Clinical evaluation of radioimmunoimaging with 131I-C0C183B2 monoclonal antibody against ovarian carcinoma by intraperitoneal injection.

Forty-eight cases subjected to radioimmunoimaging (RII) by intraperitoneal injection with 131I-C0C183B2 monoclonal antibody (MAb) prepared in our laboratory were studied. Thirteen of 14 cases of proved primary ovarian carcinoma were positive. In 11 follow-up cases of ovarian carcinoma after initial surgery and chemotherapy, 5 recurrences were positive and 6 cases without recurrence were negative; all were confirmed histopathologically after a second operation. One false negative was ovarian mucinous adenocarcinoma, which also negatively stained with C0C183B2 by the peroxidase anti-peroxidase method. Twenty of 23 cases of nonepithelial or metastatic carcinoma of the ovary, benign tumors, and benign diseases were negative. The sensitivity and specificity were 94.7 and 89.7%, respectively. If patients had complications with ascites, the MAb which positively stained with the cancer cells in the ascites was chosen for RII. For follow-up cases PAP staining with the tumor tissue from the initial surgery and the MAb should be done before RII. These are the principal factors that increase the positive rate and accuracy of RII. The intraperitoneal route seems to be a valuable method for clinical staging and tumor localization as well as for follow-up use.

[Association between essential hypertension and immunology].

Using the immunofluorescence method, autoantibodies of serum IgG was investigated in 100 consecutive patients of essential hypertension with or without family history of hypertension. 30 healthy normotensive subjects with family history of hypertension and 40 healthy normotensive subjects of the same age range were also studied. The results showed that in the treated patients, the frequency of smooth muscle antibodies (SMA), antimitochondrial antibodies (AMA), antinuclear antibodies (ANA) and heart reactive antibodies (HRA) was higher than that in healthy normotensive subjects. The frequency of these antibodies was not associated with difference in mean arterial blood pressure, sex, and clinical stage and a possible genetic predisposition is suggested. The autoantibodies may even appear before the elevation of blood pressure in patients with essential hypertension. The above-mentioned results show that lack of autoimmunity may be one of the pathogenetic factors of essential hypertension.

Antithrombotic activity of verapamil.

The antithrombotic activity of verapamil (Ver) was assessed in an arterial thrombosis model and an ex vivo thrombosis model. Ver 2 and 4 mg.kg-1 ip markedly prolonged the time of thrombotic occlusion in rat carotid artery induced by electric stimulation. The weight of thrombus formed ex vivo was reduced by Ver 0.2 mg.kg-1 iv. However, Ver showed no effects on blood viscosity.

A preliminary study of radioimmunoimaging by 131I-COC183B2 monoclonal antibody in patients with epithelial ovarian cancer.

Monoclonal antibody 131I-COC183B2, developed in our laboratory and proved to fit for human treatment was injected intraperitoneally or subcutaneously in 13 patients. In 8 cases with i.p. injection the disease corresponded with the image, i.e. 3 primary ovarian epithelial cancers showed positive images, 1 ovarian Krukenberg tumor was negative and the other 4 negative images included 1 uterine myoma and 3 ovarian teratomas. In the subcutaneous injection group, 4 cases had ovarian carcinoma, surgery and chemotherapy. Two negative images corresponded with the clinical status-in good health, another negative case had metastatic left supraclavicular lymph node due to ovarian mucinous adenocarcinoma. The last negative image in this group was a case of benign ovarian teratoma which was proved after surgery. The 1 positive case was waiting to be proved by a scheduled third operation. The computer scintigram calculation of T/NT was 5.35 to 13.7. The results suggest that this monoclonal antibody can be used for radioimmunoimaging for the localization of ovarian carcinoma, which is not only helpful for clinical staging and differential diagnosis but is also a good follow-up method.

Generation and characterization of human monoclonal antibody HMD4 against ovarian carcinoma and the study of radioimmunoimaging in nude mice.

Lymphocytes from regional lymph nodes of patients with ovarian carcinoma were immortalized by fusing them with a nonsecreting cell line of murine myeloma (Sp2/0-Ag14). By early cloning and recloning a hybrid cell line, named HMD4, was established. It has secreted human IgG for more than 15 months stably. Chromosome analysis corresponded with the characterization of human-mouse hybridoma. Large quantities of ascites were obtained after hybrid cells injection into the primed nude mice. Human IgG of light chain was detected and purified from the ascites. Twenty-six of 43 (60.5%) epithelial ovarian cancers were positively stained with HMD4 by ABC immunoperoxidase methods while nonepithelial ovarian cancers and almost all benign tumors and normal tissues were negative. The molecular weight of the antigen recognized by HMD4 was 55KDa determined by Western blotting. 131I labeled HMD4 was administered intraperitoneally to nude mice bearing human ovarian epithelial adenocarcinoma; 131I labeled normal human IgG and normal murine IgG were used as controls. Measurements of T/NT and T/B ratios of 131I-HMD4 were done. Radioimaging showed HMD4 clearly localized on tumor regions at 48 and 72 hours and the biodistribution and metabolism of the labeled HMD4 corresponded with the images. The above results indicate that HMD4 was specific to ovarian carcinoma, a hopeful clue for clinical applications.

