This novel quadruplex/duplex structure was not reported in a previous selection for DNA molecules which bind to thrombin [Bock et al. (1992) Nature 355, 564-566]. All quadruplex/duplex molecules tested bound to thrombin with higher affinity than quadruplex structures lacking the duplex structure. However, binding affinities did not always correlate with inhibitory potency since some molecules with high affinity were not potent inhibitors in vitro. 1H NMR spectroscopy studies demonstrated that the complementarity of bases in the duplex portion of a selected sequence allows it to form multimolecular structures. Constraining these molecules to the unimolecular quadruplex/duplex structure by bridging the 5' and 3' ends of the duplex motif with either triethylene glycol or disulfide bonds improved their thrombin inhibitory activity. All bridged quadruplex/duplex molecules were more potent inhibitors than molecules with only a quadruplex motif. Bridging the ends of these structures not only increased thrombin inhibition but also improved resistance to nucleases in serum more than 40-fold over the unbridged quadruplex. In addition, we have found that both the length and sequence of the duplex motif are important for inhibition.

Stabilization of double-stranded oligonucleotides using backbone-linked disulfide bridges.

A convenient, practical route to the synthesis of disulfide-bridged oligonucleotides has been developed. Aliphatic linkers with terminal thiol groups have been attached to the phosphodiester backbones of partially or fully complementary oligonucleotide sequences and oxidized to yield covalently closed oligonucleotides with disulfide bridges. This procedure has been used to prepare a duplex with disulfide bridges at both ends and stem-loop sequences with single disulfide bridges. Oxidation of a self-complementary duplex possessing terminal thiol groups produced both hairpin and duplex structures with disulfide bridges, the relative proportions of each being dependent upon the reaction conditions. These bridged hairpin and duplex structures were shown to be interconvertible by reduction and re-oxidation. The melting profiles of disulfide-bridged oligonucleotides were compared with the same sequences without bridges and with sequences possessing triethylene glycol bridges, and in all cases the introduction of disulfide bridges resulted in a considerable increase in thermal stability. EcoRI endonuclease was capable of cleaving a disulfide-bridged duplex possessing a recognition site for this enzyme, thus supporting a lack of distortion of the recognition site. The disulfide bridges could be cleaved using a large excess of DTT to regenerate the corresponding sulfhydryl compounds. A study of the serum stabilities of disulfide-bridged oligonucleotides showed that the bridged duplexes were much more stable than their unmodified counterparts, whereas the rate of degradation of the stem-loop structures was more dependent upon the size of the loop than the presence or absence of the disulfide bridge. In summary, we have described a novel methodology, employing commercially available reagents, for the stabilization of oligonucleotide duplexes or stem-loop structures by disulfide bridge formation.

Basic fibroblast growth factor in retinal development: differential levels of bFGF expression and content in normal and retinal degeneration (rd) mutant mice.

The spatial and temporal patterns of expression and content of bFGF during postnatal development of the retina were established in C57BL/6J mice. Western blot analysis, using an anti-rodent bFGF antibody, shows multiple molecular weights of 18, 20.5, and 22 kDa of bFGF protein isolated from the adult retina. A bioassay indicates that this putative basic fibroblast growth factor (bFGF) stimulates proliferation of BALB/c 3T3 fibroblasts in a dose-dependent manner identical to an authentic bFGF standard. Immunocytochemistry reveals that bFGF immunoreactivity is located primarily in the immature photoreceptors during postnatal development and is associated with the photoreceptor outer segment/interphotoreceptor matrix complex in the adult retina. bFGF mRNA expression pattern and levels were evaluated using mouse bFGF riboprobes with in situ hybridization and quantitative ribonuclease protection assay. bFGF mRNA expression is not detectable in the retina until Postnatal Day 10 (P10), although high levels of bFGF mRNA signals were consistently observed in astrocytes in the optic disc at all postnatal ages examined. From P10 to the adult stage, bFGF mRNA was localized mainly to the photoreceptor inner segments, and the bFGF mRNA levels were approximately the same at P10 and in the adult retina. The patterns of retinal bFGF expression and content during normal development established above were compared to these parameters in the retina of rd (C57BL/6J rd/rd), a spontaneous mouse mutant in which photoreceptors degenerate shortly after birth. More bFGF immunoreactivity was detected in the outer retina during photoreceptor degeneration than was present in normal photoreceptors at equivalent ages. Densitometry measurements indicate that the level of immunoreactivity is 56% to 1.8-fold higher in rd than in the normal retina between P6 and P10, respectively. This is at least partially due to elevated bFGF mRNA expression in rd retinas during photoreceptor degeneration. In situ hybridization showed more intense bFGF mRNA hybridization signals in rd photoreceptors from P10 to P15, and RNase protection assay demonstrated much higher hybridization signals in rd retinas from P6 to P10 than in the normal retinas at these stages. More bFGF mRNA hybridization signals were also present in some cells in the inner nuclear layer following photoreceptor cell death in the rd retina but were only weakly evident in the inner nuclear layer in the normal adult retina. These results provide the first evidence that a naturally occurring neuronal degeneration is accompanied by elevated expression of bFGF in degenerating neurons prior to cell death.(ABSTRACT TRUNCATED AT 400 WORDS)

