Nanofabrication of organic/inorganic hybrids of TiO2 with substituted phthalocyanine or polythiophene.

Organic photovoltaic cells, similar to Grätzel type, have been widely investigated in recent years. In the case of Grätzel-type cells, TiO2 colloids are usually spin-coated onto an electrode and then sintered. Later, such electrodes are immersed in dye solution to sensitize the TiO2 layer for fabrication of photovoltaic cells. In the current study, an attempt was made to fabricate photovoltaic cells using a layer-by-layer technique. Based on such a method, ordered substituted phthalocyanine or conducting polythiophene-sensitized TiO2 multilayers were fabricated at the molecular level. Buildup of multilayer films of copper phthalocyanine-capped TiO2 and poly(thiophene-3-acetic acid)/TiO2 was monitored by increments in the UV-visible absorption and the frequency decrease of quartz crystal microbalance. The ordered multilayers were further characterized by infrared spectroscopy, and electrochemical and photoelectrochemical measurements.

Contrast-enhanced subtraction harmonic sonography for evaluating treatment response in patients with hepatocellular carcinoma.

OBJECTIVE: Our objective was to assess the usefulness of contrast-enhanced subtraction harmonic sonography in evaluating the treatment response of patients with hepatocellular carcinoma. SUBJECTS AND METHODS: Thirty-two hepatocellular carcinoma lesions in 26 patients (age range, 44-85 years; mean age, 66 years) were examined with Levovist-enhanced intermittent harmonic imaging before and after therapy. A Toshiba Powervision 8000 was used. A subtraction image was obtained by digitally subtracting the last-frame harmonic image from the first-frame image when multishot mode was preset. Results of contrast-enhanced CT were compared with the results of subtraction harmonic imaging. RESULTS: Before therapy, an enhancement pattern of tumor vascularity was seen for 93.8% (30/32) of hepatocellular carcinoma nodules on subtraction harmonic imaging. After therapy, subtraction harmonic imaging showed 46.7% (14/30) enhancement (incomplete tumor necrosis) and 53.3% (16/30) no enhancement (complete tumor necrosis). When dynamic CT was the gold standard, the sensitivity, specificity, and accuracy of subtraction harmonic imaging were 93.3%, 100%, and 96.7%, respectively. Intratumoral flow signals in hepatocellular carcinoma after therapy on harmonic imaging were used as a guide to target additional percutaneous therapy. CONCLUSION: Digital subtraction contrast-enhanced harmonic imaging can depict tumor vascularity in hepatocellular carcinoma after therapy sensitively and accurately. Because it is easy to perform and provides real-time needle insertion guidance, it may be preferable to perform after localized therapy to monitor treatment response, which will reduce unnecessary CT scanning.

[The research of receptor-mediated drug carrier].

One way to increase the concentration of drug in diseased tissue is to attach drug to a suitable carrier such as protein or peptides, which can improve the pharmacokinetic properties of drug and decrease it's side-effect on normal tissue. The developments of receptor-mediated transporting of drug assotiated with protein or peptides are reviewed in this paper. The most frequently used receptors and drug carriers and the bonds between the drug and the carrier are introduced. The machanism of transporting and the factors that affect the efficiency of the drug carrier are discussed as well.

[Correlation between liver fibrosis stage and serum liver fibrosis markers in patients with chronic hepatitis B].

OBJECTIVE: To evaluate the diagnostic value of serum liver fibrosis markers by analyzing the correlation between liver fibrosis stage in patients with chronic hepatitis B and serum liver fibrosis markers, such as hyaluronate, type III procollagen, laminin, and type IV collagen. METHODS: There were 278 patients with chronic hepatitis B in this series. Before liver biopsy, the serum was collected to test hyaluronate, type III procollagen, laminin, and type IV collagen. Data were analyzed statistically. RESULTS: Liver fibrosis stage was correlated to inflammation degree. In patients with moderate or serious liver inflammation and fibrosis, serum hyaluronate was above normal level, and correlated to liver inflammation and fibrosis. For patients with chronic hepatitis B, serum type III procollagen, laminin, and type IV collagen increased apparently. While laminin was correlated to liver fibrosis, type III procollagen and type IV collagen correlated to liver inflammation. CONCLUSIONS: Serum hyaluronate, type III procollagen, laminin, and type IV collagen could reflect the state of liver fibrosis. Hyaluronate may be a useful marker for liver cirrhosis.

