Irritation and staining by dithranol (anthralin) and related compounds. III. Cumulative irritancy and staining during repeated chamber testing.

Irritation and staining, the side effects of dithranol and related compounds have previously been studied after a single chamber-exposure. To mimic the therapeutic situation, the chamber application was repeated for 3 weeks on the uninvolved back skin of 17 psoriasis patients. In equimolar concentrations corresponding to 0.05% dithranol, dithranol caused markedly more erythema and staining than 10-propionyl dithranol and 10-butyryl dithranol, whereas 10-butyryl dithranol irritated and stained less than 10-propionyl dithranol. To reach the irritation and staining level of dithranol, a 4-fold higher concentration of 10-butyryl dithranol was required. During the first week the irritation culminated, declining later despite repeated applications, possibly because of exhaustion of anthrone-induced inflammatory mediators.

DNA-flow-cytometric measurements on the normal, atrophic, hyperplastic and neoplastic human endometrium.

DNA distribution patterns and the fractions of the cell cycle phases were determined by means of flow-through cytometry in 87 samples of normal, atrophic, hyperplastic and carcinomatous human endometrium. The S-phase fractions vary during the normal menstrual cycle between 1 and 3% and reach a periovulatory maximum between 4.4 and 4.7%. Atrophic endometrium and regressive glandular cystic hyperplasia have little DNA synthesis (1.01% and 1.68% S-phase fractions respectively). Proliferating glandular cystic hyperplasia reveals 3.38% S-phase fraction, whereas adenomatous hyperplasia has an increased number of DNA-synthesizing cells (4.81%). The well-differentiated endometrial carcinoma shows no cytophotometrically detectable differences in comparison to adenomatous hyperplasia. All endometrial samples except for poorly differentiated endometrial carcinoma showed a diploid to tetraploid DNA distribution pattern. The poorly differentiated endometrial carcinoma displays two different types: one rapidly growing diploid-tetraploid tumor with 8.0 to 9.6% S-phase fractions, and another type with stemline deviations, polyploid nuclei and less pronounced synthetic activity.

[CT diagnostic of ovarial tumors].

The diagnostic value of computed tomography was examined in 34 patients by the clinical diagnosis of an ovarial tumor established immediately before the operation. In 78,9% of all cases, a carcinoma could be surely recognized, often only by indirect signs of malignancy. Benign processes were correctly recognized in 83,3% of all cases. The staging of carcinomas was correct in 57,9%. A disadvantage of CT lies in the fact that it does not show peritoneal disseminations of smallest metastases as well as metastatic formations in not enlarged lymph nodes, which often leads to an understaging.

A cAMP-dependent phosphorylated motility protein in bovine epididymal sperm.

A putative cAMP-dependent protein kinase substrate associated with the cAMP stimulation of bovine sperm motility has been identified. Optimum conditions for a linear, concentration-dependent incorporation of [32P]ATP into phosphoproteins of an epididymal sperm sonicate by cAMP-dependent protein kinase are described. The motility state of the sperm was reduced by incubation at 37 degrees C and reactivated with theophylline. Endogenous levels of cAMP correlated with the motility state of the sperm. The phosphorylation state of phosphoproteins was frozen by addition of NaF (100 mM final concentration). After sonication and removal of endogenous nucleotides, 32P incorporation into phosphoprotein varied inversely with motility. The inverse relationship results from the procedure monitoring the capacity for incorporation of 32P into dephosphorylated cAMP-dependent protein kinase substrates. Over 70% of the cAMP-dependent label was in the soluble fraction. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis showed one cAMP-dependent phosphoprotein from the soluble fraction which had an inverse correlation with motility. This 55,000-dalton protein did not bind [3H]cholchicine (tubulin) or [3H]cAMP (REGULATORY SUBUNIT OF PROTEIN KINASE). These data indicate the existence of a cytosolic phosphorylated motility protein.

Malignant mixed tumor of the salivary-gland type, primary in the prostate.

A previously undescribed malignant mixed tumor arising in the prostate of a 72-year-old man is herein presented. Clinically, it was manifested by slowly progressing urinary frequency and urgency, as well as by intense pain. The total serum acid phosphatase value and the prostatic fraction were within normal limits. Six months after prostatectomy the patient died, with local recurrence and roentgenologic evidence of a single pulmonary metastasis. Both light-microscopic and electron-microscopic examination showed features similar to those of a malignant mixed tumor of the salivary gland. These findings suggest that this tumor originated in seromucinous ectopic glands. The important factor in its recognition is that it is not hormone-dependent.

Initiation of sperm motility in the mammalian epididymis.

Spermatozoa acquire the capacity for motility as they traverse the mammalian epididymis. The biochemical basis for this induction of motility is still largely unknown. Current theories are discussed and recent studies from the authors' laboratory are described which indicate that two separate processes are involved and that these act synergistically. These processes are an increase in the intrasperm content of cyclic AMP during epididymal transit combined with the binding of a specific forward-motility protein. A second increase in cyclic AMP and stimulation by calcium ion is likely involved in the expression of the acquired potential for motility at the time of ejaculation.
