[The evaluation of hormone and metabolite profiles after the infusion of glucose, propionate and butyrate in cattle].

Lactational performance in high-yielding dairy cows has its limits in metabolic processes. Energy metabolism is maintained by mobilization of body reserves via hormonal regulation, resulting in lipolysis and sometimes ketoacidosis. For characterization of such conditions the intraruminal and intravascular application of glucose and volatile fatty acids was used in ruminant physiology. On the other hand the results of such experiments were correlated to actual and potential milk yield. For this investigation pairs of monozygous Holstein Friesian twins were tested as heifers and as cows by intravenous infusion of glucose, propionate and butyrate after 18 hours of feed withdrawal. Insulin, growth hormone, glucose, free fatty acids and beta-hydroxybutyrate were measured before, during and up to 4 hours after infusion of substrates. Each substrate caused a transient change in plasma concentrations of metabolites and hormones. Differences between heifers and cows are discussed with the time series of the mean concentrations. The reaction profile of each animal was characterized by different parameters. Analysis of these parameters revealed a close relation between hormones and metabolites even under the conditions of the load-test.

T4, T3 and rT3 levels in serum and cerebrospinal fluid of patients with amyotrophic lateral sclerosis.

Thyronine (T4), triiodothyronine (T3), and reverse-triiodothyronine (rT3) levels were evaluated in cerebrospinal fluid (CSF) and in serum of 12 patients with definite amyotrophic lateral sclerosis (ALS) by specific radioimmunoassays. Circulating microsomal and thyroglobulin antibodies were also evaluated. In all patients serum levels of T4, T3 and rT3 were within normal limits. In CSF, the rT3 levels were significantly elevated to 0.118 micrograms/l (mean), the T4 levels were not significantly elevated, and the T3 levels were below the detection limit of 0.03 micrograms/l. A correlation between the elevated rT3 levels in CSF and the severity or type of ALS could not be demonstrated by this study. The antithyroid antibodies (thyroglobulin antibodies, microsomal antibodies) showed normal titres and did not suggest disturbances of thyroid autoimmunity in the patients with ALS.

[Sequencing of deoxyribonucleic acid as a part of genomic analysis].

The main characteristics of the chemical structure of Deoxyribonucleic acid is described. The methods of sequencing of DNA used today are represented, the base-specific cleavage reactions used in the chemical sequencing procedure and the chain-termination reactions used in the enzymatic procedure. From own studies bovine genomic sequences are shown and some important methodical aspects explained. The application of the sequence analysis is shown with the organization of the porcine growth hormone gene and with the sequence dependence of DNA conformation.

Investigations on iodothyronine deiodinase activity in the maturing rat brain.

5-Monodeiodination of T4 and T3 and 5'-monodeiodination of T4 and rT3 were studied in brain homogenates of male Sprague-Dawley rats, aged 1-60 days. Portions of the homogenates were incubated with the substrates at 37 C for 30 min. The reaction products were estimated by specific RIAs. All of the four reactions were dependent upon time, temperature, pH, and upon the concentrations of substrate, thiol, and tissue protein. Maximal reactions were obtained between 40 and 160 mM dithioerythritol. T4 5'-deiodination proceeded optimally at pH 7.4 and 0.4 microM substrate, the other reactions at pH 8.5 and 10 microM substrate. The four reactions were inactivated by heat (56 C, 30 min) and inhibited by 10(-5) M iopanoic acid. Only rT3 5'-deiodination was inhibited by 3 X 10(-4) M propylthiouracil (greater than 95%). In cerebellum, basal ganglia, brainstem, and hypothalamus both T4 and T3 5-deiodinase activity were very high in perinatal rats [up to 5.56 pmol/(min X mg protein) in hypothalamus], and decreased rapidly with age. In cortex and olfactory bulb these enzyme activities were low after birth, followed by an increase during the growth spurt [up to 632 fmol/(min X mg protein) in olfactory bulb]. T4 and rT3 5'-deiodinase activity in all brain regions studied were at their lowest in perinatal rats. During and after the growth spurt an increase was observed [up to 457 fmol/(min X mg protein) in cerebellum]. The reciprocal course of 5- and 5'-deiodination between birth and growth spurt in most of the brain regions studied might lead to a reduced intracellular thyromimetic activity during the perinatal period.

