Cooperative activation of D1-like and D2-like dopamine receptors in the nucleus accumbens shell is required for the reinstatement of cocaine-seeking behavior in the rat.

Activation of D1-like (D1, D5) or D2-like (D1, D3, D4) dopamine receptors in the nucleus accumbens shell is sufficient to reinstate cocaine-seeking behavior in rats. The goal of these experiments was to assess whether cooperative activation of D1-like and D2-like dopamine receptors in the accumbens shell is required to promote cocaine reinstatement. Rats were initially trained to self-administer cocaine (0.25 mg, i.v.) using a fixed-ratio schedule of reinforcement for approximately 21 days. Animals subsequently underwent an extinction phase during which saline was substituted for cocaine. Once cocaine self-administration behavior was extinguished (defined as <15% of the total responses maintained during self-administration), dopamine receptor agonist-induced reinstatement of cocaine seeking was assessed. Administration of the selective D1/5 agonist R-(+)-6-chloro-7,8-dihydroxy-1-phenyl-2,3,4,5-tetrahydro-1H-3-benzazepine hydrobromide (SKF-81297) (1.0 microg) or the D2/3 receptor agonist trans-(-)-(4aR)-4,4a,5,6,7,8,8a,9-octahydro-5-propyl-1H-pyrazolo[3,4-g]quinoline hydrochloride (quinpirole) (3.0 microg) directly into the nucleus accumbens shell promoted reinstatement of cocaine seeking. In order to determine if endogenous dopamine tone in the accumbens shell is required for dopamine receptor agonist-induced reinstatement of cocaine seeking, D1/5 or D2/3 dopamine receptor antagonists were administered into the nucleus accumbens shell prior to a selective dopamine receptor agonist. Microinfusion of the D2/3 dopamine receptor antagonist sulpiride ((S)-5-aminosulfonyl-N-[(1-ethyl-2-pyrrolidinyl)methyl]-2-methoxybenzamide) (1.0 microg) into the nucleus accumbens shell 10 minutes prior to SKF-81297 (1.0 microg) blocked the ability of this D1-like dopamine receptor agonist to reinstate cocaine seeking. Similarly, administration of the selective D1/5 dopamine receptor antagonist R(+)-7-chloro-8-hydroxy-3-methyl-1-phenyl-2,3,4,5-tetrahydro-1H-3-benzazepine hydrochloride (SCH-23390) (1.0 microg) into the nucleus accumbens shell prior to quinpirole (3.0 microg) blocked reinstatement of drug-seeking behavior elicited by this D2/3 dopamine receptor agonist. Moreover, intra-accumbal shell co-administration of subthreshold doses of quinpirole (1.5 microg) and SKF-81297 (0.1 microg) promoted cocaine-seeking behavior. Collectively, these results indicate that cooperative activation of D1-like and D2-like dopamine receptors in the nucleus accumbens shell is necessary to reinstate cocaine seeking in rats.

Blood group and serum protein polymorphisms in a population group of Moldavians.

The distribution of the alleles and haplotypes for blood groups A1A2B0, MNSs, RHESUS, P1, KELL-CELLANO and biochemical markers of the alleles of loci AMY2, HPA, GC, C3, TF, BF, CP, PI (including subtypes) were studied in 125 Moldavian individuals from Karahasani settlement, Stefan-Voda District, Republic of Moldavia. The results show that the gene pool of Moldavians is similar to those of Southeastern European populations.

Haemogenetic studies in Bulgaria.

Within a study of the genetics of Bulgarian populations, the markers AB0, RH, MNS, P, HPA, GM(1), PGM1, ACP and ESD were examined in 2346 individuals from seven subpopulations of south-central and south-eastern Bulgaria. The combined data have been compared with other populations of the Balkans.

Dermatoglyphic studies in eastern and south-eastern Europe.

In nine population samples from Bulgaria, Croatia, Greece, Hungary, Republic of Macedonia (Skopje and Aromuns from Stip region), Romania, Serbia and Slovakia 12 dermatoglyphic variables have been studied. There are distinct differences between the populations and between males and females. The Macedonian Aromuns are clearly separated from the other populations.

Allele frequencies of D21S11, FGA, TH01, and VWA in populations of the Balkans.

This study is part of an extensive investigation of the genetic relationship between Balkan populations, especially the Aromuns. Allele frequencies of four STRs (D21S11, FGA, TH01, VWA) from Macedonians (Skopje), Gramostian Aromuns from the Stip region (Macedonia), Moskopolian Aromuns from Krusevo (Macedonia), and Musequiar Aromuns from Dukasi (Albania) are presented.

Phenotype and allele frequencies of some serum protein polymorphisms in populations of the Balkans.

Within a study of the genetics of Southeastern European populations seven serum protein polymorphisms (AMY2, BF, C3, CP, GC, HPA, TF) were examined in two samples of Aromuns and one reference sample (Musequiar-Aromuns from Dukasi in Albania, Moskopolian-Aromuns from Krusevo, Republic of Macedonia, and Macedonians from Skopje). The neighbor joining tree as well as the principal component analysis show results which do not correspond well to the geographic and historic background. This indicates that in the present case the serum protein polymorphisms give no clearly defined information about the relationships between the Balkan populations and to the origin of Aromuns.

