Efficacy, safety, and residue evaluation of levamisole gel formulation in sows.

Anthelmintic efficacy, safety, and residue studies were conducted in sows and gilts with a levamisole gel containing 11.5% levamisole HCl. In 12 sows and 12 gilts, 8 mg of levamisole HCl equivalent/kg of body weight orally was 100% (resinate) and 91.1% (gel) effective against 55-day-old Ascaris suum and 100% (gel) and 96.1% (resinate) effective against Oesophagostomum dentatum. In 20 sows given levamisole gel (8 mg of levamisole HCl/kg) orally just before breeding, 4 to 6 weeks after breeding, 4 to 6 weeks before farrowing, and just before farrowing, there were no adverse effects. Transient salivation was noticed in five sows after treatment. In 4 groups of 4 sows each given levamisole gel orally to provide 8, 24, 40, or 80 mg of levamisole HCl/kg, adverse clinical signs were not observed in sows treated with 8 mg/kg. Transient salivation was noticed in one sow given 24 mg/kg, two sows given 40 mg/kg, and four sows given 80 mg/kg. Multiple emesis and chomping occurred in one sow given 80 mg/kg. Levamisole residues in edible tissues from sows given 8 mg of levamisole gel/kg orally were less than 0.1 mg/kg of muscle and fat in sows killed on posttreatment day (PTD) 3 and less than 0.1 mg/kg of kidney in sows killed on PTD 5. Liver residues averaged 0.78 mg/kg in sows killed on PTD 3 and were reduced to 0.31 mg/kg in sows killed on PTD 5. The 99% upper tolerance limit with 95% confidence on the withdrawal time to assure levamisole residues of less than 0.10 mg/kg in liver tissue was 11 days.

[Vitamin C concentrations in maternal plasma, amniotic fluid, umbilical cord blood, the plasma of newborn infants, colostrum and transitory and mature breast milk].

Vitamin C concentrations have been measured in the plasma of 200 mothers and their newborns as well as in amniotic fluid and breastmilk. Out of this group 19 mother-infant-pairs were taken as normal control group with no risk factors, complications or diseases during pregnancy or delivery or in the newborn infant. Vitamin C concentrations in plasma showed great variability. This is true for both the entire study group and the normal control group. A positive correlation was found between the vitamin C concentrations in maternal plasma at the time of admission to the obstetric unit and that in the second stage of labor immediately before delivery. Cord blood and newborn plasma vitamin C concentrations were nearly twice as high compared to maternal concentrations. They too correlate with the concentrations in the maternal plasma. A further correlation was found between maternal plasma and amniotic fluid at the time of delivery (ratio about 1:3). No more significant correlations of vitamin C concentrations have been found in the normal control group. Various diseases or risk factors in mother and/or child were shown to be associated with lower vitamin C concentrations. Vitamin C concentrations were considerably lower in all biological fluids in smokers and mothers with diabetes. Other statistical correlations will be shown and possible casualties will be discussed. In this study vitamin C concentrations in groups with abnormal states are documented only with small numbers of cases and are therefore considered as a basis for further more specific investigations.

[Keratoacanthoma].

We give a brief review concerning morphogenesis, growth pattern, as well as clinical and histopathological definition of keratoacanthoma. The treatment of choice is excision of the tumor and postsurgical histological verification.

Quantitative assessment of subcutaneous fibrinolysis in the rat.

A clot lysis model in the rat was described which was responsive to manipulation of the fibrinolytic system. By employing [125I]-labeled fibrinogen in homologous, plasminogen-deficient plasma, clots were prepared in Teflon-perforated cylinders and were implanted subcutaneously. After a brief lag phase, lysis proceeded essentially linearly with 70% of the clot resolved within 12 days. Enhancement of lysis was achieved with a stimulator of fibrinolysis, stanozolol; and inhibition, with the antifibrinolytic, tranexamic acid. Other drugs affecting blood pressure, inflammation, or connective tissue growth had no effect on lysis. Interstitial fluid collected from empty cylinders and fluid obtained from the recovered clots were monitored for plasminogen, alpha 2-antiplasmin, plasminogen activator, and plasminogen activator inhibitor using sensitive and specific assays. All of these fibrinolytic proteins were quantitated relative to their plasma concentrations. The most striking differences between the two fluids and plasma were a threefold higher concentration of the plasminogen activator inhibitor and only traces of free tissue plasminogen activator in the fluids. Nevertheless, significant tissue plasminogen activator was delivered to the clot. It was concluded that lysis occurred via delivery of the fibrinolytic system in interstitial fluid. It is suggested that this model is a technically simple and quantitative screen in vivo for potentially fibrinolytic or antifibrinolytic agents.

