Mesenchymal-epithelial interactions in sex differentiation.

Development of male and female accessory sexual glands is described in terms of the respective roles of epithelium and mesenchyme. During embryonic and neonatal periods mesenchyme alone exhibits androgen receptor activity (nuclear androgen binding sites) and is the actual target and mediator of the morphogenetic effects of androgens upon the epithelium. Mesenchyme induces specific patterns of epithelial morphogenesis, cytodifferentiation, and probably also specifies the functional (biochemical) activities of the epithelium. Mesenchymal influence upon expression of epithelial characteristics occurs in the perinatal period during morphogenesis, but also plays an important role in adulthood by maintaining favorable conditions for maintenance of epithelial morphology and function. Morphogenetic processes in adult hormone-dependent organs are though to be mediated by stromal cells.

Calcified gastric leiomyoma--a case report.

A 62 year-old man was admitted with the chief complaint of a epigastric pain and difficulty of swallowing. Examination of the upper gastrointestinal tract revealed a calcified mass causing a smooth filling defect at the lesser curvature of the cardia. At laparotomy, a localized tumor was detected beneath the serosa of the stomach. The excised tumor was 4 x 4 x 1.6 cm and the pathological diagnosis was calcified leiomyoma probably due to cytoplasmic degeneration. In this paper, details of the case are described and the related literature is reviewed.

Use of cultured lymphoblastoid cells for the study of abnormal enzymes: molecular abnormality of a phosphoglycerate kinase variant associated with hemolytic anemia.

A phosphoglycerate kinase (PGKase: ATP:3-phosphoglycerate 1-phosphotransferase, EC 2.7.2.3; X chromosome-linked) variant, PGKase-Tokyo, is associated with enzyme deficiency, nonspherocytic hemolytic anemia, and neurological disturbances. Because a sufficient amount of the patient's erythrocytes was not available, the variant enzyme was purified to homogeneity from the cultured lymphoblastoid cells of the patient. The enzyme activity of the variant lymphoblastoid cells was about 16% of that of the normal lymphoblastoid cells. PGKase-Tokyo, compared to the normal enzyme, had a lower specific activity (31% of normal in the backward reaction and 15% of normal in the forward reaction), higher than normal Michaelis constants for ATP and 3-phosphoglycerate, a more acidic pH optimum, and increased thermal instability. Microscale peptide mapping analysis revealed that the structural abnormality of PGKase-Tokyo is a single amino acid substitution from valine to methionine at position 266. Thus, the use of the cultured lymphoblastoid cells is proven to be useful for the study of structural and functional abnormalities of mutant enzymes.

An excitatory response preceding the silent period in masseter electromyographs.

A masseteric excitatory reflex response preceding the silent period which appears following tapping movement of the jaw, was investigated in order to evaluate its origin. In the pre-anaesthetic state, the latency of response did not change with the intensity of tapping. However, its amplitude increased depending on the intensity of tapping. The response did not disappear even after the anaesthesia. After the anaesthesia the lighter the intensity of tapping was, the longer the latency of the response, coming up to that of the jaw-jerk reflex. The pre-anaesthetic response had an intricate wave form comparing with the post-anaesthetic one. From the above findings it was concluded that the response must be a complex one in nature originating in the muscle spindle of jaw closing muscles and in a certain receptor of the structures surrounding the tooth.

Three-dimensional regional cerebral blood perfusion images with single-photon emission computed tomography.

Twenty-seven patients were studied for cerebral blood perfusion abnormalities using krypton 81m. A solution of 81mKr was continuously infused into an internal carotid artery, and single-photon emission computed tomograms were recorded. The transverse section images using 81mKr represented the three-dimensional regional cerebral blood perfusion, and this technique seemed to be useful for evaluating the regional cerebral blood perfusion in the deep brain.

Progestin receptors in testes from various animal species.

Using a synthetic progestin, R5020 (17 alpha, 21-dimethyl-19-nor-4, 9-pregnadiene-3, 20-dione), we investigated the presence of progestin receptor(s) in testicular cytosol from various animal species including humans. After incubation of cytosol from adult chickens with [3H]R5020 in the presence or absence of various unlabeled steroids, specific progestin binding was found in the 8 S region with an equilibrium dissociation constant of 2 nM and a binding capacity of 75 fmoles/mg protein. Specific progestin binding was also found in cytosol from immature chickens sedimenting in the 4 S region with a dissociation constant of 61 nM and a binding capacity of 1 pmole/mg protein. Although 5 S binding was found in the cytosols from humans, a monkey and rats, the binding specificity was low. These results clearly indicate that the testicular cytosols from chickens contain progestin receptor(s), whereas those from the other species do not.

[Activity determination of filter sample with liquid scintillation measurement (author's transl)].

The technique for solubilization of various filter samples into a liquid scintillator has been established to overwhelm the inherent shortcomings of a disc counting technique. Conventional liquid scintillation measurement methods that employ the quenching correction methods can be adopted to the homogeneously dispersed filter samples, which consequently enable the activity of the filter samples to be determined. This technique involves a resolution of the last problem in a sample preparation technique.

Effect of sex steroids on the uptake of 3H-leucine by brain tissues of ovariectomized mice.

Effect of the sex steroids, progesterone (P) and testosterone (T), on 3H-leucine uptake by the brain cells of ovariectomized mice were examined. Animals were divided into four groups, i.e. group 1: control animals treated with sesame oil; group 2: animals treated with P; group 3: animals treated with T, and group 4: animals first treated with T and then with P. Animals in each group were given a single i.p. injection of 3H-leucine 24 hr after the last hormonal treatment, and sacrificed 2 hr later. Intensity of the uptake of the radiochemical was measured by counting the number of reduced silver grains over cell bodies in various brain regions using an autoradiographic technique. Group 1 showed a relatively high uptake in the SO, PV and SPh when compared with that in the remaining nuclei examined. Groups 2 and 3 both showed a significant enhancement of the uptake in SCH, ARC and PM when compared with that in group 1. Group 4 showed enhancement of the uptake in most of the nuclei except POL, DM and SPH when compared with that in group 1. However, only the POM, PV, SO and VM revealed a significantly higher uptake than the respective nuclei in groups 2 and 3. The uptake by cells in the EC and CC remained unchanged after the hormonal treatment. The present results suggest that in female mice P or T stimulates protein synthesis in the hypothalamic nuclei and that the effect of P on protein synthesis is greatly influenced by T-priming.

A single amino acid substitution (Asp leads to Asn) in a phosphoglycerate kinase variant (PGK München) associated with enzyme deficiency.

The structural abnormality of the phosphoglycerate kinase variant, PGK München, associated with red cell enzyme deficiency and heat instability, was elucidated by a microscale peptide-mapping method. A single amino acid substitution, from aspartic acid in the normal enzyme to asparagine in the variant enzyme, was found. From the known amino acid sequence of normal human phosphoglycerate kinase, the substitution is in the aspartic acid residue located at 268th position from the NH2-terminal of the protein.

Suppression of immune response by isolation of the bursa of Fabricius from environmental stimuli.

When the bursa of Fabricius was isolated from environmental stimuli across the bursal epithelium by clipping the bursal duct on the 19th day of incubation, marked retardation was observed both in the bursal development and in the maturation of the immune reactivity. Neither necrotic changes nor involution in the bursa was, however, observed at any stage examined after clipping the bursal duct. Lymphopoiesis in the bursa and the immune reactivity of these chickens gradually increased with age, although they were constantly lower than those of controls. These results suggest that environmental stimuli across the bursal epithelium play an important role in teh amplification mechanisms of B-cell differentiation in the bursa.
