Using oligonucleotides complementary to the conserved inter-finger region of a variety of previously described zinc finger-encoding genes, a novel mouse gene was cloned and characterized. The gene is localized on chromosome 8 and comprises five exons. Its corresponding mRNA is developmentally regulated in various tissues and includes an open reading frame encoding a protein of 72,422 daltons. It shares amino-terminal homologies with human KRAB (or FPB) boxes, and contains 13 zinc fingers of the C2-H2 type. The NK10 KRAB domains exhibit repressing activity when tested in GAL4 fusion protein assays. Cloning of putative target sequences revealed that the individual domains differentially contribute to zinc-dependent target DNA binding.
Neural-cell adhesion molecules (N-CAMs) are members of the immunoglobulin superfamily mediating homo- and heterophilic cell-cell interactions. N-CAM exists in various isoforms which are generated by alternative splicing. During embryonic development, N-CAMs are expressed in derivatives of all three germ layers, whereas in the adult animal they are predominantly present in neural tissue. Processes like neurulation, axonal outgrowth, histogenesis of the retina and development of the olfactory system are correlated with the regulated expression of N-CAMs. We show here that N-CAM-deficient mice generated by gene targeting appear healthy and fertile, but adult mutants show a 10% reduction in overall brain weight and a 36% decline in size of the olfactory bulb. N-CAM deficiency coincides with almost total loss of protein-bound alpha-(2,8)-linked polysialic acid, a carbohydrate structure thought to be correlated with neural development and plasticity. The animals showed deficits in spatial learning when tested in the Morris water maze, whereas activity and motor abilities appeared normal.

In a 6-year-old boy, we found a malignant blue nevus with metastases of the lymph nodes. After surgical therapy under melanoma conditions, he was treated with interferon. Up to now (2 years after therapy), the follow-up examinations have been negative.

This report deals with the clinical, ultrasonographical, computertomographical and morphological aspects of the rare malignant fibrous histiocytoma of the prostatic gland. A 80-year-old patient suffered from a fast growing painless tumor of the prostatic area, which, on microscopical examination, proved to be a malignant fibrous histiocytoma. The patient refused surgical treatment and died several weeks thereafter. On autopsy no metastases were found. The diagnosis of a malignant fibrous histiocytoma of the prostatic gland can only be made on microscopical grounds. The differential diagnosis includes a pleomorphic rhabdomyosarcoma or a pleomorphic liposarcoma. The clinical differential diagnosis includes, first of all, a carcinoma of the prostatic gland. Surgery is the therapy of choice. The prognosis of the malignant fibrous histiocytoma of the prostatic gland is poor.

A case report is given of a 65 year old female patient harboring a rare mucoepidermoid carcinoma of the stomach. On X-ray examination a smooth faced mass with a diameter of 3 cm was found in the corpus region of the stomach. During gastroscopy the tumor was seen to be localized in the lower third of the stomach close to the lesser curvature with an ulceration on top. On microscopic examination a cornified squamous cell carcinoma was found including interspersed mucilaginous adenocarcinoma tissue. Prognosis of mucoepidermoid carcinoma is bad, histogenesis controversial. Discussion of this case as well as of other unusual adenocarcinomas for example with trophoblastic differentiation has to include the possibility of bidirectional development of undifferentiated stem cells. However it still remains unclear, why such tumors of the stomach are almost always adenocarcinomas and not signet ring cell carcinoma.
A case report is given of a 28 year old homosexual suffering from segmental proctitis caused by Chlamydia. The patient had watery diarrhea and fever since 3 weeks. Stool specimen testing was negative in regard to Salmonella, Shigella and worm eggs. Infection with N. gonorrhoea, Herpes simplex virus, Toxoplasma gondi, Yersinia and M. tuberculosis could be excluded. Serological testing yielded results suggestive of an old lues infection successfully treated. On histological examination typical reticulocytary suppurating granuloma were found. Therapy of choice of Chlamydia-proctitis is tetracyclines.

This article reports on clinical as well as light-microscopical and electron-microscopical findings in a fibrous histiocytoma of the left external auditory canal. The patient, who was then 67 years old, complained of a space-occupying mass in the external auditory canal which resulted in partial deafness. On microscopical examination a 3 x 2 cm large tumour was found to be located at the boundary between the osseous and the cartilaginous auditory canal. The covering skin was intact. On histological examination we identified a moderately cellular, locally aggressive fibrous histiocytoma. To our knowledge, no fibrous histiocytoma of this localisation has ever been reported. The diagnosis can be made on histological grounds only. Differential diagnosis includes, besides a ceruminoma, almost all benign and malignant non-epithelial tumours of the external auditory epithelial canal. Surgical removal is the therapy of choice. One year after the operation there has been no relapse.

The clinical and morphological features of Bowen's disease of the nipple are reported. The 51 year old patient had an eczema type change of the left nipple for over 1 year. The microscopic evaluation of the extensive segmental resection showed Bowen's disease of the nipple. Paget's disease was excluded with certainty. As far as the authors know this is the first case of Bowen's disease of the nipple reported. The treatment of choice is extended segmental resection. With this treatment the prognosis of Bowen's disease of the nipple appears to be favourable.