Immune cell infiltration and persistence in the mouse trigeminal ganglion after infection of the cornea with herpes simplex virus type 1.

Following inoculation of the mouse cornea with herpes simplex virus type 1 (HSV-1), the spread of virus was investigated and the types of immune cell infiltrating the trigeminal ganglion (TG) were identified in low temperature paraffin wax sections. Virus antigen was first found on day 3 and was absent after day 14. Early presentation of antigen to T cells may occur since increased expression of major histocompatibility complex (MHC) class II antigens, including de novo expression on satellite and Schwann cells, was detected in foci of such antigen on day 3. A second large peak of such expression was detected on day 10 together with increasing numbers of B and T cells. Large numbers of these lymphocytes and extensive expression of MHC class II were seen in the TG well into the phase of virus latency; the significance of this is discussed.

Immunosensing with photo-immobilized immunoreagents on planar optical wave guides.

Immunocomplexation at wave guiding TiO2/SiO2 surfaces was investigated using an integrated optical grating coupler. For extended application of this label-free monitoring system, F(ab')2 fragments of monoclonal antibodies were photo-immobilized by photolinker polymer-mediated procedures that do not require functionalization of either the immunoreagent or the TiO2/SiO2 surface. Covalent, light-dependent binding of photolinker polymer and F(ab')2 fragments was achieved using a single-step photo-reaction. Bovine serum albumin derivatized with aryldiazirines (T-BSA) served as a photolinker polymer. T-BSA suppressed the non-specific adsorption of analytes to wave guide surfaces. Immunoreagent binding and immunological activity were analyzed and modified surfaces were investigated by scanning force microscopy. Apparent immunoreagent surface densities were 16.7 fmol F(ab')2 per mm2 sensor surface. Optical analyses revealed linear, dose-dependent antigen binding with label free analytes. Immunocompetent surfaces were regenerable by treatment at pH 2.3, rendering the immunosensing system applicable for repetitive use.

Hydrophobic contribution constants of amino acid residues to the hydrophobicities of oligopeptides.

PURPOSE: The main purpose of this study is to explore the additive-constitutive nature of common amino acids in their contribution to the partition coefficients of small peptides. METHODS: The Log P values and other physico-chemical parameters of the peptides studied are taken from the literature. The frequency of appearance (ni) of each individual amino acid is calculated as the number of the amino acids in a given peptide. RESULTS: The partition coefficients (Log P(oct./buff.) at pH 7) of 87 N-acetyl-peptide-amides have been correlated with the frequency of appearance of amino acids. From the correlation obtained, the de novo hydrophobic contribution constants of 19 amino acid residues are derived for the first time. The contribution constants are extended to 59 unmodified regular peptides with the inclusion of the pka values of both N-terminal and C-terminal amino acids. The models thus obtained have been validated with additional 27 peptides (both N-acetyl-peptide-amides and unmodified). CONCLUSIONS: The Log P of oligopeptides is very well correlated with the de novo hydrophobic contribution constants of amino acids. The models we have derived are reasonably accurate in predicting the hydrophobicities of new oligopeptides (< or = tetrapeptides) at a fixed pH (e.g., 7).

QSAR analysis of skin permeability of various drugs in man as compared to in vivo and in vitro studies in rodents.

A general mathematical model involving partition coefficient, molecular weight and hydrogen bonding has been formulated for correlating the structures and skin permeability of a wide range of compounds through human skin and through hairless mouse skin. The correlations obtained are dependent not only on the biological system but also on the vehicle used. Without the use of lipophilic vehicle, the ideal lipophilicity for maximum permeability through human skin as measured by log Po(oct/w) ranges from 2.5 to 6 (extrapolated value). When a lipophilic vehicle was used in hairless mouse skin study, the log Po(oct/w) was lowered to around 0.4 approximately 0.6. While increased M.W. always has a negative effect on the permeability, increased H-bond can have either a slight positive or a slight negative effect, depending on the experiments (absorption vs. permeability constant). Cross validations with previously unanalyzed data as well as other biological systems support the usefulness of the general model developed for passive diffusion.

