Heterogeneity of nuclear lamin A mutations in Dunnigan-type familial partial lipodystrophy.

We previously identified a novel mutation, namely LMNA R482Q, that was found to underlie Dunnigan-type partial lipodystrophy (FPLD) and diabetes in an extended Canadian kindred. We have since sequenced LMNA in five additional Canadian FPLD probands and herein report three new rare missense mutations in LMNA: V440M, R482W, and R584H. One severely affected subject was a compound heterozygote for both V440M and R482Q. The findings indicated that 1) a spectrum of LMNA mutations underlies FPLD; 2) aberrant lamin A, and not lamin C, is likely to underlie FPLD, as R584H occurs within LMNA sequence that is specific for lamin A; 3) the V440M mutation may not cause lipodystrophy on its own; 4) compound heterozygosity for V440M and R482Q is associated with a relatively more severe FPLD phenotype, but not with complete lipodystrophy; and 5) variation in the severity of the phenotype might be related to environmental factors.

Disruption of Golgi structure and function in mammalian cells expressing a mutant dynamin.

The large GTPase dynamin is a mechanoenzyme that participates in the scission of nascent vesicles from the plasma membrane. Recently, dynamin has been demonstrated to associate with the Golgi apparatus in mammalian cells by morphological and biochemical methods. Additional studies using a well characterized, cell-free assay have supported these findings by demonstrating a requirement for dynamin function in the formation of clathrin-coated, and non-clathrin-coated vesicles from the trans-Golgi network (TGN). In this study, we tested if dynamin participates in Golgi function in living cells through the expression of a dominant negative dynamin construct (K44A). Cells co-transfected to express this mutant dynamin and a GFP-tagged Golgi resident protein (TGN38) exhibit Golgi structures that are either compacted, vesiculated, or tubulated. Electron microscopy of these mutant cells revealed large numbers of Golgi stacks comprised of highly tubulated cisternae and an extraordinary number of coated vesicle buds. Cells expressing mutant dynamin and GFP-tagged VSVG demonstrated a marked retention (8- to 11-fold) of the nascent viral G-protein in the Golgi compared to control cells. These observations in living cells are consistent with previous morphological and in vitro studies demonstrating a role for dynamin in the formation of secretory vesicles from the TGN.

An alternative base-pairing of catechol-bearing nucleosides by borate formation.

A chelator-type beta-C-nucleoside having a catechol ligand as the nucleobase was found to form a stable 2:1 complex with trimethyl borate, which was characterized by 1H-NMR and electrospray ionization time-of-flight (ESI-TOF) mass spectroscopies. Both phosphoramidite and phosphotriester derivatives of this nucleoside were also prepared as synthetic precursors for DNA oligomer syntheses.

Clinical utility of HNF1A genotyping for diabetes in aboriginal Canadians.

OBJECTIVE: To determine the diagnostic performance characteristics of HNF1A genotyping for diabetes and impaired glucose tolerance (IGT) in Canadian Oji-Cree Indians. RESEARCH DESIGN AND METHODS: We studied all Oji-Cree subjects > or = 50 years of age (96 subjects) who had participated in a community-wide prevalence survey for type 2 diabetes. Subjects were classified either as having "disease," which included type 2 diabetes and IGT, or not. All subjects were genotyped for the HNF1A G319S mutation. RESULTS: The prevalence of disease in this group was 65.7%, of whom 71.4% had type 2 diabetes. For a carrier of HNF1A S319, the specificity, sensitivity, and positive and negative predictive values were 97.0, 30.1, 95.0, and 42.1%, respectively. When the pretest disease prevalence was accounted for, the probability of disease after a positive test was 97.2%, and the probability of disease after a negative test was 42.2%. The values were very similar for the subgroup of subjects with type 2 diabetes alone. CONCLUSIONS: The HNF1A genotype appears to be the most specific genetic test yet reported for the prediction of a common multifactorial disease by applying present-day standards of clinical epidemiology in molecular genetics. A positive test result had particular diagnostic value in the Oji-Cree: a subject with HNF1A S319 was virtually certain of having diabetes or IGT by 50 years of age. In contrast, a subject without HNF1A S319 had a reduced risk compared with the age-specific prevalence but was not totally risk-free. Because HNF1A S319 was not the only predisposing factor for diabetes in the Oji-Cree, subjects without HNF1A S319 were still at some risk for diabetes or IGT.

[Effects of ischemic preconditioning on apoptosis of hepatocytes in liver transplantation in rats].

