Expression of p21(Cip1/Waf1/Sdi1) and p27(Kip1) cyclin-dependent kinase inhibitors during human hematopoiesis.

Expression of p21 and p27 cyclin-dependent kinase inhibitors is associated with induced differentiation and cell-cycle arrest in some hematopoietic cell lines. However, it is not clear how these inhibitors are expressed during normal hematopoiesis. We examined various human hematopoietic colonies derived from cord blood CD34(+) cells, bone marrow, and peripheral blood cells using a quantitative reverse transcription-polymerase chain reaction assay, immunochemistry, and/or Western blot analysis. p21 mRNA was expressed increasingly over time in all of the colonies examined (granulocytes, macrophages, megakaryocytes, and erythroblasts), whereas p27 mRNA levels remained low, except for erythroid bursts. Erythroid bursts expressed both p21 and p27 mRNAs with differentiation but expressed neither protein, whereas both proteins were expressed in megakaryocytes and peripheral blood monocytes. In bone marrow, p21 was immunostained almost exclusively in a subset of megakaryocytes and p27 protein was present in megakaryocytes, plasma cells, and endothelial cells. In megakaryocytes, reciprocal expression of p27 to Ki-67 was evident and an inverse relationship between p21 and Ki-67 positivities was also present, albeit less obvious. These observations suggest that a complex lineage-specific regulation is involved in p21 and p27 expression and that these inhibitors are involved in cell-cycle exit in megakaryocytes.

Suppression of allograft rejection with anti-alphabeta T cell receptor antibody in rat corneal transplantation.

BACKGROUND: Anti-alphabeta T cell receptor monoclonal antibody (R73) has been reported to be a potent immunosuppressant. The suppressive effects of this antibody on allograft rejection after corneal transplantation are unknown. METHODS: Orthotopic rat penetrating keratoplasty was performed using Lewis rats as recipients and Brown Norway and Fisher rats as donors. The treated groups received R73 intraperitoneally until day 12 after the transplantation. In grafted rats with or without R73 treatment, cytokine expression of the aqueous humor, corneal-infiltrating cells, draining lymph nodes, and splenocytes was determined. Delayed-type hypersensitivity (DTH) responses were compared. RESULTS: All allografts in the untreated controls of Fisher-to-Lewis or BN-to-Lewis rat combinations were rejected within 14 days. In contrast, indefinite survival rates of the postoperative R73-treated group increased to 86% in the Fisher-to-Lewis and 23% in the Brown Norway-to-Lewis combinations, respectively. Interferon-y, interleukin (IL)-2 (T helper [Th]1), and IL-10 (Th2), but not IL-4 (Th2), expression of the eye and DTH responses in the control group were suppressed in the R73-treated group. Both IL-2 and IL-10 expression after mixed lymphocyte culture in the R73-treated group were significantly lower than those of the naive and untreated control group. CONCLUSIONS: alphabeta T cell receptor-targeted therapy prevents allograft rejection in rat corneal transplantation as evidenced by suppression of DTH responses. The cytokine profile after R73 treatment was characterized by low interferon-gamma, IL-2, and IL-10, and high IL-4 expression.

Telomerase reverse transcriptase mRNA expression and telomerase activity in hepatocellular carcinoma.

Human telomerase reverse transcriptase (hTERT) has been identified as the catalytic subunit of human telomerase. To clarify the clinical significance of hTERT mRNA in hepatocellular carcinoma (HCC), we investigated the relationship between telomerase activity and hTERT mRNA in human HCC and non-HCC tissues. The hTERT mRNA was detected in 17 (89.47%) of 19 livers with HCC and in 4 (21.05%) of 19 noncancerous tissues from these livers. Telomerase activity was detected in 17 of the 19 tumor tissues (89.47%) and in 4 of the 19 nontumor tissues (21.05%). The hTERT mRNA was detected in all tissues that were telomerase-positive and it was undetected in all tissues that were telomerase-negative. The correlation between the expression of hTERT mRNA and human telomerase activity in this study indicates that hTERT mRNA could be useful to diagnose cancer. Also, as telomerase production may be under the control of hTERT mRNA, the possibility is great that noncancerous liver tissue with chronic liver diseases acquires HCC when the hTERT mRNA is positive.

