Plasminogen activator in bronchoalveolar fluid.

Plasminogen activator was purified from the soluble fraction of bronchoalveolar lavage fluid, and biological and immunological characteristics of the activator were examined by electrophoretic enzymography. The purified fraction showed a single band with a molecular weight of 53,000. The enzyme activity was eliminated in the presence of DFP and did not display fibrin affinity. Immunological tests revealed that the plasminogen activator reacted with IgG of antiurokinase but not with that of tissue-type plasminogen activator. However, the plasminogen activator cleaved S-2288 to a greater extent than S-2444 in contrast to urokinase.

[Randomized study of cisplatin and doxorubicin with or without vincristine in non-small cell lung cancer].

A randomized study of anticancer chemotherapy, CDDP plus ADM with/without VCR on patients with NSCLC was carried out. Forty-six patients received injections of CDDP (75 mg/m2) and ADM (50 mg/m2) every 4 weeks (Regimen A); 39 patients were injected with the same doses of CDDP and ADM, plus VCR (1.4 mg/m2, on day 1 and 0.7 mg/m2, on day 7), every 4 weeks (Regimen B). Seven patients (15%) and 10 patients (26%) achieved a partial response by Regimens A and B, respectively. The median survival time (MST) was 8.5 months in each group. Survival time of the responders (MST; 27 months) was much more prolonged than that of the non-responders (MST; 7 months) (p less than 0.01). Both regimens were well tolerated with only moderate gastrointestinal symptoms and mild bone marrow toxicities. Although the addition of VCR to CDDP plus ADR in NSCLC fulfilled the objective tumor regression, no additive effect could be obtained with regard to survival.

Expression in E. coli of finger-domain lacking tissue-type plasminogen activator with high fibrin affinity.

Tissue-type plasminogen activator (t-PA) has a high affinity for fibrin and induces lysis of fibrin (fibrinolysis) on the surface of fibrin without degrading circulating fibrinogen. cDNA for t-PA which lacks the 'finger-domain' (the site for fibrin affinity) was isolated from Detroit 562 cells. Analysis of the nucleotide sequence revealed a lack of the sequences which code for the finger-domain. A plasmid (pDPAT 1) containing the Escherichia coli tac promoter/operator and the cDNA sequence coding for 'finger-domain lacking t-PA' was constructed for expression in E. Coli. The polypeptide so produced was a new type of t-PA lacking finger-domain, but revealed plasminogen activator activity with the function of fibrin affinity.

[A case of remission in diffuse pleural mesothelioma induced by anticancer chemotherapy with cisplatin and adriamycin].

A 56-year-old man was admitted to our hospital with complaints of dyspnea and right pleural effusion. Malignant pleural mesothelioma was diagnosed, based on the high level of hyaluronic acid in the pleural fluid and manifestation of malignant cells by pleural needle biopsy. Anticancer chemotherapy with cisplatin and intrapleural injections of adriamycin against this mesothelioma was carried out because of the presence of stage III by Butchart's classification. Regression of the tumor, decrease of pleural fluid and decrease of LDH level in effusions were observed after this chemotherapy. Cisplatin was easily transported into the pleural fluid, and its concentration was maintained for a satisfactorily long period.

Monoclonal antibody to human tissue plasminogen activator.

Hybridoma producing a monoclonal antibody IgG1 to one-chain tissue plasminogen activator (t-PA) derived from human melanoma cells was obtained by fusion of mouse myeloma cells (SP-1) and spleen cells of mice previously immunized with purified t-PA. The monoclonal antibody reacted only with t-PA derived from the human melanoma cells (Bowes), and not with plasminogen activator purified from porcine heart or from human urine. The monoclonal antibody obtained from mouse ascites demonstrated 50 times stronger antibody activity than that of polyclonal antibody obtained from mouse serum. The monoclonal antibody bound t-PA firmly, inhibited the fibrinolytic activity of t-PA, but did not inhibit the amidolytic activity of t-PA completely. The fibrin-binding ability of t-PA was not inhibited. The binding of monoclonal antibody to the non-reduced form of t-PA did not differ from that to the reduced form of t-PA.

