Evaluation of proteohormon concentrations in serum and seminal fluid and its relationship with sperm count.

The measurement of FSH, LH, and PRL in serum and seminal fluid of 105 men attending a male infertility clinic and 21 men of proven fertility (controls) did not reveal an interrelationship between the spermatozoal count and the level of the proteo-hormones in both biological fluid except for FSH, which was significantly higher in the serum of men with low as compared to normal sperm count (p less than 0.01). FSH in seminal fluid was significantly lower (p less than 0.01), and LH-like activity significantly higher (p less than 0.01) than in serum, while there was no difference between the respective levels of PRL. These results could be confirmed in 4 patients with azoospermia due to occlusive disease, and in 1 out of 2 vasectomized men. It was demonstrated for the first time that the higher concentration of "LH" in seminal fluid as compared to serum was due to the presence of HCG-beta or a HCG-like material cross-reacting with the antiserum against LH.

A comparison of the bovine cervical mucus penetration test and the postcoital test.

In 68 ejaculates an in vitro bovine cervical mucus penetration-test (Penetrak) was compared to the postcoital-test and the spermiogram. There was a positive correlation between the Penetrak-Test and concentration, motility, and morphology of the spermatozoa, however there was no correlation to the postcoital test. In connection with the PCT, the bovine mucus penetration-test seems to be a useful tool in the exploration of male infertility.

Male subfertility and the outcome of intrauterine insemination.

Seven out of 27 infertile women conceived by intrauterine insemination (IUI, 26%), one of them twice. Three other pregnancies occurred spontaneously after the discontinuation of treatment. A comparison of the data obtained from 65 treatment cycles revealed that the number of motile spermatozoa per ml of ejaculate and per ml of medium after swim-up preparation was higher in the ultimately fertile group as compared to the patients who failed to conceive. It is concluded that male subfertility affects the outcome of IUI unfavorably.

[Separation of motile spermatozoa by "Separon" as measured by a videomicrography computer system].

By means of a recently developed apparatus ("Separon") motile spermatozoa were concentrated in a small volume of culture medium (0,3 ml). Sperm concentration, progressive motility and mean velocity of 60 ejaculates were determined by a computerized videomicrography system. After "Separon" sperm concentration was reduced in an average of 30%, mean progressive motility was increased from 27% to 42% (asthenospermic ejaculates) and from 45% to 84% (control ejaculates) respectively. Mean sperm velocity was increased from 18,5 to 34,5 microns/sec. (control ejaculates).