Embryogenesis and organogenesis of Carausius morosus under spaceflight conditions.

The influence of cosmic radiation and/or microgravity on insect development was studied during the 7 day German Spacelab Mission D1. Eggs of Carausius morosus of five stages differing in sensitivity to radiation and in capacity to regeneration were allowed to continue their development in the BIORACK 22 degrees C incubator, either at microgravity conditions or on the 1 g reference centrifuge. Using the Biostack concept--eggs in monolayers were sandwiched between visual track detectors--and the 1 g reference centrifuge, we were able to separate radiation effects from microgravity effects and also from combined effects of these two factors in space. After retrieval, hatching rates, growth kinetics and anomaly frequencies were determined in the different test samples. The early stages of development turned out to be highly sensitive to single hits of cosmic ray particles as well as to the temporary exposure to microgravity during their development. In some cases, the combined action of radiation and microgravity even amplified the effects exerted by the single parameters of space. Hits by single HZE particles caused early effects, such as body anomalies, as well as late effects, such as retarded growth after hatching. Microgravity exposure lead to a reduced hatching rate. A synergistic action of HZE particle hits and microgravity was established in the unexpectedly high frequency of anomal larvae. However, it cannot be excluded, that cosmic background radiation or low LET HZE particles are also causally involved in damage observed in the microgravity samples.

World Association for the Advancement of Veterinary Parasitology (W.A.A.V.P.) guidelines for evaluating the efficacy of anthelmintics in swine.

As a follow-up to a previous guideline publication on efficacy evaluation of anthelmintics in ruminants, an expert group of the World Association for the Advancement of Veterinary Parasitology presents similar guidelines for the testing of anthelmintics in swine. The goal of the guidelines is to develop uniform testing standards and registration requirements. Data obtained from investigations according to the guidelines should be internationally accepted in the registering of drugs. Further, the use of the guidelines may contribute towards reducing costs, labor and the number of experimental animals used for drug testing.

Polyomavirus small T antigen enhances replication of viral genomes in 3T6 mouse fibroblasts.

Transfection of 3T6 cells with a cloned polyomavirus genome encoding only large T antigen resulted in DNA replication with only about 1/10 the efficiency of wild-type viral DNA coding for all three T antigens. This replication defect was at least in part overcome by the simultaneous transfections of polyomavirus genomes which allowed the expression of small T antigen. We conclude that polyomavirus small T antigen has a (probably indirect) role in replication.

Release of tissue plasminogen activator and its fast-acting inhibitor in defective fibrinolysis.

Releasable tissue plasminogen activator (t-PA) and the fast inhibitor of t-PA were measured in 18 controls and a pedigree with venous thrombosis. The functional assay was performed by a technique that destroys the t-PA inhibitor when blood is drawn. It was found that activator and inhibitor levels varied widely in the control group. One patient demonstrated inhibitor levels, on two different occasions, of 2.82 and 3.54 IU of t-PA per milliliter of plasma, as compared with a releasable activator level of 1.87 IU/mL. The t-PA antigen levels of this patient and the remainder of the pedigree were essentially normal for all seven subjects. Thus, it is suggested that the previously reported fibrinolytic disorder in this pedigree represents an imbalance between activator and inhibitor levels rather than an actual deficiency of t-PA.

[Clinical experiences with an antimycotic foam preparation with added prednisolone (Nystacorton foam)].

Twenty-five infants and children with Candida infections in the perianal, genital and inguinal areas and on the skin normally covered by the napkin or diaper, confirmed by culture tests and microscopy, were treated with a new combined foam preparation containing the active substances nystatin, chlorhexidine and prednisolone. Even partially very severe lesions with multiple, satellitelike colonies on the trunk, extremities and neck were, without exception, cured within a maximum of 13 days. After disappearing of the exudative stage, intermittent treatment with a rather more greasy excipient (Excipial Cream) containing no active substance, has proved beneficial.

[Food value of field beans for chickens and evaluation of various portions with special reference to the tannin-poor sort in broiler fattening food].

The investigations were carried out with varieties rich in tannin (Erfordia, Fribo) and one poor in tannin (White Flowering). The protein of the variety poorer in tannin is slightly richer in methionine and is slightly better digested. The studies on the varieties Erfordia, Fribo and White Flowering in quotas of 7.5...45% as component in the fattening feed for broilers resulted in the same fattening performance as in the control group (maize-soybean-fishmeal ration) and had no influence on the chemical composition of the body and the weight of the pancreas. All rations were isonitrogenous and isocaloric. The soybean oilmeal in the ration was replaced gradually resp. completely by the field bean coarse meal in connection with methionine supplements. The variety poorer in tannin did not have nutrition-physiologic advantages in the the growth experiment with young broiler chickens.