[The problem of heat activation of bacterial spores after disinfection with regard to an aerosol method of decontaminating equipment and rooms].

This paper describes investigations on disinfection of germ carriers, contaminated with an alcoholic suspension of Bacillus cereus or Bacillus subtilis. Result of disinfection is compared with that of an additional heat treatment (80 degrees C, 60 min) after disinfection. Besides Formalin, Tegodor forte (Th. Goldschmidt, Essen) and P 3 oxonia active (Henkel KG, Düsseldorf) are tested with different concentration and duration. Heat activation was possible with all three disinfectants. For illuminating the conditions of activation, circumstances of heat action after disinfection with formaldehyde-aerosol have been varied. Heat activation in dry air, moist air and distilled water was not successful. Only in Nutrient Broth (Standard I-Bouillon; Merck, Darmstadt) spores were viable again, after activation. Addition of serum as a protective cover had no influence on the result. Consequences of the results on a disinfection method with formaldehyde-aerosol are discussed.

Steric factors in the pharmacokinetics of lindane and alpha-hexachlorocyclohexane in rats.

1. The elimination of alpha-1,2,3,4,5,6-hexachlorocyclohexane (alpha-HCH) by rats, as assessed by g.l.c. determination of the chemical's disappearance from depot fat, exhibited a sex difference (males:females = 4:1) and a sizeable deuterium isotope effect (6.3 in males). 2. Lindane (gamma-HCH), by contrast, disappeared from depot fat, skeletal muscle, brain and blood at nearly the same rates in both sexes. Perdeuteration, though effective in reducing hepatic removal, did not significantly retard the overall elimination of this isomer (isotope effect in males less than 2). Partial explanation of this finding is that lindane and lindane-d6 are equally subject to dechlorination in the gut. 3. alpha-HCH distributed into cerebral white matter in preference to grey matter to a much higher degree than did lindane and the beta-isomer of HCH, and elimination from that tissue was slow. The finding is considered to indicate a stereoselective affinity of alpha-HCH to some component(s) of myelin.

[Transient ischemic attacks with intracranial tumors (author's transl)].

Transient ischemic attacks are not only the consequence of cerebral atherosclerosis. A woman of 48 years had transient ischemic attacks because of a meningeom narrowing the internal carotid artery. A steal syndrome in tumor vessels of a glioblastoma must be presumed in a man of 67 showing initial hemisyndrome with first transient, later on remaining palsy.

Self-expandable macroporous nitinol stents for transfemoral exclusion of aortic aneurysms in dogs: preliminary results.

This experimental study evaluated the feasibility of excluding artificially created aortic aneurysms in dogs with transfemorally inserted nitinol stents, serving as intraluminal bypass. Infrarenal aortic aneurysms were created in eleven dogs (28-35 kg) by end-to-end anastomosis of sacciform conduits. Single and double wire-knitted, macroporous, superelastic, self-expanding nitinol stents without any additional wrapping or in-between fabrics served as endovascular grafts. The pore size of the double-knitted stents was less than 0.7 mm2. Angiography following stent placement showed immediate exclusion of the aneurysms for the double-knitted stents. For the single-knitted stents (2 cases), partial exclusion was achieved only after considerable delay. CT studies in the dogs with the double-knitted stents revealed patency of the grafts after 3 to 5 weeks with the aneurysms totally thrombosed. So far gross pathology of 1 dog at 24 h and 1 at 12 months revealed patency of the stents. At 12 months histopathology showed a neointima covering the inner stent surface with the aneurysm filled by an organized thrombus. It is concluded, that an "open" stent with a tight meshwork appears to be a suitable device to instantly exclude artificial aneurysms of the canine aorta.