Molecular homology among the structural proteins of densonucleosis virus from silkworm, Bombyx mori.

Similarities among the four structural proteins of Bombyx densonucleosis virus (DNV) were examined by four independent techniques, peptide mapping, immunodiffusion tests, amino acid analysis and enzyme linked immunosorbent (ELISA) technique. The peptide maps produced by Staphylococcus aureus V8 protease, or chymotrypsin indicated the existence of sequence homology among the proteins. The results of immunodiffusion tests revealed that these four structural proteins share common antigens. The amino acid compositions of these proteins were also very similar to each other. However, the amount of amino acid residues (e.g., serine) in VP3 was not always sufficient to account for the amount found in the smaller structural protein, VP2. These results indicate that the four structural proteins of Bombyx DNV probably originate, at least partially, from a common DNA sequence, and that VP2 is not a direct cleavage product of VP3.

[Combination chemotherapy for small cell carcinoma of the lung: AVN-DVC therapy].

Comparative studies of alternating non-cross resistant chemotherapy, A.V.N.-D.V.C. (ACNU + VCR + PCZ-ADM + VCR + CPA) was carried out on 19 patients with small cell lung cancer. A.V.N. (A. V.F.) therapy on 45 patients and D.V.C. therapy on 19 patients were used as historical control, respectively. A.V.N. (A.V.F.) therapy was composed of ACNU (2.5 mg/kg, day 1), VCR (0.02 mg/kg, once every week) and PCZ (1 mg/kg, daily) (or FT-207 (15 mg/kg, daily], and duration of one course was 6 to 8 weeks. D.V.C. therapy was composed of ADM (1 mg/kg, day 1) VCR (0.02 mg/kg, days 1 and 5) and CPA (2 mg/kg, days 1 to 5), and repeated every 4 weeks. A.V.N.-D.V.C. therapy was done by a timely alternation of one course of A.V.N. and two course of D.V.C. Reduction rate of tumor size was 84% in A.V.N.-D.V.C. therapy, 62% in A.V.N. (A.V.F.) therapy and 26% in D.V.C. therapy, respectively. Median survival time was 13 months on A.V.N.-D.N.C. therapy, 8.5 months and A.V.N. (A. V.F.) therapy and 4 months on D.V.C. therapy. Significant prolongation of median survival time was obtained in A.V.N.-D.V.C. therapy in comparison with that of historical controls. Major toxicity in A.V.N.-D.V.C. therapy was slight bone marrow suppression.

Effects of antiplatelet agents on pulmonary metastases.

The role of platelets in cancer metastasis was studied by investigating the effects of the antiplatelet agents ticlopidine, diltiazem, dipyridamole and trapidil on artificial and spontaneous pulmonary metastases in mice. These agents were tested at their optimal inhibitory doses on adenosine diphosphate-induced platelet aggregation; namely, 100 mg/kg for ticlopidine, 2 mg/kg for diltiazem, 180 mg/kg for trapidil and 60 mg/kg for dipyridamole. At these doses, trapidil caused moderate inhibition of thrombin-induced platelet aggregation in mice, but the other agents had only slight effects. Artificial pulmonary metastasis was produced by inoculation of Lewis lung carcinoma (LLC) or B16 melanoma (B16) cells into C57BL/6 mice. For induction of spontaneous pulmonary metastases, these tumor cells were implanted subcutaneously into the footpads of mice. The resulting primary tumors of LLC and B16 were removed 9-10 and 17 days later, respectively. Artificial pulmonary metastases were inhibited significantly by all the antiplatelet agents tested. Spontaneous pulmonary metastases were markedly reduced only when these agents were given after removal of the primary tumor. The role of platelets is discussed with respect to thrombus formation in the lodgement of tumor cells and the participation of platelet-derived growth factor in the growth of metastatic foci.