OBJECTIVE: To evaluate the effects of ischemic preconditioning (IP) on hepatocyte apoptosis in ischemia/reperfusion injury of grafted liver and clarify its mechanism. METHODS: Orthotopic liver transplantation with cuff anastomosing method for the portal vein, suprahepatic vena cava and infrahepatic vena cava in the rats were divided into two groups: with and without IP treatment. The parameters of liver function were determined by auto-biochemical analyser. The concentrations of malondialdehyde (MDA) were measured with chromometry. The apoptotic hepatocytes in grafts liver were detected by flow cytometry combined with the in situ terminal deoxynucleotidyl transferase-mediated dUTP-FITC nick end-labeling (TUNEL) method. RESULTS: The levels of AST, ALT, LDH in the serum, and MDA and apoptotic cells in the liver were significantly increased compared with normal values after ischemia/reperfusion to the grafted liver. After IP treatment, the levels of AST, ALT, LDH, MDA and apoptotic cells were significantly decreased. CONCLUSION: IP can attenuate ischemia/reperfusion injury by alleviating lipid peroxide and decreasing apoptosis of hepatocytes.

[Abdominoplasty with a combined technique of lipectomy and liposuction].

OBJECTIVE: To evaluate an abdominoplasty technique combined with lipectomy and liposuction. METHODS: Twenty-five patients were undergoing for the abdominoplasty with lipectomy and liposuction in the same operation procedure. A public incision with W-shape was used for the lower abdominal lipectomy, and the upper abdomen was treated with an electronic liposuction technique. After the operation, each patient was followed up for evaluation of the results. RESULT: All of the patients got good contour of the abdomen without obvious complications. The abdominal perimeter was shrunk form 5 cm to 15 cm with over 3 months follow-ups. CONCLUSIONS: The combination of lipectomy and liposuction may be a good and safe method for abdominoplasty. The abdominal flap during the lipectomy could achieve higher survival rate than the traditional technique, due to the less injury on the neurovascular structures sustained by the liposuction procedure.

[The study on relation of human papillomavirus and P53 expression with bladder transitional cell carcinoma].

OBJECTIVE: To study the relationship between the bladder transitional cell carcinoma and the human papillomavirus (HPV) types 6, 11, 16, 18 and P53. METHODS: We used polymerase chain reaction (PCR) to test HPV type 6, 11, 16 and 18 infection from 75 cases of bladder cancer, and used immunohistochemistry method to detect P53 expression. RESULTS: The positive rates of HPV types 6, 11, 16 and 18 were 6.7%, 5.3%, 33.3% and 6.7%, respectively. The positive rate of low risk HPV type (6/11) was 9.3% (7/75), and that of high risk HPV types (16/18) was 34.7% (26/75). There were 8 cases with two or three HPV types mixed infection, accounting for 10.6%, HPV types 6,11,16,18 in tumor metastasis group were significantly higher than that in non-metastasis group. The positive rates of HPV 16, 18 types in G3 were significantly higher than that in G1 and G2. HPV type 16 was the main type in bladder cancer. Positive rate of P53 expression was 44%. CONCLUSIONS: These results implicated that the occurrence and development of bladder transitional cell carcinoma may be closely related to the high risk HPV type infection and P53 expression.

Effect of combination of valsartan with benazepril on blood pressure and left ventricular hypertrophy in SHR.

AIM: To evaluate the antihypertensive efficacy of angiotensin converting enzyme inhibitor (ACEI) benazepril in combination with AT1 receptor antagonist valsartan and their effect on left ventricular hypertrophy, renin angiotensin aldosterone system (RAAS) and endoxin in spontaneously hypertensive rat (SHR). METHODS: WKY control group (n = 6) and other 4 groups consisted of 24 SHR (14-week-old, male, n = 6): SHR control group, benazepril group, valsartan group, and combination drug therapy group. Systolic blood pressure (SBP) of SHR was measured at the beginning and at the end of 2, 4, 6, and 8 wk of drug intervention. Morphometric determination, renin activities, angiotensin II (Ang II), endoxin, and ATPase activity analysis were performed at the end of 8 week of drug intervention. RESULTS: SBP, ratio of left ventricular mass (LVM), body weight (BW) (LVM/BW), and transverse diameter of myocardial cell (TDM) of SHR were remarkably decreased after drug intervention, and this decrease was most remarkable in the combination drug therapy group. Renin activities of plasma and myocardium were remarkably increased in drug intervention groups. The levels of Ang II in plasma and myocardium were remarkably increased in valsartan group, decreased in benazepril group and combination drug therapy group. Na(+)-K(+)-ATPase activities in myocardium were remarkably increased and the level of endoxin in myocardium were remarkably decreased as SBP decreased after drug intervention. CONCLUSION: Both benazepril and valsartan can decrease SBP of SHR, and cause regression of ventricular hypertrophy. The efficacy of combination drug therapy group is most remarkable among all groups and avoids the side effects of induction of high Ang II levels in plasma and myocardium caused by long-term use of valsartan alone.