Endothelium-derived relaxing factors: a focus on endothelium-derived hyperpolarizing factor(s).

Endothelium-derived hyperpolarizing factor (EDHF) is defined as the non-nitric oxide (NO) and non-prostacyclin (PGI2) substance that mediates endothelium-dependent hyperpolarization (EDH) of vascular smooth muscle cells (VSMC). Although both NO and PGI2 have been demonstrated to hyperpolarize VSMC by cGMP- and cAMP-dependent mechanisms, respectively, and in the case of NO by cGMP-independent mechanisms, a considerable body of evidence suggests that an additional cellular mechanism must exist that mediates EDH. Despite intensive investigation, there is no agreement as to the nature of the cellular processes that mediates the non-NO/PGI2 mediated hyperpolarization. Epoxyeicosatrienoic acids (EET), an endogenous anandamide, a small increase in the extracellular concentration of K+, and electronic coupling via myoendothelial cell gap junctions have all been hypothesized as contributors to EDH. An attractive hypothesis is that EDH is mediated via both chemical and electrical transmissions, however, the contribution from chemical mediators versus electrical transmission varies in a tissue- and species-dependent manner, suggesting vessel-specific specialization. If this hypothesis proves to be correct then the potential exists for the development of vessel and organ-selective vasodilators. Because endothelium-dependent vasodilatation is dysfunctional in disease states (i.e., atherosclerosis), selective vasodilators may prove to be important therapeutic agents.

[Modulation of angelica sinensis polysaccharides on the expression of nitric oxide synthase and Bax, Bcl-2 in liver of immunological liver-injured mice].

OBJECTIVE: To investigate the modulation of Angelica Sinensis Polysaccharides on the expression of nitric oxide synthase and Bax, Bcl-2 in the liver of immunological liver injured mice. METHODS: Immunological liver injury was induced by lipopolysaccharide (ip, 2mug/10g) in bacillus calmette-guerin (ip, 1mg/10g, qd/1 d) primed mice. Angelica Sinensis Polysaccharides was administrated to the mice with 30mg/kg, 60mg/kg, respectively. The effects of Angelica Sinensis Polysaccharides on the expression of nitric oxide synthase, Bcl-2, and Bax gene were assessed by immunohistochemistry, and the nitric oxide (NO) production, alanine transaminase (ALT) activity, glutathione S-transferase (GST) activity in serum were determined. RESULTS: The levels of NO, sALT, and sGST were significantly higher in immunological liver-injured mice than controls. The contents of iNOS and Bax were 17.8 times (P<0.001) and 1.31 times (P<0.05) of control values, respectively. cNOS expression was not obviously changed, and no Bcl-2 expression was found in immunological liver-injured mice. Angelica Sinensis Polysaccharides of 30mg/kg could reduce the levels of NO, sALT, and sGST by 24.6%, 40.8%, and 18.4%, respectively; the expression of iNOS and Bax decrease by 84.2% and 37.1%; and the expression of cNOS, Bcl-2 increase by 66.8% and 3.38 times, respectively. The influence of Angelica Sinensis Polysaccharides of 60mg/kg was not so obvious as that of 30mg/kg on sGST, NO, cNOS, Bax, and Bcl-2, but was more effective on sALT and iNOS. CONCLUSIONS: NO production may play a role in the LPS-induced hepatotoxicity. Angelica Sinensis Polysaccharides can alleviate the immune liver injury by modulating the expression of cNOS, iNOS, Bax, Bcl-2.

Adenovirus-mediated expression of a truncated PDGFbeta receptor inhibits thrombosis and neointima formation in an avian arterial injury model.

