Iodine content, protein content and percentage of thyroglobulin to total protein in the thyroid glands were studied in two groups of 10 patients with Hashimoto's thyroiditis (5 euthyroid and 5 hypothyroid patients) and in 6 normal subject. Iodine content was significantly decreased in both hypothyroid (24 +/- 29 microgram/g wet weight of thyroid tissue, mean +/- S.D., p less than 0.01) and euthyroid (96 +/- 52, p less than 0.01) groups of Hashimoto's thyroiditis compared with those in controls (779 +/- 37). The value of hypothyroid group was significantly smaller than that of euthyroid group (p less than 0.05). A significant negative correlation was obtained between thyroidal iodine content and serum TSH (r = -0.68, y = -0.41 chi + 55.6, p less than 0.05). Thyroidal protein and percentage of thyroglobulin to total thyroidal protein were also decreased in Hashimoto's thyroiditis compared with controls, while there was no difference between euthyroid and hypothyroid groups in these values. The percentage of thyroglobulin-bound 131I obtained by in vitro iodination of thyroglobulin in Hashimoto's thyroiditis appeared to be decreased in 2 cases of hypothyroid group. Some immunological reaction occurring in Hashimoto's thyroiditis may change the structure of thyroglobulin and in turn the normal iodination of thyroglobulin may be disturbed.

Serum free thyroxine (T4) was measured by a solid-phase radioimmunoassay in various thyroid states and in those associated with changes in serum T4-binding proteins. And it was compared with the value obtained by product of total serum T4 and of per cent free T4 by equilibrium dialysis. Serum free T4 concentrations were found to be 1.34 +/- 0.33 ng/100 ml (mean +/- S.D.) in 51 normal subjects, 5.02 +/- 1.41 ng/100 ml in 29 hyperthyroid patients, 0.16 +/- 0.15 ng/100 ml in 12 hypothyroid patients, and the value in each individual of the latter two groups was significantly different from normal. Serum free T4 was normal of 1.23 +/- 0.22 ng/100 ml in 7 pregnant women and all had free T4 levels within the normal range. Moreover, five out of six patients with decreased thyroxine-binding globulin had normal free T4 levels, though their mean value (0.85 +/- 0.20 ng/100 ml) was significantly lower than the normal value. An excellent correlation (r = 0.93) was observed between values for serum free T4 obtained by the present method and those by the equilibrium dialysis method. The simplicity, accuracy, sensitivity, and specificity of this assay render it highly attractive for routine clinical determination.
Distribution of 5-fluorouracil (5-FU) in various tissues including transplanted tumors was examined 1, 3 (or 4), and 24 hr after a single oral dose of 5'-deoxy-5-fluorouridine (5'-DFUR), Ftorafur (FT-207), or 5-FU itself to groups of mice bearing sarcoma-180 and of rats bearing Walker carcinoma-256. The levels of 5-FU derived from 5'-DFUR were highest in the tumors among the tissues examined 3 (or 4) and 24 hr after administration. At the 1st hr after dosing the 5-FU level in the small intestine was close to, but that in other tissues was much lower than, the 5-FU level in the tumor of both types. Such a selective distribution of 5-FU in tumor tissues was not observed after dosing of either FT-207 or 5-FU itself. Since the anticancer effect of 5'-DFUR is likely to be manifested after its conversion to 5-FU, the 5'-deoxyribose moiety of 5'-DFUR may be deemed as an efficient carrier by which the 5-FU moiety is conveyed selectively to cancer tissues.