[Relationship between light and regeneration character of Huangshan pine].

The growth and physiological characters of 1-3 years old Huangshan pine seedlings in pure and mixture plots with four different transparancy were determined, and the germination percentage of seeds and their complex stressing vigour were tested. Meanwhile, the regeneration mechanism of Huangshan pine in natural forest was studied in this paper. The results showed that the plumpness of seeds was 65%, the germination percentage was 27%, and the complex stressing vigour was 26%. The lower germination percentage affected the reproduction of Huangshan pine. The photosynthetic rate was more affected by solar energy, but less by humidity and surface temperature. With high light composation, Huangshan pine's regeneration under forest was difficult. The seedlings of Huangshan pine in mixed coniferous and broadleave tree plots were at a disadvantage because of slow growth and insufficient light.

Changes in systemic and splanchnic hemodynamics after orthotopic liver transplantation in cirrhotic rats.

OBJECTIVE: To investigate early changes in systemic and splanchnic hemodynamics after orthotopic liver transplantation (OLT) in normal and cirrhotic rats. METHODS: Male Sprague-Dawley rats were divided into 4 groups: normal controls (NL, n = 10), intrahepatic portal hypertension (IHPH, n = 10) induced by injection of CCl4, normal rats with OLT (NL-OLT, n = 9) and IHPH rats with OLT (IHPH-OLT, n = 16). IHPH-OLT rats were divided into 2 subgroups: 3 days (Group A, n = 9) and 7 days (Group B, n = 7) after OLT. OLT was performed in rats using cuffs for the anastomosis of the suprahepatic inferior vena cava, infrahepatic vena cava and portal vein. Two weeks after production of IHPH rats, 7 days after NL-OLT rats, 3 days and 7 days after IHPH-OLT rats, radioactive microspheres were used in a hemodynamic study. RESULTS: There were no significant differences in hemodynamic changes between NL-OLT and NL rats, except mean arterial blood pressure (MAP). The characteristics of systemic and splanchnic hyperdynamic circulatory state, including increased cardiac output and splanchnic blood flow, decreased mean arterial blood pressure, total peripheral vascular resistance and splanchnic vascular resistance were observed in IHPH, IHPH-OLT A, and IHPH-OLT B rats. The magnitude of hyperhemodynamics was in the order of IHPH > IHPH-OLT A > IHPH-OLT B rats. Moreover, the derangement of splanchnic hyperhemodynamics was more significant than that of systemic hyperthermodynamics. CONCLUSIONS: The present study demonstrates that the persistence of early systemic and splanchnic hyperkinetic circulation after OLT may be the consequence of factors which maintain hyperhemodynamics in liver cirrhosis, where portal hypertension is not completely eliminated. Hyperhemodynamics is not induced by OLT per se.

[Epidemiological studies on the relationship between body weight and the prevalence rate of impaired glucose tolerance (IGT) in the population of urban areas of Tianjin city].

OBJECTIVE: In order to explore the relationship between body weight and the prevalence rate of IGT in a community - based population and to provide scientific informations of IGT for prevention and control. METHODS: Cluster sampling was carried out in prevalence survey, together with clinical examination and test of blood plasma glucose. RESULTS: A sample of 62,577 people was performed by cluster random sampling. Results showed that the crude prevalence rate of IGT in the whole population of three communities was 13.13 per thousand in males, 19.18 per thousand in females, and 16.23 per thousand in both sexes respectively. The IGT prevalence rate of different body weight group in community population had a marked difference. There was a trand that the IGT prevalence rate increased markedly with the increase of body weight. CONCLUSION: The incidence of IGT is related to body weight and obesity.

[Study on erzhi pill effects on neuro-endocrinologic-immuno network of animal of deficiency of yin].

OBJECTIVE: To investigate Erzhi Pill effect on Neuro-Endocrinologic-Immuno network of animal of deficiency of Yin. METHOD: In the experiment through the determination of hemotic cortisol, insulin, testosterone, beta-endorphin, cholesterol, weight of adrenal and thymus, ConA-induced, spleelymphocyte proliferation and activity of natural killer cells (NKC). The changes of the animals of deficiency of Yin and the effects of the Erzhi Pill were investigated. RESULT: It showed that rats of deficiency of Yin, their hemotic, cortisol, insulin, testosterone, beta-endorphin, weight of adrenal and thymus decreased, cholesterol increased and mouse of deficiency of Yin, their lymphocyte proliferation and activity of NKC dcreased, which might be modulated with Erzhi Pill. Erzhi Pill could improve hemotic cortisol, insulin, testosterone, beta-endorphin, weight of adrenal and thymus, reduce hemotic cholesterol, improve lymphocyte proliferation and activity of NKC. CONCLUSION: This principle could improve animals of deficiency of Yin regulating Neuro-Endocrinologic-Immuno network and keeping homeostais.