[Rugen (Germany): hemogenetic study of an island population].

24 haemogenetic markers (5 erythrocyte antigens, 7 polymorphisms of serum proteins. 12 polymorphisms of red cell enzymes) had been studied in up to 171 individuals from the island of Rügen (Germany, Baltic Sea). The cluster analysis separates clearly the ügen sample just as the islands of Hiddensee and Ummanz from the neighbouring populations. The comparison of the data with neighboured larger populations as for instance Denmark, Hamburg or Sweden clearly results in an exceptional position of the island of Rügen. The possible reasons are discussed.

Blood group frequencies in Romania: microregional and ethnic differences.

This study is based on the results of the blood group typing (AB0, MN, RH: D/d) of two samples from Romania and on literature data. The 40 population samples used are scattered on the today's territory of Romania. Some data refer to more or less endogamous villages or groups of villages which belong to the same marriage area, some data refer to towns with admixed populations. Samples of nine ethnic minorities have been included in the study. The results have been discussed on the basis of the historical and geographical background.

Influence of perfusate calcium concentration on the inotropic insulin effect in isolated guinea pig and rat hearts.

Conflicting data on the inotropic effect of insulin are present in the literature suggesting a positive inotropic property or no inotropic effect or even a negative influence. To clarify the reason for these diverging findings, dose-response curves of insulin have been performed in isolated working rat and guinea pig hearts perfused with Krebs-Henseleit buffer containing 0.9, 1.25, 2.5, and 5 mM Ca(2+) at 37 degrees C. At 1.25 mM [Ca(2+)], insulin (8 to 16 IU) regularly improved the inotropic state. LVdP/dt(max) increased significantly from 1,900 to 2,300 mm Hg/s (+21 %) in guinea pig hearts and from 3,197 to 4,345 mm Hg/s (+36 %) in rat hearts. LVEDP did not change significantly. Myocardial oxygen consumption increased parallel with contractility. Heart rate was not influenced in either species. Coronary flow increased by 16.5 % in guinea pig hearts, but decreased in rat hearts by 13.6 % (p < 0.05 each). With 0.9 mM [Ca(2+)] the positive inotropic effect of insulin did not further augment. At 2.5 mM [Ca(2+)] insulin exhibited in both species no significant change of LVdP/dt(max) but very high insulin doses depressed the heart. At 5 mM [Ca(2+)] insulin depressed the heart significantly already at lower concentrations. At 31 degrees C and 1.25 mM [Ca(2+)] the positive inotropic insulin effect was preserved. We conclude that the positive inotropic insulin effect in rat and guinea pig hearts depends on the extracellular [Ca(2+)], i.e., is maximal around 1.25 mM [Ca(2+)] and is reduced or absent at higher [Ca(2+)] or may even become negative.

Allele frequencies of alpha-1-antitrypsin (PI) in the Balkans.

The phenotype and allele frequencies of alpha-1-antitrypsin has been studied by an IEF technique (pH 4.2-4.9) in ten population samples from the Balkans. The allele frequencies varied from 0.6667 to 0.7361 (*M1), 0.1100 to 0.1793 (*M2), 0.0992 to 0.1700 (*M3), 0 to 0.0105 (*S), 0 to 0.0078 (*Z) and 0 to 0.0172 (others). The results were compared with data from South and Middle European populations from the literature. Most of the populations form a cluster with small genetic distances, and a weak relationship to geographical distributions. In contrast, the samples from Southern France, the Iberian Peninsula and Madeira form a clearly separated cluster. The differences are mainly based on high frequencies of PI*S in the latter populations.

Isolated working rat heart adaptation after abrupt changes in extracellular Ca2+ concentration.

In isolated working rat hearts, a rapid up- or downward change in perfusate Ca2+ concentration by the factors 2, 4, or 8 in the range between 1.25 and 10 mM resulted within 1 min in the well-known change of left ventricular contractility as evaluated by maximum left ventricular pressure change velocity (LVdP/dt max) and left ventricular end-diastolic pressure (LVEDP). However, within about 10 min thereafter, contractility showed an adaptive behaviour opposite to the initial change, with t 1/2 values for LVdP/dt max between 1.45 and 2.8 min. The adaptive LVdP/dt max reactions amounted to 10-35% of the initial change. With an abrupt fall from 10 mM to 1.25 mM Ca2+ (not tolerated by all hearts), LVdP/dt max decreased initially by 4.260 mmHg/s (LVEDP +9.7 mmHg) and increased thereafter by 524 mmHg/s (LVEDP -6.8 mmHg). The adaptive inotropic behaviour cannot be related to changes in heart rate or coronary flow and is not affected by thapsigargin (1 microM) or the amiloride derivative benzamil (10 microM). This suggests that sarcoplasmatic Ca2+-ATPase and sarcolemmal Na+-Ca2+ exchange do not play a decisive role in this adaptive behaviour. In conclusion, an intrinsic regulatory mechanism of the myocardium attenuates the inotropic effect of acute changes in Ca2+ concentration. This phenomenon might protect the heart against Ca2+ overload after an acute rise in catecholamine concentration.