Evaluation of endoscopic injection sclerotherapy with and without simultaneous ligation for the treatment of esophageal varices.

For more effective and simple endoscopic injection sclerotherapy (EIS) for esophageal varices, we developed an EIS procedure with ligation (EISL) that is non-invasive, in which EIS and endoscopic variceal ligation (EVL) are performed simultaneously. In this study, we compared EISL and EIS in a randomized sample of patients (n = 14 for each procedure). For EISL, EVL was performed, including the injection site, after the injection of 5% ethanolamine oleate with iopamidol (EOI) into a varix. The mean number of treatment sessions required for eradication of esophageal varices was 2.3+/-0.5 for EISL and 3.9+/-0.8 for EIS (P < 0.001); the mean number of treatment sites was 6.2+/-2.2 for EISL and 14.0+/-5.0 for EIS (P < 0.001); the mean total amount of EOI used was 13.8+/-5.2ml for EISL and 26.3+/-9.8ml for EIS (P < 0.001). There were no significant differences in rates of recurrence of varices or in bleeding between the two groups. For EISL, fewer treatment sessions and less sclerosant were sufficient, probably because the sclerosants were more effective due to the blockage of variceal blood flow by the ligation. This method should provide a novel modification of EIS.

Anatomy of the cranial nerve foramina in the Risso's dolphin (Grampus griseus).

In the present study, we examined the cranial nerve foramina of Risso's dolphin (Grampus griseus). There were two distinguishable characters in the cranial nerve foramina compared with terrestrial mammals. One was that the foramen infraorbitale was composed of several holes, but not a single hole. They should therefore be termed foramina infraorbitales. The infraorbital nerves ran through these foramina, went into the 'melon' and then branched in a complicated fashion. The facial nerve innervated the muscles surrounding the melon. A well-developed infraorbital and facial nerve complex may control the melon. Another was the presence of a porus acusticus internus and independent tympano-periotic bone. The separate ear bone forced the vestibulocochlear and facial nerves to exit from the cranial cavity through the porus acusticus internus. The independent ear bone structure may shut off the noise from the cranial bone to the periotic bone with a true receptor of hearing. It may be an adaptation for an acute sense of hearing. Compared with other dolphins, the cranial foramina of Risso's dolphin are definitely separate. The structure of the foramina is similar to that of pilot whales, but not to dolphins, so that Grampus may be closely allied to pilot whales.

Functional morphology of the locomotor system in the northern smooth-tailed tree shrew (Dendrogale murina).

The locomotor system of the northern smooth-tailed tree shrew (Dendrogale murina), one of the arboreal species of Scandentia, was examined by means of macroscopic anatomy. In this study, we describe the muscular system in the shoulder, forearm, hip and crural regions. The M. deltoideus and the acromion of the scapula were well-developed. The characteristic postscapular fossa was distinguishable in the scapula. The M. teres major and M. triceps brachii possessed large bundles and were attached to the postscapular fossa. In the hindlimb region, the M. biceps femoris was inserted into large area of distal hindlimb. The M. tenuissimus was discernible beneath the M. biceps femoris. We suggest that these findings may contribute to our understanding of arboreal locomotion in this species, and that this may typically represent the arboreal adaptational pattern of the muscular system in the Scandentia.

Morphological character of the shoulder and leg skeleton in Przewalski's horse (Equus przewalskii).