Nucleosome structural features and intrinsic properties of the TATAAACGCC repeat sequence.

Nucleosomes, the fundamental building blocks of chromatin, play an architectural role in ensuring the integrity of the genome and act as a regulator of transcription. Intrinsic properties of the underlying DNA sequence, such as flexibility and intrinsic bending, direct the formation of nucleosomes. We have earlier identified genomic nucleosome-positioning sequences with increased in vitro ability for nucleosome formation. One group of sequences bearing a 10-base pair consensus repeat sequence of TATAAACGCC had the highest reported nucleosome affinity from genomic material. Here, we report the intrinsic physical properties of this sequence and the structural details of the nucleosome it forms, as analyzed by footprinting techniques. The minor groove is buried toward the histone octamer at the AA steps and facing outwards at the CC steps. By cyclization kinetics, the overall helical repeat of the free DNA sequence was found to be 10.5 base pairs/turn. Our experiments also showed that this sequence is highly flexible, having a J-factor 25-fold higher than that of random sequence DNA. In addition, the data suggest that twist flexibility is an important determinant for translational nucleosome positioning, particularly over the dyad region.

Preparative isolation and purification of hydroxyanthraquinones from Rheum officinale Baill by high-speed counter-current chromatography using pH-modulated stepwise elution.

Analytical and preparative high-speed counter-current chromatography was successfully used for the isolation and purification of hydroxyanthraquinones from Rheum officinale Baill (Dahuang) using pH-modulated stepwise elution. Four major components including chrysophanol, emodin, physcion and aloe-emodin were isolated each at over 98% purity.

[DNA cloning and partial sequence analysis of EBV LMP1 gene isolated from human nasopharyngeal carcinoma cell line SUNE1].

OBJECTIVE: To study the variation of EBV LMP1 encoding gene isolated from human nasopharyngeal carcinoma cell line SUNE1 in Guangdong. METHODS: The EBV LMP1 gene was amplified from the SUNE1 cell genome by PCR and then the recombinant vector was constructed by inserting the PCR fragment into pcDNA3. The encoding EBV LMP1 spanning from exon1 to exon3 in recombinant vector was sequenced by the Sequence Analyzer. RESULTS: The 1,312 bp encoding EBV LMP1 pieces in 2.6 kb PCR fragments were compared with the same EBV segments in B95-8 cell line. The data indicated that the rate of nucleotide sequence homology between the two fragments is 98.5% and the rate of amino acid is 96%. The restricted enzyme site of Xho I in exon1 was deleted in SUNE1 but the 30 bp deletion at the carboxyl terminus in most Chinese NPC LMP1 gene was not present. CONCLUSION: Although the LMP1 gene derived from SUNE1 had greater tumorigenicity than that derived from B95-8 cell, the high homology rate of nucleotide and amino acid sequences between them indicated that it was not the result from the variation of certain nucleotide sites but the change in amino acid domain.

The ferric uptake regulation (Fur) repressor is a zinc metalloprotein.

The Fur protein regulates the expression of a wide variety of iron-responsive genes; however, the interaction of this repressor with its cognate metal ion remains controversial. The iron-bound form of Fur has proved difficult to obtain, and conflicting results have been published using Mn(II) as a probe for in vitro DNA-binding studies. We report here that the purified protein contains tightly bound zinc and propose that Zn(II) is bound to the protein in vivo. Upon purification, Fur retains ca. 2.1 mol of Zn(II)/mol of Fur monomer (Zn2Fur). One zinc is easily removed by treatment of Zn2Fur with zinc chelating agents, resulting in Zn1Fur with ca. 0.9 mol of Zn(II)/mol of protein. The remaining zinc in Zn1Fur can only be removed under denaturing conditions to yield apo-Fur with ca. 0.1 mol of Zn(II)/mol of protein. Our results suggest that many literature descriptions of purified Fur protein do not correspond to the apo-protein, but to Zn1Fur or Zn2Fur. Dissociation constants (Kd) of protein-DNA complexes are ca. 20 nM for both Zn2Fur and Zn1Fur as determined by electrophoretic mobility shift assays and DNase I footprinting assays. The two metalated forms, however, show qualitative differences in the footprinting assays while apo-Fur does not bind specifically to the operator. The existence of these Zn(II) binding sites in Fur may resolve some discrepancies in the literature and have implications concerning Zur, a Fur homologue in E. coli that regulates zinc-responsive genes.