Platelet-derived growth factor (PDGF) is a major mediator of neointima formation after arterial injury. We constructed a recombinant adenovirus, Ad/PDGFtr, that expresses the soluble extracellular domain of the murine PDGFbeta receptor (PDGFtr). The expressed PDGFtr was appropriately glycosylated and secreted by chicken vascular smooth muscle cells (SMCs) in vitro. The expressed PDGFtr inhibited human PDGF-BB induced receptor autophosphorylation, and also inhibited PDGF-BB induced cell proliferation without affecting PDGF-AA induced mitogenesis. In vivo transduction of balloon-injured rooster femoral arteries with Ad/PDGFtr resulted in expression and secretion of the glycosylated PDGFtr. The expressed PDGFtr significantly inhibited neointima formation compared with controls. Neointima-associated thrombus was significantly reduced in Ad/PDGFtr transduced arteries compared with controls. Thus, in addition to impacting on SMC proliferation and migration, PDGF-BB plays a role in thrombus formation in response to arterial injury. Growth factor inhibition by localized gene delivery constitutes a powerful approach to intervene in the molecular pathways involved in vascular disease.

Relationship of large multifunctional proteasome 7 gene polymorphism with susceptibility to type 1 diabetes mellitus and DR3 gene.

OBJECTIVE: To study the relationship of the large multifunctional proteasome 7 (LMP7) gene polymorphism with susceptibility to type 1 diabetes mellitus (DM-1) and the DR3 gene in south Chinese Han population. METHODS: LMP7 genotypes and the DR3 gene were identified in 71 DM-1 patients and 86 healthy persons (as controls) by polymerase chain reaction-restriction fragment length polymorphism. DM-1 patients and controls were divided into DR3-positive and DR3-negative subjects. The frequencies of LMP7 genotypes and alleles were compared between DM-1 patients and controls respectively in the random subjects and in the DR3-matched subjects. Furthermore, DM-1 patients were divided into 3 groups according to the age of diabetic onset: group A < or = 14 years, group B 15-30 years, group C > or = 31 years. RESULTS: In the random subjects, the frequency of LMP7-B/B was lower (39% vs 58%, P < 0.05) and that of LMP7-B/A was higher (54% vs 31%, P < 0.01) in DM-1 patients than that in controls. In DR3-positive subjects, the frequencies of LMP7 genotypes and alleles showed no differences between DM-1 patients and controls. In DR3-negative subjects, the frequency of LMP7-B/B was decreased (40% vs 61%) and that of LMP7-B/A was increased (55% vs 28%, P < 0.01) in DM-1 patients. The frequencies of LMP7 genotypes and alleles showed no significant differences among different ages of diabetic onset. CONCLUSIONS: LMP7-B/B may be the protective genotype, and LMP7-B/A may be the susceptible genotype of DM-1, and this may not be affected by the DR3 gene. Persons with LMP7-B/B may have a decreased risk, and those with LMP7-B/A have an increased risk suffering from DM-1. The LMP7 gene may not be associated with the age of diabetic onset.

[Transconjunctival approach to the fractures of the orbital floor and infraorbital rim].

OBJECTIVE: To study the surgical approach to the orbital floor and infraorbital rim. METHOD: 15 cases of fractures of the orbital floor and infraorbital rim were treated with the transconjunctival approach. RESULT: All fractures of the orbital floor and infraorbital rim were repositioned and fixed through transconjunctival approach. Except for 1 case with mild canthal malposition, no other complication was observed. CONCLUSION: The transconjunctival approach can provide adequate exposure of the infraorbital rim and orbital floor without scar formation in the face.

[The study on the occult metastases to cervical lymph node in squamous cell carcinoma of the larynx and hypopharynx].