The skeletal feature was observed in Przewalski's horse (Equus przewalskii) and the domestic horse (E. caballus). As results of the observations, remarkable differences were found in the scapula. The caudal border of the scapula in Przewalski's horse is curved in the caudal direction more than that of the domestic horse. Moreover, in the domestic horse, the sharp caudal border is formed by the shift of the outer muscular line (Linea muscularis) to the border. In Przewalski's horse, however, the caudal border is rounded because the outer muscular line does not shift to the border but lies beside it. We suggest that the morphological difference observed between the scapulae of these horses may lead to changes in the shoulder movement with the various attachments of the long head of the triceps muscle (Caput longum musculi tricipitis). The skeletal measurement and the measurement ratio show that the leg skeleton of Przewalski's horse is shorter and thicker than that of the domestic horse. These results show that the center of gravity of Przewalski's horse is low in position and that it has a short stride.

Magnetic fields produced by single motor units in human skeletal muscles.

OBJECTIVE: To develop new non-invasive ways of analyzing human skeletal muscle function, biomagnetic measurement was applied to the vastus lateralis and vastus medialis of 3 healthy adult males using a 64 channel superconducting quantum interference device system. METHODS: Discharges from single motor units were detected by simultaneously recorded surface electromyography. Magnetic signals were averaged 64-158 times at zero-crossing timing in the surface electromyographic signal. RESULTS: Six motor units detected in the 3 subjects produced large magnetic fields with peak-to-peak amplitudes of 1-2 pT. Magnetomyographic isofield maps showed current sources arising from motor endplate regions and propagating in directions opposite to fiber ends. The absolute intensity of current moments in muscle fibers within motor units was estimated based on dipole fitting. The estimated moment was 23.9-114.3 nAm for repolarization dipole. Dividing these moment values by the typical dipole moment of 0.286 nAm in a single muscle fiber, the number of muscle fibers in motor unit was estimated to be 84-400. CONCLUSIONS: Magnetic recording may provide a new non-invasive way of analyzing and diagnosing human muscle function.

MEG characterization of spontaneous alpha rhythm in the human brain.

The present piece of research studied the spontaneous alpha rhythm of the human brain by combining the use of a whole-cortex neuromagnetometer and Magnetic Resonance Imaging. Single trials of spontaneous brain activity were recorded from ten human subjects asked to rest, with their eyes either closed or open, in relaxed wakefulness. MEG measurements were conducted over a period of one and a half years. The replicability of the results was confirmed for eight subjects out of ten. For three subjects, the alpha rhythm did not show any reductions due to the opening of the eyes. Both field map pattern and location of the estimated source were persistently stationary during each of the bursts of oscillations of the alpha rhythm. Dipoles were concentrated in clusters, indicating the existence of several spatially distributed sources. The calcarine fissure, the parieto-occipital sulcus and the surrounding occipital and parieto-occipital areas were identified as cortical sites of the brain where the alpha rhythm may originate. For four subjects, the majority of the sources were located near or in the calcarine fissure, while for five subjects, they were located near or in the parieto-occipital sulcus and for the remaining subject they were equally divided between the two generation sites.

Morphology of the intestinal tract in the white rhinoceros (Ceratotherium simum).

The intestinal tract of the white rhinoceros (Ceratotherium simun) was dissected. We observed a simple structure for the small intestine, duodenum, jejunum and ileum, and a well-developed large intestine, cecum, colon and rectus. The cecum consisted of a small chamber, whereas the colon was much enlarged; notably larger than that of the domesticated horse, which belongs to the same order, Perissodactyla. This suggests that in the white rhinoceros the cecum may be functionally replaced by the well-developed colon which may act as the main fermentation tank in this animal.

CT examination of the head of the Baikal seal (Phoca sibirica).

We carried out a computerised tomographic (CT) examination to elucidate the modifications in the head related to orbital enlargement in the Baikal seal. Transverse CT images showed that (1) the external frontal contours and the frontal sinuses are compressed medially and ventrally by the orbital enlargement; (2) the caudal part of the nasal cavity and nasopharynx are compacted ventrally; and (3) the cranial cavity is displaced caudally. The neurocranium is obviously separated from the facial part in the transverse plane at the caudal region of the orbit. The disposition of the mandible, zygomatic arch, temporal bone, and the masseter, temporal, digastric and pterygoid muscles is changed by the enlarged orbit in the 3-dimensional reorganisation of the head in this species. It is suggested that adaptation for the Lake Baikal environment primarily resulted in orbital enlargement, and that the altered orbital design may subsequently have influenced the form and function of the masticatory and respiratory system.

