Effect of blocking protein synthesis at nonpermissive temperatures on temperature-sensitive deoxyribonucleic acid mutants of Escherichia coli.

When protein synthesis was blocked in temperature-sensitive deoxyribonucleic acid synthesis mutants of Escherichia coli at nonpermissive temperatures, it reduced the amount of apparent subsequent chain elongation to approximately half that observed in the mutants either at nonpermissive temperatures alone or when protein synthesis was blocked at the permissive temperature. Blocking protein synthesis at the nonpermissive temperatures for periods of 40 min caused the loss of ability to reinitiate deoxyribonucleic acid synthesis at the permissive temperature.

[Meniscal injuries and associated lesions seen in the light of long-term results after meniscectomy].

The authors have put together their experience of the long-term results after meniscectomy. They came to the following conclusions: 1. A knee joint does not become unstable after removal of a cartilage unless there is another connected lesion. 2. X-ray modifications are to be found on the majority of the patients after meniscectomy, but very often without any clinical sign. 3. The sooner meniscectomy is practiced the better the long-term results are to be expected. 4. A good operative technique is most important to get satisfactory long-term results.

[What advantage are there in using gentamycin-PMMA-chains in the treatment of posttraumatic osteomyelitis, osteoarthritis and soft tissue infections?].

The use of gentamicin-polymethylmethacrylate (PMMA) bead chains permits the depositing of a high dose of gentamicin at the place of the infection. Short indications on the experimental basis and hints regarding the practical application are given. It follows the discussion of the results in 34 patients with chronic posttraumatic osteomyelitis, arthritis due to infection, diabetic and arteriosclerotic gangrene and the infections of the soft tissues. Primary wound healing was achieved in 70%. Discussion of interesting cases. The advantages of the use of gentamicin-PMMA bead chains in infection cases are emphasized.

Accumulation of the capacity of initiation of deoxyribonucleic acid replication in Escherichia coli.

Several temperature-sensitive initiation mutants of Escherichia coli were examined for the ability to initiate more than one round of replication after being held at nonpermissive temperature for approximately 1.5 generation equivalents. The capacity for initiation was measured by residual synthesis experiments and rate experiments under conditions where protein synthesis and ribonucleic acid synthesis were inhibited. Results of the rate and density transfer experiments suggest that the cells may initiate more than one round of replication in the absence of protein or ribonucleic acid synthesis. This contrasts with the results of the residual synthesis experiments which suggest that, under these conditions, only one round of synthesis is achieved. These findings suggest that the total amount of residual synthesis achieved in the presence of an inhibitor may be both a function of the number of initiation events which occur and the effect of the inhibitor of protein or ribonucleic acid synthesis on chain elongation.