OBJECTIVE: To study the metastastic regularity of cervical lymph node in squamous cell carcinoma of the larynx and hypopharynx, which will provide some theoretical basis for surgeon to use selective neck dissection (SND) in some patients with carcinoma above. METHOD: Sixty-nine originally cN0 staged neck dissection specimens from 40 patients were evaluated using mutipl-level sectioning in 4 microns intervals, which were respectively processed by H&E-staining and immunostaining with an antibody to cytokeratin (CK AE3). These patients weren't given any treatment (radiotherapy or chemotherapy or surgery in neck) before surgery. All cases had been followed up 1 year or more. RESULT: The examination of lymph nodes from 40 patients revealed 31 (0.59% 31/2219) occult metastases in 14 (35% 14/40) patients. Among them, six patients with supraglottic cancer, 1 patient with transglottic cancer and 7 patients with hypopharyngeal cancer. There were no occult metastases to cervical lymph node among 9 patients with glottic cancer. All occult metastases mainly located in ipsilateral level II and/or level III. Occult metastases weren't found in nodes from level I, level IV and level V. CONCLUSION: The study further prove that metastases to cervical lymph node mainly occur in the ipsilateral level II and level III among patients with carcinoma of the larynx and hypopharynx. According to the risks of occurring occult metastases to cervical lymph node, we suggest applying to ipsilateral or bilateral SND (level II and level III be dissected) among patients with carcinoma of the larynx and hypopharynx which staged T2-T4 N0M0 clinically.

[Application of nasal endoscope in diagnosis and treatment of epistaxis].

OBJECTIVE: To investigate the indication for diagnosis and treatment of epistaxis under nasal endoscope. METHOD: The data of 226 epistaxis cases were analized. Under nasal endoscope: 126 cases were packed, 78 cases were treated with microwave, laser cautery, 42 cases were operated. RESULT: An outstanding visutization was afforded by nasal endoscope and operation conveniencly, diagnosis correctively. CONCLUSION: We think it is an excellent method to examine and treat epistaxis and worthy to be recommented.

[Research on the near-infrared (NIR) photon migration in multi-layered structures of biological tissues].

The phenomena of photon migration in multi-layered structures of biological tissue is very important in the field of non-invasive near-infrared optical diagnosis. In this paper, a two-layered structure consisting of the fat and the muscle of poke was used as the sample. Near-infrared light source and detector were located on the surface of the fat and a part of the photons reflected by the two-layered structure of the tissue were detected. The relationship between the optical current and the distance from source to detector and the relationship between the optical current and the thickness of the fat layer were given. In order to analyze the experiment results, Monte-Carlo simulation was used to calculate the photons migration traces in the uniform tissues and the Fresnel's law is used to determine the refraction and reflection components of a photon package at the boundary between the fat and the muscle. The results showed that the simulation data coincident well with the experimental results. The conclusion is that the experiment and the simulation method provided in this study may be useful to analyze the photon migration in a mult-layered tissue and to optimize the design of the transducer.

Expression and regulation of neuropilin-1 in human astrocytomas.

Vascular endothelial growth factor (VEGF), through activation of its endothelial receptors VEGFR-1 and VEGFR-2, is an important positive modulator of tumor angiogenesis and edema in solid tumors such as malignant astrocytomas. Neuropilin-1 (Npn-1) is a transmembrane receptor expressed by both endothelial and non-endothelial cells, including tumor cells. Npn-1 has been postulated to function as a co-factor in activation of the biologically relevant VEGFR-2, by the most abundant VEGF165 isoform. However, the function of Npn-1 in normal and pathological angiogenesis, its expression pattern in relation to VEGF in tumors such as astrocytomas and whether it is similarly or differentially regulated compared to VEGF remain unknown. In our study, the expression pattern of Npn-1 and VEGF by human astrocytoma cell lines and specimens was closely correlated and associated with malignant astrocytomas. Mitogens, such as epidermal growth factor and activation of p21-Ras, previously demonstrated to be relevant in astrocytoma proliferation and induction of VEGF, also induce Npn-1 expression. Hypoxia, the main physiological inducer of VEGF expression, decreased Npn-1 expression. Increased Npn-1 expression was also demonstrated in a transgenic mouse astrocytoma model. Astrocytomas are an ideal system for furthering our understanding of the functional relevance, if any, of Npn-1 in tumor angiogenesis.

Antifungal rapamycin analogues with reduced immunosuppressive activity.

Several 1,2,3,4-tetrahydro- and 7-N-hydroxycarbamate derivatives of the natural product rapamycin were prepared and assayed for their immunosuppressive and antifungal profiles. Substitutions at the 7-position indicate the possibility of a differentiated immunosuppressive to antifungal profile, whereas 40-position variants of the tetrahydro-analogues did not show similar differentiated activity.

