Electrophoresis of acid soluble proteins from rat liver nuclei and polysomes on large two-dimensional polyacrylamide gels.

Increased resolution of 0.4 N H(2)SO(4) soluble rat liver polysomal and nuclear proteins was achieved by initial electrophoresis on 15 percent polyacrylamide disc gels (25 times 0.5 cm) followed by electrophoresis on 16 percent polyacrylamide gel slabs (25 times 15 cm). With this method, it was found that the histones were resolved into 12 spots, the polysomal proteins A1 and A9 were each resolved into two components, and polysomal proteins A1, A1 and A1 with molecular weights of approximately 14,000 were separated from the other proteins by their rapid migration in the first dimension. Moreover, no histones were found in the acid extracts of the polysomes.

[Socio-psychological aspects in marriages of alcoholic women].

In a controlled study during two years we have collected psychological and sociological data on partners of chronic female alcoholics by making use of a semistandardized interview (80 items were evaluated by program Dial and the input scales of Spitzer and his decision logic), of the Freiburger-Persönlichkeits-Inventar (FPI) and of the Hawie-Dahl-test, a German abbreviated version of the Wechsler-Bellevue-Intelligence-test. Furthermore we investigated the interpersonal perception in the marriage of the female alcoholic by a semantic differential. The relatively small number of subjects and controls emphasizes the considerable problems as far to the methodial implications of such a study. Therefore we have to interpretate cautiously our results. We may plead for the hypothese, that the husband of the chronic female alcoholic already from the beginning of the marriage had fifficulties in the realisation of his male role and beyond this that the partnership of the chronic female alchoholic is evidently disturbed by distortion of the interpersonal perception. Finally the interpretation of our results is disucssed with regard to the special approach of examination.

Nucleolar phosphoproteins of normal rat liver and Novikoff hepatoma ascites cells.

The nucleolar acid-soluble proteins from normal rat liver and Novikoff hepatoma ascites cells were labeled in vivo for 2 hr after the injection of [32P]orthophosphate and in vitro with [gamma-32P]adenosine triphosphate in systems containing 0.25 M sucrose, 5 mM MgCl2, 12.5 mM NaCl, and 0.05 M Tris-HCl buffer, pH 7.3, AT 37 DEGREES. Two-dimensional polyacrylamide gel electrophoresis and autoradiography showed that approximately 40 and 20 protein spots were labeled in vivo and in vitro, respectively. There were some marked differences in labeling in vivo between normal rat liver and Novikoff hepatoma acid-soluble nucleolar proteins. By 32P analysis of gel slices, the proportion of the total 32P incorporated into protein Spot A1-4 was greater in normal liver, and the proportion of 32P incorporated into some high-molecular-weight protein spots, such C23-24 and C26-27, was greater in the Novikoff hepatoma ascites cells. With the in vitro incubation system, the 32P uptake per mg protein was about twice as high in Novikoff hepatoma nucleolar proteins as in normal rat liver nucleolar proteins but, generally, the same proteins were labeled in both tissues.

Characterization of nuclear and chromatin poly A-containing RNA.

Poly A-containing RNA was isolated from Novikoff hepatoma cell chromatin on nitrocellulose membrane filters after the cells were labeled with 32P-orthophosphate for 6 hours. Under denaturing conditions, the poly A-containing chromatin RNA had a sedimentation coefficient of approximately 8-18S. The presence of poly A tracts in the chromatin RNA fractions indicate that poly A addition occurs almost immediately after transcription. Poly A-containing chromatin RNA and nuclear RNA were approximately 1/5 as active as cytoplasmic poly A-containing RNA in the wheat germ translational system. Since chromatin poly A-containing RNA is the precursor of cytoplasmic poly A-containing RNA, further processing of this RNA prior to its attachment to polysomes may account for its increase in translational capacity.

Ultrastructural study of the effect of bleomycin A2 on the nucleolus and its possibly related cytoplasmic constituents in Novikoff hepatoma cells.

Treatment of Novikoff hepatoma ascites cells with bleomycin A2, in vivo as well as in vitro, in varying doses produced marked alterations in nucleolar and cytoplasmic ultrastructural organization. A series of changes occur including formation of fibrillar centers, fragmentation of fibrillar nucleolar elements, appearance of microspherules, and the loss of fibrillar elements from these nucleoli. A bleomycin concentration of 10 mug/ml in vitro produced an increased number of fibrillar centers with well-defined nucleolonemas. At a concentration of 50 mug/ml, there was an increase in number and fragmentation of these fibrillar centers and many microspherules were found throughout the nucleolus. Approximately one-fourth of the cells contained cytoplasmic fibrillar bodies and amorphous fibrous tufts around the nuclear envelope. At a bleomycin concentration of 100 mug/ml, the nucleoli contained more granular elements and numerous microspherules. Almost 90% of the cells contained cytoplasmic fibrillar bodies. The effects of bleomycin in vivo (10 mg/kg) closely resemble those found in vitro with concentrations of 50 and 100 mug/ml.

Biochemical effects of bleomycin A2 on Novikoff hepatoma ascites cells.

Purified nucleolar DNA was markedly degraded at a concentration of 13 mug/ml by bleomycin A2; bleomycin concentrations 20-30 times greater were required to degrade nucleoplasmic DNA. Whole nuclear DNA was degraded to only a small extent at 13 mug/ml but was markedly degraded at higher bleomycin concentrations. Treatment of the various types of DNA with high concentrations of bleomycin A2 produced low molecular weight (approximately 6S) fragments that were no longer sensitive to degradation by bleomycin A2. Hybridization studies demonstrated a loss of ribosomal DNA sequences from nucleolar DNA treated with bleomycin A2 in vitro. Studies on RNA synthesis in Novikoff hepatoma ascites cells in vitro showed there was a decreased uptake of 32Pi into high molecular weight nuclear RNA in the presence of bleomycin A2. These results indicate that nucleolar function is inhibited by a direct effect of bleomycin A2 on nucleolar DNA.
