Ovarian function in 91 dairy cows with cystic ovarian disease was assessed by rectal palpation and by plasma hormone analysis before and after treatment. Plasma analysis showed that 84% of the cysts were correctly classified clinically and only these cows are considered further. Luteinised cysts occurred in 59 cows whereas only 18 had non-luteinised cysts. The mean plasma concentrations of luteinising hormone (LH), follicular stimulating hormone (FSH), progesterone, oestradiol and testosterone were not significantly different when compared with values at relevant stages of the oestrous cycle in normal cows. Success of treatment with progesterone, a synthetic prostaglandin, human gonadotrophin (HCG), or gonadotrophin releasing hormone (GnRH) was not dependent upon prior hormone concentrations, except for the prostaglandin which required active luteal tissue. LH and FSH concentrations in cows with luteinised cysts were not significantly different before and after successful treatment with GnRH or progesterone. Normal luteal function was not always established after treatment of non-luteinised cysts with GnRH.

A radioimmunoassay system for the measurement of ovine follicle-stimulating hormone (FSH) was established. After the onset of normal oestrus, there were simultaneous surges of luteinizing hormone (LH) and FSH, and 24 h later, a surge of FSH alone. Administration of sodium pentobarbitone when the dual surge of LH and FSH was expected inhibited the release of LH, but prolonged that of FSH. Development of corpora lutea that took place immediately was normal in ewes treated with sodium pentobarbitone, but after the subsequent oestrus, the life-time of corpora lutea was abnormally short. When sodium pentobarbitone was administered at the time when the second surge of FSH was expected, no effect was observed on the level of LH or FSH. Subsequent development of corpora lutea was similar to that in the non-treated group. Administration of sodium pentobarbitone at the expected time of the dual surges of LH and FSH after infusion of oestradiol to anoestrous ewes blocked the release of both LH and FSH. Administration of sodium pentobarbitone at the expected time of the second surge of FSH after infusion of oestradiol delayed the increase in the level of FSH. In a pilot experiment after combined infusion of androstenedione and oestradiol, sodium pentobarbitone did not completely inhibit the release of FSH, but the release of LH was totally prevented.
Testosterone, oestradiol and progesterone were measured in peripheral plasma during the oestrous cycle of 6 heifers. Oestradiol and progesterone results confirmed earlier reports. Concentration of testosterone on the day of oestrus was 40+/-3 pg/ml (mean+/-S.E.M.), and two peaks were detected during the cycle, one 7 days before oestrus (1809+/-603 pg/ml) and the other (78+/- 7 pg/ml) on the day before the onset of oestrus. The concentration of progesterone declined in most cases 1 day after the maximum concentration of testosterone. Betamethasone treatment in 5 heifers extended luteal function by an average of 10 days: plasma androstenedione and oestradiol concentrations were unaltered; cortisol values were depressed for at least 16 days after treatment; testosterone concentrations were lowered by 13+/-2-4% during treatment, and except in one heifer the peak on Day -7 was abolished.
A radioimmunoassay with Sephadex chromatography for androstenedione is described. Concentrations of androstenedione in bovine peripheral blood during the oestrous cycle ranged from 80 to 100 pg/ml. There was considerable variation and no pattern suggestive of physiological significance emerged, unlike those of oestradiol and testosterone.
