A rat model for study of bilirubin conjugation by a cultured cell/ artificial capillary liver assist device.

The Gunn rat, congenitally lacking the conjugating enzyme glucuronyl transferase, provides an animal model for the study of bilirubin conjugation by an extracorporeal liver assist device (LAD) composed of cultured cells and artificial capillaries. Rat hepatoma cells of the H4-II-E cell line able to form conjugated bilirubin glucuronide from bilirubin were grown to tissue density by circumfusion culture of the LAD before symbiotic hemoperfusion experiments lasting up to 24 hours. Rat bile was analyzed by thin layer chromatography for bilirubin glucuronide which was formed by the device and then excreted by the rat.

Personal experience with tympanoplasty in children.

An analysis is presented of a series of 434 operations in chronic otitis media in children taking into account the extent of the lesion, the type of operation, the early and late functional and morphological results. In 18% of cases a radical operation was performed, in 21% atticotomy or other conservative operations, and in 61% open or closed tympanoplasty. Because of dynamic character of the cholesteatomatous process in childhood, the creation of a "safe ear", i.e. free of recurrences of cholesteatoma, is difficult by means of closed tympanoplasty operations.

Mechanism of heparin protection in endotoxin shock.

Blood clearance by phagocytosis, predominantly by the liver, presents an important mechanism for the elimination and detoxification of endotoxin. However, its effectiveness alone does not determine the outcome of an experimentally induced standardized endotoxin shock. Consequently other effects elicited by endotoxin--for instance, those on the coagulation mechanism or on an endotoxin-detoxifying enzyme system in plasma--must also play an important role in determining the ultimate fate of this condition, for it is now abundantly clear that interrupting such effects with heparin provides considerable protection.