Photochemical linkage of antibodies to silicon chips.

Antibodies and antigen binding fragments thereof were photochemically immobilized on surface-modified silicon chips of 5 x 5 mm size. Silicon surface-grafted diazirines and benzophenones formed covalent bonds with the immunoreagents on light activation. Photolithographic immobilization of monoclonal antibodies in aqueous media was achieved on silicon chips by activating surface-grafted benzophenones. The presence of bovine serum albumin during irradiation reduced nonspecific adsorption of the immunoreagents and retained the immunoactivity of the photoimmobilized molecules.

[Histopathological and immunohistochemical study of ovarian polyembryoma].

A case of polyembryoma which is rare in ovarian malignant germ cell tumors was reported. Microscopically, numerous and variable embryoid bodies, surrounded by embryonic hepatoid tissues, trophoblastic cells and multiple elements, were observed. The results of immunohistochemical analysis were: (1) alpha fetoprotein (AFP) was not only shown positive in yolk sac cells among the embryoid bodies, in the endoderm and in the margins of the ectodermal cells, but also in multiple elements in the mucinous matrix. (2) Hepatoid tissues reacted positively to AFP and alpha antitrypsin (AAT) as well as to hCG. (3) By using hCG and human placental lactogen (hPL) as markers it is proven that among trophoblastic cells of the tumor, not only syncytiotrophoblastic cells but also the intermediate cells and the cytotrophoblastic cells could be identified.

[Isolation of the lower esophagus and fundus of the stomach by specially designed forceps to avoid ectopic embolization during the process of coronary venous embolizing therapy].

To avoid ectopic embolization occasionally occuring in embolization therapy of coronary vein of the stomach, the authors designed forceps to temporarily isolate the lower esophagus and the gastric fundus. A series of 61 patients with portal hypertension were treated with this method from march 1990 to march 1994 with no ectopic embolization. The operative mortality was 1.6%. The patients were followed up for six months to 4.5 years, with a rebleeding rate of 6%, and an improved varices in 85.4% of all cases. This is a reliable method to avoid ectopic embolization and the individualization of embolitic dosage.

[Effects of 24 Chinese medicinal herbs on nucleic acid, protein and cell cycle of human lung adenocarcinoma cell].

Using flow cytometry the authors analysed the effect of 24 Chinese medicinal herbs (CMH) in compound recipe on proliferation index (PI), DNA index, protein index and ratio of various phases in cell cycle of human lung adenocarcinoma cell (SPC-A-1), The PI were more than 20%, in 4 CMH, while 3 CMH such as Gynostemma pentaphyllum, Glehnia littoralis, Panax Ginseng, could strengthen the body resistance. That suggested using CMH of strengthening body resistance not only served as conventional tonice but also as tumor cell inhibitor. Meanwhile the action point of 24 CMH on cell cycle were different. Therefore according to these results a new recombined Chinese recipe would be more effectively used for clinical practice.

[Electroacupuncture promotes enlargement of adrenals and enhances level of blood corticosterone in ovariectomized rats].

The present article reports EA with special acupoints may promote the enlargement of bilateral adrenals and enhance the level of blood corticosterone in ovariectomized (OV) rats. The mean of adrenal weight (M +/- SE, mg) as following, OV + EA group 71 +/- 2.1, OV group 57.4 +/- 4.7, EA group 57.6 +/- 1.1 and control group (CT group) 54 +/- 2.8 respectively. For observation of a self control, the detection of blood corticosterone contents was performed in the same animals before and after ovariectomy. The results showed that before ovariectomy the contents of blood corticosterone in the rats were 3.27 +/- 0.7 micrograms/100ml before EA and 3.63 +/- 0.9 micrograms/100ml after EA. No significant difference of blood corticosterone level was found before and after EA. After the animals ovariectomized the level of blood corticosterone increased from 4.38 +/- 0.62 micrograms/100ml to 6.02 +/- 0.77 micrograms/100ml following EA treatment (P < 0.01). No similar effect of EA treatment with control acupoints in OV rats was seen. These results suggest that EA may regulate the abnormal function of endocrine and this effect of EA possesses relative specificity of acupoint.
