Lymphocyte subpopulations and reactivity in breast cancer recurrence following adjuvant postoperative chemotherapy.

The size of various subset of blood lymphocytes and stimulations with PHA and allogeneic cells (MLC) were examined in 45 women with breast cancer receiving postoperative adjuvant cyclic chemotherapy with chlorambucil, methotrexate, and 5-fluorouracil (LMF). The patients who remained disease-free (n = 19) and those who relapsed (n = 26) during a 4.5 to 6-year follow-up period had similar initial values; and similar kinetics of the examined immunological parameters during and following completion of treatment.

Influence of adjuvant radiation therapy in breast cancer on PWM induced Ig-secretion by blood lymphocytes in vitro.

The capacity of PWM (poke-weed mitogen) to stimulate Ig-secretion by blood lymphocytes was examined before and at various times after local radiation therapy (46.0 Gy) for breast cancer. It was observed that the secretion of IgM, IgA and IgG were significantly reduced at completion of radiation therapy. Secretion of IgM was reduced to the highest relative extent (approximately 10% of the pretreatment value). Thereafter there was a recovery of the Ig-secreting capacity which, however, remained below the pretreatment level, 12-18 months after completion of radiation therapy. The capacity of PWM-stimulated blood lymphocytes to secrete specific antibodies against morbilli and herpes simplex virus was also significantly impaired after radiation therapy. The reduction of Ig-synthesis in vitro was not correlated with the increase in the ratio between numbers of monocytes and lymphocytes in peripheral blood after radiation therapy. Possible explanations for these results are discussed.

Increased reduction of nitroblue tetrazolium by human blood monocytes following post-operative radiation therapy for breast cancer.

Reduction of nitroblue tetrazolium by blood monocytes was significantly increased following post-operative radiation therapy for breast cancer. Attachment and ingestion of yeast particles by monocytes was not affected. Exposure of purified monocytes in vitro to 13 Gy did not significantly affect NBT reduction of yeast particle attachment, whereas ingestion of yeast particles was slightly increased.

Dual effects of folinic acid in 5-fluorouracil induced killing of human tumor cell lines in vitro.

Folinic acid (leucovorin) is frequently used to augment and modulate the clinical activity of 5-fluorouracil (5-FU) in patients with advanced gastrointestinal (Gl) cancer. However, there are conflicting opinions concerning the optimal doses for these patients, and whether folinic acid modulates the clinical activity of 5-FU in patients with non-Gl cancer. To elucidate these questions, model experiments have been performed on human tumor cell lines in vitro to determine the modulatory activity of various concentrations of folinic acid on 5-FU mediated cytotoxicity using a clonogenic assay. Three cell lines of colon cancer and 3 of glioblastoma origin were exposed to 5-FU alone or with folinic acid for 24 hours. It was observed that relatively low concentrations of folinic acid enhanced the cytotoxicity of 5-FU against the colon cancer lines whereas higher concentrations were less effective. Folinic acid did not enhance the 5-FU mediated killing of the glioma cell lines at any concentration (0.01-100 micrograms/ml). On the contrary, folinic acid seemed to counteract the cytotoxic effect of 5-FU in a reasonably dose-dependent fashion. These results may suggest that the value of folinic acid in the treatment of non-Gl cancer with 5-FU should be evaluated within the framework of controlled clinical trials, and that high doses of folinic acid may not necessarily be more effective than low.