A new MALDI-TOF based mini-sequencing assay for genotyping of SNPS.

A new MALDI-TOF based mini-sequencing assay termed VSET was developed for genotyping of SNPs. In this assay, specific fragments of genomic DNA containing the SNP site(s) are first amplified, followed by mini-sequencing in the presence of three ddNTPs and the fourth nucleotide in the deoxy form. In this way, the primer is extended by only one base from one allele, while it is typically extended by two bases from another allele. The products are then analyzed using MALDI-TOF mass spectrometry. The genotype of the SNP site is identified based on the number of nucleotides added. This assay has been examined using both synthetic and genomic DNA samples. In addition, multiplexed assays were successfully performed to genotype four SNP sites in a single tube. The main aspect of this assay is that it can overcome the key problems associated with the currently used mini-sequencing methods. First, it significantly reduces the stringent high-resolution and extensive desalting requirements that are essential to the pinpoint assay. Second, it avoids the long extension problem associated with the PROBE assay.

Nucleotide binding activity of SecA homodimer is conformationally regulated by temperature and altered by prlD and azi mutations.

SecA ATPase is critical for protein translocation across the Escherichia coli inner membrane. To understand this activity further, the high affinity nucleotide binding activity of SecA was characterized. We found that at 4 degrees C SecA homodimer binds one ADP molecule with high affinity. This nucleotide binding activity was conformationally regulated by temperature: at low temperature SecA affinity for ADP was high with a slow exchange rate, whereas at high temperature the converse was true. Azi- and PrlD-SecA proteins that confer azide-resistant and signal sequence suppressor phenotypes were found to have reduced affinity for ADP and accelerated exchange rates compared with wild type SecA. Consistent with this observation, fluorescence and proteolysis studies indicated that these proteins had a conformationally relaxed state at a reduced temperature compared with SecA. The level of Azi- and PrlD-SecA protein was also elevated in inverted membrane vesicles where it displayed higher membrane ATPase activity. These results provide the first direct evidence for conformational regulation of the SecA-dependent nucleotide cycle, its alteration in azi and prlD mutants, and its relevance to in vivo protein export.

Direct nitric oxide signal transduction via nitrosylation of iron-sulfur centers in the SoxR transcription activator.

Nitric oxide (NO) has diverse roles in intercellular communication and (at higher levels) in immune-mediated cell killing. NO reacts with many cellular targets, with cell-killing effects correlated to inactivation of key enzymes through nitrosylation of their iron-sulfur centers. SoxR protein, a redox-sensitive transcription activator dependent on the oxidation state of its binuclear iron-sulfur ([2Fe-2S]) centers, is also activated in Escherichia coli on exposure to macrophage-generated NO. We show here that SoxR activation by NO occurs through direct modification of the [2Fe-2S] centers to form protein-bound dinitrosyl-iron-dithiol adducts, which we have observed both in intact bacterial cells and in purified SoxR after NO treatment. Functional activation through nitrosylation of iron-sulfur centers contrasts with the inactivation typically caused by this modification. Purified, nitrosylated SoxR has transcriptional activity similar to that of oxidized SoxR and is relatively stable. In contrast, nitrosylated SoxR is short-lived in intact cells, indicative of mechanisms that actively dispose of nitrosylated iron-sulfur centers.

Quasiparticles in the superconducting state of Bi(2)Sr(2)CaCu(2)O(8+delta)

Recent improvements in momentum resolution lead to qualitatively new angle-resolved photoemission spectroscopy results on the spectra of Bi(2)Sr(2)CaCu(2)O(8+delta) (Bi2212) along the (pi,pi) direction, where there is a node in the superconducting gap. We now see the intrinsic line shape, which indicates the presence of true quasiparticles at all Fermi momenta in the superconducting state, and lack thereof in the normal state. The region of momentum space probed here is relevant for charge transport, motivating a comparison of our results to conductivity measurements by infrared reflectivity.