[Helgoland (Germany): hemogenetic study of an island population].

24 haemogenetic markers (5 erythrocyte antigenes, 6 polymorphisms of serum proteins, 12 polymorphisms of red cell enzymes) had been studied in up to 80 individuals from the island of Helgoland (Germany). The cluster analysis separates clearly the Helgoland sample from the neighbouring populations as well as from European standard data. This special position is interpreted partly by genetic peculiarities developed in the course of time, partly as a consequence of genetic drift.

Isonymy, consanguinity and repeated pairs of surnames in Aromun populations.

The Aromuns represent a small and almost unknown people that live scattered over the Balkan Peninsula. Due to their language, that is very similar to classical Latin, they are in a special position. The Aromuns settled only in more recent times. Until now they lived as shepherds, as caravan guides and merchants and lead a semi-nomadic life. We are currently carrying out studies to determine the genetic structure of this population. To facilitate the interpretation of these data, we are also trying to obtain other important parameters that pertain to migration processes and the genealogical structure of this populations. The data arise from three areas in Albania, the Republic of Macedonia and Romania. The inbreeding coefficient and the proportion of repeated pairs of surnames was calculated through the use of genealogies and the isonymy method. The difference between these three populations are due primarily to confounding by selection of mates and family composition.

Population genetic studies in the Balkans. I. Serum proteins.

Within a study of the genetics of Southeastern European populations seven serum protein polymorphisms (AMY2, BF, C3, CP, GC, HPA, TF) were examined in three samples of Aromuns (Albania: the village of Andon Poci, province Gjirocaster, Republic of Macedonia: Stip region, Romania: the village Kogalniceanu, province Dobruja) and four reference samples (Albanians: Tirana, Romanians: Constanta and Ploiesti as well as Greeks (Northeastern Greece)). The Aromun samples from Albania and Romania form one separate cluster and the reference samples together with the Aromuns from Macedonia (Stip region) form a second one.

Population genetic studies in the Balkans. II. DNA-STR-systems.

Within a study of the genetics of Southeastern European populations four DNA-STR-systems (D21S11, FGA, TH01, VWA) were examined in seven samples (samples of three Aromuns and four other Balkan populations). The results have been compared to data from four samples from literature (Austrians, Germans, Hungarians, Slovenians). The results show three clusters: a) the Aromuns from Albania (Andon Poci) and Macedonia (Stip region), b) the Romanian Aromuns (Kogalniceanu), Romanians (Constanta, Ploiesti) and Albanians (Tirana) und c) the data from literature. A sample of Northeastern Greece clearly differs from these three clusters. Including seven serum protein polymorphisms (without the populations from literature) results in two clusters: a) the three Aromun populations and b) Albanians and Romanians. Again the sample of Northeastern Greece clearly differs from these clusters.

[Variability of blood group frequencies in Romania].

This paper presents the results of a study of two population samples from Dobruja and Muntenia (Romania) as well as data from the literature (AB0, MN, Rhesus (D/d) and HPA). The data have been pooled according to the different regions of Romania. Interestingly, the Carpathian Mountains do not form a genetical barrier: there are clear genetical similarities between the provinces east and west as well as between the provinces south and north from the mountains. The differences and similarities between Romania as a whole and other populations in this region reflect the historical processes of the last centuries.

DNA-PCR systems TH01 and VWA31: population data from Albania and Romania.

The microsatellite systems TH01 and VWA31 have been typed and analyzed for three populations from Albania (city population of Tirana, and Aromuns from a small-sized village, named Andon Poci, near Gjirokaster) and Romania (city population of Constanta). The genetic data have been compared with other, particularly South-Eastern European population.

Interbeat interval variability in isolated working rat hearts at various dynamic conditions and temperatures.

This study quantifies the effect of afterload and preload changes and of temperature on interbeat interval variability of the intact isolated heart. Ventricular pressure pulse records were obtained from isolated working rat hearts. The variability of interbeat intervals (BIs) was quantified by C90, the central 90% range of the BIs during 10 min periods; predominant frequencies were searched for by power spectral analysis. At 37 degrees C the BI lengths oscillated pseudo-randomly with BI variability C90< or =4 ms. Alternating signs of consecutive BI differences were predominant, and no peaks. were seen in the power spectra. Changes in end-diastolic and aortic pressure had little effect. From 37 degrees C down to 27 degrees C the variability increased about sevenfold, run phase length became randomly distributed, and individual, time-variant peaks occurred in the power spectra. BI variability vanished during atrial pacing. We conclude that: (1) effective mutual synchronization with minimal fluctuation happens within the sino-atrial node of intact rat hearts at body temperature, and synchronization is not affected even by extreme changes in pre- and afterload, (2) the sino-atrial node is the sole source of BI variability in the intact isolated rat heart, (3) low temperature hampers this functional organization which can be reestablished by sinus node accelerating agents (isoprenaline, theophylline), (4) decreasing frequency by N6-Cyclopentyladenosine at normothermia also increases BI variability but less pronouncedly than hypothermia does.