CT examination of the manipulation system in the giant panda (Ailuropoda melanoleuca).

The manipulation mechanism of the giant panda (Ailuropida melanoleuca) was examined by means of CT (computed tomography) and 3-dimensional (3-D) Volume Rendering techniques. In the 3-D images of the giant panda hand, not only the bones but also the muscular system was visualised. Sections of the articulated skeleton were obtained. It was demonstrated that the hand of the panda is equipped with separately moulded manipulation units as follows: (1) the radial sesamoid (RS), the radial carpal, and the first metacarpal (R-R-M) complex; and (2) the accessory carpal (AC) and the ulnar (A-U) complex. When the giant panda grasps anything, the R-R-M complex strongly flexes at the wrist joint, the RS becomes parallel with the AC, and the phalanges bend and hold the object. It is shown that the well-developed opponens pollicis and abductor pollicis brevis muscles envelop and fix the objects between the R-R-M complex and the phalanges during grasping.

Spatio-temporal analysis of cortical activity evoked by gustatory stimulation in humans.

Gustatory activated regions in the cerebral cortex have not been identified precisely in humans. In this study we recorded the magnetic fields from the brain in response to two tastants, 1 M NaCl and 3 mM saccharin. We estimated the location of areas activated sequentially after the onset of stimulation with magnetic source imaging. We investigated the primary gustatory area (area G) precisely, and found it at the transition between the parietal operculum and the insular cortex. The central sulcus was activated less frequently than area G but with almost the same latency in cases of NaCl stimulation. Following area G, we found activation in several cortical regions, e.g. both the frontal operculum and the anterior part of the insula, the hippocampus, the parahippocampal gyrus and the superior temporal sulcus.

Immunohistochemical analysis of PAI-2 (plasminogen activator inhibitor type 2) and p53 protein in early gastric cancer patients with recurrence: a preliminary report.

BACKGROUND: High levels of urokinase-type plasminogen activator (u-PA) were demonstrated in gastric carcinomas along with inhibitors of plasminogen activators (PAI-1 and PAI-2). They may influence the ability to invade and metastasize and therefore be of importance to the risk of recurrence of stomach neoplasms after curative operation. This also appears to be the case for p53 mutations and p53 protein overexpression. METHODS: Six patients, all differentiated cancer cases who developed recurrent disease 5-10 years after curative operations for early gastric cancers (recurrence group), were studied in comparison with 49 patients who had no recurrence more than 10 years after similar surgery (control group). The expression of u-PA, PAI-1, PAI-2 and p53 was compared immunohistochemically in the recurrence and control groups. RESULTS: The expression of PAI-2 was significantly more frequent in the recurrence group, being found in five (83.3%) patients vs eight (16.3%) in the control group. p53 was expressed in five (83.3%) patients in the recurrence group and in 15 (30.6%) in the control group; the rate was again significantly higher in the former. CONCLUSION: The results suggest that PAI-2 and p53 expressed in differentiated early gastric cancers are possible indices of the risk of recurrence.

Endotoxin activity measured by limulus assay.

The binding activity of endotoxin (Escherichia coli 0111) to dentin powder pre-treated with 10% sodium hypochlorite and with 3% hydrogen peroxide was investigated. This was carried out for periods of 30, 10 and 1 min. The endotoxin was diluted into 100 ng/ml solutions. Dentin powder suspended in a 34 micrograms/ml protein concentration was used in the study. The limulus amoebocyte lysate test was used to determine the amount of endotoxin binding, which was measured by the microtiter plate reader. The binding activity level of that concentration of endotoxin was significantly lower in dentin powder treated with 10% sodium hypochlorite than in that treated with 3% hydrogen peroxide at 1 and 10 min. Endotoxin binding activity measured the lowest in the 30 min test period.