A human monoclonal antibody HMD4 against ovarian carcinoma associated antigen.

Lymphocytes from regional lymph nodes of patients with ovarian carcinoma were immortalized by fusing them with a nonsecreting cell line of murine myeloma (Sp2/0-Ag14). The fusion rate was 0-87.5%. By early cloning and recloning, a hybrid cell line, named HMD4, was established. It has stably secreted human IgG for more than 15 months. Chromosome analysis showed the characteristics of human-mouse hybridoma. The cells of HMD4 were injected into the abdominal cavities of nude mice and 2-3 weeks later large quantities of ascites were obtained. Human IgG of lambda light chain was detected and purified from the ascites. The specificity of HMD4 human McAb was tested by ABC or PAP immunoperoxidase stainings of paraffin-embedded tissue sections, cryostat sections, cell smears of various tissues and different cancer cell lines. 60.5% (26/43) of epithelial ovarian cancers was positive, while nonepithelial ovarian cancers, most cancers from other organs and almost all nonmalignant and normal tissues were negative. The molecular weight of the antigen recognized by HMD4 was 55 KDa determined by Western blotting. The problems of maintaining the IgG secreting function of human-mouse hybridoma and its screening were also discussed.

[Biodistribution, pharmacokinetics and imaging of 131I-labeled HMD4 human McAb in human ovarian carcinoma xenograft in nude mice].

HMD4, a human IgG (lambda) monoclonal antibody against ovarian carcinoma, was purified by affinity chromatography of CNBr-activated Sepharose 4 B coupled with goat antihuman IgG, 131I-HMD4 was prepared by chloramin-T or Iodogen method. The nude mice bearing human ovarian carcinoma (OSC-2) were intraperitoneally injected with 131I-HMD4, while 131I-labeled normal human IgG and normal murine IgG were used as controls. Imaging was performed daily using a gamma camera. For analysis of distribution and metabolism of 131I-HND4 in the nude mouse models, T/NT and T/B ratios were calculated. The HMD4 was specifically localized at the tumor masses in nude mice and the best imaging was obtained at 72 hours after injection. Total positive rate of imaging was 82%, while negative rate in the controls was 91%. Radioimmunoimaging index was highest (4.17) at 48 hours after injection. The average half life of 131I-HMD4 in the blood was about 30 hours.

Generation and characterization of three monoclonal antibodies to human ovarian epithelial adenocarcinomas.

Three murine monoclonal antibodies COCL66-9, COC183A5 and COC183B2 were raised against human epithelial ovarian adenocarcinoma with soluble antigens. The monoclonal antibodies were stable after culture and stored under 80 C for 9 to 20 months. The subclass and titre were all examined and the reactivities were tested on various paraffin embedded tissue sectionism by immunoperoxidase stainings. The results of positive staining against ovarian epithelial adenocarcinoma were 77.7%, 87.2% and 75.6% for COC166-9, COC183A5 and COCL83B2 respectively. Human ovarian carcinoma xenograft in nude mice were radioimaged after 131I-COC183B2 administration. Gamma scintigraphy demonstrated specific localization of the radiolabeled monoclonal antibody on these tumors. The results showed a bright future for immunodiagnosis and immunotherapy.

[The distribution, characteristics and effects of ir-BNP in central and peripheral tissues in rat].

The present study first investigated the distribution, biochemical characteristics, receptor binding and biological effects of immunoreactive brain natriuretic peptide (ir-BNP), in the central nervous system and some peripheral tissues in rats, using highly specific radioimmunoassay, radioreceptor assay and immunohistochemical method. The results suggest that BNP may be a novel neurotransmitter or circulatory hormone, which is widely distributed in various tissues and involved in the regulation of water electrolyte balance and cardiovascular activity.

Efficient expression of unfused human alpha D-interferon in Escherichia coli using overlapping termination and initiation codons (TGATG) in its signal sequence.

A plasmid carrying the lambda PL promoter was constructed to express efficiently unfused human alpha D-interferon (HuIFN-alpha D) in Escherichia coli using a TGATG site in its signal sequence, which occurs also in the lambda DNA sequence. The unfused nature of HuIFN-alpha D expressed by pBV867 in E. coli (BMH 71-18) was confirmed by the following evidence: first, the purified IFN showed a single band of 19.5K in sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE); a 27K band, representing lambda N-HuIFN-alpha D fusion protein, was not detected. Second, the peak of IFN activity coincided with the 19.5K protein. Third, the peak of absorbent material for human leukocyte IFN antibody coincided with that of IFN activity. Finally, amino-terminal sequencing of purified IFN demonstrated an unfused HuIFN-alpha D. This suggests that E. coli is able to process the signal sequence of HuIFN-alpha D. Studies on the mechanism of "translational coupling" initiation of gene expression were carried out by the construction of two hybrid plasmids and titration of the IFN activities produced by them. The level of expression by the ATG-TGATG initiation mode was found to be six times higher than that of the single ATG mode.
