In vitro effect of cadmium on primary antibody response to T-cell independent antigen (DNP-Ficoll).

The effect of various concentrations of cadmium on the in vitro primary antibody response to T-cell independent antigen (DNP-Ficoll) was investigated. Anti DNP-Ficoll antibody response in mice was significantly enhanced by 8 microM cadmium, but suppressed by 20 and 40 microM cadmium. However, cell viabilities were decreased by cadmium exposure with dose-dependent relationships. The results suggest that in vitro exposure to cadmium produces different effects on antibody responses.

Suppression of IgE antibody production after exposure to ozone in mice.

The effect of ozone exposure on IgE antibody production with aerosolized ovalbumin (OA) administration was investigated in Balb/c mice. Mice were continuously exposed to 0.8 ppm ozone for 1, 2, or 4 weeks, respectively, and subsequently aerosolized OA was administered through the respiratory tract for 6 min with a nebulizer. The mice were then immunized intraperitoneally 1 week later with OA. IgE antibody production was suppressed in ozone-exposed mice. However, no significant difference in the primary IgE antibody production by intraperitoneal immunization alone with OA was observed between ozone-exposed mice and nonexposed mice. In order to elucidate the suppressive mechanism of IgE antibody production, hapten-carrier antigenic system was used. It is shown that the induction of helper T cell function was suppressed if the aerosolized carrier protein was administered before intraperitoneal immunization in the mice exposed to ozone. These results suggest that ozone exposure has the effect on the stage of administration of inhaled antigen and the quite insignificant effect on the IgE antibody production after intraperitoneal immunization with OA.

[Clinical study on unmodified intravenous dried ion-exchange resin treated human normal immunoglobulin, SM-4300].

Unmodified intravenous dried ion-exchange resin treated human normal immunoglobulin, SM-4300, was administered to 4 patients with severely intractable infectious diseases. Obvious antipyretic effect, opsonic effect and and investigational findings on healing are not observed. However, in both subjective and objective clinical effects, we obtained the following test results, that is, 1 case of effective and 2 cases of rather effective. No side effects by SM-4300 were observed.

Enhancement of antibody response in Japanese quails by acute NO2 exposure.

The effect of acute exposure to nitrogen dioxide (NO2) on anti-SRBC antibody response in two lines of Japanese quails, high and low responder, was investigated. Mean survival time of 11- to 13-week-old quails after exposure to 20 ppm NO2 was 12.0 hr. The high responder line lived longer than the low responder line. Changes of spleen weights of both lines were not observed in 10- and 20-ppm NO2 exposure for 4 hr. Anti-SRBC antibody response in high responder quail was significantly enhanced by 20-ppm NO2 exposure for 4 hr and the response in low responder quail was slightly enhanced. A similar, although somewhat less pronounced, tendency was observed also after 10-ppm NO2 exposure. These results suggest that acute NO2 exposure enhances the antibody response of Japanese quails.

Effect of short-term exposure to O3 on antibody response in mice.

The effect of short-term exposure to ozone (O3) on antibody response in mice was investigated. BALB/c mice were continuously exposed to 0.4 and 0.8 ppm O3 for 1, 3, 7, and 14 days. Lung weights increased significantly after 3, 7, and 14 days O3 exposure. Spleen weights of mice exposed for 1 and 3 days were decreased compared to those of controls. However, exposure for 7 and 14 days showed no alterations in spleen weight. In the thymus, all O3 exposures markedly decreased its weights. Primary antibody response to sheep red blood cells (SRBC) in the spleens of mice exposed to 0.8 ppm O3 was suppressed in all exposure periods. However, no suppression was observed in primary antibody response to DNP-Ficoll of exposed spleens. These results suggest that the impairment of T-lymphocyte function in spleen and thymus was induced by short-term exposure to 0.8 ppm O3.

Adjuvant effects of aluminum silicate on IgE and IgG1 antibody production in mice.

The adjuvant effects of aluminum silicate on IgE and IgG1 antibody production were investigated. BALB/c mice were immunized intraperitoneally with 10 micrograms ovalbumin (OA) adsorbed on 0.2, 2, or 20 mg aluminum silicate. The enhancement of anti-OA IgE antibody production was observed in the mice injected with aluminum silicate and antigen compared with the mice injected with antigen alone. Anti-OA IgE antibody production with 2 and 20 mg aluminum silicate was greater than that with aluminum hydroxide (alum) as an adjuvant. Similar adjuvant effects with 2 mg aluminum silicate or alum were observed in AKR and C57BL/6 mice using 10 micrograms OA, and in BALB/c mice using 2 micrograms DNP-KLH (dinitrophenyl keyhole limpet hemocyanin): IgE antibody production induced by aluminum silicate adjuvant persisted for weeks in these experiments. The enhancement of IgG1 antibody production to OA mixed with aluminum silicate was also demonstrated. However, no difference between aluminum silicate and alum was observed on the IgG1 antibody production.

Effects of single exposure to cadmium on the primary humoral antibody response.

Effects of a single intraperitoneal (i.p.) injection of cadmium (Cd) on the primary humoral antibody responses against sheep red blood cells (SRBC) in mice were studied by assaying splenic plaque forming cells (PFC). PFC responses in mice were suppressed when exposed to Cd 2 days after immunization, and inconsistently stimulated when exposed before immunization. Dose-response relationships were observed in the suppressive effect of Cd exposure 2 days after immunization, but not consistently in the stimulative effect of Cd exposure before immunization. Thymus weights and cell numbers decreased markedly 4 days after Cd exposure with or without the antigenic stimulus. Splenic weights increased 2 days after Cd exposure, while the number of spleen cells was dramatically decreased 1 days after Cd exposure and still remained below normal 2 days after exposure.

Inhibition of delayed hypersensitivity reaction in mice by cadmium.

The effect of a single exposure to Cd on DH was studied through exposure to Cd at different time intervals in relation to antigen administration. Exposure to Cd significantly suppressed the DH reaction, with a dose-response relationship, when administered 2 or 3 days after immunization of BALB/c and DBA/2 mice with sheep red blood cells. A strain difference in sensitivity to Cd was also demonstrated. These results suggest that a single exposure to Cd impairs the cellular immune response in mice.

Enhancement of IgE antibody production by ovalbumin aerosol in mice.

The effect of aerosolized ovalbumin (OA) on the induction of IgE antibody production was investigated in BALB/c mice. Enhancement of anti-OA IgE antibody production was obtained after the administration of 10 micrograms OA in mice preexposed to 1% aerosolized OA for 6 or 30 min, and after the administration of 1 microgram OA in mice preexposed to aerosolized OA for 30 min. IgE antibody production could also be induced by preexposure to aerosolized OA at a concentration as low as 0.1%. A carrier effect was observed by preexposure to aerosolized OA using a hapten-carrier system. In the latter system, whole body irradiation (200 R) could not enhance IgE response, suggesting that irradiation-sensitive suppressor T cells might be generated in insufficient amount or not at all by aerosolized OA. Preexposure to aerosolized dinitrophenol-OA could not enhance anti-dinitrophenol IgE response. These results suggest that the exposure to aerosolized antigen primed preferentially T helper cells.

Modulated cytokine production from cervical lymph node cells treated with B[a]P and PCB.

To investigate the effects of Benzo[a]pyrene (B[a]P) and polychlorinated biphenyls (PCB) on in vitro cytokine production, we isolated cervical lymph node cells from ovalbumin (OA)-primed mice and the lymph node cells were cultured with various concentrations of B[a]P or PCB in the presence of antigen presenting cells (APC) and OA. B[a]P significantly increased the levels of interleukin-4 (IL-4) and -6(IL-6) in the culture supernatants. However, the levels of interferon-gamma (IFN-gamma) were decreased by treatment with PCB. These findings indicate that in vitro treatment with B[a]P or PCB modulate cytokine production from lymph node cells.

Novel crystalloid structures in suprasellar paraganglioma.

A-52-year-old woman was admitted to a hospital because of 2-year history of abnormal behavior and impaired visual acuity. Magnetic resonance imaging delineated a sizable mass at the suprasellar region. The partially removed tumor was arranged in irregular lobules composed of an admixture of clusters of cobblestone-like small cells and process-bearing cells with ovoid nuclei, surrounded by a fine, neuropil-like matrix. The Zellballen structure was inconspicuous, and mitosis was absent. Immunohistochemically, the tumor cells were positive for chromogranin A, synaptophysin, class III beta-tubulin and neurofilament, while negative for glial fibrillary acidic protein, cytokeratin and all 6 pituitary hormones. S100 protein expression was limited to cells adjacent to stroma. The MIB-1 labeling index was 0.5%. Histopathological diagnosis was paraganglioma of abortive architecture. Ultrastructurally, numerous dense-cored vesicles were found within the processes and cytoplasm. Synapse formation was not demonstrated. Interestingly, crystalloids up to 3 microm in size were frequently found. They had hexagonal or quadrilateral architecture without limiting membranes. The interval between periodically arranged fibrils was variable, ranging from approximately 20 - 50 nm. Retrospective examination by light microscopy failed to reveal corresponding structures. Crystalloids are rare manifestation of paragangliomas, yet undescribed in those of intracranial origin. Furthermore, the ultrastructure of the present case differs from those of previous cases.

Comparison of the effect of cadmium on lymphocytes of young and adult mice.

The influence of cadmium on in vitro primary plaque-forming cells (pfc) response of young (3 weeks old) and adult (5 mon old) mice to sheep red blood cells (SRBC) was investigated. Spleen cells of young and adult mice were cultured with various concentrations of cadmium chloride. Cadmium of 4 microM stimulated primary pfc response in cells of both groups, but 40 microM Significantly suppressed those of spleen cells from both groups. In the pre-treatment with 4 microM cadmium for 24 hr, spleen cells from young mice was also stimulated but hose from adult mice remained normal. As to cell viability of T and B cells from both groups after incubation with 4 microM cadmium for 24 hr, T cells from both groups and B cells from young mice were reduced by cadmium exposure. These results showed that young mice were more sensitive to the effect of cadmium on the antibody response than adult mice.
